Detection of Arah1 (a major peanut allergen) in food using an electrochemical gold nanoparticle-coated screen-printed immunosensor

A gold nanoparticle-coated screen-printed carbon electrode was used as the transducer in the development of an electrochemical immunosensor for Ara h 1 (a major peanut allergen) detection in food samples. Gold nanoparticles (average diameter=32 nm) were electrochemically generated on the surface of screen-printed carbon electrodes. Two monoclonal antibodies were used in a sandwich-type immunoassay and the antibody–antigen interaction was electrochemically detected through stripping analysis of enzymatically (using alkaline phosphatase) deposited silver. The total time of the optimized immunoassay was 3 h 50 min. The developed immunosensor allowed the quantification of Ara h 1 between 12.6 and 2000 ng/ml, with a limit of detection of 3.8 ng/ml, and provided precise (RSD <8.7%) and accurate (recovery >96.6%) results. The immunosensor was successfully applied to the analysis of complex food matrices (cookies and chocolate), being able to detect Ara h 1 in samples containing 0.1% of peanut.

Bacillus cereus infection outbreak in captive psittacines

This study reports an uncommon epizootic outbreak of Bacillus cereus that caused the sudden death of 12 psittacines belonging to the species Anodorhynchus hyacinthinus (1 individual), Diopsittaca nobilis (1 individual), Ara severe (1 individual) and Ara ararauna (9 individuals) in a Brazilian zoo. Post-mortem examination of the animals reveled extensive areas of lung hemorrhage, hepatic congestion, hemorrhagic enteritis and cardiac congestion. Histopathological examination of the organs showed the presence of multiple foci of vegetative cells of Gram-positive bacilli associated with discrete and moderate mononuclear inflammatory cell infiltrate. Seventeen B. cereus strains isolated from blood and sterile organs of nine A. ararauna were analyzed in order to investigate the genetic diversity (assessed by Rep-PCR) and toxigenic profiles (presence of hblA, hblC and hblD; nheA, nheB and nheC as well as cytK, ces and entFM genes) of such strains. Amplification of genomic DNA by Rep-PCR of B. cereus strains generated two closely related profiles (Rep-PCR types A and B) with three bands of difference. All strains were classified as belonging to the toxigenic profile I which contained HBL and NHE gene complexes, entFM and cytK genes. Altogether, microbiological and histopathological findings and the evidence provided by the success of the antibiotic prophylaxis, corroborate that B. cereus was the causative agent of the infection that killed the birds. Preservação da qualidade pós-colheita de araçá-vermelho através do tratamento com 1-metilciclopropeno e do acondicionamento em embalagens plásticas, sob refrigeração

O objetivo deste trabalho foi estudar o padrão respiratório e de produção de etileno, e os efeitos do tratamento com diferentes doses de 1-metilciclopropeno (1-MCP) e do acondicionamento em diferentes embalagens plásticas, associado à refrigeração, na preservação da qualidade pós-colheita de araçá-vermelho. Os frutos apresentaram comportamento climatérico de respiração e produção de etileno. Frutos tratados com doses crescentes de 1-MCP (0; 100; 300; 600 e 1.200 nL L-1) e armazenados a 10±1C/90±5% UR, durante 10 dias, apresentaram retardo substancial no amadurecimento, através da preservação da textura e inibição na mudança de cor da epiderme. Frutos acondicionados com diferentes filmes (polietileno de baixa densidade, policloreto de vinila e à base de náilon) apresentaram, em termos gerais, melhor preservação da textura e menores mudanças na cor da epiderme e perdas de massa fresca, durante armazenamento refrigerado (5C e 10C). Os resultados demonstram que os frutos de araçá-vermelho apresentam elevada perecibilidade, caracterizada pelas elevadas taxas respiratórias e de produção de etileno, sendo mais bem preservados quando refrigerados, e tratados com 1-MCP ou acondicionados em embalagens plásticas.

Induction of topoisomerase I cleavage complexes by 1-β-d-arabinofuranosylcytosine (ara-C) in vitro and in ara-C-treated cells

1-β-d-Arabinofuranosylcytosine (Ara-C) is a nucleoside analog commonly used in the treatment of leukemias. Ara-C inhibits DNA polymerases and can be incorporated into DNA. Its mechanism of cytotoxicity is not fully understood. Using oligonucleotides and purified human topoisomerase I (top1), we found a 4- to 6-fold enhancement of top1 cleavage complexes when ara-C was incorporated at the +1 position (immediately 3′) relative to a unique top1 cleavage site. This enhancement was primarily due to a reversible inhibition of top1-mediated DNA religation. Because ara-C incorporation is known to alter base stacking and sugar puckering at the misincorporation site and at the neighboring base pairs, the observed inhibition of religation at the ara-C site suggests the importance of the alignment of the 5′-hydroxyl end for religation with the phosphate group of the top1 phosphotyrosine bond. This study also demonstrates that ara-C treatment and DNA incorporation trap top1 cleavage complexes in human leukemia cells. Finally, we report that camptothecin-resistant mouse P388/CPT45 cells with no detectable top1 are crossresistant to ara-C, which suggests that top1 poisoning is a potential mechanism for ara-C cytotoxicity.

Genetic evaluation of the mating system in the blue-and-yellow macaw (Ara ararauna, Aves, Psittacidae) by DNA fingerprinting

More than 90% of birds are socially monogamous, although genetic studies indicate that many are often not sexually monogamous. In the present study, DNA fingerprinting was used to estimate the genetic relationships between nestlings belonging to the same broods to evaluate the mating system in the socially monogamous macaw, Ara ararauna. We found that in 10 of 11 broods investigated, the nestlings showed genetic similarity levels congruent with values expected among full-sibs, suggesting that they shared the same parents. However, in one brood, the low genetic similarity observed between nestlings could be a result of intraspecific brood parasitism, intraspecific nest competition or extra-pair paternity. These results, along with available behavioral and life-history data, imply that the blue-and-yellow macaw is not only socially, but also genetically monogamous. However, the occurrence of eventual cases of extra-pair paternity cannot be excluded.

Detection of the peanut allergen Ara h 6 in foodstuffs using a voltammetric biosensing approach

A voltammetric biosensor for Ara h 6 (a peanut allergen) detection in food samples was developed. Gold nanoparticle-modified screen-printed carbon electrodes were used to develop a sandwich-type immunoassay using two-monoclonal antibodies. The antibody-antigen interaction was detected through the electrochemical detection of enzymatically deposited silver. The immunosensor presented a linear range between 1 and 100 ng/ml, as well as high precision (inter-day RSD ≤9.8 %) and accuracy (recoveries ≥96.7 %). The detection and quantification limits were 0.27 and 0.88 ng/ml, respectively. It was possible to detect small levels of Ara h 6 in complex food matrices.

CARACTERIZAÇÃO FÍSICA E QUÍMICA DOS FRUTOS DE ARAÇÁ-PERA (Psidium acutangulumD.C.)

Frutos de araçá-pera (Psidium acutanguium D.C.) em estádio de amadurecimento comercial foram avaliados quanto às características físicas, fisico-químicas e químicas. Houve grande variação no peso dos frutos (47,89 a 138,34 g) e no rendimento em polpa (55,01 a 75,98%). O araçá-pera é um fruto suculento (85,85% de umidade), com baixa relação Brix/ acidez (5,88), baixo pH (3,0) e acidez elevada (1,87% de ácido citrico). Destaca-se como excelente fonte de vitamina C total (389,34 mg l00g-1 de polpa integral). O baixo grau de doçura é fator limitante para o consumo "in natura". Apresenta potencial para industrialização, e para esta finalidade, seus atributos de qualidade são a uniformidade de formato,(levemente arredondado), alto rendimento em polpa, baixo pH e elevadas concentrações de acidez e vitamina C total.
