Foi avaliada a atividade inibidora do diflubenzuron na ecdise de larvas do Aedes aegypti, visando à utilização desse produto no controle desse mosquito. Além disso, conhecer a interação do produto com o tipo de criadouro e a suscetibilidade do mosquito. Os bioensaios foram realizados em um fundo de quintal de residência, em sete tipos potenciais de criadouros artificiais: pneu, vidro, cimento-amianto, cimento, lata, plástico e cerâmica. Para cada tipo de criadouro colocaram-se 20 larvas de cada estádio do Aedes aegypti. O mesmo número de larvas foi utilizado para o controle. Foram feitas nove réplicas e as leituras de mortalidade foram em intervalos de 24 horas, após o início dos experimentos, até atingir o índice de 100%. Isto foi obtido a 1 ppm. Não houve diferença significativa entre os períodos médios de sobrevivência das larvas e nem entre os diferentes tipos de criadouros. Houve diferenças significativas entre os estádios, sendo o 3° o mais tolerante. Constatou-se também que as concentrações não interagiram com os estádios e tipos de criadouros, ao nível de 5%.

Aedes aegypti (L) (Diptera: Culicidae) was reared in several concentrations of diflubenzuron and methoprene under laboratory conditions in Uberlândia, State of Minas Gerais, southeastern Brazil. Characteristics such as LC50 and LC95, the susceptibility of immature stages of different ages to these insect growth regulators and their residual effects were studied. The LC50 and LC95 of diflubenzuron and methoprene were 5.19 and 12.24 ppb; 19.95 and 72.08 ppb, respectively. While diflubenzuron caused great mortality in all larval instars, methoprene was more effective when the mosquito was exposed from the start of the fourth larval instar onwards. Commercial concentrations of these two insect growth regulators close to LC95 presented greater residual activity than did their respective technical formulations. The parameters were compared with those obtained elsewhere. The characteristics investigated here indicate that these insect growth regulators are effective alternatives for controlling the dengue vector in the Uberlândia region.

Horn fly immatures were raised in media containing different concentrations of diflubenzuron in order to verify their susceptibility to this insect growth regulator (IGR). The 50% and 95% lethal concentrations of diflubenzuron for the population (LC50, LC95) were determined as well as the effect of this IGR on the different immature horn fly stages. The tests were performed using the progeny of adults collected in the field. The immatures were maintained in a growth chamber at 25.0 ± 0.5ºC and 12-12 h photoperiod. IGR concentrations of 300 ppb, 100 ppb and 50 ppb were lethal for 100% of the sample. Pupae malformation occurred in the breeding media containing different diflubenzuron concentrations. Values for LC50 , LC95 (± 95% fiducial limits) and the slope of the regression line were respectively, 25.521 ± 1.981 ppb, 34.650 ± 2.001 ppb and 12.720 ± 1.096. The third larval instar was more sensitive to the sub-lethal concentration of the product than the first and second ones were. The results indicate that this IGR can be an important tool for controlling horn fly populations as well as for managing horn fly resistance to conventional insecticides against Haematobia irritans in Uberlândia, State of Minas Gerais.

This work aimed to adapt the analysis of methemoglobin recommended by Evelyn - Malloy (visible spectrophotometry), in order to facilitate its application in the field, or to analysis in clinical laboratory, of existing sites of diflubenzuron application. The parameters changed included: centrifuge rotation speed; time between the collection of biological sample and analysis, and storage temperature of the samples; and the volume of reagents. The comparison of the rotation speed (rpm) of the reference methodology with the rpm of a "clinical centrifuge" did not reveal a statistically significant difference in the levels of methemoglobin. The time between the collection of biological sample and analysis was extended for a period of up to 48 hours for both conservation by refrigeration and ambient temperature, producing no statistically significant difference when compared to the standard duration of 2 hours. Regarding the reagents, the reference methodology already uses the volume necessary to ensure complete reaction, whereas a wider range from the recommended volume to a 5-fold reduction in comparison to the reference methodology could be used. It was concluded that the proposed changes to the methodology for adapting the analysis are applicable to studies of field / workplace exposure and ensure the reliability of results. The adapted methodology was inter-laboratory validated and the parameters changed can be selected according to the requirements of the laboratory at which the methemoglobin is to be measured.

Studies assessing the characteristics of active ingredients (AIs) of toxic baits for leaf-cutting ants are still scarce, although the need for a delayed action of these compounds on adult workers has been well accepted (mortality <= 15% at 24 h and <= 90% at 21 days). Therefore, we determined the insecticidal action of two AIs used in commercial baits, diflubenzuron and dechlorane, over time in workers, and discussed the control of colonies in relation to the existing literature. Dechlorane presented excellent insecticidal activity with a delayed action at all concentrations tested, although its commercial use has been prohibited due to its organochlorine nature. In contrast, diflubenzuron did not cause significant mortality or symptoms of intoxication, indicating that the successful cases reported with the use of baits containing this AI were in fact due to an accidental contamination with dechlorane. We comment about the ineffectiveness of diflubenzuron on alternative targets, i.e., young forms and the mutualistic fungus, supporting the concept that the AI needs to a have a delayed action on adult workers.

Avaliou-se, neste experimento, a eficácia in vitro e in vivo do diflubenzuron a 25% para uso em bovinos, no controle da infestação por Haematobia irritans. Para o teste in vitro, ovos de moscas-dos-chifres foram mantidos em recipientes contendo fezes de animais não-tratados ou tratados com diflubenzuron a 25%, e acompanhados até emergência dos adultos. No teste in vivo, foram utilizadas 40 fêmeas aneloradas, divididas em dois grupos: controle (C) e tratado (T) com intensidade parasitária equivalente. Durante o experimento, o grupo C recebeu apenas suplementação mineral, enquanto o grupo T recebeu suplementação mineral e diflubenzuron a 25%. A contagem de moscas nos animais foi realizada na região dorsal, desde a nuca até as pontas da anca de cada animal, no início e ao final de um período de cinco meses. Na avaliação in vitro, o grupo controle apresentou média de emergência de 86% (± 8,4%), enquanto o grupo cultivado em fezes de bovinos tratados com diflubenzuron a 25% apresentou taxa de emergência média de 1% (± 0,2%), sendo a eficácia calculada de 98,83%. No teste in vivo, não foi observada redução significativa na contagem de moscas no grupo C, porém, no grupo T houve significativa redução da infestação por H. irritans (t = 16,46, p < 0,0001). A eficácia do produto, em condições de campo, foi de 99,20%. O diflubenzuron a 25% adicionado ao sal mineral mostrou-se eficaz contra H. irritans, sendo indicado para esse fim.

Podisus nigrispinus (Dallas) (Heteroptera: Pentatomidae) is a common natural predator of defoliating caterpillars in agricultural and forest systems. Insecticides acting as growth regulators of insect pests can indirectly affect their predators through consumption of contaminated prey. We examined the reproductive performance of P. nigrispinus fed on caterpillars of Anticarsia gemmatalis Hubner (Lepidoptera: Noctuidae) reared on soybean leaves exposed to the chitin synthesis inhibitor, diflubenzuron. Caterpillars of A. gemmatalis were fed for 12 h with treated soybean leaves and offered to adults of the predator P. nigrispinus over five consecutive days. The fertility of P. nigrispinus was reduced when feeding on diflubenzuron treated caterpillars, especially at the beginning of the reproductive period, but recovered 3 weeks later. The effects of diflubenzuron ingestion on the life table parameters of P. nigrispinus included an increase in the period taken to double the population size, and reductions in the intrinsic rate of population increase, generation duration, and net reproductive rate. Diflubenzuron therefore had an indirect negative effect on the reproduction and the population dynamics of the non-target predator P. nigrispinus. Clearly, its use in integrated pest management requires further evaluation.

Neste trabalho, avaliou-se a eficácia antiparasitária do praziquantel, levamisol e diflubenzuron administrados via oral, adicionados à ração, para pacus (Piaractus mesopotamicus) infectados por Anacanthorus penilabiatus e Dolops carvalhoi. Foram utilizadas 19 caixas d'água de 300 L de capacidade, comportando 28 peixes cada. Os tratamentos foram feitos misturando os princípios ativos nas dietas. A intensidade parasitária e eficácia foram avaliadas 1 dia antes e 3, 7 e 15 dias após o início da alimentação com ração contendo diflubenzuron, levamisol e praziquantel isolados ou associados em diferentes concentrações por 7 dias. Os resultados da eficácia terapêutica sugerem que, isoladamente ou associado com levamisol e praziquantel, o diflubenzuron é eficiente contra o crustáceo D. carvalhoi, demonstrando que a eficácia dos tratamentos nos dias 3, 7 e 15 variou de 96,2 a 100%. Contra os monogenóides, as drogas não apresentaram eficácia satisfatória. Os resultados sugerem o uso do diflubenzuron para o controle de D. cavalhoi em peixes de cativeiro e em condições de quarentenário.

Difubenzuron (DFB), an insecticide and acaricide insect growth regulator, can be used in agriculture against insect predators and in public health programs, to control insects and vectors, mainly Aedes aegypti larvae. Due to the lack of toxicological assessments of this compound, the objective of the present study was to evaluate the toxicological effects of subacute exposure to the DFB insecticide in adult male rats. Adult male rats were exposed (gavage) to 0, 2, 4, or 8 mg/kg of DFB for 28 days. No clinical signs of toxicity were observed in the DFB-treated animals of the experimental groups. However, there was an increase in serum levels of alanine aminotransferase in the group that received 8 mg/Kg/DFB/day and urea at doses of 4 and 8 mg/Kg/DFB/day, without altering other biochemical or hematological parameters. The subacute exposure to the lowest dose of DFB caused significant decrease in testis weight, daily sperm production, and in number of sperm in the epididymis in relation to the control group. However, no alterations were observed in the sperm morphology, testicular, epididymis, liver and kindney histology, or testosterone levels. These findings unveiled the hazardous effects of DFB on male reproduction after the subacute exposure and special attention should be addressed to the effects of low doses of this pesticide.

Resumo: Predição da concentração de baixo risco de diflubenzuron para organismos aquáticos e avaliação da argila e brita na redução da toxicidade. O diflubenzuron é um inseticida que além de ser usado agricultura, tem sido amplamente empregado na piscicultura, apesar do seu uso ser proibido nesta atividade. Este composto não consta na lista da legislação brasileira que estabelece limites máximos permissíveis em corpos de água para a proteção das comunidades aquáticas. No presente trabalho, a partir da toxicidade do diflubenzuron em organismos não-alvo, foi calculada a concentração de risco para somente 5% das espécies (HC5). O valor deste parâmetro foi estimado em aproximadamente 7 x 10-6 mg L-1 . Este baixo valor é devido à extremamente alta toxicidade do diflubenzuron para dafnídeos e à grande variação de sensibilidade entre as espécies testadas. Dois matérias de relativamente baixo custo se mostraram eficientes na remoção da toxicidade do diflubenzuron de soluções contendo este composto. Dentre esses materiais, a argila expandida promoveu a redução em aproximadamente 50% da toxicidade de uma solução contendo diflubenzuron. Os resultados podem contribuir para políticas públicas no Brasil relacionadas ao estabelecimento de limites máximos permissíveis de xenobióticos no compartimento aquático. Também, para a pesquisa de matérias inertes e de baixo custo com potencial de remoção de xenobióticos presentes em efluentes da aquicultura ou da agricultura. Abstract: Diflubenzuron is an insecticide that, besides being used in the agriculture, has been widely used in fish farming. However, its use is prohibited in this activity. Diflubenzuron is not in the list of Brazilian legislation establishing maximum permissible limits in water bodies for the protection of aquatic communities. In this paper, according toxicity data of diflubenzuron in non-target organisms, it was calculated an hazardous concentration for only 5% of the species (HC5) of the aquatic community. This parameter value was estimated to be about 7 x 10 -6 mg L -1 . The low value is due to the extreme high toxicity of diflubenzuron to daphnids and to the large variation in sensitivity among the species tested. Two relatively low cost and inert materials were efficient in removing the diflubenzuron from solutions containing this compound. Among these materials, expanded clay shown to promote reduction of approximately 50% of the toxicity of a solution containing diflubenzuron. The results may contribute to the establishment of public policies in Brazil associated to the definition of maximum permissible limits of xenobiotics in the aquatic compartment. This study is also relevant to the search of low cost and inert materials for xenobiotics removal from aquaculture or agricultural effluents.
