The present work emphasizes the use of chirality as an efficient tool to synthesize new types of second order nonlinear materials. Second harmonic generation efficiency (SHG) is used as a measure of second order nonlinear response. Nonlinear optical properties of polymers have been studied theoretically and experimentally. Polymers were designed theoretically by ab initio and semiempirical calculations. All the polymeric systems have been synthesized by condensation polymerization. Second harmonic generation efficiency of the synthesized systems has been measured experimentally by Kurtz and Perry powder method

Six polyesters were synthesised from 4,4 ' -oxy-bis(benzoyl chloride) and 1,4-, 1,5-, 1,6-, 2,3-, 2,6-, and 2,7-naphthalenediol isomers. The structures of the polyesters were characterised by means of IR, inherent viscosities in tetrachloroethane (TCE), solutions at 303 K and thermal analysis. The glass transition temperatures were in the range of 425-494 K by DSC thermal analysis. All of the polyesters were irradiated in an AECL Gammacell 220 unit at a dose rate of approximately 6.7 kGy/h to doses in the range of 0-15 kGy at 77 and 300 K. ESR spectroscopy was used to examine the radicals formed during radiolysis and to measure their yields. The G-values for radical formation in the polyesters were found to be in the range 0.18-1.41 at 77 K and 0.19-0.78 at 300 K. At 77 K, up to 15% of the radicals formed on radiolysis were found to be photo-bleachable anion radicals. Annealing experiments were carried out in order to identify the neutral radicals, which were assigned to naphthyl- or phenyl- and phenoxyl-type radicals. (C) 2001 Elsevier Science Ltd. Os autores apresentam, os resultados do ensaio terapêutico de um nôvo antihelmíntico, o triclorofenolpiperazina (Cl.416), sobre Ascaris lumbricoides, ancilostomídeos e Trichuris trichiura. Diversos esquemas terapêuticos foram ensaiados, visando avaliar a tolerância e eficácia da droga. Concluem os autores, em vista dos resultados apresentados, ser o C1-416 um antihelmíntico eficaz para o tratamento da ascaridiase e da ancilostomíase, especialmente quando utilizadas as doses de 2 a 3 g/dia pelo período de 4 dias consecutivos e 50 mg/kg e 80 mg/kg/dia durante 3 a 4 dias. A dose única de 3 a 5 g promoveu resultados significativos, especialmente se considerada a redução, da carga parasitária. A tolerância ao medicamento pode ser considerada boa, principalmente quando empregadas doses de 2 a 3 g ao dia e mesmo a dose única de 3-5 g; em nenhum caso se tornou necessária a interrupção do curso terapêutico.

O autor tratou 40 pacientes portadores de diversas helmintíases intestinais com um, nôvo sal de piperazina - Bis (2, 4, 5 - Triclorofenol) piperazina na dose única de 50 mg/kg de pêso corporal em jejum ou duas horas após a refeição matinal. Obteve com uma série de tratamento cura parasitológica em 25 pacientes (62,5%) infectados por Ancilostomídeos e mais 6 (15%) com um segundo curso da medicação, totalizando 31 pacientes curados parasitològicamente (77,5% dos casos). À tolerância ao produto foi boa, com exceção de dois casos (5%) que referiram vômitos logo após a ingestão da substância. Três enfermos relataram expulsão de exemplares adultos mortos de Ascaris lumbricoides pelas fezes. Com o esquema terapêutico empregado, o autor observou pequena atividade ãa droga com respeito à infecção por Ascaris lumbricoides e nula sôbre o Trichuris trichiura. Conclui que o nôvo antihelmíntico é bastante ativo contra a infestação determinada pelos Ancilostomídeos.

En este proyecto se propone: 1- Formular y analizar los problemas actuales en las técnicas de inyección de fallas para estimar SER (Single Event Response) en los circuitos integrados, aplicandolas luego para evaluar la tolerancia a fallos de diferentes circuitos integrados analógicos/digitales. El objetivo general que se persigue es proporcionar una solución que permita realizar, de forma rápida, eficaz y a bajo costo, la inyección de fallos en los circuitos analógicos y digitales. 2- Estudiar una aproximación no intrusita de detección de fallos en CI, combinando técnicas de hardware y software para detectar errores transitorios en circuitos analógicos y digitales. Este tipo de fallos transitorios tienen una influencia importante en sistemas de microprocesadores, que afectan al flujo de datos y a la etapa de control. Con el fin de proteger el sistema, un módulo de hardware orientado a la aplicación se generará automáticamente, reconfigurándose en el sistema durante el tiempo de ejecución. Cuando se combina esto con técnicas de tolerancia a fallas basadas en programación (Software), esta solución ofrece una protección total del sistema contra fallos transitorios. La campaña de inyección de fallas se planea realizar en un microprocesador MIPS, ejecutando algún programa de evaluación, con ayuda de una plataforma genérica y versátil desarrollada en TIMA (Francia). 3- Comparar los resultados obtenidos del estudio de las técnicas de inyección con los resultados experimentales, a partir de ensayos de radiación (aceleradores de partículas, micro rayos, etc.) al exponer a los circuitos a posibles fuentes de fallas.

Methicillin resistant Staphylococcus aureus (MRSA) bacteria have emerged in the early 1980's in numerous health care institutions around the world. The main transmission mechanism within hospitals and healthcare facilities is through the hands of health care workers. Resistant to several antibiotics, the MRSA is one of the most feared pathogens in the hospital setting since it is very difficult to eradicate with the standard treatments. There are still a limited number of anti-MRSA antibiotics but the first cases of resistance to these compounds have already been reported and their frequency is likely to increase in the coming years. Every year, the MRSA infections result in major human and financial costs, due to the high associated mortality and expenses related to the required care. Measures towards a faster detection of resistant bacteria and establishment of appropriate antibiotic treatment parameters are fundamental. Also as part as infection prevention, diminution of bacteria present on the commonly touched surfaces could also limit the spread and selection of antibiotic resistant bacteria. During my thesis, projects were developed around MRSA and antibiotic resistance investigation using innovative technologies. The thesis was subdivided in three main parts with the use of atomic force microscopy AFM for antibiotic resistance detection in part 1, the importance of the bacterial inoculum size in the selection of antibiotic resistance in part 2 and the testing of antimicrobial surfaces creating by sputtering copper onto polyester in part 3. In part 1 the AFM was used two different ways, first for the measurement of stiffness (elasticity) of bacteria and second as a nanosensor for antibiotic susceptibility testing. The stiffness of MRSA with different susceptibility profiles to vancomycin was investigated using the stiffness tomography mode of the AFM and results have demonstrated and increased stiffness in the vancomycin resistant strains that also paralleled with increased thickness of the bacterial cell wall. Parts of the AFM were also used to build a new antibiotic susceptibility-testing device. This nano sensor was able to measure vibrations emitted from living bacteria that ceased definitively upon antibiotic exposure to which they were susceptible but restarted after antibiotic removal to which they were resistant, allowing in a matter of minute the assessment of antibiotic susceptibility determination. In part 2 the inoculum effect (IE) of vancomycin, daptomycin and linezolid and its importance in antibiotic resistance selection was investigated with MRSA during a 15 days of cycling experiment. Results indicated that a high bacterial inoculum and a prolonged antibiotic exposure were two key factors in the in vitro antibiotic resistance selection in MRSA and should be taken into consideration when choosing the drug treatment. Finally in part 3 bactericidal textile surfaces were investigated against MRSA. Polyesters coated after 160 seconds of copper sputtering have demonstrated a high bactericidal activity reducing the bacterial load of at least 3 logio after one hour of contact. -- Au cours des dernières décennies, des bactéries multirésistantes aux antibiotiques (BMR) ont émergé dans les hôpitaux du monde entier. Depuis lors, le nombre de BMR et la prévalence des infections liées aux soins (IAS) continuent de croître et sont associés à une augmentation des taux de morbidité et de mortalité ainsi qu'à des coûts élevés. De plus, le nombre de résistance à différentes classes d'antibiotiques a également augmenté parmi les BMR, limitant ainsi les options thérapeutiques disponibles lorsqu'elles ont liées a des infections. Des mesures visant une détection plus rapide des bactéries résistantes ainsi que l'établissement des paramètres de traitement antibiotiques adéquats sont primordiales lors d'infections déjà présentes. Dans une optique de prévention, la diminution des bactéries présentes sur les surfaces communément touchées pourrait aussi freiner la dissémination et l'évolution des bactéries résistantes. Durant ma thèse, différents projets incluant des nouvelles technologies et évoluant autour de la résistance antibiotique ont été traités. Des nouvelles technologies telles que le microscope à force atomique (AFM) et la pulvérisation cathodique de cuivre (PCC) ont été utilisées, et le Staphylococcus aureus résistant à la méticilline (SARM) a été la principale BMR étudiée. Deux grandes lignes de recherche ont été développées; la première visant à détecter la résistance antibiotique plus rapidement avec l'AFM et la seconde visant à prévenir la dissémination des BMR avec des surfaces crées grâce à la PCC. L'AFM a tout d'abord été utilisé en tant que microscope à sonde locale afin d'investiguer la résistance à la vancomycine chez les SARMs. Les résultats ont démontré que la rigidité de la paroi augmentait avec la résistance à la vancomycine et que celle-ci corrélait aussi avec une augmentation de l'épaisseur des parois, vérifiée grâce à la microscopie électronique. Des parties d'un AFM ont été ensuite utilisées afin de créer un nouveau dispositif de test de sensibilité aux antibiotiques, un nanocapteur. Ce nanocapteur mesure des vibrations produites par les bactéries vivantes. Après l'ajout d'antibiotique, les vibrations cessent définitivement chez les bactéries sensibles à l'antibiotique. En revanche pour les bactéries résistantes, les vibrations reprennent après le retrait de l'antibiotique dans le milieu permettant ainsi, en l'espace de minutes de détecter la sensibilité de la bactérie à un antibiotique. La PCC a été utilisée afin de créer des surfaces bactéricides pour la prévention de la viabilité des BMR sur des surfaces inertes. Des polyesters finement recouverts de cuivre (Cu), connu pour ses propriétés bactéricides, ont été produits et testés contre des SARMs. Une méthode de détection de viabilité des bactéries sur ces surfaces a été mise au point, et les polyesters obtenus après 160 secondes de pulvérisation au Cu ont démontré une excellente activité bactéricide, diminuant la charge bactérienne d'au moins 3 logio après une heure de contact. En conclusion, l'utilisation de nouvelles technologies nous a permis d'évoluer vers de méthodes de détection de la résistance antibiotique plus rapides ainsi que vers le développement d'un nouveau type de surface bactéricide, dans le but d'améliorer le diagnostic et la gestion des BMR.

An overview is presented of the results obtained with biodegradable sustained release devices (SRDs) containing a mixture of polymers and either isometamidium (ISMM) or ethidium. Under controlled laboratory conditions (monthly challenge with tsetse flies infected with Trypanosoma congolense) the protection period in SRD treated cattle could be extended by a factor 2.8 (for ethidium) up to 4.2 (for ISMM) as compared to animals treated intramuscularly with the same drugs. Using a competitive drug ELISA ISMM concentrations were detected up to 330 days after the implantation of the SRDs, whereas after i.m. injection the drug was no longer present three to four months post treatment. Two field trials carried out in Mali under heavy tsetse challenge showed that the cumulative infection rate was significantly lower in the ISMM-SRD implanted cattle than in those which received ISMM intramuscularly. Using ethidium SRD, however, contradictory results were obtained in field trials in Zambia and in Mali. The potential advantages and inconvenients of the use of SRDs are discussed and suggestions are made in order to further improve the currently available devices.

La qualitat d’un producte elaborat és un factor important, tant pels consumidors, com pels òrgans reguladors que en defineixen normatives cada cop més estrictes. Iniciatives com la del PAT (Process Analytical Technology) en el sector farmacèutic, responen a aquestes necessitats. El PAT afavoreix la implantació de noves tècniques analítiques que facilitin el monitoratge i el control de paràmetres clau in-/on-line durant els processos de producció. En aquest sentit, el NIR-CI (Near Infrarred-Chemical Imaging) podria ser una eina molt útil en la millora de la qualitat de la indústria farmacèutica, ja que aprofita les avantatges del NIR com a tècnica analítica (ràpid, no invasiu, no destructiu) i les aplica a tota la superfície espacial de la mostra. És una tècnica capaç de proporcionar una gran quantitat d’informació, tant espectral com espacial, en una sola imatge. L’objectiu d’aquest treball és avaluar la capacitat de la tècnica NIR-CI, com a eina pel control de paràmetres de qualitat de comprimits farmacèutics. Concretament, s’han analitzat quantitativament la concentració i la distribució dels components (principi actiu i excipients) d’un comprimit farmacèutic amb i sense recobriment. A més, també s’ha determinat el gruix de la pel·lícula de laca de recobriment i la seva distribució a la superfície del comprimit. Per obtenir aquesta informació, es parteix d’imatges NIR-CI hiperespectrals dels comprimits. Per a l’extracció de les dades d’interès s’ha usat l’algoritme PLS en les diferents versions dels softwares Isys 5.0 i Unscrambler 9.8. La versió de l’Isys permet determinar la contribució de cada component pur a cada punt de la imatge, emprant únicament l’espectre del component en estudi. Amb la de l’Unscrambler, en canvi, es construeix un model de calibratge que, a partir d’unes mostres de referència, prediu la distribució del gruix de recobriment sobre la superfície del comprimit.
