The exchangeable cation compositions of organic-poor terrigenous sediments containing smectite as primary ion exchanger from a series of holes along ODP Leg 168 transect on the eastern flank of the Juan de Fuca Ridge have been examined as a function of distance from the ridge axis and burial depth. The total cation exchange capacity (CEC) values of the sediments ranged from 2 to 59 meq/100 g, increasing with increases in the wt.% smectite. At the seafloor, the exchangeable cation compositions involving Na, K, Mg, and Ca, expressed in terms of equivalent fraction, are nearly constant regardless of the different transect sites: XNa = 0.21 ± 0.04, XK = 0.08 ± 0.01, XMg = 0.33 ± 0.09, and XCa = 0.38 ± 0.09. The calculated selectivity coefficients of the corresponding quaternary exchange reactions, calculated using porewater data, are in log units -5.45 ± 0.39 for Na, 1.97 ± 0.49 for K, 0.42 ± 0.41 for Mg, and 3.06 ± 0.69 for Ca. The exchangeable cation compositions below the seafloor change systematically with distance from the ridge crest and burial depth, conforming to the trends of the same cations in the porewaters. The selectivities for Na and Mg are roughly constant at temperatures from 2 to 66°C, indicating that the equivalent fractions of these two cations are independent of sediment alteration taking place on the ridge flank. Unlike Na and Mg, the temperature influence is significant for K and Ca, with Ca-selectivity decreases being coupled with increases in K-selectivity. Although potentially related to diagenetic and/or hydrothermal mineral precipitation or recrystallization, no evidence of such alteration was detected by XRD and TEM. In sites where upwelling of hydrothermal fluids from basement is occurring, the K-selectivity of the sediment is appreciably higher than at the other sites and corresponds to the formation of (Fe, Mg) rich smectite and zeolites. Our study indicates that local increases in K-selectivity at hydrothermal sites are caused by the formation of these authigenic minerals.

The geochemical implications of thermally driven flow of seawater through oceanic crust on the mid-ocean ridge flank have been examined on a well-studied 80 km transect across the eastern flank of the Juan de Fuca Ridge at 48°N, using porewater and basement fluid samples obtained on ODP Leg 168. Fluid flow is recognised by near-basement reversals in porewater concentration gradients from altered values in the sediment section to seawater-like values in basaltic basement. In general, the basement fluids become more geochemically evolved with distance from the ridge and broadly follow basement temperature which ranges from not, vert, similar16° to 63°C. Although thermal effects of advective heat exchange are only seen within 20 km east of where basement is exposed near the ridge crest, chemical reactivity extends to all sites. Seawater passing through oceanic crust has reacted with basement rocks leading to increases in Ca2+ and decreases in alkalinity, Mg2+, Na+, K+, SO42- and delta18O. Sr isotope exchange between seawater and oceanic crust off axis is unequivocally demonstrated with endmember 87Sr/86Sr ~ 0.707. Evidence of more evolved fluids is seen at sites where rapid upwelling of fluids through sediments occurs. Chlorinities of the basement fluids are consistent with post-glacial seawater and thus a short residence time in the crust. Rates of lateral flow have been by estimated by modelling porewater sulphate gradients, using Cl as a glacial chronometer, and from radiocarbon dating of basal fluids. All three methods reveal fluid flow with 14C ages less than 10,000 yr and particle velocities of ~1-5 m/yr, in agreement with thermally constrained volumetric flow rates through a ~600 m thick permeable layer of ~10% porosity. Delta(element)/Delta(heat) extraction ratios are similar to values for ridge-crest hydrothermal systems.

Pore water was collected from each of 10 sites during Ocean Drilling Program (ODP) Leg 168 on the eastern flank of the Juan de Fuca Ridge. These ten sites delineate a transect perpendicular to the present ridge axis and span a crustal age of 0.86-3.59 Ma. At nine of the ten sites the entire sediment section, which ranged from 41.3 to 613.8 m thick, was cored and attempts were made to recover at least one whole round of sediment per section of core for extraction of pore water. Several (2-5) whole-round sediment samples were taken from the uppermost and lowermost cores to constrain the chemical gradient near the sediment/water and sediment/basalt interfaces, respectively. Pore water was extracted from whole-round sediment core sections by squeezing only the most pristine sediment in a titanium squeezer designed by Manheim and Sayles (1974). Two additional water samples were collected in situ using the water-sampler temperature probe (WSTP; Barnes, 1988, doi:10.2973/odp.proc.ir.110.104.1988). Both of these samples were collected in the cased section of the open borehole from ODP Hole 1026B. Formation fluids were flowing up the cased hole into the overlying deep seawater (Fisher et al., 1997, doi:10.1029/97GL01286). beta-1,3-1,4-Glucanases (E.C. 3.2.1.73) hydrolyze linked beta-D-glucans, such as lichenan and barley beta-glucan. Recombinant beta-1,3-1,4-glucanase from Bacillus subtilis expressed in Escherichia coil and purified by Ni-NTA chromatography exhibited optimum activity at 50 degrees C and pH 6.0. The catalytic half-life at 60 degrees C decreased from 90 to 5 min when the enzyme was incubated in the presence and absence of Ca(2+) respectively. The kinetic parameters of lichenan hydrolysis were 2695, 3.1 and 1220 for V(max) (mu mol/min/mg), K(m) (mg mL(-1)) and K(cat) (s(-1)), respectively. Analysis by DLS, AUC and SAXS demonstrated the enzyme is monomeric in solution. Chemical denaturation monitored by ITFE and far-UV CD yielded Delta G(H2O) values of 9.6 and 9.1 kcal/mol, respectively, showing that the enzyme has intermediate stability when compared with other Bacillus beta-1,3-1,4-glucanases. The crystal structure shows the anti-parallel jelly-roll beta-sheet conserved in all GH16 beta-1,3-1,4-glucanases, with the amino acid differences between Bacillus sp. enzymes that are likely determinants of stability being distributed throughout the protein. Sequence analysis reveals that the Bacillus subtilis 168 tuaABCDEFGH operon encodes enzymes required for the polymerization of teichuronic acid as well as for the synthesis of one of its precursors, the UDP-glucuronate. Mutants deficient in any of the tua genes, grown in batch cultures under conditions of phosphate limitation, were characterized by reduced amounts of uronate in their cell walls. The teichuronic acid operon belongs to the Pho regulon, as phosphate limitation induces its transcription. Placing the tuaABCDEFGH operon under the control of the inducible Pspac promoter allowed its constitutive expression independently of the phosphate concentration in the medium; the level of uronic acid in cell walls was dependent on the concentration of the inducer. Apparently, owing to an interdependence between teichoic and teichuronic acid incorporation into the cell wall, in examined growth conditions, the balance between the two polymers is maintained in order to insure a constant level of the wall negative charge.

Sequence homologies suggest that the Bacillus subtilis 168 tagO gene encodes UDP-N-acetylglucosamine:undecaprenyl-P N-acetylglucosaminyl 1-P transferase, the enzyme responsible for catalysing the first step in the synthesis of the teichoic acid linkage unit, i.e. the formation of undecaprenyl-PP-N-acetylglucosamine. Inhibition of tagO expression mediated by an IPTG-inducible P(spac) promoter led to the development of a coccoid cell morphology, a feature characteristic of mutants blocked in teichoic acid synthesis. Indeed, analyses of the cell-wall phosphate content, as well as the incorporation of radioactively labelled precursors, revealed that the synthesis of poly(glycerol phosphate) and poly(glucosyl N-acetylgalactosamine 1-phosphate), the two strain 168 teichoic acids known to share the same linkage unit, was affected. Surprisingly, under phosphate limitation, deficiency of TagO precludes the synthesis of teichuronic acid, which is normally induced under these conditions. The regulatory region of tagO, containing two partly overlapping sigma(A)-controlled promoters, is similar to that of sigA, the gene encoding the major sigma factor responsible for growth. Here, the authors discuss the possibility that TagO may represent a pivotal element in the multi-enzyme complexes responsible for the synthesis of anionic cell-wall polymers, and that it may play one of the key roles in balanced cell growth.

OBJETIVO: Avaliar o atendimento ao paciente portador de traumatismo torácico. MÉTODO: Estudo retrospectivo de 168 casos de trauma do tórax, com ênfase na abordagem inicial, conduta operatória e cuidados pós-operatórios. RESULTADOS: Dos 168 pacientes, 120 eram do sexo masculino e a média de idade encontrada foi de 35,5 anos. Dez pacientes foram toracotomizados de urgência, quatro por ferimento cardíaco, quatro devido à lesão de vasos pulmonares, um por lesão do saco pericárdico e outro do pedículo pulmonar. Os demais tiveram suas condições clínicas corrigidas através de simples drenagem torácica do hemitórax atingido. Ocorreram dois óbitos não cirúrgicos e um devido à insuficiência respiratória. CONCLUSÕES: O traumatismo torácico, além de ser muito freqüente, é na maioria das vezes, de fácil resolução. Podem ocorrer, no entanto, lesões torácicas graves envolvendo órgãos vitais que merecem intervenção mais agressiva e perícia técnica, com suporte hospitalar de alto nível.
