An Insight to the Dynamics of Conserved Water Molecular Triad in IMPDH II (Human): Recognition of Cofactor and Substrate to Catalytic Arg 322

Inosine 5' monophosphate dehydrogenase (IMPDH II) is a key enzyme involved in the de novo biosynthesis pathway of purine nucleotides and is also considered to be an excellent target for cancer inhibitor design. The conserve R 322 residue (in human) is thought to play some role in the recognition of inhibitor and cofactor through the catalytic D 364 and N 303. The 15 ns simulation and the water dynamics of the three different PDB structures (1B3O, 1NF7, and 1NFB) of human IMPDH by CHARMM force field have clearly indicated the involvement of three conserved water molecules (W-L, W-M, and W-C) in the recognition of catalytic residues (R 322, D 364, and N 303) to inhibitor and cofactor. Both the guanidine nitrogen atoms (NH1 and NH 2) of the R 322 have anchored the di- and mono-nucleotide (cofactor and inhibitor) binding domains via the conserved W-C and W-L water molecules. Another conserved water molecule W-M seems to bridge the two domains including the R 322 and also the W-C and W-L through seven centers H-bonding coordination. The conserved water molecular triad (W-C - W-M - W-L) in the protein complex may thought to play some important role in the recognition of inhibitor and cofactor to the protein through R 322 residue.

Representantes indígenas fizeram uma avaliação do substitutivo do relator Bernardo Cabral (PMDB-AM). O Capítulo sobre os índios, não chegou a ser votado na Sistematização. Os índios querem que o Plenário da Constituinte alterem dois artigos: o que tira os diretos dos índios em elevado estágio de aculturação, sem especificar o que seja esse estágio, e o que considera os índios incapazes. Os indígenas deram vários depoimentos e apresentaram a carta de Manaus com reivindicações. Pedem a proibição da mineração em suas terras, mas a principal questão que querem ver na Constituição é a demarcação, como comenta Terêncio Luiz da Silva, índio Macuxi. De acordo com o Deputado Virgílio Guimarães (PT-MG), a questão indígena é pior abordada no texto do que em Constituições anteriores. Segundo Senador Ronan Tito (PMDB-MG) o índio precisa de mais terra do que moradores comuns, devido sua identidade total com a natureza. Jovens do PMDB querem garantir os avanços conquistados na Constituinte: adoção do voto aos 16 anos, redução da maioridade para 18 anos para candidatura a a cargos eletivos e garantia do ensino público gratuito. De acordo com Pedro Longo, Presidente da Juventude Nacional do PMDB, o trabalho da Sistematização é satisfatório, mas há a preocupação de que os avanços obtidos sejam alterados na votação em Plenário. O Deputado Hermes Zaneti (PMDB-RS), autor da emenda do voto aos 16 anos, alerta jovens sobre o movimento de Constituintes de direita que podem reverter o quadro no Plenário da Constituinte. O Senador Irapuam Costa Junior (PMDB-GO) e o Deputado Nilson Gibson (PMDB-PE) assinaram o documento do Centrão para mudar sistema de votação no Plenário. O Senador considera que o texto não deve ser rejeitado em sua totalidade. Já o Deputado, afirma que há alguns assuntos no texto do Centrão dos quais discorda. A Mesa da Constituinte se reunirá para definir como será o Plenário da Constituinte. O Deputado José Melo (PMDB-AC) espera que seja alterado o regimento permitindo que sejam apresentadas em Plenário emendas ao Substitutivo aprovado pela Comissão de Sistematização. O Deputado Maguito Vilela (PMDB-GO) espera que o Plenário mantenha os avanços conquistados pela Sistematização.

Some abundant Tendipedid species (Diptera, Tendipedidae) of the Amu-Darya drainage basin. Acalcarella shilova, gen.n. only [Translation from: Ent.Obozr. 34 313-322, 1955]

This document is a partial translation of taxonomic notes on two Diptera species (Acalcarella Shilova, Acalcarrella nucus) which were abundant in a survey of Tendipedid species (Diptera, Tendipedidae) of the Amu Darya drainage basin. These taxonomic notes of the larva, pupa and adults include drawings of the Acalcarella nucus larva.

Rates for repair of pBR 322 DNA radicals by thiols as measured by the gas explosion technique: evidence that counter-ion condensation and co-ion depletion are significant at physiological ionic strength.

Rates of rapair of pBR 322 plasmid DNA radicals by thiols of varying net charge (Z) at pH 7 and physiological ionic strength were measured using the oxygen explosion technique. The extent of conversion of supercoiled to relaxed circular plasmid was measured by HPLC as a function of the time of oxygen exposure before or after irradiation, the time-courses being fitted by a pseudo-first-order kinetic expression with k1 = k2[RSH]. Values of k2 (M-1 S-1) were: 2.1 x 10(5) (GSH, Z = -1), 1.4 x 10(6) (2-mercaptoethanol, Z = 0), 1.2 x 10(7) (cysteamine, Z = +1), 6.6 x 10(7) (WR-1065 or N-(2-mercaptoethyl)-1,3-diamino?? propane, Z = +2). The approximately 6-fold increase in rate with each unit increase in Z is attributed to concentration of cationic thiols near DNA as a consequence of counter-ion condensation and reduced levels of anionic thiols near DNA owing to co-ion depletion. The results are quantitatively consistent with chemical repair as a significant mechanism for radioprotection of cells by neutral and cationic thiols under aerobic conditions, but indicate that repair by GSH will compete effectively with oxygen only at low oxygen tension.
