Lo studio è stato effettuato su esemplari di Palinurus elephas, decapodi appartenenti alla famiglia dei Palinuridae. Tali organismi oltre ad essere importanti dal punto di vista bio-ecologico, data l’ampia distribuzione geografica e il ruolo critico che rivestono in numerose reti trofiche marine, costituiscono una risorsa commerciale considerevole in tutto il Mediterraneo. Questo studio nasce dall’assunzione che il rumore di natura antropica potrebbe avere effetti negativi sugli stock di aragoste, andando a mascherare sia il suono del fondo marino, avente un ruolo di orientamento nelle fasi larvali di molti decapodi, sia i suoni d’importanza bio-ecologica prodotti da conspecifici e non. Negli ultimi anni si sono indagati i meccanismi d’emissione acustica di Palinurus elephas dimostrando che questi stimoli sonori, chiamati Rasp (range di frequenza = 2-75 kHz), avvengono in associazione a comportamenti anti-predatoriali, ossia in tutti quei comportamenti che implicano un movimento dell’apparato antennale. Tuttavia ben poco si conosce sull’ecologia di questa specie, in particolare per quanto concerne la comunicazione intraspecifica e l’abilità di questi animali nella percezione di segnali acustici (inclusi quelli emessi da conspecifici) a diverse frequenze. Nel corso di questo studio si sono volute indagare le risposte comportamentali di questo crostaceo a suoni di diversa natura: Sweep Basso = 0.1-20 kHz; Sweep Alto = 20-80 kHz; Rasp = 2-75 kHz, desumendo la sensibilità di questa specie a spettri sonori diversi in frequenza e ampiezza. Le alterazioni del comportamento sono state monitorate e analizzate attraverso filmati audio-visivi. Nella fase sperimentale, sono state valutate 6 diverse variabili comportamentali: Distanza Percorsa, Mobilità, Velocità, Movimento, Tail Flip ed Emissioni Acustiche, al fine di evidenziare eventuali differenze significative tra i gruppi esposti a uno dei tre segnali e il gruppo di controllo.

In vielen Arthropoden wird Sauerstoff mittels des Kupferproteins Hämocyanin transportiert. In der vorliegenden Arbeit wurde das Hämocyanin einiger Vertreter der Arthropoden molekularbiologisch untersucht. Bei den Crustaceen (Homarus americanus und Palinurus elephas) konnten vier Untereinheiten isoliert werden, bei denen es sich um Hämocyanine des a-Typs handelt. Bei den Myriapoden (Chilopoda, Scutigera coleoptrata und Diplopoda, Spirostreptus spec.) konnten drei unterschiedliche Hämocyaninuntereinheiten gefunden werden. Erst seit einiger Zeit ist das Hämocyanin bei Myriapoden biochemisch charakterisiert und bei diesen Hämocyaninsequenzen handelt es sich um die ersten von Myriapoden. Bei den Onychophoren (Epiperipatus spec.) konnte ebenfalls erstmals ein Hämocyanin isoliert und sequenziert werden. Hierbei handelt es sich um den ersten Hinweis, dass Onychophoren über ein respiratorisches Protein verfügen. In einer phylogenetischen Analyse der Hämocyaninsequenzen konnte ein Stammbaum der Hämocyaninsuperfamilie erstellt werden. Innerhalb der Crustacea ordnen sich die verschiedenen Hämocyaninuntereinheiten in distinkten Ästen an. Die Hämocyanine der Myriapoda sind monophyletisch, wobei die Auftrennung in distinkte Untereinheiten bereits vor der Trennung der Chilopoden und Diplopoden erfolgte; die phylogenetische Stellung der Myriapoda kann anhand der Hämocyaninsequenzen nicht zuverlässig aufgelöst werden. Eine gemeinsame Anordnung mit den Hexapoda ('Tracheata'-Hypothese) kann jedoch mit hoher Sicherheit ausgeschlossen werden. Die Onychophora stehen im Arthropodenstammbaum an basaler Position und können somit als Proarthropoda angesprochen werden. Es konnte gezeigt werden, dass die Hämocyaninevolution der Arthropodenevolution entspricht.

In der vorliegenden Arbeit wurden die bioinformatischen Methoden der Homologie-Modellierung und Molekularen Modellierung dazu benutzt, die dreidimensionalen Strukturen verschiedenster Proteine vorherzusagen und zu analysieren. Experimentelle Befunde aus Laborversuchen wurden dazu benutzt, die Genauigkeit der Homologie-Modelle zu erhöhen. Die Ergebnisse aus den Modellierungen wurden wiederum dazu benutzt, um neue experimentelle Versuche vorzuschlagen. Anhand der erstellten Modelle und bekannten Kristallstrukturen aus der Protein-Datenbank PDB wurde die Struktur-Funktionsbeziehung verschiedener Tyrosinasen untersucht. Dazu gehörten sowohl die Tyrosinase des Bakteriums Streptomyces als auch die Tyrosinase der Hausmaus. Aus den vergleichenden Strukturanalysen der Tyrosinasen resultierten Mechanismen für die Monophenolhydroxylase-Aktivität der Tyrosinasen sowie für den Import der Kupferionen ins aktive Zentrum. Es konnte der Beweis geführt werden, daß die Blockade des CuA-Zentrums tatsächlich der Grund für die unterschiedliche Aktivität von Tyrosinasen und Catecholoxidasen ist. Zum ersten Mal konnte mit der Maus-Tyrosinase ein vollständiges Strukturmodell einer Säugetier-Tyrosinase erstellt werden, das dazu in der Lage ist, die Mechanismen bekannter Albino-Mutationen auf molekularer Ebene zu erklären. Die auf der Basis des ermittelten 3D-Modells gewonnenen Erkenntnisse über die Wichtigkeit bestimmter Aminosäuren für die Funktion wurde durch gerichtete Mutagenese an der rekombinant hergestellten Maus-Tyrosinase getestet und bestätigt. Weiterhin wurde die Struktur der Tyrosinase des Krebses Palinurus elephas durch eine niedrigaufgelöste 3D-Rekonstruktion aus elektronenmikroskopischen Bildern aufgeklärt. Der zweite große Themenkomplex umfasst die Strukturanalyse der Lichtsammlerkomplexe LHCI-730 und LHCII. Im Falle des LHCII konnte der Oligomerisierungszustand der LHCMoleküle mit diskreten Konformationen des N-Terminus korreliert werden. Auch hier kam eine Kombination von Homologie-Modellierung und einer experimentellen Methode, der Elektronen-Spin-Resonanz-Messung, zum Einsatz. Die Änderung des Oligomerisierungszustands des LHCII kontrolliert den Energiezufluß zu den Photosystemen PS I und PS II. Des Weiteren wurde ein vollständiges Modell des LHCI-730 erstellt, um die Auswirkungen gerichteter Mutagenese auf das Dimerisierungsverhalten zu untersuchen. Auf Basis dieses Modells wurden die Wechselwirkungen zwischen den Monomeren Lhca1 und Lhca4 evaluiert und potentielle Bindungspartner identifiziert.

In dieser Dissertation wurden die Methoden Homologiemodellierung und Molekulardynamik genutzt, um die Struktur und das Verhalten von Proteinen in Lösung zu beschreiben. Mit Hilfe der Röntgenkleinwinkelstreuung wurden die mit den Computermethoden erzeugten Vorhersagen verifiziert. Für das alpha-Hämolysin, ein Toxin von Staphylococcus aureus, das eine heptamere Pore formen kann, wurde erstmalig die monomere Struktur des Protein in Lösung beschrieben. Homologiemodellierung auf Basis verwandter Proteine, deren monomere Struktur bekannt war, wurde verwendet, um die monomere Struktur des Toxins vorherzusagen. Flexibilität von Strukturelementen in einer Molekulardynamiksimulation konnte mit der Funktionalität des Proteines korreliert werden: Intrinsische Flexibilität versetzt das Protein in die Lage den Konformationswechsel zur Pore nach Assemblierung zu vollziehen. Röntgenkleinwinkelstreuung bewies die Unterschiede der monomeren Struktur zu den Strukturen der verwandten Proteine und belegt den eigenen Vorschlag zur Struktur. Überdies konnten Arbeiten an einer Mutante, die in einer sogenannten Präporenkonformation arretiert und nicht in der Lage ist eine Pore zu formen, zeigen, dass dieser Übergangszustand mit der Rotationsachse senkrecht zur Membran gelagert ist. Eine geometrische Analyse beweist, dass es sterisch möglich ist ausgehend von dieser Konformation die Konformation der Pore zu erreichen. Eine energetische und kinetische Analyse dieses Konformationswechsels steht noch aus. Ein weiterer Teil der Arbeit befasst sich mit den Konformationswechseln von Hämocyaninen. Diese wurden experimentell mittels Röntgenkleinwinkelstreuung verfolgt. Konformationswechsel im Zusammenhang mit der Oxygenierung konnten für die 24meren Hämocyanine von Eurypelma californicum und Pandinus imperator beschrieben werden. Für eine Reihe von Hämocyaninen ist nachgewiesen, dass sie unter Einfluss des Agenz SDS Tyrosinaseaktivität entfalten können. Der Konformationswechsel der Hämocyanine von E. californicum und P. imperator bei der Aktivierung zur Tyrosinase mittels SDS wurde experimentell bestätigt und die Stellung der Dodekamere der Hämocyanine als wesentlich bei der Aktivierung festgestellt. Im Zusammenhang mit anderen Arbeiten gilt damit die Relaxierung der Struktur unter SDS-Einfluss und der sterische Einfluss auf die verbindenden Untereinheiten b & c als wahrscheinliche Ursache für die Aktivierung zur Tyrosinase. Eigene Software zum sogenannten rigid body-Modellierung auf der Basis von Röntgenkleinwinkelstreudaten wurde erstellt, um die Streudaten des hexameren Hämocyanins von Palinurus elephas und Palinurus argus unter Einfluss der Effektoren Urat und Koffein strukturell zu interpretieren. Die Software ist die erste Implementierung eines Monte Carlo-Algorithmus zum rigid body-Modelling. Sie beherrscht zwei Varianten des Algorithmus: In Verbindung mit simulated annealing können wahrscheinliche Konformationen ausgefiltert werden und in einer anschließenden systematischen Analyse kann eine Konformation geometrisch beschrieben werden. Andererseits ist ein weiterer, reiner Monte Carlo-Algorithmus in der Lage die Konformation als Dichteverteilung zu beschreiben.

Background: Molecular tools may help to uncover closely related and still diverging species from a wide variety of taxa and provide insight into the mechanisms, pace and geography of marine speciation. There is a certain controversy on the phylogeography and speciation modes of species-groups with an Eastern Atlantic-Western Indian Ocean distribution, with previous studies suggesting that older events (Miocene) and/or more recent (Pleistocene) oceanographic processes could have influenced the phylogeny of marine taxa. The spiny lobster genus Palinurus allows for testing among speciation hypotheses, since it has a particular distribution with two groups of three species each in the Northeastern Atlantic (P. elephas, P. mauritanicus and P. charlestoni) and Southeastern Atlantic and Southwestern Indian Oceans (P. gilchristi, P. delagoae and P. barbarae). In the present study, we obtain a more complete understanding of the phylogenetic relationships among these species through a combined dataset with both nuclear and mitochondrial markers, by testing alternative hypotheses on both the mutation rate and tree topology under the recently developed approximate Bayesian computation (ABC) methods. Results: Our analyses support a North-to-South speciation pattern in Palinurus with all the South-African species forming a monophyletic clade nested within the Northern Hemisphere species. Coalescent-based ABC methods allowed us to reject the previously proposed hypothesis of a Middle Miocene speciation event related with the closure of the Tethyan Seaway. Instead, divergence times obtained for Palinurus species using the combined mtDNA-microsatellite dataset and standard mutation rates for mtDNA agree with known glaciation-related processes occurring during the last 2 my. Conclusion: The Palinurus speciation pattern is a typical example of a series of rapid speciation events occurring within a group, with very short branches separating different species. Our results support the hypothesis that recent climate change-related oceanographic processes have influenced the phylogeny of marine taxa, with most Palinurus species originating during the last two million years. The present study highlights the value of new coalescent-based statistical methods such as ABC for testing different speciation hypotheses using molecular data.

La presente Tesi di Laurea si colloca nell’ambito di un progetto strategico di ateneo chiamato OpIMA. Tale progetto mira a sviluppare codificatori innovativi per segnali di tipo impulsivo, discreti nei livelli, con riferimento soprattutto all’attuazione, alla sintesi di forme d’onda e all’amplificazione audio. L’obiettivo del progetto è di sviluppare nuove generazioni di codificatori, simili nell’uso a quelli tradizionali ed in qualche modo compatibili con essi e pure basati su principi operativi radicalmente diversi. La Tesi di Laurea è stata svolta presso il Dipartimento di Ingegneria Elettrica (DIE) nel laboratorio di Compatibilità Elettromagnetica (LACEM). Il lavoro tratta di uno studio nel dominio tempo-frequenza sulle emissioni condotte prodotte da un convertitore statico DC/AC progettato presso il DIE. Tale studio risulta utile per capire come e quando vengono generati questi disturbi e per migliorare la compatibilità elettromagnetica del convertitore. La Tesi inizialmente richiama con il Capitolo 1 i concetti che stanno alla base della compatibilità elettromagnetica, soprattutto per quanto riguarda le emissioni condotte. Con il Capitolo 2 si descrive in dettaglio il banco prova in ogni sua parte; nel Capitolo 3 vengono riportati i dati teorici di carica e scarica del BUS DC del convertitore. Nel Capitolo 4, 5, 6 vengono riportati i risultati delle prove effettuate sul convertitore per quanto riguarda le emissioni condotte; in particolare nel Capitolo 4 le prove sono state eseguite sul convertitore originale, nel Capitolo 5 si sono ripetute le prove separando elettricamente il circuito di potenza del convertitore da quello di controllo (ovviamente in questa fase abbiamo utilizzato due fonti di alimentazione separate) e nel Capitolo 6 si è voluto eliminare il rumore generato dall’alimentatore presente all’interno del circuito di controllo e per far ciò sono stati costruiti cinque alimentatori lineari esterni con determinate caratteristiche. Infine nell’ultimo Capitolo si sono tratte le conclusioni sui risultati delle misure effettuate.

Elephants use vocalizations for both long and short distance communication. Whereas the acoustic repertoire of the African elephant (Loxodonta africana) has been extensively studied in its savannah habitat, very little is known about the structure and social context of the vocalizations of the Asian elephant (Elephas maximus), which is mostly found in forests. In this study, the vocal repertoire of wild Asian elephants in southern India was examined. The calls could be classified into four mutually exclusive categories, namely, trumpets, chirps, roars, and rumbles, based on quantitative analyses of their spectral and temporal features. One of the call types, the rumble, exhibited high structural diversity, particularly in the direction and extent of frequency modulation of calls. Juveniles produced three of the four call types, including trumpets, roars, and rumbles, in the context of play and distress. Adults produced trumpets and roars in the context of disturbance, aggression, and play. Chirps were typically produced in situations of confusion and alarm. Rumbles were used for contact calling within and among herds, by matriarchs to assemble the herd, in close-range social interactions, and during disturbance and aggression. Spectral and temporal features of the four call types were similar between Asian and African elephants.

Asian elephants (Dephas maximus), prominent ``flagship species'', arelisted under the category of endangered species (EN - A2c, ver. 3.1, IUCN Red List 2009) and there is a need for their conservation This requires understanding demographic and reproductive dynamics of the species. Monitoring reproductive status of any species is traditionally being carried out through invasive blood sampling and this is restrictive for large animals such as wild or semi-captive elephants due to legal. ethical, and practical reasons Hence. there is a need for a non-invasive technique to assess reproductive cyclicity profiles of elephants. which will help in the species' conservation strategies In this study. we developed an indirect competitive enzyme linked immuno-sorbent assay (ELISA) to estimate the concentration of one of the progesterone-metabolites i.e, allopregnanolone (5 alpha-P-3OH) in fecal samples of As elephants We validated the assay which had a sensitivity of 0.25 mu M at 90% binding with an EC50 value of 1 37 mu M Using female elephants. kept under semi-captive conditions in the forest camps of Mudumalar Wildlife Sanctuary, Tamil Nadu and Bandipur National Park, Karnataka, India. we measured fecal progesterone-metabolite (5 alpha-P-3OH) concentrations in six an and showed their clear correlation with those of scrum progesterone measured by a standard radio-immuno assay. Statistical analyses using a Linear Mixed Effect model showed a positive correlation (P < 0 1) between the profiles of fecal 5 alpha-P-3OH (range 0 5-10 mu g/g) and serum progesterone (range: 0 1-1 8 ng/mL) Therefore, our studies show, for the first time, that the fecal progesterone-metabolite assay could be exploited to predict estrus cyclicity and to potentially assess the reproductive status of captive and free-ranging female Asian elephants, thereby helping to plan their breeding strategy (C) 2010 Elsevier Inc.All rights reserved.

Asian elephants in the wild live in complex social societies; in captivity, however, management often occurs in solitary conditions, especially at the temples and private places of India. To investigate the effect of social isolation, this study assessed the social group sizes and the presence of stereotypies among 140 captive Asian elephants managed in 3 captive systems (private, temple, and forest department) in Tamil Nadu, India, between 2003 and 2005. The majority of the facilities in the private (82%) and temple (95%) systems held a single elephant without opportunity for social interaction. The forest department managed the elephants in significantly larger groups than the private and temple systems. Among the 3 systems, the proportion of elephants with stereotypies was the highest in temple (49%) followed by private system (26%) and the forest department facility (6%); this correlates with the social isolation trend observed in the 3 systems and suggests a possible link between social isolation and abnormal elephant behavior separate from other environmental factors. The results of this study indicate it would be of greater benefit to elephant well being to keep the patchily distributed solitary temple and private elephants who are socially compatible and free from contagious diseases in small social groups at ocommon elephant houseso for socialization.

This study examines the population genetic structure of Asian elephants (Elephas maximus) across India, which harbours over half the world's population of this endangered species. Mitochondrial DNA control region sequences and allele frequencies at six nuclear DNA microsatellite markers obtained from the dung of free-ranging elephants reveal low mtDNA and typical microsatellite diversity. Both known divergent clades of mtDNA haplotypes in the Asian elephant are present in India, with southern and central India exhibiting exclusively the β clade of Fernando et al. (2000), northern India exhibiting exclusively the α clade and northeastern India exhibiting both, but predominantly the α clade. A nested clade analysis revealed isolation by distance as the principal mechanism responsible for the observed haplotype distributions within the α and β clades. Analyses of molecular variance and pairwise population FST tests based on both mitochondrial and microsatellite DNA suggest that northern-northeastern India, central India, Nilgiris (in southern India) and Anamalai-Periyar (in southern India) are four demographically autonomous population units and should be managed separately. In addition, evidence for female philopatry, male-mediated gene flow and two possible historical biogeographical barriers is described.

This study updates the status and conservation of the Endangered Asian elephant Elephas maximus in Cat Tien National Park, Vietnam. Line transect indirect surveys, block surveys for elephant signs, village surveys of elephant-human conflict incidents, guard-post surveys for records of sightings, and surveys of elephant food plants were undertaken during the dry and wet seasons of 2001. A minimum of 11 elephants and a maximum of 15-17 elephants was estimated for c. 500 km2 of the Park and its vicinity. The elephants are largely confined to the southern boundary of the Park and make extensive use of the adjoining La Nga State Forest Enterprises. During the dry season the elephants depend on at least 26 species of wild and cultivated plants, chiefly the fruits of cashew. Most of the villages surveyed reported some elephant-human conflict. Two adult male elephants seem to cover a large area to raid crops, whereas the family groups restrict themselves to a few villages; overall, the conflict is not serious. Since 2001 there have been no reports of any deaths or births of elephants in the Park. We make recommendations for habitat protection and management, increasing the viability of the small population, reducing elephant-human conflicts, and improving the chances of survival of the declining elephants of this Park. The Government has now approved an Action Plan for Urgent Conservation Areas in Vietnam that calls for the establishment of three elephant conservation areas in the country, including Cat Tien National Park.

Reproductive management of the Asian elephant (Elephas maximus) is important for its conservation. To monitor its estrous cyclicity, we earlier used an indirect ELISA to show that levels of fecal progesterone (P(4))-metabolite (allopregnanolone: 5 alpha-P-3OH) in semi-captive females sampled randomly positively correlated with serum P(4) levels [12]. In this longitudinal study (51 weeks), we measured levels of fecal 5 alpha-P-3OH and serum P(4) in seven semi-captive female elephants. Females exhibited three types of hormonal profiles. Four females showed cyclical patterns of fecal 5 alpha-P-3OH and serum P(4) typical of normal estrous cycles, two showed acyclic pattern while one showed high values indicative of a pregnant animal. Values for anestrous or follicular phases were <= 0.3 mu g g(-1), (5 alpha-P-3OH) and <= 0.3 ng mL(-1) (P(4)); for luteal phase 0.32-11.09 mu g g(-1) (5 alpha-P-3OH) and 0.32-1.48 ng mL(-1) (P(4)); for pregnancy 1.41-7.38 mu g g(-1) (5 alpha-P-3OH) and 0.39-1.6 ng mL(-1) (R(4)). A positive correlation (t = 8.8, p < 0.01, n = 321) between levels of fecal 5 alpha-P-3OH and serum P4 was observed. A random sample of 30 free-ranging female elephants showed fecal 5 alpha-P-3OH values of 0.06-23.4 mu g g(-1), indicating them to be in different stages of estrous cyclicity. This study is the first to assess the reproductive phases of female Asian elephants based on the correlative-patterns of both the fecal 5 alpha-P-3OH and serum P(4) values over multiple estrous cycles. Chemical signaling is a prominent mode of male-female communication among elephants, especially during their sexually active periods. Studies on the Asian elephant in zoos have shown the significance of a urinary pheromone (Z7-12:Ac) in conveying the reproductive status of a female toward the opposite sex. We investigated the additional possibility of an inter-sexual chemical signal being conveyed through dung. Sixteen semi-captive adult male elephants were presented with dung samples of three female elephants in different reproductive phases. Each male was tested in 3 separate trials, within an interval of 1-3 days. The trials followed a double-blind pattern as the male and female elephants used in the trials were strangers, and the observer was not aware of the reproductive status of females during the period of bioassays. Males responded preferentially (P < 0.005), in terms of higher frequency of sniff, check and place behavior toward the dung of females close to pre-ovulatory period (follicular-phase) as compared to those in post-ovulatory period (luteal-phase). The response toward the follicular phase samples declined over repeated trials though was still significantly higher than the corresponding response toward the non-ovulatory phase in each of the trials performed. This is the first study to show that male Asian elephants were able to distinguish the reproductive phase of the female by possibly detecting a pre-ovulatory pheromone released in dung.
