Autotransplantation of spleen tissue has been done, in the past ten years, in children with schistosomiasis mansoni with bleeding varices. The purposes of this investigation were: (1) to study the morphology and function of the remnant spleen tissue; (2) to quantify the production of tuftsin; and (3) to assess the immune response to pneomococcal vaccine of these patients. Twenty three children, who underwent splenectomy and autologous implantation of spleen tissue into the greater omentum were included in this investigation. The average postoperative follow-up is five years. Splenosis was proved by colloid liver-spleen scans. Search for Howell-Jolly bodies assessed the filtration function. Tuftsin and the titer of pneumococcal antibodies were quantified by ELISA. Splenosis was evident in all children; however, it was insufficient in two. Howell-Jolly bodies were found only in these two patients. The mean tuftsin serum concentration (335.0 ± 29.8 ng/ml) was inside the normal range. The immune response to pneumococcal vaccination was adequate in 15 patients; intermediate in four; and inadequate in four. From the results the following conclusions can be drawn: splenosis was efficient in maintaining the filtration splenic function in more than 90% and produced tuftsin inside the range of normality. It also provided the immunologic splenic response to pneumococcal vaccination in 65% of the patients of this series.

The present investigation was carried out on a sample of 840 children (5 to 16 years old) from ten small towns of the State of Bahia in northeastern Brazil. The objetive was to study, by using a cross sectional methodology, the evolution of schistosomiasis morbidity (hepatic and splenic enlargement) in children, and the role of the intensity of S. mansoni infection in this process. The children were analised in three age groups (5 to 8, to 12 and 13 to 16 years old) and classified as uninfected, mildly infected, moderately infected and heavily infected according to the number of eggs in the stool. In children aged 5 to 8 years, increasing egg counts were not associated with increasing frequencies of hepatic or splenic enlargement. In the 9 to 12 years old group and association was observed with the prevalence of hepatic enlargement, but not with the prevalence of spleen enlargement. In the oldest group, 13 to 16 years old, an association was observed with the prevalence of enlargement of both organs. It was evident that in this population schistosomiasis morbidity develops in the early period of life as a gradual process starting with liver enlargement and followed by spleen enlargement some years later. It was found that the intensity of infection has a fundamental role in this process, although there is a latent period of some years before clinical splenomegaly appears in moderate-heavily infected children. The Authors suggest that the prevalence of splenomegaly in the 13 to 16 years old group is a good measure of the community level of schistosomiasis morbidity and could be used to measure the impact of control programs.

Cytokines are important in the cell-mediated response to Schistosoma mansoni eggs. We have found that Th2 cytokine responses (e.G. IL-4 and IL-5) are argumented after egg laying begins while the response (IL-2 and IFN-*) are down regulated in S. mansoni infected mice. Treatment of mice with anti-IL-5 monoclonal antibodies (Mab) suppressed the eosinophil response almost completley but did not affect granuloma size and slightly increased hepatic fibrosis. Anti-IL-4 treatment abolished IgE responses in infected mice and decreased hepatic fibrosis slightly. Anti-IFN-* treatment had no effect on hepatic pathology. Anti-IL-2 treatment decreased granuloma size significantly and decreased hepatic fibrosis markedly. Anti-IL-2 treatment dramatically decreased IL-5 secretion by splenic cells in vitro and decreased peripheral blood and tissue eosinophilia. In contrast IL-4 secretion was unaffected and serum IgE was normal or increased. IL-2 and IFN-* secretion by splenic cells of treated mice were slightly but not significantly increased suggesting that anti-IL-2 treatment affecting Th2 rather than Th1 responses.

The zinc finger motifs (Cys2His2) are found in several proteins playing a role in the regulation of transcripton. SmZF1, a Schistosoma mansoni gene encoding a zinc finger protein was initially isolated from an adult worm cDNA library, as a partial cDNA. The full sequence of the gene was obtained by subcloning and sequencing cDNA and genomic fragments. The collated gene sequence is 2181 nt and the complete cDNA sequence is 705 bp containing the full open reading frame of the gene. Analysis of the genome sequence revealed the presence of three introns interrupting the coding region. The open reading frame theoretically encodes a protein of 164 amino acids, with a calculated molecular mass of 18,667Da. The predicted protein contains three zinc finger motifs, usually present in transcription regulatory proteins. PCR amplification with specific primers for the gene allowed for the detection of the target in egg, cercariae, schistosomulum and adult worm cDNA libraries indicating the expression of the mRNA in these life cycle stages of S. mansoni. This pattern of expression suggests the gene plays a role in vital functions of different life cycle stages of the parasite. Future research will be directed to elucidate the functional role of SmZF1.

The study of the Schistosoma mansoni genome, one of the etiologic agents of human schistosomiasis, is essential for a better understanding of the biology and development of this parasite. In order to get an overview of all S. mansoni catalogued gene sequences, we performed a clustering analysis of the parasite mRNA sequences available in public databases. This was made using softwares PHRAP and CAP3. The consensus sequences, generated after the alignment of cluster constituent sequences, allowed the identification by database homology searches of the most expressed genes in the worm. We analyzed these genes and looked for a correlation between their high expression and parasite metabolism and biology. We observed that the majority of these genes is related to the maintenance of basic cell functions, encoding genes whose products are related to the cytoskeleton, intracellular transport and energy metabolism. Evidences are presented here that genes for aerobic energy metabolism are expressed in all the developmental stages analyzed. Some of the most expressed genes could not be identified by homology searches and may have some specific functions in the parasite.

Cytochrome p450s (cyp450s) are a family of structurally related proteins, with diverse functions, including steroid synthesis and breakdown of toxins. This paper reports the full-length sequence of a novel cyp450 gene, the first to be isolated from the tropical freshwater snail Biomphalaria glabrata, an important intermediate host of Schistosoma mansoni. The nucleotide sequence is 2291 bp with a predicted amino acid sequence of 584aa. The sequence demonstrates conserved cyp450 structural motifs, but is sufficiently different from previously reported cyp450 sequences to be given a new classification, CYP320A1. Initially identified as down-regulated in partially resistant snails in response to S. mansoni infection, amplification of this gene using RT-PCR in both totally resistant or susceptible snail lines when exposed to infection, and all tissues examined, suggests ubiquitous expression. Characterization of the first cyp450 from B. glabrata is significant in understanding the evolution of these metabolically important proteins.

Foi focalizado, pela primeira vez o encontro de B. straminea no Estado de São Paulo. Esta espécie vem juntar-se aos planorbídeos já assinalados em nosso Estado. Foram descritos os criadouros, onde a B. straminea foi coletada, localizados em tanques de criação de peixes nas Estações de Piscicultura de Barra Bonita e Americana, Estado de São Paulo, e em um aquário particular na capital dêsse Estado. Fêz-se referência ao transporte de peixes oriundos de zonas do país onde ocorre aquela espécie, Amazonas e Ceará, como responsável pela introdução daquele molusco no Estado. Destacou-se êsse achado pelo perigo que representa a distribuição de peixes da maneira como vem sendo feita atualmente em nosso país, tendo sido julgado necessário o estabelecimento de quarentena para aquêles vindos de zonas infestadas por espécies hospedeiras intermediárias do S. mansoni. Foram relatadas as medidas de combate aos caramujos efetuadas imediatamente após aquela descoberta e os resultados obtidos. Conclui-se que a dispersão passiva da B. straminea pelo transporte de peixes, deve ampliar a distribuição geográfica dêsse planorbídeo, já assinalado na Venezuela, Guianas e no Brasil, sendo que neste último ocorre em tôdas as Unidades Federativas, exceto, no Rio Grande do Sul, Santa Catarina, Rio de Janeiro e Territórios.

Foram analisados os dados referentes ao desenvolvimento de cercárias de S. mansoni das linhagens de Belo Horizonte (MG) e de São José dos Campos (SP), Brasil. Concluiu-se que após a penetração das cercárias pelo tegumento dos camundongos, não houve diferença significativa quanto ao número de vermes adultos que se desenvolveram, pertencentes às duas cepas estudadas.

Concluiu-se haver diferenças significativas referentes às medidas de comprimento do verme, distância entre as ventosas oral e acetabular, distância entre a parte anterior dos vermes e a extremidade distal das gônadas e quanto ao número de testículos.

Utilizando um esquema de seleção individual em progenies autofecundadas, foram obtidas, após quatro gerações, populações de Biomphalaria tenagophila e de Biomphalaria glabrata, altamente suscetíveis às linhagens do Schistosoma mansoni do Vale do Rio Paraíba do Sul, SP e de Belo Horizonte, MG (Brasil), respectivamente. Os rápidos ganhos genéticos obtidos confirmam ser a suscetibilidade de moluscos à infecção esquistossomótica, uma característica de alta herdabilidade, sendo aparentemente condicionada por um pequeno número de genes.

Foram feitas observações no laboratório e no campo, em Belo Horizonte, MG, Brasil, com a finalidade de se obter informações biológicas e ecológicas sobre Pomacea haustrum (Reeve, 1856), molusco pilídeo, competidor-predador de hospedeiros intermediários de Schistosoma mansoni Sambon 1907.
