We characterized four Brazilian trypanosomes isolated from domestic rats and three from captive nonhuman primates that were morphologically similar to T. lewisi, a considered non-pathogenic species restricted to rodents and transmitted by fleas, despite its potential pathogenicity for infants. These isolates were identified as T. lewisi by barcoding using V7V8 SSU rDNA sequences. In inferred phylogenetic trees, all isolates clustered tightly with reference T. lewisi and T. lewisi-like trypanosomes from Europe, Asia and Africa and despite their high sequence conservation formed a homogeneous clade separate from other species of the subgenus T. (Herpetosoma). With the aim of clearly resolving the relationships between the Brazilian isolates from domestic rats and primates, we compared sequences from more polymorphic ITS rDNA. Results corroborated that isolates from Brazilian rats and monkeys were indeed of the same species and quite close to T. lewisi isolates of humans and rats from different geographical regions. Morphology of the monkey isolates and their behaviour in culture and in experimentally infected rats were also compatible with T. lewisi. However, infection with T. lewisi is rare among monkeys. We have examined more than 200 free-ranging and 160 captive monkeys and found only three infected individuals among the monkeys held in captivity. The findings of this work suggest that proximity of monkeys and infected rats and their exposure to infected fleas may be responsible for the host switching of T. Iewisi from their natural rodent species to primates. This and previous studies reporting T. lewisi in humans suggest that this trypanosome can cause sporadic and opportunistic fleaborne infection in primates. (C) 2010 Elsevier B.V. This case study from North Spain, highlights the importance of the collection of mites in addition to insects, from crime scenes or corpses subjected to environmental constraints that reduce or minimise insect activity, such as hanged corpses. In addition, this analysis highlights the relevance of arthropods’ collection in the field, even after the corpse has been moved away for autopsy. Four species of mites, phoretic on carrion (Silphidae) and rove (Staphylinidae) beetles, complemented and reinforced the autopsy analysis as well as the scarce information provided by insect activity. Poecilochirus carabi Canestrini & Canestrini, 1882 and Poecilochirus (Physoparasitus) davydovae Hyatt, 1980 (Mesostigmata: Parasitidae) were found in association with two Silphidae, Nicrophorus Fabricius, 1775 and Necrodes Leach, 1815, only when sampled in the autopsy room; this is suggestive of host-switching of mites and was likely due to the lack of availability of specific carriers in the field. The interpretation of the activity of Parasitidae mites both in the field and the autopsy room allows a better understanding of the timing and circumstances of decomposition. Phoretic deutonymphs of Pelzneria Scheucher 1957 (Astigmata: Histiostomatidae) were highly abundant, mostly P. crenulata Oudemans, 1909 and are reported for the first time on a Staphylinidae rove beetle, Creophilus maxillosus (L., 1758). Surprisingly, in this case study no Pelzneria were associated with the Silphidae found, which are their most common hosts, such as Necrodes littoralis (L., 1758) and Nicrophorus interruptus (Stephens, 1830). All histiostomatids were removed from the staphylinid (rove beetle) collected from the soil, at the scene of death, suggesting a recent arrival of the beetle. The occurrence of Staphylinidae beetles and their associated mites, such as Parasitidae and Pelzneria, and the information they provided would have been easily overlooked or lost if only the autopsy sampling would have been considered in the analysis of the case. The four mite species are reported for the first time for the Iberian Peninsula.

It is thought that speciation in phytophagous insects is often due to colonization of novel host plants, because radiations of plant and insect lineages are typically asynchronous. Recent phylogenetic comparisons have supported this model of diversification for both insect herbivores and specialized pollinators. An exceptional case where contemporaneous plant-insect diversification might be expected is the obligate mutualism between fig trees (Ficus species, Moraceae) and their pollinating wasps (Agaonidae, Hymenoptera). The ubiquity and ecological significance of this mutualism in tropical and subtropical ecosystems has long intrigued biologists, but the systematic challenge posed by >750 interacting species pairs has hindered progress toward understanding its evolutionary history. In particular, taxon sampling and analytical tools have been insufficient for large-scale cophylogenetic analyses. Here, we sampled nearly 200 interacting pairs of fig and wasp species from across the globe. Two supermatrices were assembled: on an average, wasps had sequences from 77% of 6 genes (5.6 kb), figs had sequences from 60% of 5 genes (5.5 kb), and overall 850 new DNA sequences were generated for this study. We also developed a new analytical tool, Jane 2, for event-based phylogenetic reconciliation analysis of very large data sets. Separate Bayesian phylogenetic analyses for figs and fig wasps under relaxed molecular clock assumptions indicate Cretaceous diversification of crown groups and contemporaneous divergence for nearly half of all fig and pollinator lineages. Event-based cophylogenetic analyses further support the codiversification hypothesis. Biogeographic analyses indicate that the present-day distribution of fig and pollinator lineages is consistent with a Eurasian origin and subsequent dispersal, rather than with Gondwanan vicariance. Overall, our findings indicate that the fig-pollinator mutualism represents an extreme case among plant-insect interactions of coordinated dispersal and long-term codiversification.

Trypanosomes from crocodilians were obtained by haemoculturing. Growth behaviour, morphology, and ultrastructural features complement the molecular description of two new species strongly supported by phylogenetic analyses. Results The inferred phylogenies disclosed a strongly supported crocodilian-restricted clade comprising three subclades. The subclade T. grayi comprised the African Trypanosoma grayi from Crocodylus niloticus and tsetse flies. The subclade T. ralphi comprised alligatorid trypanosomes represented by Trypanosoma ralphi n. sp. from Melanosuchus niger, Caiman crocodilus and Caiman yacare from Brazilian river basins. T. grayi and T. ralphi were sister subclades. The basal subclade T. terena comprised alligatorid trypanosomes represented by Trypanosoma terena n. sp. from Ca. yacare sharing hosts and basins with the distantly genetic related T. ralphi. This subclade also included the trypanosome from Ca. crocodilus from the Orinoco basin in Venezuela and, unexpectedly, a trypanosome from the African crocodilian Osteolaemus tetraspis. Conclusion The close relationship between South American and African trypanosomes is consistent with paleontological evidence of recent transoceanic dispersal of Crocodylus at the Miocene/Pliocene boundaries (4–5 mya), and host-switching of trypanosomes throughout the geological configuration of South American hydrographical basins shaping the evolutionary histories of the crocodilians and their trypanosomes.

The aphid parasitoid Lysiphlebus testaceipes is a potentially valuable biological control agent of Aphis gossypii a major worldwide pest of cotton. One means of increasing the abundance of a biological control agent is to provide an alternative host habitat adjacent to cropping, from which they can provide pest control services in the crop. Host selection and parasitism rate of an alternative host aphid, Aphis craccivora by L. testaceipes were studied in a series of experiments that tested its host suitability relative to A. gossypii on cotton, hibiscus and mungbean. Host acceptance, as measured by number of ovipositions was much greater in A. craccivora compared to A. gossypii, while more host aphids were accepted on mungbean than cotton. When given a choice L. testaceipes attacks more 4th instar and adult stages (63% and 70%, respectively) of both hosts than 2nd instar nymphs (47%). In a switching (host choice) experiment, L. testaceipes preferentially attacked A. craccivora on mungbean over A. gossypii on cotton. Observations of parasitoid contact with A. gossypii cornicle secretion suggest it provides a useful deterrent against parasitoid attack. From these experiments it appears L. testaceipes has a preference for A. craccivora and mungbean compared to A. gossypii and cotton, in this respect using A. craccivora and mungbean as alternative habitat may not work as the parasitoid is unlikely to switch away from its preferred host. © 2012.

Bulk Ge(15)Te(85 - x)Sn(x) and Ge(17)Te(83 - x)Sn(x) glasses, are found to exhibit memory type electrical switching. The switching voltages (V(t)) and thermal stability of Ge(15)Te(85 - x)Sn(x) and Ge(17)Te(83 - x)Sn(x) glasses are found to decrease with Sn content. The composition dependence of v, has been understood on the basis of the decrease in the OFF state resistance and thermal stability of these glasses with tin addition. X-ray diffraction studies reveal that no elemental Sn or Sn compounds with Te or Ge are present in thermally crystallized Ge-Te-Sn samples. This indicates that Sn atoms do not interact with the host matrix and form a phase separated network of its own, which remains in the parent glass matrix as an inclusion. Consequently, there is no enhancement of network connectivity and rigidity. The thickness dependence of switching voltages of Ge(15)Te(85 - x)Sn(x) and Ge(17)Te(83 - x)Sn(x) glasses is found to be linear, in agreement with the memory switching behavior shown by these glasses. In this letter, the uniform lying helix (ULH) liquid crystal texture, required for the flexoelectro-optic effect, is polymer stabilized by the addition of a small percentage of reactive mesogen to a high-tilt-angle (φ>60°) bimesogenic chiral nematic host. The electro-optic response is measured for a range of reactive mesogen concentration mixtures, and compared to the large-tilt-angle switch of the pure chiral nematic mixture. The optimum concentration of reactive mesogen, which is found to provide ample stabilization of the texture with minimal impact on the electro-optic response, is found to be approximately 3%. Our results indicate that polymer stabilization of the ULH texture using a very low concentration of reactive mesogen is a reliable way of ruggedizing flexoelectro-optic devices without interfering significantly with the electro-optics of the effect, negating the need for complicated surface alignment patterns or surface-only polymerization. The polymer stabilization is shown to reduce the temperature dependence of the flexoelectro-optic response due to "pinning" of the chiral nematic helical pitch. This is a restriction of the characteristic thermochromic behavior of the chiral nematic. Furthermore, selection of the temperature at which the sample is ultraviolet cured allows the tilt angle to be optimized for the entire chiral nematic temperature range. The response time, however, remains more sensitive to operating temperature than curing temperature. This allows the sample to be cured at low temperature and operated at high temperature, providing simultaneous optimization of these two previously antagonistic performance aspects. © 2006 American Institute of Physics.

We present a study on a series of dye guest-host mixtures using fluorescent perylene-based molecules as the guest dye in an organosiloxane host. These hosts have temperature-independent switching, at room temperature, through 90° for fields of the order of 10 Vrms/μm. Perylene molecules have been grafted onto the organosiloxane moiety via an alkyl spacer producing novel and rugged fluorescent dyes that are readily miscible in the host. Micro-separation of the low molar mass siloxane groups in the mesophases tend to form smectic phases. These planes produce an effective two-dimensional polymer backbonethat engenders the rugged mechanical properties of polymeric liquid crystals onto these low molar mass ferroelectric liquid crystals. In this study we show how the introduction of the dye molecules affects the electro-optic properties of the organosiloxane host. © 2001 OPA (Overseas Publishers Association) N.V. Problème: Ma thèse porte sur l’identité individuelle comme interrogation sur les enjeux personnels et sur ce qui constitue l’identification hybride à l’intérieur des notions concurrentielles en ce qui a trait à l’authenticité. Plus précisément, j’aborde le concept des identifications hybrides en tant que zones intermédiaires pour ce qui est de l’alternance de codes linguistiques et comme négociation des espaces continuels dans leur mouvement entre les cultures et les langues. Une telle négociation engendre des tensions et/ou apporte le lien créatif. Les tensions sont inhérentes à n’importe quelle construction d’identité où les lignes qui définissent des personnes ne sont pas spécifiques à une culture ou à une langue, où des notions de l’identité pure sont contestées et des codes communs de l’appartenance sont compromis. Le lien créatif se produit dans les exemples où l’alternance de code linguistique ou la négociation des espaces produit le mouvement ouvert et fluide entre les codes de concurrence des références et les différences à travers les discriminations raciales, la sexualité, la culture et la langue. Les travaux que j’ai sélectionnés représentent une section transversale de quelques auteurs migrants provenant de la minorité en Amérique du Nord qui alternent les codes linguistiques de cette manière. Les travaux détaillent le temps et l’espace dans leur traitement de l’identité et dans la façon dont ils cernent l’hybridité dans les textes suivants : The Woman Warrior de Maxine Hong Kingston (1975-76), Hunger of Memory de Richard Rodriguez (1982), Comment faire l’amour avec un nègre sans se fatiguer de Dany Laferrière (1985), Borderlands/La Frontera de Gloria Anzalduá (1987), Lost in Translation de Eva Hoffman (1989), Avril ou l’anti-passion de Antonio D’Alfonso (1990) et Chorus of Mushrooms de Hiromi Goto (1994). Enjeux/Questions La notion de l’identification hybride est provocante comme sujet. Elle met en question l’identité pure. C’est un sujet qui a suscité beaucoup de discussions tant en ce qui a trait à la littérature, à la politique, à la société, à la linguistique, aux communications, qu’au sein même des cercles philosophiques. Ce sujet est compliqué parce qu’il secoue la base des espaces fixes et structurés de l’identité dans sa signification culturelle et linguistique. Par exemple, la notion de patrie n’a pas les représentations exclusives du pays d’origine ou du pays d’accueil. De même, les notions de race, d’appartenance ethnique, et d’espaces sexuels sont parfois négativement acceptées si elles proviennent des codes socialement admis et normalisés de l’extérieur. De tels codes de la signification sont souvent définis par l’étiquette d’identification hétérosexuelle et blanche. Dans l’environnement généralisé d’aujourd’hui, plus que jamais, une personne doit négocier qui elle est, au sens de son appartenance à soi, en tant qu’individu et ce, face aux modèles locaux, régionaux, nationaux, voire même globaux de la subjectivité. Nous pouvons interpréter ce mouvement comme une série de couches superposées de la signification. Quand nous rencontrons une personne pour la première fois, nous ne voyons que la couche supérieure. D’ailleurs, son soi intérieur est caché par de nombreuses couches superposées (voir Joseph D. Straubhaar). Toutefois, sous cette couche supérieure, on retrouve beaucoup d’autres couches et tout comme pour un oignon, on doit les enlever une par une pour que l’individualité complète d’une personne soit révélée et comprise. Le noyau d’une personne représente un point de départ crucial pour opposer qui elle était à la façon dont elle se transforme sans cesse. Sa base, ou son noyau, dépend du moment, et comprend, mais ne s’y limite pas, ses origines, son environnement et ses expériences d’enfance, son éducation, sa notion de famille, et ses amitiés. De plus, les notions d’amour-propre et d’amour pour les autres, d’altruisme, sont aussi des points importants. Il y a une relation réciproque entre le soi et l’autre qui établit notre degré d’estime de soi. En raison de la mondialisation, notre façon de comprendre la culture, en fait, comment on consomme et définit la culture, devient rapidement un phénomène de déplacement. À l’intérieur de cette arène de culture généralisée, la façon dont les personnes sont à l’origine chinoises, mexicaines, italiennes, ou autres, et poursuivent leur évolution culturelle, se définit plus aussi facilement qu’avant. Approche Ainsi, ma thèse explore la subjectivité hybride comme position des tensions et/ou des relations créatrices entre les cultures et les langues. Quoique je ne souhaite aucunement simplifier ni le processus, ni les questions de l’auto-identification, il m’apparaît que la subjectivité hybride est aujourd’hui une réalité croissante dans l’arène généralisée de la culture. Ce processus d’échange est particulièrement complexe chez les populations migrantes en conflit avec leur désir de s’intégrer dans les nouveaux espaces adoptés, c’est-à-dire leur pays d’accueil. Ce réel désir d’appartenance peut entrer en conflit avec celui de garder les espaces originels de la culture définie par son pays d’origine. Ainsi, les références antérieures de l’identification d’une personne, les fondements de son individualité, son noyau, peuvent toujours ne pas correspondre à, ou bien fonctionner harmonieusement avec, les références extérieures et les couches d’identification changeantes, celles qu’elle s’approprie du pays d’accueil. Puisque nos politiques, nos religions et nos établissements d’enseignement proviennent des représentations nationales de la culture et de la communauté, le processus d’identification et la création de son individualité extérieure sont formées par le contact avec ces établissements. La façon dont une personne va chercher l’identification entre les espaces personnels et les espaces publics détermine ainsi le degré de conflit et/ou de lien créatif éprouvé entre les modes et les codes des espaces culturels et linguistiques. Par conséquent, l’identification des populations migrantes suggère que la « community and culture will represent both a hybridization of home and host cultures » (Straubhaar 27). Il y a beaucoup d’écrits au sujet de l’hybridité et des questions de l’identité et de la patrie, toutefois cette thèse aborde la valeur créative de l’alternance de codes culturels et linguistiques. Ce que la littérature indiquera Par conséquent, la plate-forme à partir de laquelle j’explore mon sujet de l’hybridité flotte entre l’interprétation postcoloniale de Homi Bhabha concernant le troisième espace hybride; le modèle d’hétéroglossie de Mikhail Bakhtine qui englobent plusieurs de mes exemples; la représentation de Roland Barthes sur l’identité comme espace transgressif qui est un modèle de référence et la contribution de Chantal Zabus sur le palimpseste et l’alternance de codes africains. J’utilise aussi le modèle de Sherry Simon portant sur l’espace urbain hybride de Montréal qui établit un lien important avec la valeur des échanges culturels et linguistiques, et les analyses de Janet Paterson. En effet, la façon dont elle traite la figure de l’Autre dans les modèles littéraires au Québec fournisse un aperçu régional et national de l’identification hybride. Enfin, l’exploration du bilinguisme de Doris Sommer comme espace esthétique et même humoristique d’identification situe l’hybridité dans une espace de rencontre créative. Conséquence Mon approche dans cette thèse ne prétend pas résoudre les problèmes qui peuvent résulter des plates-formes de la subjectivité hybride. Pour cette raison, j’évite d’aborder toute approche politique ou nationaliste de l’identité qui réfute l’identification hybride. De la même façon, je n’amène pas de discussion approfondie sur les questions postcoloniales. Le but de cette thèse est de démontrer à quel point la subjectivité hybride peut être une zone de relation créatrice lorsque l’alternance de codes permet des échanges de communication plus intimes entre les cultures et les langues. C’est un espace qui devient créateur parce qu’il favorise une attitude plus ouverte vis-à-vis les différents champs qui passent par la culture, aussi bien la langue, que la sexualité, la politique ou la religion. Les zones hybrides de l’identification nous permettent de contester les traditions dépassées, les coutumes, les modes de communication et la non-acceptation, toutes choses dépassées qui emprisonnent le désir et empêchent d’explorer et d’adopter des codes en dehors des normes et des modèles de la culture contenus dans le discours blanc, dominant, de l’appartenance culturelle et linguistique mondialisée. Ainsi, il appert que ces zones des relations multi-ethniques exigent plus d’attention des cercles scolaires puisque la population des centres urbains à travers l’Amérique du Nord devient de plus en plus nourrie par d’autres types de populations. Donc, il existe un besoin réel d’établir une communication sincère qui permettrait à la population de bien comprendre les populations adoptées. C’est une invitation à stimuler une relation plus intime de l’un avec l’autre. Toutefois, il est évident qu’une communication efficace à travers les frontières des codes linguistiques, culturels, sexuels, religieux et politiques exige une négociation continuelle. Mais une telle négociation peut stimuler la compréhension plus juste des différences (culturelle ou linguistique) si des institutions académiques offrent des programmes d’études intégrant davantage les littératures migrantes. Ma thèse vise à illustrer (par son choix littéraire) l’identification hybride comme une réalité importante dans les cultures généralisées qui croissent toujours aujourd’hui. Les espaces géographiques nous gardent éloignés les uns des autres, mais notre consommation de produits exotiques, qu’ils soient culturels ou non, et même notre consommation de l’autre, s’est rétrécie sensiblement depuis les deux dernières décennies et les indicateurs suggèrent que ce processus n’est pas une tendance, mais plutôt une nouvelle manière d’éprouver la vie et de connaître les autres. Ainsi les marqueurs qui forment nos frontières externes, aussi bien que ces marqueurs qui nous définissent de l’intérieur, exigent un examen minutieux de ces enjeux inter(trans)culturels, surtout si nous souhaitons nous en tenir avec succès à des langues et des codes culturels présents, tout en favorisant la diversité culturelle et linguistique. The human malaria parasite Plasmodium falciparum expresses erythrocyte-surface directed variant antigens which are important virulence factors Many are transcribed from multigene families and presumably their mode of expression is strictly controlled to guarantee immune evasion in the human host. In order to elucidate the dynamics of rif transcription and to investigate if rif switching is comparable to var switching we monitored rif variant gene expression in parasites with different cytoadhesive properties as well as after a number of reinvasions. We found identical transcripts in parasite lines with different adhesive phenotypes suggesting that rif genes do not have a critical role in determining the cytoadhesion specificity of infected erythrocytes. We show for the first time that rif genes may show a conserved mode of transcription, maintaining the previously dominant rif transcript in subsequent reinvasions, but also observed rapid switching at rates up to 45% per generation, much higher than for the var gene family. The ferroelectric and the dielectric behaviors of binary blends formed by an equi-molar Poly(vinylidene fluoride trifluoroethylene) copolymer [P(VDF-TrFE)] and Poly(methyl methacrylate) [PMMA] were investigated, for several PMMA compositions. For 40 wt.% or more PMMA contents, the blends are completely amorphous. Below this value, they crystallize in the usual Cm2m polar structure of P(VDF-TrFE). The ferroelectric switching characteristics and the dielectric response of the blends demonstrate the formation of dynamically stable ferroelectric domains. Moreover, the blended films are highly transparent in the optical region. Therefore, thin films of these binary blends are good candidates as host materials for nonlinear optical applications.

Sustained growth of solid tumours can rely on both the formation of new and the co-option of existing blood vessels. Current models suggest that binding of angiopoietin-2 (Ang-2) to its endothelial Tie2 receptor prevents receptor phosphorylation, destabilizes blood vessels, and promotes vascular permeability. In contrast, binding of angiopoietin-1 (Ang-1) induces Tie2 receptor activation and supports the formation of mature blood vessels covered by pericytes. Despite the intense research to decipher the role of angiopoietins during physiological neovascularization and tumour angiogenesis, a mechanistic understanding of angiopoietin function on vascular integrity and remodelling is still incomplete. We therefore assessed the vascular morphology of two mouse mammary carcinoma xenotransplants (M6378 and M6363) which differ in their natural angiopoietin expression. M6378 displayed Ang-1 in tumour cells but no Ang-2 in tumour endothelial cells in vivo. In contrast, M6363 tumours expressed Ang-2 in the tumour vasculature, whereas no Ang-1 expression was present in tumour cells. We stably transfected M6378 mouse mammary carcinoma cells with human Ang-1 or Ang-2 and investigated the consequences on the host vasculature, including ultrastructural morphology. Interestingly, M6378/Ang-2 and M6363 tumours displayed a similar vascular morphology, with intratumoural haemorrhage and non-functional and abnormal blood vessels. Pericyte loss was prominent in these tumours and was accompanied by increased endothelial cell apoptosis. Thus, overexpression of Ang-2 converted the vascular phenotype of M6378 tumours into a phenotype similar to M6363 tumours. Our results support the hypothesis that Ang-1/Tie2 signalling is essential for vessel stabilization and endothelial cell/pericyte interaction, and suggest that Ang-2 is able to induce a switch of vascular phenotypes within tumours.
