Hydroxyurea is used for sickle-cell disease patients in order to increase fetal hemoglobin synthesis and consequently decrease the severity of pain episodes. Fetal hemoglobin, which is formed by gamma-globin chains A and G, is present in a constant composition throughout fetal development: about 75% of Ggamma and 25% of Agamma. In contrast, adult red cells contain about 40% of Ggamma and 60% of Agamma. In the present study, we analyzed the effect of hydroxyurea induction on the gamma chain composition of fetal hemoglobin in 31 sickle-cell disease patients treated with hydroxyurea. The control group was composed of 30 sickle-cell disease patients not treated with hydroxyurea in clinical steady state. The patients were older than 13 years and were not matched for age. All patients were seen at Hemocentro/UNICAMP and Boldrini Infantile Center, Campinas, SP, Brazil. The levels of total hemoglobin were significantly higher in patients treated with hydroxyurea (mean ± SD, 9.6 ± 2.16 g/dl) than in untreated patients (8.07 ± 0.91 g/dl). Fetal hemoglobin levels were also higher in treated patients (14.16 ± 8.31%) than in untreated patients (8.8 ± 4.09%), as was the Ggamma/Agamma ratio (1.45 ± 0.78 vs 0.98 ± 0.4, P < 0.005). The increase in the Ggamma/Agamma ratio in patients treated with hydroxyurea suggests the prevalence of a pattern of fetal hemoglobin synthesis, whereas patients not treated with hydroxyurea maintain the adult pattern of fetal hemoglobin synthesis. Because no correlation was observed between the Ggamma/Agamma ratio and total hemoglobin or fetal hemoglobin levels, the increase in Ggamma chain synthesis may not imply a higher production of hemoglobin.

Fetal hemoglobin (HbF), encoded by the HBG2 and HBG1 genes, is the best-known genetic modulator of sickle cell anemia, varying dramatically in concentration in the blood of these patients. This variation is partially associated with polymorphisms located in the promoter region of the HBG2 and HBG1 genes. In order to explore known and unknown polymorphisms in these genes, the sequences of their promoter regions were screened in sickle cell anemia patients and correlated with both their HbF levels and their βS-globin haplotypes. Additionally, the sequences were compared with genes from 2 healthy groups, a reference one (N = 104) and an Afro-descendant one (N = 98), to identify polymorphisms linked to the ethnic background.The reference group was composed by healthy individuals from the general population. Four polymorphisms were identified in the promoter region of HBG2 and 8 in the promoter region of HBG1 among the studied groups. Four novel single nucleotide polymorphisms (SNP) located at positions -324, -317, -309 and -307 were identified in the reference group. A deletion located between -396 and -391 in the HBG2 promoter region and the SNP -271 C→T in the HBG1 promoter region were associated with the Central African Republic βS-globin haplotype. In contrast, the -369 C→G and 309 A→G SNPs in the HBG2 promoter region were correlated to the Benin haplotype. The polymorphisms -396_-391 del HBG2, -369 SNP HBG2 and -271 SNP HBG1 correlated with HbF levels. Hence, we suggest an important role of HBG2 and HBG1 gene polymorphisms on the HbF synthesis.

Foi determinada a concentração plasmática de zinco em crianças pertencentes a famílias de baixa renda que residiam em três bairros da periferia de Ribeirão Preto, SP (Brasil), e verificada uma possível correlação desses níveis com algumas medidas antropométricas. Foram estudadas 126 crianças de 2 a 7 anos. Os níveis plasmáticos de zinco foram inferiores a 70 µg% em 13% das crianças estudadas. As medidas antropométricas mostraram que 42,9% das crianças estudadas apresentavam peso, 6,3% apresentavam altura e 9,5% apresentavam peso/altura inferiores a 90% do percentil 50 do National Center for Health Statistics (NCHS). A prega tricipital e a circunferência braqueal e muscular foram inferiores a 90% do percentil 50 do NCHS em respectivamente 65,8%, 11% e 7,5% das crianças estudadas. Não se observou correlação significante entre os níveis plasmáticos de zinco e os parâmetros antropométricos estudados.

De uma plantação de almeirão (Chicorium intybus c.v. Folha Larga) foram coletadas 30 plantas com 4 repetições aos 25, 35, 45 e 55 dias após a emergência. O material coletado foi dividido em folhas e raízes e analisados por N, P, K, Ca, Mg, S, B, Cu, Fe, Mn, Zn, pelos métodos convencionais de laboratório. O elemento extraído em maior quantidade foi o nitrogênio, seguido de potássio, cálcio, fósforo, magnésio e enxofre. Dentre os micronutrientes o elemento extraído em maiores quantidades foi o ferro, seguido do manganês, zinco, boro e finalmente pelo cobre. Estabelece-se o nível crítico para os nutrientes os seguintes valores: 4,39% N, 0,47% P, 2,93 K, 1,00 % Ca, 0,35% Mg, 0,20% S, 59 ppm B, 15 ppm Cu, 2,926 ppm Fe, 117 ppm Mn e 80 ppm Zn. A extração total por folhas e raízes foi de 5.000 plantas/100 m² e de 92,3 g N, 13,3 g P, 83,2 g K, 28,8 g Ca, 8,6 g Mg, 5,45 g S, 154 mg B, 38 mg Cu, 7,979 mg Fe, 301 mg Mn e 214 mg Zn.

The metabolic derangement caused by diabetes mellitus may potentially affect bone mineral metabolism. In the present study we evaluated the effect of diabetes metabolic control on parathyroid hormone (PTH) secretion during stimulation with EDTA infusion. The study was conducted on 24 individuals, 8 of them normal subjects (group N: glycated hemoglobin - HbA1C = 4.2 ± 0.2%; range = 3.5-5.0%), 8 patients with good and regular metabolic control (group G-R: HbA1C = 7.3 ± 0.4%; range = 6.0-8.5%), and 8 patients with poor metabolic control (group P: HbA1C = 12.5 ± 1.0%; range: 10.0-18.8%). Blood samples were collected at 10-min intervals throughout the study (a basal period of 30 min and a 2-h period of EDTA infusion, 30 mg/kg body weight) and used for the determination of ionized calcium, magnesium, glucose and intact PTH. Basal ionized calcium levels were slightly lower in group P (1.19 ± 0.01 mmol/l) than in group N (1.21 ± 0.01 mmol/l) and group G-R (1.22 ± 0.01 mmol/l). After EDTA infusion, the three groups presented a significant fall in calcium, but with no significant difference among them at any time. Basal magnesium levels and levels determined during EDTA infusion were significantly lower (P<0.01) in group P than in group N. The induction of hypocalcemia caused an elevation in PTH which was similar in groups N and G-R but significantly higher than in group P throughout the infusion period (+110 min, N = 11.9 ± 2.1 vs G-R = 13.7 ± 1.6 vs P = 7.5 ± 0.7 pmol/l; P<0.05 for P vs N and G-R). The present results show that PTH secretion is impaired in patients with poorly controlled diabetes.

The production and regulation of interleukin (IL) IL-13, IL-4 and interferon-gamma was evaluated in different clinical forms of human schistosomiasis. The mechanisms of immune regulation are apparently different in the various clinical stages of the disease, some of them being antigen specific.

Gamma-glutamyltranspeptidase (GGT-EC 2.3.2.2) activity and glutathione (GSH) content were measured in livers of female weanling Wistar rats (N = 5-18), submitted to rice-and-bean diets (13 and 6% w/w protein), both supplemented or not with DL-methionine (0.5 and 0.23 g/100 g dry diet, respectively). After 28 days, the rats on the rice-and-bean diets showed significantly higher levels (four times higher) of liver GGT activity and a concomitant 50% lower concentration of liver GSH in comparison with control groups feeding on casein. The addition of DL-methionine to rice-and-bean diets significantly increased the liver GSH content, which reached levels 50% higher than those found in animals on casein diets. The increase in GSH was accompanied by a decrease in liver GGT activity, which did not reach levels as low as those observed in the control groups. No significant correlation could be established between GGT and GSH changes under the present experimental conditions. Linear correlation analysis only revealed that in animals submitted to unsupplemented rice-and-bean diets GSH concentration was positively associated (P<0.05) with weight gain, food intake and food efficiency. GGT, however, was negatively correlated (P<0.05) with food intake only, and exclusively for supplemented rice-and-bean diets. The high levels of GGT activity observed in the present study for rats receiving a rice-and-bean mixture could be a result of the poor quality of these diets associated with their deficiency in sulfur amino acids. The results also suggest that diet supplementation with methionine could be important in the reduction of the deleterious effects of GSH depletion by restoring the intracellular concentration of this tripeptide.

The objective of the present study was to evaluate the production of cytokines, interferon-g (INF-g) and interleukin-10 (IL-10), in cultures of peripheral blood mononuclear cells (PBMC) from type 1 and type 2 diabetic patients and to correlate it with inadequate and adequate metabolic control. We studied 11 type 1 and 13 type 2 diabetic patients and 21 healthy individuals divided into two groups (N = 11 and 10) paired by sex and age with type 1 and type 2 diabetic patients. The PBMC cultures were stimulated with concanavalin-A to measure INF-g and IL-10 supernatant concentration by ELISA. For patients with inadequate metabolic control, the cultures were performed on the first day of hospitalization and again after intensive treatment to achieve adequate control. INF-g levels in the supernatants of type 1 diabetic patient cultures were higher compared to type 2 diabetic patients with adequate metabolic control (P < 0.001). Additionally, INF-g and IL-10 tended to increase the liberation of PBMC from type 1 and 2 diabetic patients with adequate metabolic control (P = 0.009 and 0.09, respectively). The increased levels of INF-g and IL-10 released from PBMC of type 1 and 2 diabetic patients with adequate metabolic control suggest that diabetic control improves the capacity of activation and maintenance of the immune response, reducing the susceptibility to infections.

Experimental murine L. major infection is characterized by the expansion of distinct CD4+ T cell subsets. The Th1 response is related to production of IFN-g and resolution of infection, whereas Th-2 response with production of IL-4 and IL-10 and dissemination of infection. The objective of this study was to measure the circulating levels of IFN-g, IL-10 and TNF-a in patients with visceral leishmaniasis (VL) before, during and at the end of therapy and to examine the association between cytokine levels and activity of VL. Fifteen patients with VL were evaluated. The cytokine determinations were done by using the enzyme-linked immunoassay (ELISA) before, during and at the end of therapy. At baseline, we detected circulating levels of IFN-g in 13 of 15 patients (median = 60 pg/ml); IL-10 in 14 of 15 patients (median = 141.4 pg/ml); and TNF-a in 13 of 14 patients (median = 38.9 pg/ml). As patients improved, following antimonial therapy, circulating levels of IL-10 showed an exponential decay (y = 82.34 e–0,10367x, r = –0.659; p < 0.001). IFN-g was no longer detected after 7/14 days of therapy. On the other hand, circulating levels of TNF-a had a less pronounced decay with time on therapy, remaining detectable in most patients during the first seven days of therapy (y = 36.99-0.933x, r = –0.31; p = 0.05). Part of the expression of a successful response to therapy may, therefore, include reduction in secretion of inflammatory as well as suppressive cytokines. Since IL-10 and IFN-g are both detected prior to therapy, the recognized cellular immune depression seen in these patients may be due to biological predominance of IL-10 (type 2 cytokine), rather than lack of IFN-g (type 1 cytokine) production.

Isolation of Leishmania parasite and species identification are important for confirmation and to help define the epidemiology of the leishmaniasis. Mice are often used to isolate pathogens, but the most common mouse strains are resistant to infection with parasites from the Leishmania (Viannia) subgenus. In this study we tested the inoculation of interferon gamma knockout (IFNγ KO) mice with biopsy macerates from Leishmania-infected patients to increase the possibility of isolating parasites. Biopsies from twenty five patients with clinical signs of leishmaniasis were taken and tested for the presence of parasites. Immunohistochemical assay (IHC) and conventional histopathology detected the parasite in 88% and 83% of the patients, respectively. Leishmania sp. were isolated in biopsy macerates from 52% of the patients by culture in Grace's insect medium, but 13% of isolates were lost due to contamination. Inoculation of macerates in IFNγ KO mice provides isolation of parasites in 31.8% of the biopsies. Most isolates belong to L. (Viannia) subgenus, as confirmed by PCR, except one that belongs to L. (Leishmania) subgenus. Our preliminary results support the use of IFNγ KO mice to improve the possibility to isolate New World Leishmania species.

A total of 123 stool specimens collected in Teresina, Piauí between 1994 and 1996, from 0 to 2-year-old children with diarrhea, were used for this study. Molecular characterization of the G and P rotavirus genotypes was performed using the reverse transcriptase polymerase chain reaction. The following results were obtained for the P genotypes: P[8] (17. 1%), P[1] (4. 9%), P[4] (3. 3%), P[6, M37] (2. 4%) and mixtures (27. 6%). The P[1]+P[8] mixture was found in 19. 5% of the samples. For the G genotypes, the results were: G1 (25. 2%), G5 (13. 8%), G2 (2. 5%), G4 (2. 5%), G9 (0. 8%) and mixtures (41. 5%). G1+G5 was the mixture most frequently found (12. 1%). Our results showed unusual combinations such as P[1]G5 and P[1]+P[8]G5. The high percentage of mixtures and unusual combinations containing mixtures of human and animal rotavirus genotypes strongly suggests the possibility of gene reassortment and interspecies transmission.

Aproximadamente 2500 alevinos de matrinchã com comprimento padrão de 0,8 a 5,0cm foram coletados no lago Catalão, margem direita do rio Negro e no rio Solimões, próximo a ilha da Marchantaria, Estado do Amazonas. Uma subamostra de 136 peixes foi retirada para exames parasitológicos e o restante foi colocado em tanques em uma estação de piscicultura, durante 30 dias. Após esse período uma segunda amostra foi retirada e examinada. Dos 136 peixes: 49 (36%) estavam parasitados com metacercárias de trematódeos; 32 (23,5%) com nematóides Spirocamallanus inopinatus; 26 (19%) com monogenóideos Jainus amazonensis; 3 (2%) com protozoários Trichodina sp.; 1 (0,7%) com acantocéfalo Echinorhynchus sp. e 2 (1,5%) com estágios larvais de cestóides. Dos peixes mantidos nos tanques, após 30 dias foi examinada uma subamostra de 60 peixes: 42 (70%) estavam parasitados com J. amazonensis; 14 (23%) com S. inopinatus; 9 (15%) com metacercárias de trematódeos e 2 (3%) com Echinorhynchus sp. A intensidade média e abundância média foram maiores em J. amazonensis (342 e 65), seguido por cestóides (70 e 1), Trichodina sp. (13 e 0,3), trematódeos (5 e 2), S. inopinatus (1 e 0,3) e Echinorhynchus sp. (1 e 0,007). Na estação de piscicultura a intensidade média e abundância média também foram maiores para o J. amazonensis (222 e 115) em relação aos demais grupos. A espécie J. amazonensis apresentou maior intensidade média e menor prevalência na natureza. Houve variação na composição da fauna parasitógica e nos índices de infestação, após 30 dias de permanência na estação de piscicultura.

Foram estudados a dieta, a composição corporal e o valor de energia bruta do conteúdo estomacal, filé e fígado, bem como alguns frutos e sementes que fazem parte da dieta do matrinxã, relacionando-se estes parâmetros com as flutuações do nível da água durante um ciclo anual. Observou-se que a espécie se alimenta de sementes, frutos, flores, restos de vegetais e insetos, apresentando hábito alimentar onívoro. Estômagos vazios foram frequentes (68,5%) nos diferentes períodos hidrológicos. As análises proximais e valor da energia bruta do filé e fígado dos peixes analisados não apresentaram diferenças significativas entre os sexos (P>0,05). No entanto, observou-se diferenças entre os períodos de águas baixas e altas. As médias das composições centesimais e os valores de energia bruta do conteúdo estomacal, expressos com base na matéria seca revelaram, na enchente, um baixo teor de proteína (8,9%) e cinza (1,3%), alto valor de lipídios (70,2%) e energia (775 Kcal EB/100g). Na seca, o teor de proteína (24,9%) e cinza (6,3%) foram maiores, porém os teores de lipídios (13,2%) e de energia (416,9 Kcal EB 100g) foram mais baixos que na cheia. Foi verificado abundante depósito de gordura cavitária o ano todo, diminuindo em janeiro, mês em que o matrinxã realiza a desova.

The volatile constituents obtained from the pentane extract, using simultaneous distillation-extraction of the flowers of Encyclia vespa and E. fragrans were analysed by GC/ MS. The main volatile components identified in the flowers of E. vespa were terpinen-4-ol (20.3%), verbenone (14.8%), trans-verbenol (13.6%) and x-pinene (11.8%). The major volatiles of the flowers of E. fragrans were terpinen-4-ol (18.3%), (2Z,6E)-farnesol (15.4%) and trans-verbenol (10.2%).
