One of the most significant challenges confronting orchid researchers is the lack of specific molecular markers, mainly for species in the Neotropics. Here we report the first set of specific chloroplast microsatellite primers (cpSSR) developed for Neotropical orchids. In total, nine polymorphic cpSSR loci were isolated and characterized in four species occurring in the Brazilian Atlantic Rainforest: Epidendrum cinnabarinum, E. denticulatum, E. fulgens and E. puniceoluteum. Levels of intraspecific polymorphism were characterized using two populations for each species, with 13-20 individuals each. Allele numbers varied from two to three per locus, while the number of haplotypes ranged from three to six per species. Extensive differentiation among the taxa was detected. All markers were successfully cross-amplified in eight other different genera. These cpSSRs markers will enable novel insights into the evolution of this important Neotropical genus.

There is a great demand for simpler and less costly laboratory techniques and for more accessible procedures for orchid breeders who do not have the necessary theoretical basis to use the traditional seed and clone production methods of orchids in vitro. The aim of this study was to assess the use of sodium hypochlorite (NaClO) as a decontaminant in the process of inoculating adult orchid explants of Arundina bambusifolia and Epidendrum ibaguenses. Solutions of NaClO (1.200, 2.400, 3.600, 4.800 and 6.000 mg L-1 - equivalent to 50, 100, 150, 200 and 250 mL L-1 of commercial bleach - CB) were sprayed on the explants (1.0 mL) and the culture medium (GB5), in the presence or absence of activated charcoal (2 g L-1). The explants used were nodal segments of field-grown adult plants. The procedures for inoculating the explants were conducted outside the laminar flow chamber (LFC), except for the control treatment (autoclaved medium and explant inoculation inside the LFC). The best results for fresh weight yield, height and number of shoots were obtained using NaClO in solution at 1.200 mg L-1 (equivalent to 50 mL L-1 commercial bleach) with activated charcoal in the culture medium. Fresh weight figures were 1.10 g/jar for Arundina bambusifolia and 0.16 g/jar for Epidendrum ibaguenses. Spraying the NaClO solutions controls the contamination of the culture medium already inoculated with the explants.

Fungos micorrízicos rizoctonioides Epulorhiza spp. têm sido isolados de orquídeas do gênero Epidendrum e vêm sendo utilizados na germinação simbiótica das sementes de orquídeas. Epidendrum secundum é uma orquídea largamente distribuída em campos de altitude do Parque Estadual da Serra do Brigadeiro (PESB), Minas Gerais, e pouco se sabe sobre a associação micorrízica dessa espécie nesse parque. O objetivo deste trabalho foi avaliar a diversidade morfológica dos fungos micorrízicos rizoctonioides isolados de quatro populações de E. secundum em três regiões de um campo de altitude localizado na subserra Totem Deitado, PESB. Vinte e seis isolados fúngicos foram obtidos, todos pertencentes ao gênero Epulorhiza. As características morfológicas qualitativas e quantitativas avaliadas revelaram, de modo geral, baixa variabilidade entre os isolados obtidos de uma mesma população e de populações localizadas na mesma região, porém grande variabilidade foi observada entre os isolados obtidos das populações de diferentes regiões. Com base nessas características morfológicas, os isolados foram divididos em quatro grupos: o primeiro constituído pelos fungos obtidos das populações I e II da região A, o segundo pelos fungos da população III da região B, o terceiro pelo isolado M61 da população II da região A, e o quarto pelo único isolado obtido na população IV da região C. A variabilidade morfológica observada é um indicativo da diversidade dos fungos Epulorhiza spp. associados a E. secundum no PESB.

In the best cultivation methods of orchids, in particular of the genus Phalaenopsis, liming is a common practice. The objective of this study was to evaluate the influence of lime rates (0.0; 1.0; 2.0; 3.0; 4.0; and 5.0 g dm-3 of substrate) applied to the cultivation substrate (xaxim) on the growth of Epidendrum ibaguense seedlings. In a greenhouse, 1-L plastic pots filled with 0.8 dm³ of xaxim were irrigated such that no leachate was lost during the experiment. N, P, K, Ca, Mg, S, Fe, Zn, B, and Mn contents in roots, stems and leaves were measured. Leachate was collected by applying a sufficient water volume to obtain 25 mL from each pot. Fourteen days after lime application of 3 g dm-3, the pH of the collected leachate reached values above 7 and a value of 6.29 with the highest lime rate at the end of the experiment. The lime rate did not influence plant height, probably due to a Zn deficiency at high pH levels and a Ca deficiency in the control. Nevertheless, there was a large increase in leaf production, for number as well as for dry matter mass. There was no statistical difference between treatments in root dry matter production. Maximum dry matter production was obtained at a lime rate of 4.09 g dm-3. Zinc concentrations diminished linearly with increasing lime rates; the concentrations in all treatments were below the levels suggested as adequate in the literature (25-200 mg kg-1). Nutrient concentrations in leaves indicated deficiency of N, S, and B at the highest lime rates (4.0 and 5.0 g dm-3), and of Ca in the treatment without liming.

O objetivo deste trabalho foi avaliar a influência do inibidor da síntese de etileno aminoetoxivinilglicina (AVG) aplicado na forma de solução de condicionamento e pulverização sobre a abscisão e longevidade de inflorescências de Epidendrum ibaguense Kunth. As hastes foram colhidas e imediatamente condicionadas em solução com 0, 0,5, 1, 1,5 e 2 mM de AVG por 6, 12, 18 e 24 horas. Os mesmos tratamentos foram aplicados na forma de pulverização até o molhamento completo da inflorescência. O experimento foi conduzido em um esquema fatorial entre tempo de aplicação e doses de AVG, mais doses de AVG aplicadas em pulverização nas flores, e o delineamento foi em blocos ao acaso, com cinco repetições, com três hastes por unidade experimental. Independentemente do modo de aplicação do AVG, as concentrações utilizadas promoveram aumento da longevidade das flores em aproximadamente 70% em comparação ao controle, com resposta máxima nas concentrações de 1,5 e 2 mM. Apercentagem de abscisão de flores foi reduzida em todos os tratamentos com AVG, principalmente quando se utilizou pulverização, com decréscimo na abscisão acumulada superior a 80% nas concentrações entre 1 e 2 mM de AVG.A aplicação de AVG prolongaa longevidadee reduz a abscisãode flores de Epidendrum ibaguense.

Epidendrum paniculatum Ruiz & Pavón ocorre na América Central e do Sul. As populações estudadas desenvolvem-se como rupícola em matas semidecíduas de altitude, na Serra do Japi (Judiaí-SP). Suas flores são verdes e brancas, e fragrantes durante o dia e à noite. Na Serra do Japi, os polinizadores são borboletas da subfamília Ithomiinae e mariposas diurnas da família Arctiidae, ambas conhecidas por coletar alcalóides em flores. Os testes realizados, no entanto, demonstraram que alcalóides estão ausentes em flores de E. paniculatum, que atrai seus polinizadores provavelmente pela produção de néctar e fragrância. Os tratamentos realizados revelaram um alto grau de auto-incompatibilidade para a espécie. No entanto, devido à tendência de muitos lepidópteros permanecerem em seus micro-hábitats, muitas autopolinizações ocorrem. O alto grau de auto-incompatibilidade, somado à ineficiência por parte dos agentes polinizadores, provoca grande perda de pólen e, conseqüentemente, uma baixa produção de frutos. Mesmo com a baixa frutificação, os frutos produzidos são derivados de polinizações cruzadas. Este fato, adicionado à produção de milhares de sementes dispersas pelo vento em cada fruto, pode ser suficiente para a manutenção do número de indivíduos da espécie na região, bem como para a promoção de maior variabilidade genética e da ampla distribuição de E. paniculatum.

The goal of this paper was to test the presence of mimicry in Asclepias curassavica L., Epidendrum fulgens Brong., and Lantana camara L. The study was carried out at the Parque Estadual de Itapeva, RS, southern Brazil, from 2004 to 2006. Flowering period of each of the three species was followed up; focal observations of butterflies visiting flowers, from fixed point and during random walks were carried out. We also estimated the frequency of pollinaria removal in the orchid, as well as its mode of reproduction. All these variables were important for testing the mimicry hypothesis. Despite some temporal coincidences in the flowering period of two plants in the system, there was no statistical association among the three plants as to flowering period. Twenty-nine species of butterflies, as potential pollinators, were recorded, particularly Agraulis vanillae maculosa, Dryas iulia alcionea, Urbanus simplicius, Tegosa claudina, and Heliconius erato phyllis, which were the more frequent visitors of the three plants. There was association between the number of visits to L. camara and E. fulgens, based on Pearson correlation (r = 0.4603; n = 19; P = 0.0473). Pollinaria removal of E. fulgens was low, as measured by the percentage of removal (range: 0 - 10%). The analysis of the mode of reproduction of this orchid showed its pollinator-dependence, since no fruits were formed by spontaneous self-pollination. In contrast, the percentage of fruit set that resulted from geitonogamy and xenogamy was, in average, 86%. The results here shown are not conclusive as to the occurrence of a mimicry system among the three plants.

Epidendrum L. is the largest genus of Orchidaceae in the Neotropical region; it has an impressive morphological diversification, which imposes difficulties in delimitation of both infrageneric and interspecific boundaries. In this study, we review infrageneric boundaries within the subgenus Amphiglottium and try to contribute to the understanding of morphological diversification and taxa delimitation within this group. We tested the monophyly of the subgenus Amphiglottium sect. Amphiglottium, expanding previous phylogenetic investigations and reevaluated previous infrageneric classifications proposed. Sequence data from the trnL-trnF region were analyzed with both parsimony and maximum likelihood criteria. AFLP markers were also obtained and analyzed with phylogenetic and principal coordinate analyses. Additionally, we obtained chromosome numbers for representative species within the group. The results strengthen the monophyly of the subgenus Amphiglottium but do not support the current classification system proposed by previous authors. Only section Tuberculata comprises a well-supported monophyletic group, with sections Carinata and Integra not supported. Instead of morphology, biogeographical and ecological patterns are reflected in the phylogenetic signal in this group. This study also confirms the large variability of chromosome numbers for the subgenus Amphiglottium (numbers ranging from 2n = 24 to 2n = 240), suggesting that polyploidy and hybridization are probably important mechanisms of speciation within the group.

The hypothesis of gene flow between species with large differences in chromosome numbers has rarely been tested in the wild, mainly because species of different ploidy are commonly assumed to be reproductively isolated from each other because of instantaneous and strong postzygotic barriers. In this study, a broad-scale survey of molecular variation was carried out between two orchid species with different ploidy levels: Epidendrum fulgens (2n = 2x = 24 chromosomes) and Epidendrum puniceoluteum (2n = 4x = 52 chromosomes). To test the strength of their reproductive barriers, we investigated the distribution of genetic variation in sympatric and allopatric populations of these two species and conducted crossing experiments. Nuclear and plastid microsatellite loci were used to genotype 463 individuals from eight populations across the geographical range of both species along the Brazilian coastal plain. All six sympatric populations analysed presented hybrid zones, indicating that hybridization between E. fulgens and E. puniceoluteum is a common phenomenon. Bayesian assignment analysis detected the presence of F(1) and F(2) individuals and also signs of introgression, demonstrating a high potential for interspecific gene flow. Introgression occurs preferentially from E. fulgens to E. puniceoluteum. Pure parental individuals of both species display strong genotype-habitat associations, indicating that environment-dependent selection could be acting in all hybrid zones. This study suggests that hybridization and introgression are evolutionary processes playing a role in the diversification of Epidendrum and indicates the importance of investigations of hybrid zones in understanding reproductive barriers and speciation processes in Neotropical orchid species.

Epidendrum fulgens has a patchy distribution along the Atlantic Rainforest in the Brazilian coast, due to the destruction of its native habitat. Here, we report on both the development of nine new microsatellite markers isolated from this species and the characterization of their allele variability in two distant and unrelated populations. The number of alleles observed for each locus ranged from 2 to 17 with an average of 6.4 alleles per locus. These microsatellites should be valuable tools for studying the effect of habitat fragmentation on the genetic structure of E. fulgens populations.

Epidendrum puniceoluteum is an endemic orchid of Atlantic Rainforest, restricted to few populations only due to the destruction and fragmentation of its native habitat. Here, we report on the development of 10 microsatellite markers isolated from this orchid species. Genetic variability was characterized in two distant populations from Brazil coast. The number of alleles observed for each locus ranged from two to 12 and with an average of 6.4 alleles per locus. These microsatellites should be valuable tools for studying both fine-scale genetic structure of scattered E. puniceoluteum population and patterns will be useful genetic markers for other closely related taxa.

Under greenhouse conditions, Epidendrum nocturnum Jacq. plants produce fruits by both self-fertilization and cleistogamy. Although adapted to these reproductive processes the species respond also to cross-pollination. Seeds without embryos and with one embryo are usual but occasionally seeds with two, three or four embryos are produced. Multiple embryos are formed by polyembryony and apomixis. © 1985 Annals of Botany Company.

Phylogeographic studies provide an important framework for investigating the mechanisms operating during the earliest stages of speciation, as reproductive barriers can be examined among divergent lineages in a geographic context. We investigated the evolution of early stages of intrinsic postmating isolation among different populations and lineages of Epidendrum denticulatum, a Neotropical orchid distributed across different biomes in South America. We estimated genetic diversity and structure for both nuclear and plastid markers, using a haplotype network, differentiation tests, Bayesian assignment analysis, and divergence time estimates of the main lineages. Reproductive barriers among divergent lineages were examined by analyzing seed viability following reciprocal crossing experiments. Strong plastid phylogeographic structure was found, indicating that E. denticulatum was restricted to multiple refuges during South American forest expansion events. In contrast, significant phylogeographic structure was not found for nuclear markers, suggesting higher gene flow by pollen than by seeds. Large asymmetries in seed set were observed among different plastid genetic groups, suggesting the presence of polymorphic genic incompatibilities associated with cytonuclear interactions. Our results confirm the importance of phylogeographic studies associated with reproductive isolation experiments and suggest an important role for outbreeding depression during the early stages of lineage diversification. © 2013 The Society for the Study of Evolution.
