The present study intended to analyze the seroprevalence of Helicobacter pylori, IgG, and its relation to dyspepsia in a population from the western Amazon region. During the "Projeto Bandeira Científica", a University of São Paulo Medical School program, in Monte Negro's rural areas, state of Rondônia, 266 blood samples were collected from volunteers. The material was tested for IgG antibodies anti-Helicobacter pylori by ELISA method and the participants were also interviewed on dyspepsia, hygiene and social aspects. Participants aged between five and 81 years old (34 years on average), 149 (56%) were female and 117 (44%) male. We found 210 (78.9%) positive, 50 (18.8%) negative and six (2.3%) undetermined samples. Dyspeptic complaints were found in 226 cases (85.2%). There was no statistical association between dyspepsia and positive serology for H. pylori. We concluded that the seroprevalence in all age categories is similar to results found in other studies conducted in developing countries, including those from Brazil. On the other hand, the seroprevalence found in Monte Negro was higher than that reported in developed countries. As expected, there was a progressive increase in the positivity for H. pylori in older age groups.

Five human volunteers were vaccinated with a live avirulent strain of Trypanosoma cruzi and followed-up for one year. Except for a few cases of questionable results presented by only one Laboratory, ali the other clinicai, parasitological and serological tests remained negative during that period.

Clinical studies of the immunological effects of methionine enkephalin in normal volunteers, cancer, and AIDS patients are summarized. The major immunology changes seen were increases in T cell subsets, natural killer activity, as well as mitogen blastogenesis. Clinically, the cancer and ARC patients did not develop infections.

The design of malarial vaccine based on the circumsporozoite (CS) protein, a majuor surface antigen of the sporozoite stage of the malaria parasite, requires the identification of T and B cell epitopes for inclusion in recombinant or synthetic vaccine candidates. We have investigated the specificity and function of a series of T cell clones, derived from volunteers immunized with Plasmodium falciparum sporozoites in an effort to identify relevant epitopes in the immune response to the pre-erythrocytic stages of the parasite. CD4+ T cell clones were obtained wich specifically recognized a repetitive epitope located in the 5'repeat region of the CS protein. This epitope, when conjugated to the 3'repeat region in a synthetic MAPs construct, induced high titers of antisporozoite antibodies in C57B1 mice. A second T cell epitope, which mapped to aa 326-345 of the carboxy terminal, was recognized by lytic, as well as non-lytic, CD4+ T cells derived from the sporozoite-immunized volunteers. The demonstration of CD4+ CTL in the volunteers, and the recent studies inthe rodent model (Renia et al., 1991; Tsuji et al., 1990), suggested that CS-specific CD4+ T cells, in addition to their indirect role as helper cells in the induction of antibody and CD8 + effector cells, may also play a direct role in protection against sporozoite challenge by targeting EEF within the liver.

Due to the narrow therapeutic range of theophyline, plasma concentrations of this drug are monitored in patients undergoing chronic therapy. Slow-release preparations avoid the fluctuations in plasma levels and improve patient compliance. In this study, we have compared the pharmacokinetic profiles of a theophylline slow-release tablet and a syrup form, when administered in multiple doses to healthy adult volunteers. The classification based upon releasing patterns is confirmed.

A Phase 1 double-blind placebo-controlled study was performed to evaluate a vaccine against American tegumentary leishmaniasis in 61 healthy male volunteers. Side effects and the immune response to the vaccine were evaluated, with 1- and 2- dose schemes, with intervals of 7 or 21 days, each dose containing 1440 mg of protein N antigen of a single strain of Leishmania amazonensis (PH 8) diluted in merthiolated saline (1:10,000). Merthiolated saline and an inert substance were used as placebos. No significant clinical alterations were found following the respective injections in the vaccinated individuals as compared to the placebos, except for local pain, which was associated significantly with injection of the vaccine. The laboratory alterations we observed bore no association with the clinical findings and were unimportant. We observed no differences between the groups with regard to seroconversion or the Montenegro skin test. However, the group that received a single dose of the vaccine and the one that received two doses with a 21-day interval displayed cutaneous induration significantly larger than in the control group, with 100%, 100%, and 66% conversion in the skin test, respectively. We concluded that the vaccine does not present any major side effect that would contraindicate its use in healthy individuals.

Sexually transmitted diseases (STD) are very frequent in the whole world. Males who do not use a condom during their sexual relations are at great risk. We report cases of STD during six months of observation, among homosexual/bisexual males who participate in the Project Horizonte. There were 16 cases of genital warts, 6 cases of human immunodeficiency virus infection, 24 cases of unspecific urethritis, 28 cases of herpes simplex virus infection, 30 cases of syphilis, 58 cases of gonorrhea and 84 cases of pediculosis. We concluded that a condom must be used in all sexual relations and new counseling techniques are needed, to avoid this situation.

In the light of emerging and overlooked infectious diseases and widespread drug resistance, diagnostics have become increasingly important in supporting surveillance, disease control and outbreak management programs. In many low-income countries the diagnostic service has been a neglected part of health care, often lacking quantity and quality or even non-existing at all. High-income countries have exploited few of their advanced technical abilities for the much-needed development of low-cost, rapid diagnostic tests to improve the accuracy of diagnosis and accelerate the start of appropriate treatment. As is now also recognized by World Healt Organization, investment in the development of affordable diagnostic tools is urgently needed to further our ability to control a variety of diseases that form a major threat to humanity. The Royal Tropical Institute's Department of Biomedical Research aims to contribute to the health of people living in the tropics. To this end, its multidisciplinary group of experts focuses on the diagnosis of diseases that are major health problems in low-income countries. In partnership we develop, improve and evaluate simple and cheap diagnostic tests, and perform epidemiological studies. Moreover, we advice and support others - especially those in developing countries - in their efforts to diagnose infectious diseases.

Pediculosis seems to have afflicted humans since the most ancient times and lice have been found in several ancient human remains. Examination of the head hair and pubic hair of the artificial mummy of Ferdinand II of Aragon (1467-1496), King of Naples, revealed a double infestation with two different species of lice, Pediculus capitis, the head louse, and Pthirus pubis, the pubic louse. The hair samples were also positive for the presence of mercury, probably applied as an anti-pediculosis therapy. This is the first time that these parasites have been found in the hair of a king, demonstrating that even members of the wealthy classes in the Renaissance were subject to louse infestation.

O objetivo deste trabalho foi avaliar a intensidade e a qualidade da radiação solar disponibilizada às plantas e os seus impactos sobre a fotossíntese, rendimento e qualidade dos frutos, em macieiras 'Royal Gala', cobertas ou não com telas antigranizo nas cores branca e preta. A tela preta provocou redução maior na densidade de fluxo de fótons fotossinteticamente ativos acima do dossel das plantas (24,8%), em comparação à tela branca (21,2%). O interior do dossel das plantas sob tela preta recebeu menores valores de radiação ultravioleta, azul, verde, vermelho e vermelho distante, bem como da relação vermelho:vermelho distante, em relação às plantas descobertas. Estas alterações na quantidade e qualidade da luz sob tela preta aumentaram o teor de clorofila total e a área específica nas folhas, e reduziram a taxa fotossintética potencial, o peso de frutos por cm² de seção transversal de tronco e a coloração vermelha dos frutos. As telas antigranizo branca e preta reduziram a incidência de queimadura de sol, porém não tiveram efeito sobre a severidade de "russeting" e sobre o número de sementes por fruto.
'Royal Gala' apple quality stored under ultralow oxygen concentration and low temperature conditions

The objective of this work was to evaluate the interaction of ultralow oxygen concentrations (ULO) with storage temperatures and carbon dioxide partial pressures and its influence on fruit quality preservation and on the occurrence of physiological disorders in 'Royal Gala' apples. The experiment was carried out in a completely randomized design, with four replicates 25-fruit. ULO conditions (1.0 kPa O2 + 2.0 kPa CO2; 0.8 kPa O2 + 1.5 kPa CO2; 0.8 kPa O2 + 1.0 kPa CO2; 0.6 kPa O2 + 1.5 kPa CO2; and 0.6 kPa O2 + 1.0 kPa CO2) were tested at 0, 0.5 and 1.0°C, in a 5x3 factorial arrangement. Fruit quality and ripening analyses were performed after eight-month storage plus seven days of shelf-life at 20°C. Oxygen partial pressures below 0.8 kPa increased the occurrence of internal breakdown and mealiness. The best ULO condition was 1.0 kPa O2 + plus 2.0 kPa CO2 at 1.0°C. The interaction of ULO conditions and storage temperatures shows the need of increasing O2 partial pressure at higher storage temperatures.

O experimento foi conduzido com o objetivo de avaliar o efeito da temperatura de armazenamento e condições de atmosfera controlada sobre a manutenção da qualidade da maçã cv. Royal Gala. O delineamento experimental utilizado foi o inteiramente casualizado, arranjado em um esquema bifatorial, com 4 repetições e a amostra composta por 25 frutos. Os oito tratamentos originaram-se da combinação de dois níveis do fator temperatura (-0.5ºC e 0ºC) com quatro níveis do fator condição de atmosfera controlada (1kPa O2/2kPa CO2, 1,2kPa O2/2kPa CO2, 1kPa de O2/3kPa CO2 e 1,2kPa O2/3kPa CO2). As avaliações foram realizadas após nove meses de armazenamento, na abertura das câmaras, e após sete dias de exposição dos frutos à temperatura de 20ºC. De acordo com os resultados obtidos, as temperaturas testadas não diferiram estatisticamente com relação à acidez titulável na saída da câmara, sendo que a melhor combinação de gases foi 1,2 kPa de O2 e 2 kPa de CO2. Já após sete dias de armazenamento a 0ºC, não houve diferença estatística com relação à concentração de gases. A temperatura de 0ºC apresentou maior firmeza de polpa, menor incidência de frutos com polpa farinhenta e degenerescência da polpa após sete dias a 20ºC. A combinação de gases 1,0kPa de O2 e 2 e 3 kPa de CO2 apresentou melhor manutenção da acidez titulável e menores valores de ocorrência de podridões e distúrbios fisiológicos. Concluiu-se que a melhor temperatura de armazenamento para a cv. Royal Gala é de 0(0)C e a melhor combinação de gases é de 1kPa de O2 e 2 e 3 kPa de CO2.

Maçãs 'Royal Gala', colhidas em dois estádios de maturação, foram tratadas com 1-MCP (1-metilciclopropeno) nas concentrações de 0; 625 e 1250 nL.L-1, durante 48 horas, à temperatura de 20 ± 2 ºC. Após este tratamento, foram armazenadas em dois diferentes sistemas: em atmosfera controlada à temperatura de 0 ± 0,5 ºC, 2,1 a 2,3 kPa de CO2 , 1,8 a 2,0 kPa de O2 e umidade relativa de 95 ± 5 %, durante 9 meses. Em ar refrigerado a 0,5 ± 0,5 ºC e 95 ± 5 % de umidade relativa, durante 8 meses. As análises realizadas demonstraram a eficiência do 1-MCP em controlar a maturação das frutas durante o armazenamento em ambas as condições e pontos de colheita. A aplicação do 1-MCP reduziu drasticamente a produção de etileno, diminuiu a perda de firmeza, além de preservar a acidez total titulável.

Avaliou-se o efeito do 1-metilciclopropeno (1-MCP) no controle da maturação de maçãs 'Royal Gala'. As frutas foram colhidas em dois estádios de maturação caracterizados como: estádio de maturação 1, com frutas apresentando firmeza de polpa de 82,14 N, acidez total titulável de 0,36 % de ácido málico, teor de sólidos solúveis totais de 14,10 ºBrix e produção de etileno de 2,00 nL.g-1.h-1, e estádio de maturação 2, com maçãs apresentando firmeza de polpa de 79,96 N, acidez total titulável de 0,33 % de ácido málico, teor de sólidos solúveis totais de 13,74 ºBrix e produção de etileno de 17,11 nL.g-1.h-1. Foram testadas quatro concentrações de 1-MCP: 0; 312; 625 e 1250 nL.L-1. A aplicação foi efetuada durante 48 horas, à temperatura de 20 ± 2 ºC, em câmaras com capacidade para 300 L. Após a aplicação, as frutas foram armazenadas a 20 ± 2 ºC, por 60 dias. A cada 10 dias, avaliaram-se a firmeza de polpa, a acidez total titulável, o teor de sólidos solúveis totais e a produção de etileno. O 1-MCP reduziu a produção de etileno nas maçãs e retardou a ocorrência do pico climatérico, independentemente do estádio de maturação e da concentração do produto. Em conseqüência, preservou maiores valores de firmeza de polpa e acidez total titulável.
