Stability of minimally processed radicchio (Cichorium intybus L.) was evaluated under modified atmosphere (2% O2, 5% CO2, and 93% N2) on 3, 5, 7 and 10 days of storage at 5°C. The samples were hygienized in sodium hypochlorite or hydrogen peroxide solutions to identify the most effective sanitizing solution to remove microorganisms. Microbiological analysis was conducted to identify the presence of coliforms at 35°C and 45°C, mesophilic microorganisms, and yeast and mold. Physicochemical analyses of mass loss, pH, soluble solids, and total acidity were conducted. The color measurements were performed using a Portable Colorimeter model CR-400. The antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl and 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic methods. The sensory evaluation was carried out using a hedonic scale to test overall acceptance of the samples during storage. The sodium hypochlorite (150 mg.L-1) solution provided greater safety to the final product. The values of pH ranged from 6.17 to 6.25, total acidity from 0.405 to 0.435%, soluble solids from 0.5 to 0.6 °Brix, mass loss from 1.7 to 7.2%, and chlorophyll from 1.068 to 0.854 mg/100g. The antioxidant activity of radicchio did not show significant changes during the first 3 days of storage. The overall acceptance of the sample stored in the sealed package without modified atmosphere was 70%, while the fresh sample was obtained 77% of approval. Although the samples packaged under modified atmosphere had a higher acceptance score, the samples in sealed packages had satisfactory results during the nine days of storage. The use of modified atmosphere, combined with cooling and good manufacturing practices, was sufficient to prolong the life of minimally processed radicchio, Folha Larga cultivar, for up to ten days of storage.

O conhecimento do coeficiente de cultura (Kc) permite a estimativa das necessidades hídricas de uma cultura, sendo fundamental para o manejo da irrigação. Nesta pesquisa, conduzida na área experimental do Departamento de Recursos Naturais da Faculdade de Ciências Agronômicas – FCA/UNESP, Campus de Botucatu, utilizou-se, para medida da evapotranspiração da cultura (ETc), 4 evapotranspirômetros de nível freático constante. Desses, 2 foram cobertos com plstico transparente rente ao solo, a fim de se evitar a evaporação. A evapotranspiração de referência (ETo) foi estimada pelo método de Penman-Monteith, sendo o coeficiente de cultura da chicória (Cichorium endivia L.) dado pela relação: , em suas diversas fases de desenvolvimento. Os valores de (Kc) obtidos evidenciaram um comportamento crescente durante todo o período vegetativo, com a influência da cobertura plstica na fase inicial quando a evaporação do solo representa uma fração considerável da evapotranspiração da cultura.

Variations in the inulin contents have been detected in rhizophores of Vernonia herbacea during the phenological cycle. These variations indicate the occurrence of active inulin synthesis and depolymerization throughout the cycle and a role for this carbohydrate as a reserve compound. 1-Fructan exohydrolase (1-FEH) is the enzyme responsible for inulin depolymerization, and its activity has been detected in rhizophores of sprouting plants. Defoliation and low temperature are enhancer conditions of this 1-FEH activity. The aim of the present work was the cloning of this enzyme. Rhizophores were collected from plants induced to sprout, followed by storage at 5C. A full length 1-FEH cDNA sequence was obtained by PCR and inverse PCR techniques, and expressed in Pichia pastoris. Cold storage enhances FEH gene expression. Vh1-FEH was shown to be a functional 1-FEH, hydrolyzing predominantly -2,1 linkages, sharing high identity with chicory FEH sequences, and its activity was inhibited by 81 in the presence of 10 mM sucrose. In V. herbacea, low temperature and sucrose play a role in the control of fructan degradation. This is the first study concerning the cloning and functional analysis of a 1-FEH cDNA of a native species from the Brazilian Cerrado. Results will contribute to understanding the role of fructans in the establishment of a very successful fructan flora of the Brazilian Cerrado, subjected to water limitation and low temperature during winter.

Chicory (Cichorium intybus L.) is an important broadleaves vegetable, consumed in salads and recognized by nutritional, pharmacological properties and its low calorie value. With the objective of assessing weeds interference periods over chicory crop in indirect sowing, two experiments were carried out at Garça County, São Paulo State, by using “Folhas Amarelas - Radiche” and “Folha Larga” cultivars and spacing of 0.25×0.25 m. Treatments consisted of checks with and without weeds and infestation control periods, so that crop was maintained in presence or absence of coexistence with weeds until 7, 14, 21, 28 and 35 days after seedlings transplant (DAST). Results demonstrated that “Folhas amarelas - Radiche” and “Folha Larga” chicory cultivars transplanted on winter, allowed occurrence of initial period of cohabitation with weeds (6 and 5 IPCW) greater than final period (14 and 9 FPCW), conferring, respectively, the establishment of critical periods for weed control (CPWC) in intervals of 6th to 14th and 5th to 9th days after crop transplant. Medium reduction of yield in function of weeds interference during the whole crop cycle was about 52.0% and 54.4%, respectively, for “Folhas amarelas - Radiche” and “Folha Larga” cultivars. It is important to mention that these results indicated the real need for conducting early weed control in chicory crop cultivation, even when carried out in indirect sowing system, as well as it characterizes the importance of a greater number of regional information to successfully consolidate management alternatives, less costly and more efficient in order to guarantee superior yields.

The leaves and especially the roots of chicory (Cichorium intybus L.) contain high concentrations of bitter sesquiterpene lactones such as the guianolides lactupicrin, lactucin, and 8-deoxylactucin. Eudesmanolides and germacranolides are present in smaller amounts. Their postulated biosynthesis through the mevalonate-farnesyl diphosphate-germacradiene pathway has now been confirmed by the isolation of a (+)-germacrene A synthase from chicory roots. This sesquiterpene cyclase was purified 200-fold using a combination of anion-exchange and dye-ligand chromatography. It has a Km value of 6.6 μm, an estimated molecular mass of 54 kD, and a (broad) pH optimum around 6.7. Germacrene A, the enzymatic product, proved to be much more stable than reported in literature. Its heat-induced Cope rearrangement into (−)-β-elemene was utilized to determine its absolute configuration on an enantioselective gas chromatography column. To our knowledge, until now in sesquiterpene biosynthesis, germacrene A has only been reported as an (postulated) enzyme-bound intermediate, which, instead of being released, is subjected to additional cyclization(s) by the same enzyme that generated it from farnesyl diphosphate. However, in chicory germacrene A is released from the sesquiterpene cyclase. Apparently, subsequent oxidations and/or glucosylation of the germacrane skeleton, together with a germacrene cyclase, determine whether guaiane- or eudesmane-type sesquiterpene lactones are produced.

Em 2001, plantas de chicória (Cichorium endivia) originárias do município de Catalo-GO apresentando sintomas de manchas e queima foliar foram recebidas na Clnica Fitopatolgica da Embrapa Hortaliças (CNPH). Tecidos submetidos à câmara úmida produziram esporulação fúngica, de onde foi obtida cultura monospórica em BDA + cloranfenicol. Para o teste de patogenecidade, o fungo foi multiplicado em meio de cultura suco de tomate ágar (ST) e inoculado em plantas de chicória da cultivar comercial AG-3804, em casa-de-vegetação. Após dez dias de incubação, verificou-se nas plantas inoculadas a presença de sintomas, semelhantes àqueles observados inicialmente. O patógeno foi reisolado a partir destas lesões, completando os postulados de Koch. Em seguida, visando testar a possibilidade deste patógeno infetar outras plantas da família Asteraceae, conídios produzidos em ST e atomizados em plantas de alface (Lactuca sativa) (três variedades), almeirão (Cichorium intybus) (duas variedades), Catalonha (Cichorium intybus) folha fina, serralha (Sonchus oleraceus) e uma outra variedade de chicória. Houve infecção em todas as plantas inoculadas, sendo que os sintomas variaram nas espécies e nas variedades testadas. Em chicória os sintomas surgiram mais cedo e se desenvolveram rapidamente. O fungo foi caracterizado morfolgica e morfometricamente permitindo identificá-lo como sendo Alternaria cichorii. Não se encontra registro desta espécie de Alternaria infetando plantas da família Asteraceae no Brasil. Neste trabalho relata-se pela primeira vez, a ocorrência de A. cichorii infetando chicória no Brasil e o potencial deste fungo como patógeno de outras espécies da família Asteraceae.

El objetivo de esta investigación fue determinar el efecto fitotóxico de un extracto obtenido con n-hexano de la corteza de Drimys winteri sobre la germinación y el crecimiento de Convolvulus arvensis, Setaria pumila, Daucus carota y Cichorium intybus. El efecto fitotóxico del extracto sobre la germinación de las malezas se determinó mediante bioensayos en placas de Petri y la aplicación del extracto en el medio de germinación en concentraciones de 100 a 1.000 mg L-1. En bioensayos en macetas, se determinó el efecto fitotóxico del extracto sobre la emergencia y crecimiento de las plntulas de malezas, en los cuales se aplicó el extracto mezclado con el sustrato de cultivo en concentraciones de 500 a 4.000 mg kg-1. También se determinó el efecto fitotóxico del extracto asperjado sobre plantas de malezas en concentraciones de 800 a 6.400 mg L-1. En los ensayos de germinación, el extracto retrasó la germinación y redujo el crecimiento de tallo y radícula de todas las malezas, el porcentaje de germinación disminuyó en todas las malezas, excepto en C. arvensis. La aplicación del extracto al sustrato de cultivo disminuyó y retrasó la emergencia de todas las malezas. Las plantas tratadas con extracto presentaron menor altura y hubo disminución de la biomasa acumulada. De igual modo, la aplicación del extracto al follaje de las malezas redujo la altura y la biomasa acumulada y llegó a causar la muerte de C. intybus, S. pumila y D. carota.

Endive (Cichorium intybus) is one of the vegetables least studied in Brazil with respect to the behavior of the most commercialized cultivars, but it is known that the number of plants per area is one of the factors that most influences the productivity of various crops. Thus, the objective of this work was to evaluate the behavior of endive using four spacings between rows and five spacings between plants, in the region of Jaboticabal-SP. The experiment was conducted in the field in the period of August 12, 2008 to November 6, 2008, in the experimental area of the Sector of Vegetable Crops and Aromatic-Medicinal Plants, Department of Crop Sciences, School of Agricultural and Veterinary Sciences - UNESP-FCAV. The experimental design utilized was a randomized block design with a 4 × 5 factorial scheme, i.e., four spacings between rows (0.15, 0.20, 0.25 and 0.30 m) and five spacings between plants (0.10, 0.15, 0.20, 0.25 and 0.30 m), with three repetitions. The endive cultivar studied was 'Pão-de-Acucar', obtained from the company Tecnoseed. Seedlings were established utilizing the sowing system in 200-cell Styrofoam trays containing the substrate Plantmax Vegetables® HA. The seedlings were transplanted at 37 days after planting. After harvesting, the following characteristics were determined: height (cm), diameter of the plant in rows (cm), diameter of the plant in between rows (cm), number of leaves, fresh weight (g) and total estimated productivity (t/ha). There were no interactions between the factors studied. Based on the data, we conclude that spacings of 0.15 m between rows and of 0.15 m between plants produce plants of higher quality and with greater productivity.

Rhizophores of Vernonia herbacea, an Asteraceae found in the Brazilian Cerrado, store high amounts of fructans that vary in composition over the phenological cycle. Fructan 1-exohydrolase (1-FEH) activity is detectable during the sprouting phase, mainly in the proximal regions of rhizophores, of plants induced to sprout by defoliation and/or cold storage. We found an increase in 1-FEH gene expression during natural and induced sprouting and further enhancement through low-temperature treatment. Furthermore, a comparative analysis of 1-FEH gene expression in different regions of the rhizophores during the transition from dormancy to sprouting is presented. Transcripts were detected mainly in the proximal region, coinciding with high 1-FEH activity and a high concentration of free fructose. Low temperature promoted the accumulation of fructans of a low degree of polymerization (DP) and enhanced 1-FEH activity and gene expression. It is hypothesized that a set of 1-FEH proteins acts in two different ways during fructan mobilization: (1) by hydrolyzing fructo-oligosaccharides and -polysaccharides in sprouting plants (naturally or induced) for carbon supply and (2) by hydrolyzing preferably fructo-polysaccharides under low temperature to maintain the oligosaccharide pool for plant cold acclimation. The author studied in this paper effect of chicory (Chicorium intybus L.), kikuyugrass (Pennisetum clandestinum Hoahst) and a variety of Bermuda grass (Cynodon dactilon Pers.) named "Grama seda" as green feeds in growth of White Leghorn chickens (0-8 weeks). Males and females were se-pareted by feathering and development of comb. The results obtained can be summarized as follows : a) - Without separation of sex, the chickens that received green feeds showed better development than that did not receive green suplements. b) - The chicory showed to be something better that kikuyugrass and Bermuda grass, which had a similar effect. c) - The green feeds used had little influence in development of males. d) - The females in the lots suplemented by green feeds showed to be superior to the females in the lots that did not receive it. e) - The author think that the presence of some priciples having action in the female hormony function is responsable for the better growth in the lot that received green feeds.

O uso de testes de vigor para avaliação da qualidade dos lotes de sementes é uma ferramenta imprescindível para produção e comercialização. No entanto, para sementes de alface e almeirão as pesquisas relacionadas à avaliação de sua qualidade ainda são escassas. Para sementes de alface, apenas o teste de germinação é considerado padronizado, enquanto os demais devem ser aprimorados visando fornecer informações mais precisas. Com intuito de adequar a metodologia do teste de envelhecimento acelerado para avaliar a qualidade de lotes de sementes de alface e almeirão, foram utilizados quatro lotes de sementes de alface das cultivares Maravilha e Verdinha e quatro lotes de sementes de almeirão das cultivares Catalonha e Pão-de-Açúcar. Na condução do teste de envelhecimento acelerado as sementes foram submetidas ao envelhecimento tradicional, e ao envelhecimento com solução saturada e diluída de NaCl; usaram-se três períodos distintos: 48, 72 e 96 horas. Para caracterizar a qualidade dos lotes foi determinado o grau de umidade das sementes, e realizados os testes de germinação, primeira contagem de germinação e emergência de plntulas em casa de vegetação. Concluiu-se que o teste de envelhecimento acelerado aos 41 °C possibilita a separação dos lotes em diferentes níveis de vigor, podendo ser utilizado em sementes de alface e almeirão, considerando-se o período de envelhecimento de 48 horas pelo método tradicional ou o de 72 horas pelo método com solução saturada de NaCl. Quanto ao método modificado pelo uso da solução diluída de NaCl, o período de 48 horas é adequado para as sementes de alface e o de 72 horas, para as de almeirão.

Background and Aims Ptilotus polystachyus (green mulla mulla; ptilotus) is a short-lived perennial herb that occurs widely in Australia in arid and semi-arid regions with nutrient poor soils. As this species shows potential for domestication, its response to addition of phosphorus (P) and nitrogen (N) was compared to a variety of the domesticated exotic perennial pasture herb Cichorium intybus (chicory), ‘Puna’. Methods Pots were filled with 3 kg of an extremely nutrient-deficient sterilized field soil that contained 3 mg kg−1 mineral N and 2 mg kg−1 bicarbonate-extractable P. The growth and P and N accumulation of ptilotus and chicory in response to seven rates of readily available phosphorus (0–300 mg P pot−1) and nitrogen (N) (0–270 mg N pot−1) was examined. Key Results Ptilotus grew extremely well under low P conditions: shoot dry weights were 23, 6 and 1·7 times greater than for chicory at the three lowest levels of P addition, 0, 15 and 30 mg P pot−1, respectively. Ptilotus could not downregulate P uptake. Concentrations of P in shoots approached 4 % of dry weight and cryo-scanning electron microscopy and X-ray microanalysis showed 35–196 mm of P in cell vacuoles in a range of tissues from young leaves. Ptilotus had a remarkable tolerance of high P concentrations in shoots. While chicory exhibited symptoms of P toxicity at the highest rate of P addition (300 mg P pot−1), no symptoms were present for ptilotus. The two species responded in a similar manner to addition of N. Conclusions In comparison to chicory, ptilotus demonstrated an impressive ability to grow well under conditions of low and high P availability. Further study of the mechanisms of P uptake and tolerance in ptilotus is warranted.

Because hydroponic production of vegetables is becoming more common, the carotenoid composition of hydroponic leafy vegetables commercialized in Campinas, Brazil, was determined. All samples were collected and analyzed in winter. Lactucaxanthin was quantified for the first time and was found to have concentrations similar to that of neoxanthin in the four types of lettuce analyzed. Lutein predominated in cress, chicory, and roquette (75.4 ± 10.2, 57.0 ± 10.3, and 52.2 ± 12.6 μg/g, respectively). In the lactucaxanthin-containing lettuces, β-carotene and lutein were the principal carotenoids (ranging from 9.9 ± 1.5 to 24.6 ± 3.1 μg/g and from 10.2 ± 1.0 to 22.9 ± 2.6 μg/g, respectively). Comparison of hydroponic and field-produced curly lettuce, taken from neighboring farms, showed that the hydroponic lettuce had significantly lower lutein, β-carotene, violaxanthin, and neoxanthin contents than the conventionally produced lettuce. Because the hydroponic farm had a polyethylene covering, less exposure to sunlight and lower temperatures may have decreased carotenogenesis.
