Biochemical and structural characterization of a beta-1,3-1,4-glucanase from Bacillus subtilis 168

beta-1,3-1,4-Glucanases (E.C. 3.2.1.73) hydrolyze linked beta-D-glucans, such as lichenan and barley beta-glucan. Recombinant beta-1,3-1,4-glucanase from Bacillus subtilis expressed in Escherichia coil and purified by Ni-NTA chromatography exhibited optimum activity at 50 degrees C and pH 6.0. The catalytic half-life at 60 degrees C decreased from 90 to 5 min when the enzyme was incubated in the presence and absence of Ca(2+) respectively. The kinetic parameters of lichenan hydrolysis were 2695, 3.1 and 1220 for V(max) (mu mol/min/mg), K(m) (mg mL(-1)) and K(cat) (s(-1)), respectively. Analysis by DLS, AUC and SAXS demonstrated the enzyme is monomeric in solution. Chemical denaturation monitored by ITFE and far-UV CD yielded Delta G(H2O) values of 9.6 and 9.1 kcal/mol, respectively, showing that the enzyme has intermediate stability when compared with other Bacillus beta-1,3-1,4-glucanases. The crystal structure shows the anti-parallel jelly-roll beta-sheet conserved in all GH16 beta-1,3-1,4-glucanases, with the amino acid differences between Bacillus sp. enzymes that are likely determinants of stability being distributed throughout the protein. Teichuronic acid operon of Bacillus subtilis 168.

Sequence analysis reveals that the Bacillus subtilis 168 tuaABCDEFGH operon encodes enzymes required for the polymerization of teichuronic acid as well as for the synthesis of one of its precursors, the UDP-glucuronate. Mutants deficient in any of the tua genes, grown in batch cultures under conditions of phosphate limitation, were characterized by reduced amounts of uronate in their cell walls. The teichuronic acid operon belongs to the Pho regulon, as phosphate limitation induces its transcription. Placing the tuaABCDEFGH operon under the control of the inducible Pspac promoter allowed its constitutive expression independently of the phosphate concentration in the medium; the level of uronic acid in cell walls was dependent on the concentration of the inducer. Apparently, owing to an interdependence between teichoic and teichuronic acid incorporation into the cell wall, in examined growth conditions, the balance between the two polymers is maintained in order to insure a constant level of the wall negative charge.

tagO is involved in the synthesis of all anionic cell-wall polymers in Bacillus subtilis 168.

Sequence homologies suggest that the Bacillus subtilis 168 tagO gene encodes UDP-N-acetylglucosamine:undecaprenyl-P N-acetylglucosaminyl 1-P transferase, the enzyme responsible for catalysing the first step in the synthesis of the teichoic acid linkage unit, i.e. the formation of undecaprenyl-PP-N-acetylglucosamine. Inhibition of tagO expression mediated by an IPTG-inducible P(spac) promoter led to the development of a coccoid cell morphology, a feature characteristic of mutants blocked in teichoic acid synthesis. Indeed, analyses of the cell-wall phosphate content, as well as the incorporation of radioactively labelled precursors, revealed that the synthesis of poly(glycerol phosphate) and poly(glucosyl N-acetylgalactosamine 1-phosphate), the two strain 168 teichoic acids known to share the same linkage unit, was affected. Surprisingly, under phosphate limitation, deficiency of TagO precludes the synthesis of teichuronic acid, which is normally induced under these conditions. The regulatory region of tagO, containing two partly overlapping sigma(A)-controlled promoters, is similar to that of sigA, the gene encoding the major sigma factor responsible for growth. Here, the authors discuss the possibility that TagO may represent a pivotal element in the multi-enzyme complexes responsible for the synthesis of anionic cell-wall polymers, and that it may play one of the key roles in balanced cell growth.

The European Board of Physical and Rehabilitation Medicine : the past, the present and the future [Europejska Rada Medycyny Fizykalnej i Rehabilitacji: przeszłość, teraźniejszość i perspektywy]

Right from the beginning of the development of the medical specialty of Physical and Rehabilitation Medicine (PRM) the harmonization of the fields of competence and the specialist training across Europe was always an important issue. The initially informal European collaboration was formalized in 1963 under the umbrella of the European Federation of PRM. The European Academy of PRM and the UEMS section of PRM started to contribute in 1969 and 1974 respectively. In 1991 the European Board of Physical and Rehabilitation Medicine (EBPRM) was founded with the specific task of harmonizing education and training in PRM in Europe. The EBPRM has progressively defined curricula for the teaching of medical students and for the postgraduate education and training of PRM specialists. It also created a harmonized European certification system for medical PRM specialists, PRM trainers and PRM training sites. European teaching initiatives for PRM trainees (European PRM Schools) were promoted and learning material for PRM trainees and PRM specialists (e-learning, books and e-books, etc.) was created. For the future the Board will have to ensure that a minimal specific undergraduate curriculum on PRM based on a detailed European catalogue of learning objectives will be taught in all medical schools in Europe as a basis for the general medical practice. To stimulate the harmonization of national curricula, the existing postgraduate curriculum will be expanded by a syllabus of competencies related to PRM and a catalogue of learning objectives to be reached by all European PRM trainees. The integration of the certifying examination of the PRM Board into the national assessment procedures for PRM specialists will also have to be promoted.

Cereais: Da análise realizada nos dados do 4º trimestre de 2012, o abastecimento no conjunto dos cereais, aponta um ligeiro declíneo no stock final e stock médio, e aumentos na importação, e a disponibilidade média. ■ Neste mesmo periodo a disponibilidade total do conjunto dos cereais é de 32.296 tons, sendo 16.241 tons referente ao stock inicial, e 16.055 tons referente as importações do mesmo conjunto. A disponibilidade média mensal no trimestre é de 20.803 tons e cobre largamente as necessidades previstas do consumo médio mensal aparente que é de 7.056 tons por mês. Relativamente ao trimestre anterior registou-se um decréscimo das disponibilidades médias mensais do conjunto dos cereais da ordem do 18,8%. Este decréscimo teve maior destaque no arroz pela grande descida num valor de 32,7% no trimestre ■ O conjunto dos cereais, no final do 4º trimestre de 2012, totalizaram um stock de 16.241 tons, que vai coresponder um prazo de cobertura de 4,6 meses. Regista-se uma variação absoluta negativa entre o stock inicial e final no 4º trimestre de 2012 na ordem de 760 tons, no conjunto dos cereais, esta descida registou, no milho e no trigo, que apresentaram uma redução 4.412 tons e 1.352 tons respectivamente. ■ No 4º trimestre de 2012, a importação dos grandes operadores no conjunto dos cereais é de 16.055 tons, sendo no milho com 8.686 tons que representa a maior quantidade importada no ano 2012. Em relação ao trimestre anterior houve um aumento de 6.008 tons no conjunto dos cereais. ■ O consumo aparente do conjunto dos cereais neste periodo totalizou-se 21.168 tons, o arroz contribui com 7.171 tons para o total (33,9%) seguido do trigo com 5.842 tons do total (27,6%). Comparado com o trimestre anterior, registou-se uma queda de 1,0% no consumo dos cereais. O grau de utilização média mensal do trimestre das disponibilidades dos grandes operadores é de 33,9% contra o valor médio mensal de 28,2% verificado no trimestre anterior. ■ A distribuição inter-ilhas do conjunto dos cereais totalizou num valor de 7.927 tons, no qual 1.118 tons de milho, 1.798 tons do arroz e 5.012 tons da farinha de trigo ■ Neste trimestre, o preço do conjunto dos cereais teve ligeiros declíneos, com uma baixa no milho de 5,1% e no arroz de 1,7%. Na farinha de trigo registou-se um aumento de 3,2%. Açúcar: Com base nos dados do 4º trimestre de 2012, considera-se que o abastecimento do açúcar segue uma tendência favorável, com aumentos dos stoks, da importação e das disponibilidades médias . ■ Neste trimestre a disponibilidade total do açúcar é de 5.071 tons, sendo 2.618 tons do stock inicial e 2.453 tons da importação e cobre largamente às necessidades do consumo aparente que é de 2.095 tons. A disponibilidade média mensal do açúcar no trimestre é de 3.378 tons e em relação ao trimestre anterior verifica-se uma baixa de 4,5%. ■ No final do trimestre o stock do açúcar é de 2.976 tons e o prazo de cobertura é de 3,4 meses. A variação entre o stock inicial e final é de 358 tons. ■ O grau de utilização média mensal do trimestre das disponibilidades do açúcar dos grandes operadores é de 43,9%, estima-se que o consumo aparente médio é de 698 tons mensal. ■ Neste trimestre, a distribuição inter-ilhas total do açúcar é de 484 tons. Esta quantidade foi distribuida para todas as ilhas. ■ O preço de açúcar teve redução de 1,7% em relação ao trimestre anterior, e continua apontar uma diminuição para o próximo trimestre.

Metric Short Course for the Office of Bridges and Structures, HR-378, 1995

This metric short course was developed in response to a request from the Office of Bridges and Structures to assist in the training of engineers in the use of metric units of measure which will be required in all highway designs and construction after September 30, 1996 (CFR Presidential Executive Order No. 12770). The course notes which are contained in this report, were developed for a half-day course. The course contains a brief review of metrication in the U.S., metric units, prefixes, symbols, basic conversions, etc. The unique part of the course is that it presents several typical bridge calculations (such as capacity of reinforced concrete compression members, strength of pile caps, etc.) worked two ways: inch-pound units throughout with end conversion to metric and initial hard conversion to metric with metric units throughout. Comparisons of partial results and final results (obtained by working the problems the two ways) are made for each of the example problems.

El estreno de la película Ágora de Alejandro Amenábar en octubre de 2009 puso nuevamente de moda la figura de la célebre filósofa y matemática Hipatia (a quien en rigor deberíamos llamar Hipacia) y generó la aparición de un gran número de monografías, novelas y artículos de todo tipo acerca de su figura. De entre toda esta producción destaca el libro de Clelia Martínez Maza, profesora titular de Historia Antigua de la Universidad de Málaga. Como pone de manifiesto su producción científica, la autora es una gran conocedora del fenómeno religioso y de los conflictos sociales vividos durante la Antigüedad tardía, algo que también puede observarse en la presente obra. Nos hallamos ante una monografía de divulgación culta, como manifiesta la abundante documentación que la autora maneja, aunque el libro carezca de notas tanto al final del volumen como a pie de página, algo normal en estos casos.

Traumatismo torácico: estudo retrospectivo de 168 casos

OBJETIVO: Avaliar o atendimento ao paciente portador de traumatismo torácico. MÉTODO: Estudo retrospectivo de 168 casos de trauma do tórax, com ênfase na abordagem inicial, conduta operatória e cuidados pós-operatórios. RESULTADOS: Dos 168 pacientes, 120 eram do sexo masculino e a média de idade encontrada foi de 35,5 anos. Dez pacientes foram toracotomizados de urgência, quatro por ferimento cardíaco, quatro devido à lesão de vasos pulmonares, um por lesão do saco pericárdico e outro do pedículo pulmonar. Os demais tiveram suas condições clínicas corrigidas através de simples drenagem torácica do hemitórax atingido. Ocorreram dois óbitos não cirúrgicos e um devido à insuficiência respiratória. CONCLUSÕES: O traumatismo torácico, além de ser muito freqüente, é na maioria das vezes, de fácil resolução. Podem ocorrer, no entanto, lesões torácicas graves envolvendo órgãos vitais que merecem intervenção mais agressiva e perícia técnica, com suporte hospitalar de alto nível.
