Six-week-old lambs were inoculated orally with 10(9) cfu of an antibiotic-resistance marked four-strain mixture of enterohaemorrhagic Escherichia coli (EHEC) O157:H7 to investigate faecal excretion and intestinal colonisation. In the first experiment, three E. coli O157:H7 isolates were not detected in the faeces of any lambs beyond day 8 post inoculation (pi), or from any of the tissues derived from inoculated animals. One strain, 140065 Nal(r), was isolated from the caecum and colon of one lamb on day 9 pi, from the rectum of another on day 22 pi and persisted in the faeces for up to 28 days pi. All animals remained clinically normal throughout the study period and histological evidence of adhesion of E. coli O157:H7 to the intestinal mucosa was not found. In a separate experiment, four 6-week-old lambs were inoculated orally with 10(9) efu of E. coli O157:H7 strain 140065 Nal(r) alone. Faecal samples were positive for this strain until the end of the experiment (day 19 pi). This strain was also recovered from the gastrointestinal tract of lambs on days 6, 18 and 19 pi, but was not isolated at day 17 pi. When sampled separately, rectum and terminal colon contents contained higher numbers of the inoculated strain than the intestinal tissue at these sites. Animals inoculated with O157:117 strain 140065 Nal(r) alone produced soft faeces from day 5 pi onwards. Although attaching and effacing lesions were observed in the caecum, proximal colon and rectum in one animal on day 18 pi, the adherent bacteria did not stain with antiserum raised against the O157 antigen.

Four 1-week trials were conducted to determine the effects of feeding rates on growth performance and body proximate composition of white sturgeon larvae during each of the first 4 weeks after initiation of feeding. Feeding rates (% body weight day(-1)) were 10, 20, 30, 40, 50, and 60 for trial I; 5, 10, 15, 20, 25, and 30 for trial II; and 2.5, 5.0, 7.5, 10.5, 12.5, and 15.0 for trials III and TV Four tanks with 200 larvae each were randomly assigned to each of the six feeding rates. Average initial body weights of the larvae were 49, 94, 180, and 366 mg, respectively, for trials I-IV. The larvae were kept at 19-20 degreesC in circular tanks and fed continuously one of two commercial salmonid soft-moist feeds using automatic feeders. Proximate composition (%) of the feeds for trials I-III and IV were 13.9 and 14.9 moisture, 52.5 and 50.0 crude protein, 10.3 and 12.9 crude fat, and 8.1 and 8.7 ash, respectively. Except mortality in trial I, gain per food fed in trial III, and body ash in all trials, growth performance and body composition were significantly (P<0.05) affected by all feeding rates. Broken line analysis on specific growth rates indicated the optimum feeding rates of white sturgeon larvae to be 26%, 13%, 11%, and 6% body weight day-respectively, for weeks 1, 2, 3, and 4 after initiation of feeding. Perante uma sociedade em célere envelhecimento demográfico e permanente avanço tecnológico, justifica-se a aposta em estudos que potenciem a ação comunicativa e a diminuição do isolamento social decorrente das perdas biopsicossociais associadas à idade sénior. Esta tese possui quatro objetivos de estudo: i) pretende-se investigar qual é o impacto da utilização das Tecnologias de Informação e Comunicação (TIC) no autoconceito (AC), no ânimo e na qualidade de vida (QV) de um grupo de seniores; ii) perceber se existe e qual a relação entre as variáveis independentes sexo, idade, estado civil, escolaridade, profissão, IPSS, regime de frequência, tempo na IPSS, orientação para frequentar a IPSS, visita de familiares e visita de amigos e as variáveis dependentes AC, ânimo, QV e respetivos fatores e domínios, nos momentos de pré e pós-teste; iii) perceber se a sua participação no processo de conceptualização de um serviço de comunicação assíncrona, email, influencia a sua usabilidade ao nível das componentes eficácia, eficiência e satisfação; iv) e sugerir a componente política da comunidade online sénior em desenvolvimento no âmbito do Projeto SEDUCE. Para o desenvolvimento do estudo estabeleceram-se parcerias com quatro Instituições Particulares de Segurança Social do concelho de Aveiro, integradas no âmbito do projeto SEDUCE. Os instrumentos utilizados para a avaliação do autoconceito, do ânimo e da qualidade de vida foram o Inventário Clínico de Auto-Conceito, a Escala de Ânimo do Centro Geriátrico de Philadelphia e a Escala de Qualidade de Vida da Organização Mundial de Saúde WHOQOL-Bref, respetivamente. No processo de conceptualização do serviço de email e da componente política da comunidade online utilizou-se a observação participativa e o contextual design. O estudo envolveu a participação de 42 seniores distribuídos por duas condições experimentais: 22 seniores do grupo experimental utilizaram as TIC duas vezes por semana (em sessões de 90 minutos cada, num total de 80 sessões) e 19 seniores do grupo de controlo passivo não experimentaram qualquer intervenção. Para a avaliação das variáveis psicossociais realizaram-se dois momentos de avaliação, antes e depois de 11 meses de intervenção, de Agosto de 2011 a Julho de 2012. Ao longo das sessões de envolvimento com as TIC observou-se que os seniores apresentam, continuamente, dificuldades em: manipular o rato e percecionar a sua ação no monitor; fazer a distinção entre teclas (enter, spacebar, delete, caps lock entre outras); em utilizar duplas teclas para colocar pontuação e acentuação; iniciar atividades no Microsoft Office Word; selecionar a informação disponibilizada em motores de pesquisa; perceber quais as zonas clicáveis; falta de confiança em efetivar ações; receio em iniciar nova atividades, pela falta de conhecimento e pelo medo de errar; memorizar endereços de email e passwords; e dar continuidade às tarefas. Na utilização do serviço de email consideram importante receber resposta quando enviam uma mensagem, assim como responder sempre aos remetentes; raramente colocam assunto nas mensagens; e expressam grande satisfação ao receber mensagens de familiares e/ou amigos. O processo de desenvolvimento de serviços com a participação ativa dos seniores revela-se exequível mas é necessário adaptar as práticas: os processos devem ser iterativos; evitar linguagem formal; clarificar o objetivo; deixar os seniores pensar em voz alta; dar-lhes tempo; mantê-los focados e não conduzi-los nas tarefas. Os resultados sugerem que houve aumento significativo do domínio físico da qualidade de vida do grupo experimental. Os participantes que exprimiram maiores níveis de satisfação ao utilizar as TIC apresentam uma perspetiva mais positiva sobre a maturidade psicológica e menos solidão e insatisfação. No grupo experimental e no grupo de controlo passivo verificam-se relações entre as variáveis independentes e as variáveis dependentes, quer no momento de pré-teste como de pós-teste. Conclui-se que a participação dos seniores no processo de conceptualização do serviço de email permitiu fomentar a componente eficácia da usabilidade mas não a satisfação ao utilizar o mesmo. Os resultados sobre a eficiência são inconclusivos. Sobre a componente política os seniores validam a existência de termos de utilização que orientem o comportamento de todos os utilizadores, assim como de uma política de privacidade. A área de registo proposta é adequada ao utilizador sénior.

Ruminants are regarded as a primary reservoir for Escherichia coli O157:H7, an important human pathogen. Intimin, encoded by the Locus of Enterocyte Effacement by E. coli O157:H7 organisms, has been cited as one bacterial mechanism of colonisation of the gastrointestinal tract. To confirm this and to test whether a non-toxigenic E. coli O157:H7 strain would colonise and persist in a sheep model, E. coli O157:H7 strain NCTC12900, that lacks Shiga toxin (stx) genes, was evaluated for use in a sheep model of persistence. Following oral inoculation of six-week-old sheep, persistent excretion of NCTC12900 was observed for up to 48 days. E. coli O157-associated attaching-effacing (AE) lesions were detected in the caecum and rectum of one six-week-old lamb, one day after inoculation. This is the first recorded observation of AE lesions in orally inoculated weaned sheep. Also, mean faecal excretion scores of NCTC12900 and an isogenic intimin (eae)-deficient mutant were determined from twenty-four six-week-old orally inoculated sheep. The eae mutant was cleared within 20 days and had lower mean excretion scores at all time points after day one post inoculation compared with the parental strain that was still being excreted at 48 days. Tissues were collected post mortem from animals selected at random from the study groups over the time course of the experiment. The eae mutant was detected in only 1/43 samples but the parental strain was recovered from 64/140 samples primarily from the large bowel although rumen, duodenum, jejunum, and ileum were culture positive especially from animals that were still excreting at and beyond 27 days after inoculation.

Prospective measurement of nutrition, cognition, and physical activity in later life would facilitate early detection of detrimental change and early intervention but is hard to achieve in community settings. Technology can simplify the task and facilitate daily data collection. The Novel Assessment of Nutrition and Ageing (NANA) toolkit was developed to provide a holistic picture of an individual's function including diet, cognition and activity levels. This study aimed to validate the NANA toolkit for data collection in the community. Forty participants aged 65 years and over trialled the NANA toolkit in their homes for three 7-day periods at four-week intervals. Data collected using the NANA toolkit were compared with standard measures of diet (four-day food diary), cognitive ability (processing speed) and physical activity (self-report). Bland–Altman analysis of dietary intake (energy, carbohydrates, protein fat) found a good relationship with the food diary and cognitive processing speed and physical activity (hours) were significantly correlated with their standard counterparts. The NANA toolkit enables daily reporting of data that would otherwise be collected sporadically while reducing demands on participants; older adults can complete the daily reporting at home without a researcher being present; and it enables prospective investigation of several domains at once

O experimento foi realizado com o objetivo de verificar o melhor programa de fornecimento de ração para matrizes de corte pós - pico de produção de ovos. Três mil, cento e sessenta e oito fêmeas e 352 machos matrizes de corte da linhagem Cobb foram distribuídos em um delineamento experimental inteiramente casualizado com quatro tratamentos e oito repetições por tratamento, sendo 99 fêmeas e 11 machos por unidade experimental. Os programas avaliados consistiram em: Programa 1 - Redução de 1 g de ração/ave/dia/semana para cada redução de 0,5 pontos na massa de ovos; Programa 2 - Redução de 1 g de ração/ave/dia/semana após o pico até o final do período de produção; Programa 3 - Redução de 3 g de ração/ave/dia/semana na 1ª semana após o pico, 2 g de ração/ave/dia/semana na 2ª semana e 1 g de ração/ave/dia/semana nas semanas subsequentes; Programa 4 - Redução de 5 g de ração/ave/dia/semana na 1ª semana após o pico, 3 g de ração/ave/dia/semana na 2ª semana e 1 g de ração/ave/dia/semana nas semanas subseqüentes. de acordo com os resultados obtidos para consumo de ração por ovo produzido, característica de importância na avaliação do desempenho das matrizes de corte, o melhor programa foi o programa 1, seguido pelo 3, 2 e 4. Para matrizes de corte da linhagem Cobb que atingem pico de postura no inverno, período no qual foi realizado este experimento, recomenda-se o programa 1 para alimentação após pico por ter proporcionado uma boa relação entre consumo de ração e produção de ovos.

This study was undertaken to characterize the effects of monotonous training at lactate minimum (LM) intensity on aerobic and anaerobic performances; glycogen concentrationsin the soleus muscle, the gastrocnemius muscle and the liver; and creatine kinase (CK), free fatty acids and glucose concentrations in rats. The rats were separated into trained (n =10), baseline (n = 10) and sedentary (n=10) groups. The trained group was submitted to the following: 60 min/day, 6 day/week and intensity equivalent to LM during the 12-week training period. The training volume was reduced after four weeks according to a sigmoid function. The total CK (U/L) increased in the trained group after 12 weeks (742.0±158.5) in comparison with the baseline (319.6±40.2) and the sedentary (261.6+42.2) groups. Free fatty acids and glycogen stores (liver, soleus muscle and gastrocnemius muscle) increased after 12 weeks of monotonous training but aerobic and anaerobic performances were unchanged in relation to the sedentary group. The monotonous training at LM increased the level of energy substrates, unchanged aerobic performance, reduced anaerobic capacity and increased the serum CK concentration; however, the rats did not achieve the predicted training volume.

A participant observation method was employed :in the study of a 20-week stoppage at Ansells Brewery Limited, a constituent company of Allied Breweries (U.K.). The strike, :involving 1,000 workers, began :in opposition to the implementation of a four-day working week and culminated in the permanent closure of the brewery. The three main phases of the strike's development (i.e., its :initiation, maintenance and termination) were analysed according to a social-cognitive approach, based on the psychological imagery, beliefs, values and perceptions underlying the employees' behaviour. Previous psychological treatments of strikes have tended to ignore many of the aspects of social definition, planning and coordination that are an integral part of industrial action. The present study is, therefore, unique in concentrating on the thought processes by which striking workers .make sense of their current situation and collectively formulate an appropriate response. The Ansells strike provides an especially vivid illustration of the ways in which the seminal insights of a small number of individuals are developed, via processes of communication and:influence, into a consensual interpretation of reality. By adopting a historical perspective, it has been possible to demonstrate how contemporary definitions are shaped by the prior history of union-management relations, particularly with regard to: (a) the way that previous events were subjectively interpreted, and (b) the lessons that were learned on the basis of that experience. The present approach is psychological insofar as it deals with the cognitive elements of strike action. However, to the extent that it draws from relevant sections of the industrial relations, organizational behaviour, sociology, anthropology and linguistics literatures, it can claim to be truly interdisciplinary.

Editorial: The 2015 BCLA annual conference was another fantastic affair. It was the first time the conference was held in the beautiful city of Liverpool. The venue was great and the programme was excellent. The venue overlooked the River Mersey and many of the hotels were local boutique hotels. I stayed in one which was formerly the offices of White Star Liners—where the RMS Titanic was originally registered. The hotel decor was consistent with its historic significance. The BCLA gala dinner was held in the hugely impressive Anglican Cathedral with entertainment from a Beatles tribute band. That will certainly be a hard act to follow at the next conference in 2017. Brian Tompkins took the reigns as the new BCLA president. Professor Fiona Stapleton was the recipient of the BCLA Gold Medal Award. The winner of the poster competition was Dorota Szczesna-Iskander with a poster entitled ‘Dry Contact lens poor wettability and visual performance’. Second place was Renee Reeder with her poster entitled ‘Abnormal Rosacea as a differential diagnosis in corneal scarring’. And third place was Maria Jesus Gonzalez-Garcia with her poster entitled ‘Dry Effect of the Environmental Conditions on Tear Inflammatory Mediators Concentration in Contact Lens Wearers’. The photographic competition winner was Professor Wolfgang Sickenberger from Jena in Germany. The Editorial Panel of CLAE met at the BCLA conference for their first biannual meeting. The journal metrics were discussed. In terms of number of submissions of new papers CLAE seems to have plateaued after seeing a rapid growth in the number of submissions over the last few years. The increase over the last few years could be attributed to the fact that CLAE was awarded an impact factor for the first time in 2012. This year it seems that impact factors across nearly all ophthalmic related journals has dropped. This could in part be due to the fact that last year was a ‘Research Exercise Framework (REF) year for UK universities, where they are judged on quality of their research output. The next REF is in 2020 so we may see changes nearing that time. Looking at article downloads, there seems to be a continued rise in figures. Currently CLAE attracts around 85,000 downloads per year (this is an increase of around 10,000 per year for the last few years) and the 2015 prediction is 120,000! With this in mind and with other contributing factors too, the BCLA has decided to move to online delivery of CLAE to its members starting from issue 5 of 2015. Some members do like to flick through the pages of a hard copy of the journal so members will still have the option of receiving a hard copy through the post but the default journal delivery method will now be online. The BCLA office will send various alerts and content details to members email addresses. To access CLAE online you will need to log in via the BCLA web page, currently you then click on ‘Resources’ and then under ‘Free and Discounted Publications’ you will see CLAE. This actually takes you to CLAE’s own webpage (www.contactlensjournal.com) but you need to log in via the BCLA web page. The BCLA plans to change these weblinks so that from the BCLA web page you can link to the journal website much more easily and you have the choice of going directly into the general website for CLAE or straight to the current issue. In 2016 you will see an even easier way of accessing CLAE online as the BCLA will launch a CLAE application for mobile devices where the journal can be downloaded as a ‘flick-book’. This is a great way of bringing CLAE into the modern era where people access their information in newer ways. For many the BCLA conference was part of a very busy conference week as it was preceded by the International Association of Contact Lens Educators’ (IACLE) Third World Congress, held in Manchester on the 4 days before the BCLA conference. The first and second IACE World Congresses were held in Waterloo, Canada in 1994 and 2000 respectively and hosted by Professor Des Fonn. Professor Fonn was the recipient of the first ever IACLE lifetime achievement award. The Third IACLE World Congress saw more than 100 contact lens educators and industry representatives from around 30 countries gather in the UK for the four-day event, hosted by The University of Manchester. Delegates gained hands-on experience of innovations in teaching, such as learning delivery systems, the use of iPads in the classroom and for creating ePub content, and augmented and virtual reality technologies. IACLE members around the world also took part via a live online broadcast. Work time spread across the entire week, rather than the conventional five day working week, has meant that workers are now less able to utilise longer stretches of recreation time especially in gaining access to a full two-day break over a weekend. This paper explores the issues contributing to workers' acquisition of longer recreation time. It seeks to determine the effects of this acquisition on the quality of working and non-working time for the employee through a study of work-life balance in the construction industry. It finds that weekends are more important to achieving work-life balance than shorter days over a six-day week when working long hours. Further, 'personal time' is a key element in achieving satisfactory work-life balance for employees, and this type of 'time' is often forgone in trying to integrate the necessary and desired non-work activities in the shorter time available to workers.
