The karyotype, the histological structure of the testis and the synaptonemal complex (SC) of the mammalian species Tayassu tajacu, Tayassu pecari and of an interspecific male hybrid captured in nature were analysed. The specimens of T. tajacu (2n = 30) and T. pecan (2n = 26) exhibited seminiferous tubules with germ cells in all sperma to genesis stages. In the SC studies both species had a regular structure, easily identified in the autosomes and in the sex chromosomes. The hybrid (2n = 28) had seminiferous tubules with almost all germinal cells in the spermatogonium stage and only a few cells in the primary spermatocyte stage. Gross abnormalities in SC were observed. A few lateral elements showed regular or partially regular synapsis, other lateral elements were synapsed as multivalents, and most axial elements remained unsynapsed. The results suggest that the karyotypes of the parental species have sufficient differentiation to avoid chromosome synapsis. Alternatively, the hypothesis of the existence of genetic incompatibilities between the parental species is discussed.

The surface-spreading technique for visualization of whole cell synaptonemal complex (SC) complements was employed to study the chromosome synapsis in spermatocytes of P. mesopotamicus, C. macropomum and in their interspecific hybrid (tambacu) with the main objective to analyze possible errors in chromosome pairing that could result in hybrid sterility. SC analysis showed that the parental species P. mesopotamicus and C. macropomum have 27 bivalents homogeneously synapsed. The SC in spermatocytes from the hybrid tambacu showed gross meiotic configurations in all cells analyzed. The spermatocytes exhibited a few chromosomes or well synapsed chromosome segments, while many chromosome segments did not have any synapsis. This result, allied to other genetical and cytogenetical evidence, reinforces the hypothesis that the hybrid tambacu is sterile. Further studies involving other aspects, such as behavior and physiology, should be conduced before the introduction of these hybrids in rivers and lakes.

The genus Passiflora L. consists of approximately 530 widely distributed species, including Passiflora edulis, which has drawn interest because of its commercial and agronomic value. Passiflora cincinnata is another important species owing to its long flowering period and resistance or tolerance to diseases and pests. In the present study, the meiotic segregation and pollen viability of an interspecific hybrid (P. edulis x P. cincinnata) and its parents were analyzed. The genomic contents were characterized using chromomycin A3 (CMA3)/40-60-diamidino-2-phenylindole (DAPI) staining, fluorescent in situ hybridization with 5S/45S ribosomal DNA (rDNA), genomic in situ hybridization (GISH), and inter-simple sequence repeat (ISSR) markers. The results indicated the diploid chromosome number for the parents and interspecific hybrid was 2n = 18. We also observed regular meiosis, one pair of S rDNA sites, and two pairs of 45S rDNA sites that colocalized with two pairs of CMA3 /DAPI- bands. The GISH data revealed three distinct chromosomal groups in the hybrid. The genetic origins of the interspecific hybrid, and its relationship with its parents were also confirmed using ISSR markers.

Three species of the genus Eucalyptus (E. dunni, E. grandis, E. saligna) and interspecific hybrid were studied cytogenetically. The Eucalyptus species and the hybrid showed a symmetrical karyotype with 2n=22 chromosomes, with chromosome length ranging from 0.67 to 1.39 μm. Karyotypic analysis indicated a homogenous morphology and chromosome number for the species and the hybrid studied here. Based on the karyotype asymmetry data, together with the chromosome morphology results, the hybrid presented close similarity to E. saligna, suggesting that the latter is one of the parental species involved in the production of the hybrid.

Cucumber mosaic virus (CMV) and tomato aspermy virus (TAV) belong to the Cucumovirus genus. They have a tripartite genome consisting of single-stranded RNAs, designated 1, 2, and 3. Previous studies have shown that viable pseudorecombinants could be created in vitro by reciprocal exchanges between CMV and TAV RNA 3, but exchanges of RNAs 1 and 2 were replication deficient. When we coinoculated CMV RNAs 2 and 3 along with TAV RNAs 1 and 2 onto Nicotiana benthamiana, a hybrid quadripartite virus appeared that consisted of TAV RNA 1, CMV RNAs 2 and 3, and a distinctive chimeric RNA originating from a recombination between CMV RNA 2 and the 3′-terminal 320 nucleotides of TAV RNA 2. This hybrid arose by means of segment reassortment and RNA recombination to produce an interspecific hybrid with the TAV helicase subunit and the CMV polymerase subunit. To our knowledge, this is the first report demonstrating the evolution of a new plant or animal virus strain containing an interspecific hybrid replicase complex.

Ploidy: triploid interspecific hybrid (3n = 27 chromosomes). Plant: habit prostrate, creeping, type mat-forming, height very short, longevity perennial, spreading laterally by stolons and rhizomes. Stolon: compound nodes with up to 3 leaves, internode length very short, internode thickness very thin, colour grey-brown (RHS N199A) when exposed to sunlight. Culms: length very short. Leaf blade: shape linear-triangular, length short, width narrow, colour dark green (RHS 137B). Ligule: dense row of short white hairs. Inflorescence: digitate with 3(-4) very short spicate racemes, peduncle very short. (All RHS colour chart numbers refer to 2001 edition.) Cucumber mosaic cucumovirus (CMV) infects a very wide range of plant species (>1000 species). We recently demonstrated that a previously undescribed gene (2b) encoded by RNA 2 of the tripartite RNA genome of CMV is required for systemic virus spread and disease induction in its hosts. Herein we report that when this CMV gene is replaced by its homologue from tomato aspermy cucumovirus (TAV), the resultant hybrid virus is significantly more virulent, induces earlier onset of systemic symptoms, and accumulates to a higher level in seven host species from three families than either of the parents. Our results indicate that CMV and the TAV 2b protein interact synergistically despite the fact that no synergism occurs in double infections with the two parental viruses. To our knowledge, this is the first example of an interspecific hybrid made from plant or animal RNA viruses that is more efficient in systemic infection of a number of hosts than the naturally occurring parents. As CMV and the hybrid virus accumulated to a similar level in the infected tobacco protoplasts, the observed synergistic responses most likely resulted from an increased efficacy of the hybrid virus in systemic spread in host plants provided by the TAV 2b protein. The relevance of our finding to the application of pathogen-derived resistance is discussed.

As a part of an overall project on fishculture development techniques in Tanzania, hybridization between Tilapia zillii and Tilapia andersonii was carried out at the Freshwater Fisheries Institute, Nyegezi, Tanzania. T. andersonii, a plankton feeder, is not indigenous to Tanzania but was introduced in 1968 from Zambia for certain specific purpose. T. zillii, a macrovegetation feeder, is present locally and is common. In the present studies T. zillii (245.0 mm/260.0 g) female was hybridized with T. andersonii (288.0 mm/350.0 g) male. Under cement cistern conditions it was only after about four months of acclimatization that hybridization between the two occurred. About 1,637 interspecific hybrid fry were produced in a single brood. Eggs were adhesive and parental care shown by the female, the male being driven away. Growth under cistern conditions was slow, attaining a size of 134.8 mm/44.3 g in 10 months. But this growth rate need not be taken as ideal. In body shape, colouration and other morphometric characters the hybrids had inherited from both parents. The number of gill rakers among the hybrids was eighteen which was intermediate between T. zillii (12) and T. andersonii (27). Among one hundred and seventy two specimens (106.0 mm - 168.0 mm) cut and examined the sex ration was hundred per cent males and all of them were between II and IV stages of maturity. This is the first report of fish hybridization from Tanzania and possibly the first report on hybridization between T. zillii and T. andersonii. The full significanoe of the findings and its role in African fishculture is discussed.
