A parasitological survey was carried out on 222 inhabitants of five farms in Holambra, located 30 km north of Campinas, São Paulo, Brazil, on October 1992. Approximately 70% of the inhabitants were found to be infected with at least one species of intestinal parasite. The positive rates of 6 helminths and 7 protozoan species detected are as follows: 5.4% Ascaris lumbricoides; 8.6% Trichuris trichiura; 19.8% Necator americanus; 10.4% Strongyloides stercoralis; 14% Enterobius vermicularis; 0.9% Hymenolepis nana; 3.2% Entamoeba histolytica; 2.7% E. hartmanni; 9.9% E. coli; 14.0% Endolimax nana; 2.3% Iodamoeba butschlii; 10.4% Giardia lamblia; 37.8% Blastocystis hominis. The positive rates of helminth infection were generaly higher in the younger-group under 16 years-old than those in the elder group aged 16 or more, whereas the infection rates of protozoan species were higher in the elder group. The infection rate of Strongyloides was found to be 10.4% by a newly developed sensitive method (an agarplate culture methods).

Prevalence of Strongyloides stercoralis infection in three areas of Brazil was surveyed by a recently developed faecal culture method (an agar plate culture). The Strongyloides infection was confirmed in 11.3% of 432 subjects examined. The diagnostic efficacy of the agar plate culture was as high as 93.9% compared to only 28.5% and 26.5% by the Harada-Mori filter paper culture and faecal concentration methods, when faecal samples were examined simultaneously by these three methods. Among the 49 positive samples, about 60% were confirmed to be positive only by the agar plate culture. These results indicate that the agar plate culture is a sensitive new tool for the correct diagnosis of chronic Strongyloides infection.

Between 1992 and 1997, 790 blood donors with anti-HCV EIA-2 strongly reagent (relationship between the sample optical density/cut-off > 3) detected at the blood bank serological screening, were evaluated in ambulatory environment. They were all negative for Chagas disease, syphilis, hepatitis B (HBsAg) and AIDS. Blood samples were collected at the first ambulatorial evaluation, for hemogram, biochemical tests and new serological tests for HCV (anti-HCV EIA-2). In blood samples of 226 repeatedly reagent anti-HCV EIA-2 blood donors, supplementary "immunoblot" test for HCV (RIBA-2) was used. In 209 donors, the presence of HCV-RNA was investigated by the PCR test. The abdominal ultrasonography was realized in 366 donors. In 269 patients liver biopsy was performed for the histopathological study. The follow-up of blood donors showed that 95.6% were repeatedly EIA-2 reagent, 94% were symptomless and denied any hepatitis history, with only 2% mentioning previous jaundice. In 47% of this population at least one risk factor has been detected for the HCV transmission, the use of intravenous drugs being the main one (27.8%). Blood transfusion was the second factor for HCV transmission (27.2%). Hepatomegaly was detected in 54% of the cases. Splenomegaly and signs of portal hypertension have seldom been found in the physical examination, indicating a low degree of hepatic compromising in HCV. Abdominal ultrasound showed alterations in 65% of the subjects, being the steatosis the most frequent (50%). In 83.5% of the donors submitted to the liver biopsy, the histopathological exam showed the presence of chronic hepatitis, usually classified as active (89%) with mild or moderate grade in most of the cases (99.5%). The histopathological exam of the liver was normal in 1.5% of blood donors. The RIBA-2 test and the HCV-RNA investigation by PCR were positive in respectively 91.6 and 75% of the anti-HCV EIA-2 reagent donors. The HCV-RNA research was positive in 82% of the RIBA-2 positive subjects, in 37.5% of the indeterminate RIBA-2 donors and in 9% of the negative RIBA-2 donors. Chronic hepatitis has also been observed in 50% of the histopathological exams of the anti-HCV EIA-2 reagent donors which were indeterminate RIBA-2. Among 18 blood donors with minimal changes histopathological exam 11 (61%) were HCV-RNA positive. Our blood donors anti-HCV reagent generally had clinical, laboratorial and histopathological features observed in patients with chronic HCV hepatitis and a high proportion could be identified in interviews and medical evaluation realized in blood blanks. Generally, these HCV infected donors are identified and discharged only by the serological tests results.

A Embrapa-Centro de Pesquisa Agropecuária dos Cerrados (CPAC), a Embrapa-Centro Nacional de Pesquisa de Trigo (CNPT), a Empresa de Pesquisa Agropecuária de Minas Gerais (Epamig), a Empresa Goiana de Pesquisa Agropecuária (Emgopa, atual Emater-GO) e a Cooperativa Agropecuária Mista do Programa de Assentamento Dirigido do Alto Paranaíba (Coopadap-MG), recomendaram uma nova cultivar de trigo -- EMBRAPA 22 -- para plantio no sistema de cultivo irrigado por aspersão nos estados de Goiás e Minas Gerais e no Distrito Federal. Recentemente, a cultivar foi recomendada também para os estados de Mato Grosso e Bahia. EMBRAPA 22 é originária de cruzamentos realizados no Centro Internacional de Melhoramento de Milho e Trigo (CIMMYT), situado no México. Foi selecionada na Embrapa-CPAC entre diversas linhas avançadas enviadas pelo CIMMYT e identificada em ensaios da rede experimental da região central do Brasil como linhagem CPAC 841153. A cultivar possui glúten forte e estabilidade alta, que conferem a ela uma classificação superior quanto à qualidade industrial, além de apresentar ainda ciclo precoce, alta produtividade, resistência à ferrugem-do-colmo e ferrugem-da-folha, em condições de campo, tolerância à debulha e moderada tolerância ao acamamento. Em experimentos conduzidos em Goiás e Distrito Federal, a EMBRAPA 22 produziu 6% a mais que a testemunha Anahuac, e em Minas Gerais, 4%.

O objetivo deste trabalho foi estimar o tamanho da parcela para avaliação da produtividade de grãos de trigo, sob condições irrigadas, em sistema de plantio direto e convencional. Cada experimento foi formado por 30 fileiras de 30 m de comprimento, no espaçamento de 20 cm e densidade de 380 sementes/m². A colheita foi efetuada em unidades básicas (ub) de 1,0 m de fileira (0,2 m²), colhendo-se os 12 m centrais das 24 fileiras centrais, totalizando 288 ub. O tamanho da parcela foi estimado pelos métodos da máxima curvatura, máxima curvaturamodificada, comparação de variâncias e método de Hatheway. Os tamanhos de parcela apresentaram grande variação em razão do método utilizado na sua estimativa. Parcelas menores, com maior número de repetições, foram mais eficientes no uso da área experimental do que parcelas maiores com menor número de repetições. Parcelas com tamanho variando entre 1,6 e 2,4 m² de área útil possibilitaram adequada avaliação da produtividade de grãos nas diferentes condições estudadas.

The objective of this work was to identify genomic regions that underlie resistance to Fusarium tucumaniae sp. nov., the causing agent of sudden death syndrome (SDS) in soybean in South America, using a population with a genetic background different from that previously reported for Fusarium virguliforme sp. nov. (F. solani f. sp. glycines), also responsible for SDS in soybean. Although major genes and quantitative trait loci (QTL) for SDS resistance have been identified, little is known about the same disease caused by Fusarium tucumaniae sp. nov., in South America. To identify genetic factors related to resistance to F. tucumaniae and DNA markers associated with them, a QTL analysis was performed using recombinant inbred lines. The map locations of the four loci, here identified, differed from those SDS resistance QTL previously described. It was screened a residual heterozygous line (RHL), which was heterozygous around the most effective QTL, RSDS1, and homozygous for the other genomic regions. The genetic effect of RSDS1 was confirmed using near-isogenic lines (NIL) derived from the RHL. The line which was homozygous for the Misuzudaizu genotype showed resistance levels comparable with that of the line homozygous for the Moshidou Gong 503 genotype.

O objetivo deste trabalho foi estimar a adaptabilidade e estabilidade de genótipos de trigo irrigado, nas condições do Cerrado do Brasil Central. O rendimento de grãos de cinco cultivares e nove linhagens de trigo irrigado foram aferidos no ensaio de valor de cultivo e uso. Os ensaios foram conduzidos nos anos de 2005 e 2006, em seis locais em Minas Gerais, três em Goiás e um no Distrito Federal. Os dados de rendimento de grãos foram submetidos à análise de variância individual e à análise de variância conjunta, para a estimativa dos parâmetros de adaptabilidade e estabilidade. Verificaram-se diferenças significativas, entre os genótipos, em 14 dos 16 ambientes avaliados. Nenhum dos genótipos avaliados apresentou o comportamento ideal preconizado pelo método de Cruz e colaboradores. Destacaram-se a cultivar BRS 264 e a linhagem CPAC 02167, pela superioridade em todas as condições do Cerrado do Brasil Central (adaptabilidade geral). Entre os demais genótipos avaliados, destacaram-se as cultivares BRS 207 e EMBRAPA 22, responsivas à melhoria das condições ambientais, e as cultivares BRS 254 e EMBRAPA 42, pela adaptabilidade específica a ambientes desfavoráveis ao cultivo de trigo irrigado.

The immuno-reaction has been successfully employed for the biosensor development. A focus on biosensor based on immunoassay coupled to amperometric transducer is presented.

Affinity reactions have been used for specific detection of their complementary partners and an enormous variety of enzyme-linked immunosorbent assay (ELISA) formats are used in research and in routine serological tests. With the advent of the atomic force microscopy (AFM) technique, the immune reactions have been monitored by these devices. In the present article we focus on applications of AFM to immunoassays. After introducing the basic concepts of AFM, a brief discussion on the monitoring of the interactions between antigens and antibodies through both topographic image and biosensor systems is presented.

Fast, selective, reproducible and reliable detections have been carried out by using enzymatic biosensors in several areas. The enzymatic biosensors based on the inhibition represent an important role in analytical chemistry. Enzymes like cholinesterases, peroxidases, tyrosinases, etc. have been immobilized on electrochemical and optical transducers and the enzymatic activity decreasing in the presence of the inhibitor is related with its concentrations. This article presents a review on the enzymes used on the construction of these sensors, emphasizing the respective applications.

Pharmaceutical compounds have been detected in sewage treatment plant (STP) effluents, surface waters and, less frequently, in groundwater and drinking water, all over the world. Different sources are responsible for their appearance in the aquatic environment, however, it is widely accepted that the main sources of this type of pollutant are STP effluents. The adverse effects of pharmaceuticals in the environment include aquatic toxicity, development of resistance in pathogenic bacteria, genotoxicity and endocrine disruption. Thus, the discharge of these compounds to the environment in STP effluents should be minimized.

An analytical method for the determination of the anti-inflammatory drug 5-aminosalicylic acid (5-ASA) in pharmaceutical formulations using square wave voltammetry at pencil graphite electrodes was developed. After the optimization of the experimental conditions, calibration curves were obtained in the linear concentration range from 9.78 à 10-7 to 7.25 à 10-5 mol L-1 resulting in a limit of detection of 2.12 ± 0.05 x 10-8 mol L-1. Statistical tests showed that the concentrations of 5-ASA in commercial tablets and enemas obtained with the proposed voltammetric method agreed with HPLC values at a 95% confidence level.

Neste trabalho procurou-se descrever detalhadamente o procedimento de preparo de um arranjo de eletrodos com geometria em forma de microdiscos (20,2 < r e < 25,7 mim) e estudar o comportamento eletroquímico desse conjunto de microeletrodos. Foram aplicadas diferentes equações descritas na literatura para a condição de estado estacionário e quase-estacionário e equações para a simulação de voltamogramas em regime estacionário. Para baixas velocidades, v <= 0,1 mV s-1, observou-se uma excelente sobreposição dos voltamogramas cíclicos experimental e simulado, entretanto, velocidades desta ordem de grandeza são pouco práticas do ponto de vista experimental. Observou-se, também para todos os microeletrodos estudados, uma transição da condição de corrente estacionária para quase-estacionária, com o aumento dos valores de v. Equações que permitem avaliar a contribuição das correntes radial e difusional, para a corrente total, ajustam-se perfeitamente as curvas I/E em todo o intervalo de raios dos eletrodos e valores de v empregados. Os voltamogramas obtidos para o arranjo de eletrodos operando na condição de curto circuito apresentou um fator de amplificação de 19 vezes, referente ao sinal obtido a partir de um único microeletrodo. Tal fator confirma a condição de ausência de sobreposição das camadas de difusão dos eletrodos e, portanto, reflete a contribuição individual de cada eletrodo para a corrente total.

Tartrazina e índigo carmim são corantes alimentícios amplamente utilizados na indústria alimentícia. Ambos os corantes apresentam ondas voltamétricas de redissolução catódica bem definidas no intervalo de pH 2 a 12, quando pré-acumulados sobre eletrodo de gota pendente de mercúrio. Curvas de calibração para tartrazina e índigo carmim são obtidas no intervalo de 5x10-9mol.L-1 a 1x10-7mol.L-1, em tempos de acúmulo de 30s e potencial de acúmulo de 0V e -0,10V, em tampão acetato pH 4,5 e carbonato pH 10, respectivamente. O método proposto foi aplicado para determinação direta de ambos os corantes em amostras individuais ou misturas em meio aquoso, e também, em amostras comerciais de suco artificial em pó e balas.

In order to study the affinity reaction between the anti-atrazine antibody and atrazine, an enzyme was incorporated, as a marker, to an atrazine carboxylic derivative. The hapten and conjugate were synthesized and characterized by MS, IR and NMR. The interaction between monoclonal antibodies and hapten-HRP conjugate was investigated by enzyme linked immunosorbent assay (ELISA).
