Reduced glutathione (GSH) protects cells against injury by oxidative stress and maintains a range of vital functions. In vitro cell cultures have been used as experimental models to study the role of GSH in chemical toxicity in mammals; however, this approach has been rarely used with fish cells to date. The present study aimed to evaluate sensitivity and specificity of three fluorescent dyes for measuring pro-oxidant-induced changes of GSH contents in fish cell lines: monochlorobimane (mBCl), 5-chloromethylfluorescein diacetate (CMFDA) and 7-amino-4-chloromethylcoumarin (CMAC-blue). Two cell lines were studied, the EPC line established from a skin tumour of carp Cyprinus carpio, and BF-2 cells established from fins of bluegill sunfish Lepomis macrochirus. The cells were exposed for 6 and 24 h to low cytotoxic concentrations of pro-oxidants including hydrogen peroxide, paraquat (PQ), copper and the GSH synthesis inhibitor, L-buthionine-SR-sulfoximine (BSO). The results indicate moderate differences in the GSH response between EPC and BF-2 cells, but distinct differences in the magnitude of the GSH response for the four pro-oxidants. Further, the choice of GSH dye can critically affect the results, with CMFDA appearing to be less specific for GSH than mBCl and CMAC-blue.

OBJETIVO: Comparar os limites inferiores (L.inf.) e superiores (L.sup.) da prescrição de treinamento físico aeróbio determinada pelo teste ergométrico convencional (60-70% do VO2máx estimado ou 70-85% da FCmáx atingida), com a prescrição obtida pelo teste ergoespirométrico [limiar anaeróbio (LA) e ponto de compensação respiratória (PCR)]. MÉTODOS: Realizaram teste ergoespirométrico progressivo até a exaustão 47 homens (30±5 anos), divididos em subgrupos, de acordo com a velocidade da esteira durante o teste (4 ou 5mph) e a capacidade física medida [baixa (BCF) e moderada (MCF)]. RESULTADOS: Os L.inf. de prescrição indireta apresentaram valores de VO2 e FC significantemente maiores que os valores de VO2 e FC no LA (4mph= 34,4±4,5 vs 19,6±4,6 e 5mph= 28,9±2 vs 18,9±5,4, e BCF= 32,0±4,1 vs 17,2±2,8 e MCF= 31,6±4,9 vs 21,1±5,7(mlO2.kg-1.min-1) e (4mph = 128,9±7,8 vs 113,1± 15,6 e 5mph= 130,3±5,2 vs 114,1± 18,9, e BCF= 127,6±7,2 vs 109,3±13,2 e MCF= 131,2± 5,7 vs 117,4± 19,2bpm). Os L.sup. de prescrição indireta no grupo de 4mph e BCF apresentaram valores de VO2 significantemente maiores que os valores medidos no PCR (40,1±5,3 vs 32,2±4,3 e 37,4±4,8 vs 30,6±2,5 mlO2.kg-1.min-1, respectivamente), e valores de FC semelhantes aos medidos no PCR. CONCLUSÃO: Os L.inf. da prescrição indireta de treinamento físico superestimam o LA, enquanto os L.sup. parecem adequados somente para indivíduos ativos com MCF.

Hidden Markov models (HMMs) are probabilistic models that are well adapted to many tasks in bioinformatics, for example, for predicting the occurrence of specific motifs in biological sequences. MAMOT is a command-line program for Unix-like operating systems, including MacOS X, that we developed to allow scientists to apply HMMs more easily in their research. One can define the architecture and initial parameters of the model in a text file and then use MAMOT for parameter optimization on example data, decoding (like predicting motif occurrence in sequences) and the production of stochastic sequences generated according to the probabilistic model. Two examples for which models are provided are coiled-coil domains in protein sequences and protein binding sites in DNA. A wealth of useful features include the use of pseudocounts, state tying and fixing of selected parameters in learning, and the inclusion of prior probabilities in decoding. Al consultar los pliegos de Artemisia L. contenidos en el Herbario BCF, con motivo de la revisión de dicho género en la Península Ibérica, que estamos realizando como tesis doctoral, hemos hallado dos de ellos (BCF n.° 7473 y n.° 7475) recolectados por don Salvador Rivas Goday, el 17 de septiembre de 1948, en Titulcia (MADRID) y determinados como Artemisia variabilis Ten.

Breve aportación al catálogo provincial de RIGUAL (Flora y Vegetación de la provincia de Alicante, Inst. Est. Alicantinos, 1972), a la par que se recuerda la estancia de ROUY en este enclave (Excursions botaniques en Espagne. Bull. Soc. Bot. France 31:33-41, 52-56. 1884). En el trabajo de Rouy, que no menciona el señor Rigual, se recogen no pocas novedades, algunas de indudable interés para la flora alicantina. De las mismas, aquí nos limitaremos a transcribir las que hemos podido testificar en el campo. Señalamos con un asterisco (*) las novedades al mencionado catálogo. Salmonella enterica sérovar Typhi (S. Typhi) est l’agent responsable de la fièvre typhoïde et cause environ 200 000 morts et 27 millions de cas annuellement. C’est un pathogène entérique dont le réservoir est restreint à l’Homme. Les raisons de cette restriction d’hôte sont méconnues et pourraient dépendre de l’expression de facteurs d’adhésion à des étapes importantes au cours de la pathogenèse. L’annotation bioinformatique du génome de S. Typhi identifie 12 fimbriae de type chaperon-placier (FCP), un curli ainsi qu’un pilus de type IV. L’objectif de ce projet de recherche est d’étudier ces systèmes d’adhésion peu caractérisés. D’abord, le niveau d’expression de ces gènes a été évalué dans différentes conditions de culture in vitro en utilisant une approche de gènes rapporteurs. L’expression des 14 systèmes d’adhésion a été détectée. Nos résultats indiquent qu’une carence en fer favorise l’expression des opérons bcf et csg. Indépendamment du fer, l’expression de bcf, csg, pil, sef, sta, stc, stg et sth est influencée par la richesse nutritive du milieu. L’incubation en milieu LB liquide favorise l’expression de la plupart des systèmes d’adhésion par rapport à un milieu LB liquide sans agitation ou un milieu LB solide. En somme, l’expression des systèmes d’adhésion de S. Typhi a été observée et est influencée par des conditions environnementales. Dans un second volet, nous avons tent de surexprimer les différents systèmes d’adhésion chez une souche d’E. coli ou de S. Typhi afimbriaire. Avec cette approche, nous avons été en mesure de démontrer que l’opéron tcf encode pour un fimbria fonctionnel que l’on a pu observer en microscopie électronique. L’expression de tcf chez une souche afimbriaire d’E. coli et S. Typhi a également diminué leur capacité d’adhésion à des cellules épithéliales intestinales humaines lors d’essais in vitro. Nos observations démontrent que l’expression des systèmes d’adhésion retrouvés chez S. Typhi est influencée par les conditions enviroi9onnementales. Au moins un de ces systèmes est fonctionnel. Ceci suggère une contribution des systèmes d’adhésion retrouvés chez S. Typhi lors de l’interaction de ce pathogène avec l’humain.

Se describe y analiza cómo la importancia geoestratégica de Ucrania genera una percepción de seguridad y relevancia económica en Rusia, por esta razón la Federación utiliza el gas natural como mecanismo de presión frente al Estado ucraniano para satisfacer sus intereses nacionales en función de los elementos mencionados anteriormente.Todo lo anterior a través de la Teoría del Heartland desarrollada por Halford Mackinder y El Concepto de Área Pivote de Zbigniew Brzezinski para determinar cómo se genera la importancia del Estado ucraniano frente a Rusia

Gross cystic breast disease (GCBD) is the most common benign breast disorder, but the molecular basis of cyst formation remains to be identified. If the use of aluminium-based antiperspirant salts is involved in the etiology of gross breast cyst formation, it might be expected that aluminium would be at elevated levels in human breast cyst fluid (BCF). Aluminium was measured by ICP-MS in 48 samples of BCF, 30 samples of human blood serum and 45 samples of human breast milk at different stages of lactation (colostrum, intermediate, mature). The median level of aluminium in apocrine type I BCF (n:= 27, 150 mu g I-1) was significantly higher than in transuclative type II BCF (n = 21, 32 mu g I-1; P < 0.0001). By comparison, aluminium measurements gave a median concentration of 6 mu g I-1 in human serum and 25 mu g I-1 in human breast milk, with no difference between colostrum, intermediate and mature milk. Levels of aluminium were significantly higher in both types of BCF than in human serum (P < 0.0001). However when compared with human breast milk, aluminium levels were only significantly higher in apocrine type I BCF (P < 0.0001) and not in transudative type II BCF (P = 0.152). It remains to be identified why such high levels of aluminium were found in the apocrine type I BCF and from where the aluminium originated. However, if aluminium-based antiperspirants are found to be the source and to play any causal role in development of breast cysts, then it might become possible to prevent this common breast disorder. We have performed microarray hybridization studies on 40 clinical isolates from 12 common serovars within Salmonella enterica subspecies I to identify the conserved chromosomal gene pool. We were able to separate the core invariant portion of the genome by a novel mathematical approach using a decision tree based on genes ranked by increasing variance. All genes within the core component were confirmed using available sequence and microarray information for S. enterica subspecies I strains. The majority of genes within the core component had conserved homologues in Escherichia coli K-12 strain MG1655. However, many genes present in the conserved set which were absent or highly divergent in K-12 had close homologues in pathogenic bacteria such as Shigella flexneri and Pseudomonas aeruginosa. Genes within previously established virulence determinants such as SPI1 to SPI5 were conserved. In addition several genes within SPI6, all of SPI9, and three fimbrial operons (fim, bcf, and stb) were conserved within all S. enterica strains included in this study. Although many phage and insertion sequence elements were missing from the core component, approximately half the pseudogenes present in S. enterica serovar Typhi were conserved. Furthermore, approximately half the genes conserved in the core set encoded hypothetical proteins. Separation of the core and variant gene sets within S. enterica subspecies I has offered fundamental biological insight into the genetic basis of phenotypic similarity and diversity across S. enterica subspecies I and shown how the core genome of these pathogens differs from the closely related E. coli K-12 laboratory strain.

Combining the data from conventional semen analysis with oocyte penetration assays should improve the assessment of the fertilizing ability of a semen sample. Thus, the objective of the present study was to evaluate the prognostic value of various semen parameters on the in vitro interactions between frozen-thawed canine sperm and homologous oocytes. Ten ejaculates from five stud dogs (two ejaculates/dog) were collected by digital manipulation. Semen samples were evaluated, extended in Tris-egg yolk-glycerol, frozen and stored in liquid nitrogen, and thawed several weeks later. Samples were evaluated for motility and sperm populations by computer-aided semen analysis (CASA), plasma membrane integrity (carboxy-fluorescein diacetate and propidium iodide), and sperm morphology (Bengal Rose). Thawed spermatozoa were also incubated with homologous oocytes for 18 h in an atmosphere of 5% CO2 and 95% air at 38 degrees C and sperm-oocyte interactions were evaluated. Simple linear regression models were calculated, with sperm parameters as independent variables and sperm-oocyte interactions as the dependent variable. There were significant associations between: percentage of oocytes bound to spermatozoa and beat cross frequency (BCF; R-2 = 63%); percentage of oocytes that interacted with spermatozoa and BCF (R-2 = 73%); and number of penetrated spermatozoa and velocity average pathway (VAP; R-2 = 64%) and velocity straight line (VSL; R-2 = 64%). Although plasma membrane integrity and sperm morphology had little prognostic value for in vitro interactions between canine frozen-thawed sperm and homologous oocytes, some motility patterns (evaluated by CASA) were predictive of in vitro sperm-oocyte interactions.
