Fourteen commonly occurring species of marine benthic algae, i.e., Colpomenia sinuosa (Mertens ex Roth) Derbes et Solier, Dictyopteris australis (Sonder) Askenasy, Dictyota dichotoma (Hudson) Lamouroux D. dumosa Børgesen, D. hauckiana Nizamuddin, D. indica Sonder, D. maxima Zanardini, Hincksia mitchelliae (Harvey) Silva, Jolyna laminarioides Guimardes in Guimarâes et al., Padina tetrastromatica Hauck, Sargassum tenerrinum J. Agardh, Spatoglossum variabille Figari et De Notaris, Stoechospermum marginatum (C. Agardh) Kültzing and Stokeyia indica Thivy et Doshi, belonging to all three classes of Phaeophyta, were collected from coastal areas near Karachi, Pakistan and their taxonomy determined. Although all are taxonomically known species, Dictyota dichotoma, D. dumosa, D. hauckiana, D. indica, D. maxima, Jolyna laminarioides, Padina tetrastromatica, Sargassum tenerritnum and Stokeyia indica are described for the first time from the coast of Pakistan.

Cultivation of an endophytic fungus Aspergillus niger EN-13 that was isolated from the inner tissue of the marine brown alga Colpomenia sinuosa resulted in the characterization of a new naphthoquinoneimine derivative, namely, 5,7-dihydroxy-2-[1-(4methoxy-6-oxo-6H-pyran-2-yl)-2-phenylethylaniino]-[1,4]naphthoquinone. The structure of the new compound was established on the basis of various NMR spectroscopic analyses including 2D NMR techniques, EI-MS, and HR-ESI-MS. Ergosterimide (1), a natural Diels-Alder adduct of ergosteroid and maleimide, was characterized from the culture extract of Aspergillus niger EN-13, an endophytic fungus isolated from the marine brown alga Colpomenia sinuosa. In addition, four known steroids including (22E,24R)-ergosta-5,7,22-trien-3 beta-ol (2), (22E,24R)-ergosta-4,6,8(14),22-tetraen-3one (3), (22E,24R)-5 alpha,8 alpha-epidioxyergosta-6,22-dien-3 beta-ol (4), and (22E,24R)-ergosta-7,22dien-3 beta,5 alpha,6 beta-triol. (5) were also isolated and identified. The structures of these compounds were elucidated by extensive analysis of 1D and 2D NMR and IR spectra and MS data. The plausible biosynthetic pathway of 1 was also discussed. To the best of our knowledge, 1 is the first natural Diels-Alder adduct of steroid and maleimide reported so far. (C) 2007 Elsevier Inc. Asperamides A (1) and B (2), a sphingolipid and their corresponding glycosphingolipid possessing a hitherto unreported 9-methyl-C-20-sphingosine moiety, were characterized from the culture extract of Aspergillus niger EN-13, an endophytic fungus isolated from marine brown alga Colpomenia sinuosa. The structures were elucidated by spectroscopic and chemical methods as (2S,2'R,3R,3'E,4E,8E)-N-(2'-hydroxy-3'-hexadecenoyl)-9-methyl-4,8-icosadien-1,3-diol (1) and 1-O-beta-D-glucopyranosyl-(2S,2'R,3R,3'E,4E,8E)-N-(2'-hydroxy-3'-hexadecenoyl)-9-methyl-4,8-icosadien-1,3-diol (2). In the antifungal assay, asperamide A (1) displayed moderate activity against Candida albicans.

以褐藻裙带菜(Undaria pinnatifida)、萱藻(Scytosiphon lomentarius)、海蒿子(Sargassum confusum)、叉开网地藻(Dictyopteris divaricata)、海带(Laminaria japonica)、囊藻(Colpomenia sinuosa)、鼠尾藻(Sargassum thunbergii)、水云(Ectocarpus confervoides)为材料,对其色素-蛋白质复合物的分离技术及其特性进行了系统研究。通过对裙带菜色素-蛋白质复合物分离技术及其影响因素的研究,确立了褐藻的PAGE分离方法。采用Tris-Gly电泳分离系统,以非离子去污剂DMG或DIG为增溶剂(DMG: Chl=20: 1, DIG: Chl=50: 1, 4 ℃增溶1 h),10%的分离胶浓度,丙烯酰胺与甲叉双丙烯酰胺的比例为30: 0.8,从裙带菜中成功地分离出8条含色素的蛋白质复合物带。采用Anderson命名系统,以高等植物菠菜为参照,将其命名为CPIa, CPI, CPa, LHC_1, LHC_2, LHC_3, LHC_4和LHC_5。游离色素较少。通过对三种褐藻的色素-蛋白质复合物的表观分子量测定、光谱学研究以及对裙带菜色素-蛋白质复合物的多肽组成分析,揭示了褐藻各种色素-蛋白质复合物的特征。CPIa是褐藻分子量较大的PSI复合物,为墨角藻黄素-叶绿素 a/c-蛋白质复合物。CPI是褐藻的PSI核心复合物,为P700-叶绿素 a-蛋白质复合物。CPa是褐藻的PSII复合物,为墨角藻黄素-叶绿素 a/c-蛋白质复合物。其余5条为捕光色素-蛋白质复合物,LHC_1和LHC_3是墨角藻黄素-叶绿素 a/c-蛋白质复合物,LHC_2, LHC_4和LHC_5是叶绿素 a/c-蛋白质复合物。根据裙带菜色素-蛋白质复合物的多肽分析结果并与高等植物比较,提出褐藻PSI和PSII的结构模型。褐藻PSI和PSII的反应中心多肽与高等植物相同,捕光复合物明显区别。褐藻LHCI和LHCII的多肽组成相同,都是由单一的20 kDa多肽组成。褐藻叶状体、叶绿体、类囊体膜和PSI复合物的77 K 荧光发射光谱具有特异性,缺少高等植物PSI特征的730 nm 荧光峰。叶状体的77 K 荧光发射光谱按荧光主峰的波长分为两种类型,一种类型的荧光主峰在690 nm,另一种类型的在705-720 nm。三种褐藻PSI复合物的77 K 荧光发射光谱相同,有两个分别们于680 nm和715 nm 的发射峰。褐藻的77 K 荧光特异是由PSI的结构决定的。根据褐藻PSI复合物的荧光特性以及去污剂增溶动力学分析结果,推动F715来自褐藻核心色素-蛋白质复合物,F680来源于PSI复合物中的捕光复合物。褐藻PSI复合物中缺少高等植物发射730 nm 荧光的LHCIb复合物的能量传递模型。

海洋微生物拥有丰富多样的次生代谢途径,其中海洋生物内生真菌次生代谢产物研究日益受到天然产物化学界的重视。本论文以菌丝体生物量、发酵产物重量、抗菌与细胞毒活性、薄层色谱分析结果以及高效液相色谱分析结果等为评价依据对采自青岛沿海的13株海藻内生真菌在四种液体培养基上的静置发酵产物进行了综合评价,并从中选择了黑曲霉Aspergillus niger EN-13(分离自褐藻囊藻Colpomenia sinuosa)和杂色曲霉A. versicolor EN-7(分离自褐藻鼠尾藻Sargassum thunbergii)两株真菌进行了30升规模发酵(分别采用GPYM培养基和PDB培养)和化学成分的研究,对分离得到的大部分化合物进行了初步的生物活性筛选。 发酵提取物采用常规的硅胶柱层析、反相硅胶柱层析,凝胶Sephadex LH-20柱层析、制备薄层层析、半制备高效液相色谱以及重结晶等分离手段,得到单体化合物。利用各种现代波谱技术(IR、UV、EI-MS、FAB-MS、HR-ESI-MS、1H-NMR、13C-NMR、DEPT、1H-1H COSY、HSQC、HMBC等)并结合化学方法从两种菌株发酵提取物中鉴定了55个化合物的结构。其中从菌株A. niger EN-13分离鉴定了31个化合物,发现9个新化合物,包括2个鞘酯类化合物(AN-1~2)、3个萘并-γ-吡喃酮类化合物(AN-3~5)、3个苯乙基取代的α-吡喃酮类化合物(AN-17, AN-19~20)和1个甾体Diels-Alder加成产物(AN-21),另有1个新的天然环二肽(AN-27)被分离鉴定;从菌株A. versicolor EN-7分离鉴定了24个化合物,发现2个新化合物,为蒽醌AV-12与AV-17,另外,从前一菌株(A. niger EN-13)中鉴定的2个新鞘酯类化合物(AN-1~2)在A. versicolor EN-7中也被再次分离到。 对大部分单体化合物进行了抗菌活性、DPPH自由基清除活性和细胞毒活性测试。结果显示新化合物AN-1、AN-5和AN-20具有弱或中等强度的抑制白色念珠菌生长的活性,AN-4、AN-5、AN-21显示了弱或中等强度的抑制黑曲霉生长的活性,AV-12、AV-17显示了弱的抑制大肠杆菌生长的活性。在DPPH自由基清除活性筛选中,AN-5显示了中等强度的活性,其EC50为109.3 mM,与阳性对照BHT相近(EC50为81.8 mM)。其它部分已知化合物在抗菌和DPPH自由基清除活性的筛选中也显示了弱或中等强度的活性。在针对人肝癌细胞株SMMC-7721和人肺腺癌细胞株A549的体外细胞毒活性筛选中,所测样品均未显示显著活性。

After detachment from benthic habitats, the epibiont assemblages on floating seaweeds undergo substantial changes, but little is known regarding whether succession varies among different seaweed species. Given that floating algae may represent a limiting habitat in many regions, rafting organisms may be unselective and colonize any available seaweed patch at the sea surface. This process may homogenize rafting assemblages on different seaweed species, which our study examined by comparing the assemblages on benthic and floating individuals of the fucoid seaweeds Fucus vesiculosus and Sargassum muticum in the northern Wadden Sea (North Sea). Species richness was about twice as high on S. muticum as on F. vesiculosus, both on benthic and floating individuals. In both seaweed species benthic samples were more diverse than floating samples. However, the species composition differed significantly only between benthic thalli, but not between floating thalli of the two seaweed species. Separate analyses of sessile and mobile epibionts showed that the homogenization of rafting assemblages was mainly caused by mobile species. Among these, grazing isopods from the genus Idotea reached extraordinarily high densities on the floating samples from the northern Wadden Sea, suggesting that the availability of seaweed rafts was indeed limiting. Enhanced break-up of algal rafts associated with intense feeding by abundant herbivores might force rafters to recolonize benthic habitats. These colonization processes may enhance successful dispersal of rafting organisms and thereby contribute to population connectivity between sink populations in the Wadden Sea and source populations from up-current regions.

The first part presents a general picture of the composition of the coastal plankton near Mar del Plata (Buenos Aires prov., Argentina) between August 1961-August 1962. Especially noted is the abundance of Noctiluca from October to the end of March. The second part deals with the plankton collected during a short cruise, named Operación Mar del Plata 1, planned to survey coastal waters. In the third part the author gives the descriptions of some new or interesting species. A chain-forming Gymnodinium is reported as G. catenatum; some differences with Graham description, could be due to the fact that he studied only preserved material. Oblea baculifera is a new species belonging to the new genus Oblea, of the Diplopsalis group, created for some species previ¬ously known as Peridiniopsis. A key of the included species is given. The other new species are Peridinium aspidiotum, P. anomaloplaxum, P. lipopodium and Gonyaulax fratercula, all of them carefully described and depicted. Sticholonche zanclea (Radiolaria), Calsiosolenia sinuosa (Coccolithophoridae) and Tintinnopsis levigata (Tintinnida) are reported for the first time in the South-Atlantic. For T. levigata ?, Balech, 1939, a new name has been created: T. tregouboffi. Some unknown details of the protoplasm of the silicoflagellate Dictyocha octonaria are described. In the fourth part the author compares the plankton taken in Mar del Plata with the previously studied at Pto. Quequén. The plankton of March-April points to an invasion of oceanic warm waters from the Brazilian Current System.

Understanding how invasive species spread is of particular concern in the current era of globalisation and rapid environmental change. The occurrence of super-diffusive movements within the context of Lévy flights has been discussed with respect to particle physics, human movements, microzooplankton, disease spread in global epidemiology and animal foraging behaviour. Super-diffusive movements provide a theoretical explanation for the rapid spread of organisms and disease, but their applicability to empirical data on the historic spread of organisms has rarely been tested. This study focuses on the role of long-distance dispersal in the invasion dynamics of aquatic invasive species across three contrasting areas and spatial scales: open ocean (north-east Atlantic), enclosed sea (Mediterranean) and an island environment (Ireland). Study species included five freshwater plant species, Azolla filiculoides, Elodea canadensis, Lagarosiphon major, Elodea nuttallii and Lemna minuta; and ten species of marine algae, Asparagopsis armata, Antithamnionella elegans, Antithamnionella ternifolia, Codium fragile, Colpomenia peregrina, Caulerpa taxifolia, Dasysiphonia sp., Sargassum muticum, Undaria pinnatifida and Womersleyella setacea. A simulation model is constructed to show the validity of using historical data to reconstruct dispersal kernels. Lévy movement patterns similar to those previously observed in humans and wild animals are evident in the re-constructed dispersal pattern of invasive aquatic species. Such patterns may be widespread among invasive species and could be exacerbated by further development of trade networks, human travel and environmental change. These findings have implications for our ability to predict and manage future invasions, and improve our understanding of the potential for spread of organisms including infectious diseases, plant pests and genetically modified organisms.
