O pensamento lean, assim como os conceitos e práticas que lhe estão associados, são, hoje em dia, vistos pelas organizações como meios que promovem a busca pela excelência empresarial. A aplicação dos princípios lean ultrapassou já a barreira do domínio produtivo, conhecendo-se, cada vez mais, outros ramos de utilização, como é o caso do setor dos serviços, que tem demonstrado um potencial enorme no que diz respeito à exploração desta filosofia. No entanto, as experiências de aplicação destes princípios a outras áreas, que não a indústria de transformação, têm ainda poucos resultados concretos espelhados na literatura. Este projeto surge no sentido de contribuir para alcançar processos mais eficientes, aplicando ferramentas e metodologias da filosofia lean a processos de gestão da informação, tendo o caso prático decorrido no recém-criado Departamento de Custeio Global da ECCO. Por se tratar de um novo departamento, ergue-se a necessidade de construir e modificar ferramentas e práticas em toda a estrutura da empresa. Deste modo, o lean será utilizado como um possível caminho para promover a melhoria contínua dos vários processos, tendo em vista a obtenção de uma melhor relação entre os resultados obtidos e os recursos despendidos. Assim, serão apresentadas várias propostas de melhoria para o departamento em causa, no âmbito da gestão da informação e que, partindo de uma análise inicial baseada nos princípios lean, pretendem agregar mais valor aos processos, minimizando os desperdícios que lhes estão associados e, consequentemente, promovendo o seu melhoramento contínuo.

The obduction of equatorial 13 degrees C Water in the Pacific is investigated using a simulated passive tracer of the Consortium for Estimating the Circulation and Climate of the Ocean (ECCO). The result shows that the 13 degrees C Water initialized in the region 8 degrees N-8 degrees S, 130 degrees-90 degrees W enters the surface mixed layer in the eastern tropical Pacific, mainly through upwelling near the equator, in the Costa Rica Dome, and along the coast of Peru. Approximately two-thirds of this obduction occurs within 10 years after the 13 degrees C Water being initialized, with the upper portion of the water mass reaching the surface mixed layer in only about a month. The obduction of the 13 degrees C Water helps to maintain a cool sea surface temperature year-round, equivalent to a surface heat flux of about -6.0 W m(-2) averaged over the eastern tropical Pacific (15 degrees S-15 degrees N, 130 degrees W-eastern boundary) for the period of integration (1993-2006). During El Nino years, when the thermocline deepens as a consequence of the easterly wind weakening, the obduction of the 13 degrees C Water is suppressed, and the reduced vertical entrainment generates a warming anomaly of up to 10 W m(-2) in the eastern tropical Pacific and in particular along the coast of Peru, providing explanations for the warming of sea surface temperature that cannot be accounted for by local winds alone. The situation is reversed during La Nina years.

“Sounds of the City” is a large-scale community project and exhibition commissioned by the Metropolitan Arts Centre (MAC) and led by artists from the Sonic Arts Research Centre (SARC), Queen’s University Belfast. Over a four-month period, the artists worked together with two intergenerational groups in Belfast with the aim of addressing specific sound qualities of places, events and stories. Themes that surfaced from this process constitute the basis for the exhibition which promotes listening as a form of intersecting daily life, identity and memory. Five installations address aural contexts ranging from Belfast’s industrial heritage to the local family home. These are shaped by present and past experiences of workshop participants at Dee Street Community Centre in East Belfast and Tar Isteach in North Belfast. The themes and contents of these installations center upon the relationship between sound and memory, sound and place, and the documentation of everyday personal auditory experience.
All materials exhibited have emerged through workshops, interviews and field-recording sessions. Workshops acted as a basis from which to inform each group about the project’s aims, methods of listening, methods of documenting sound and the wider areas of soundscape studies and acoustic ecology. They also provided a central point allowing participants to organize outside activities and share material for exhibition.
“Sounds of the City” explores the relationship between sound and community through everyday life and presents a dynamic and ever-changing soundscape that shapes Belfast’s identity.

Sounds of the City has been exhibited at the MAC, Belfast 2012 and at Espaço Ecco, Brasilia 2013.

Retroviruses are both powerful evolutionary forces and dangerous threats to genome integrity. As such, they have imposed strong selective pressure on their hosts, notably triggering the emergence of restriction factors, such as TRIM5 alpha, that act as potent barriers to their cross-species transmission. TRIM5 alpha orthologues from different primates have distinct retroviral restriction patterns, largely dictated by the sequence of their C-terminal PRYSPRY domain, which binds the capsid protein of incoming virions. Here, by combining genetic and functional analyses of human and squirrel monkey TRIM5 alpha, we demonstrate that the coiled-coil domain of this protein, thus far essentially known for mediating oligomerization, also conditions the spectrum of antiretroviral activity. Furthermore, we identify three coiled-coil residues responsible for this effect, one of which has been under positive selection during primate evolution, notably in New World monkeys. These results indicate that the PRYSPRY and coiled-coil domains cooperate to determine the specificity of TRIM5 alpha-mediated capture of retroviral capsids, shedding new light on this complex event.

Although estimation of turbulent transport parameters using inverse methods is not new, there is little evaluation of the method in the literature. Here, it is shown that extended observation of the broad scale hydrography by Argo provides a path to improved estimates of regional turbulent transport rates. Results from a 20 year ocean state estimate produced with the ECCO v4 non-linear inverse modeling framework provide supporting evidence. Turbulent transport parameter maps are estimated under the constraints of fitting the extensive collection of Argo profiles collected through 2011. The adjusted parameters dramatically reduce misfits to in situ profiles as compared with earlier ECCO solutions. They also yield a clear reduction in the model drift away from observations over multi-century long simulations, both for assimilated variables (temperature and salinity) and independent variables (bio-geochemical tracers). Despite the minimal constraints imposed specifically on the estimated parameters, their geography is physically plausible and exhibits close connections with the upper ocean ocean stratification as observed by Argo. The estimated parameter adjustments furthermore have first order impacts on upper-ocean stratification and mixed layer depths over 20 years. These results identify the constraint of fitting Argo profiles as an effective observational basis for regional turbulent transport rates. Uncertainties and further improvements of the method are discussed.

In the Hydrocarbon exploration activities, the great enigma is the location of the deposits. Great efforts are undertaken in an attempt to better identify them, locate them and at the same time, enhance cost-effectiveness relationship of extraction of oil. Seismic methods are the most widely used because they are indirect, i.e., probing the subsurface layers without invading them. Seismogram is the representation of the Earth s interior and its structures through a conveniently disposed arrangement of the data obtained by seismic reflection. A major problem in this representation is the intensity and variety of present noise in the seismogram, as the surface bearing noise that contaminates the relevant signals, and may mask the desired information, brought by waves scattered in deeper regions of the geological layers. It was developed a tool to suppress these noises based on wavelet transform 1D and 2D. The Java language program makes the separation of seismic images considering the directions (horizontal, vertical, mixed or local) and bands of wavelengths that form these images, using the Daubechies Wavelets, Auto-resolution and Tensor Product of wavelet bases. Besides, it was developed the option in a single image, using the tensor product of two-dimensional wavelets or one-wavelet tensor product by identities. In the latter case, we have the wavelet decomposition in a two dimensional signal in a single direction. This decomposition has allowed to lengthen a certain direction the two-dimensional Wavelets, correcting the effects of scales by applying Auto-resolutions. In other words, it has been improved the treatment of a seismic image using 1D wavelet and 2D wavelet at different stages of Auto-resolution. It was also implemented improvements in the display of images associated with breakdowns in each Auto-resolution, facilitating the choices of images with the signals of interest for image reconstruction without noise. The study of nutrient uptake is of fundamental importance to plant nutrition, as well as tell the time (growth stage) being absorbed by the culture, also signals to the levels of fertilizer to be used. Aiming to establish the uptake of nitrogen for cultivation of cotton (Gossypium hirsutum L. r. latifolium Hutch.) led to an experiment in a Alfissol under randomized block design with two treatments, sowing conventional tillage and after tillage on the straw of previous crop (millet), with five replicates. Samples were taken according to the phenology and plants development. The dry matter accumulation in cotton is linear and increasing until the period of large boll, being more intense with the appearance of flowers and boll, or 50 to 100 days after emergence. The nitrogen content is higher in newly emerged plants, having peaks at the beginning of the appearance of squares and flowers, and from the appearance of small boll, the nitrogen content in the plant decreases linearly. The extraction of nitrogen is bigger from the appearance of flowers (55-60 days after emergence).

Background: Rhodococcus equi is associated with pyogranulomatous infections, especially in foals, and this bacterium has also emerged as a pathogen for humans, particularly immunocompromised patients. R. equi infections in pigs, wild boar (Sus scrofa) and humans are mainly due to strains carrying the intermediate virulence (VapB) plasmid. In Brazil, R. equi carrying the VapB type 8 plasmid is the most common type recovered from humans co-infected with the human immunodeficiency virus (HIV). R. equi infection in pigs and wild boar is restricted predominantly to the lymphatic system, without any reports of pulmonary manifestations. Findings. This report describes the microbiological and histopathological findings, and molecular characterization of R. equi in two bronchopneumonia cases in wild boar using PCR and plasmid profile analysis by digestion with restriction endonucleases. The histological findings were suggestive of pyogranulomatous infection, and the plasmid profile of both R. equi isolates enabled the characterization of the strains as VapB type 8. Conclusions: This is the first report of bronchopneumonia in wild boar due to R. equi. The detection of the VapB type 8 plasmid in R. equi isolates emphasize that wild boar may be a potential source of pathogenic R. equi strains for humans. © 2013 de Vargas et al.; licensee BioMed Central Ltd.

O objetivo desse estudo foi avaliar diferentes métodos de secagem de folhas para três diferentes frutíferas (maracujá, pêssego e abacate), com relação à determinação da matéria seca e os teores foliares de macronutrientes. Foram coletadas amostras de folhas recém expandida de três culturas, do pomar da fazenda de ensino e pesquisa da FCAV-UNESP, câmpus de Jaboticabal, no mês de janeiro de 2010, coletando-se para cada cultura 12 amostras com 25 folhas cada. Os tratamentos constituíram-se por dois métodos de secagem, estufa de circulação de ar forçada regulada a uma temperatura de 70°C e o forno microondas (FMO). Avaliou-se a massa da matéria seca e os teores foliares de macronutrientes. Os resultados sugerem que os dois métodos de secagem testados se assemelham na determinação de matéria seca e nos teores foliares de macronutrientes para as culturas análisadas, exceto os teores de cálcio na cultura do pêssego.

Both adalimumab [ADA] and infliximab [IFX] seem to be effective in the prevention of early postoperative endoscopic recurrence [EPER] after ileocaecal resection in Crohn's disease [CD] patients. There is lack of data with direct comparison between the two agents in the postoperative scenario. The aim of this study was to compare the rates of EPER in patients treated with ADA and IFX after ileocaecal resection for CD. This was a multicentre retrospective analysis of EPER rates in CD patients after ileocaecal resections, from seven referral centres in three countries. Endoscopic recurrence was defined as Rutgeerts' score ≥ i2. The patients were allocated according to treatment to two groups: ADA or IFX. The EPER rates were compared between the two treatment groups. Among the 168 patients included in the database, 96 received anti-tumour necrosis factor [TNF] agents after resection [37 in the ADA and 59 in the IFX groups] and were included in this comparative study. The groups were comparable in all baseline characteristics, mainly age, gender, previous resections, perianal CD, and mono or combination therapy. EPER was identified in 9/37 [24.32%] in the ADA group vs 16/59 [27.12%] in the IFX group [p = 0.815]. In this retrospective direct comparison between ADA and IFX therapy after ileocaecal resection, there was no significant difference between the two anti-TNF agents in terms of EPER rates. However, prospective randomised studies are needed to confirm these data and better define the role of each agent in the prevention of EPER.

Tuberculosis (TB) is a major infectious disease caused by Mycobacterium tuberculosis (Mtb). According to the World Health Organization (WHO), about 1.8 million people die from TB and 10 million new cases are recorded each year. Recently, a new series of naphthylchalcones has been identified as inhibitors of Mtb protein tyrosine phosphatases (PTPs). In this work, 100 chalcones were designed, synthesized, and investigated for their inhibitory properties against MtbPtps. Structure-activity relationships (SAR) were developed, leading to the discovery of new potent inhibitors with IC50 values in the low-micromolar range. Kinetic studies revealed competitive inhibition and high selectivity toward the Mtb enzymes. Molecular modeling investigations were carried out with the aim of revealing the most relevant structural requirements underlying the binding affinity and selectivity of this series of inhibitors as potential anti-TB drugs.

[ES] El Banco Español de Algas (BEA) es un servicio nacional de I+D+i adscrito a la Fundación Parque Científico Tecnológico de la Universidad de Las Palmas de Gran Canaria, que tiene como objetivos generales el aislamiento, identificación, caracterización, conservación y suministro de microalgas y cianobacterias. La acreditación ante el Gobierno de España como autoridad internacional para el depósito de microorganismos confiere al BEA la función de depositario de algas con fines de reconocimiento de la propiedad intelectual ante la Organización Mundial de la Propiedad Intelectual y la Oficina Española de Patentes y Marcas. El BEA es miembro de la Organización Europea de Colecciones de Cultivo (ECCO), de la Federación Mundial de Colecciones de Cultivo (WFCC) y está incluido en el Centro Mundial de Datos sobre Microorganismos (WFCC-MIRCEN). Además de estas funciones, el BEA es un servicio que facilita el desarrollo de un nuevo sector bioindustrial basado en el cultivo y las aplicaciones de las microalgas. En la actualidad mantiene una colección de, aproximadamente, 1200 cepas (21 Clases; 135 Géneros) agrupadas en una serie de catálogos constituidos en función del origen (tipo de ambiente, campaña de bioprospección, etc.), estado de axenitud y sus aplicaciones reales y/o potenciales. Sin embargo, y de acuerdo con datos científicos contrastados, sólo el 20% de las cepas han sido descritas como cultivables y/o fuente de algún tipo de metabolito de interés biotecnológico. Esto implica que el 80% restante, más de 900 cepas de la colección, son objeto potencial de estudio científico-tecnológico con posibilidades en los campos de (1) alimentación humana (nutracéutica) y animal; (2) sustancias bioactivas; (3) cosmética; y (4) biocombustibles.
