Lead (Pb)-induced hypertension is characterized by an increase in reactive oxygen species (ROS) and a decrease in nitric oxide (NO). In the present study we evaluated the effect of L-arginine (NO precursor), dimercaptosuccinic acid (DMSA, a chelating agent and ROS scavenger), and the association of L-arginine/DMSA on tissue Pb mobilization and blood pressure levels in plumbism. Tissue Pb levels and blood pressure evolution were evaluated in rats exposed to: 1) Pb (750 ppm, in drinking water, for 70 days), 2) Pb plus water for 30 more days, 3) Pb plus DMSA (50 mg kg-1 day-1, po), L-arginine (0.6%, in drinking water), and the combination of L-arginine/DMSA for 30 more days, and 4) their respective matching controls. Pb exposure increased Pb levels in the blood, liver, femur, kidney and aorta. Pb levels in tissues decreased after cessation of Pb administration, except in the aorta. These levels did not reach those observed in nonintoxicated rats. All treatments mobilized Pb from the kidney, femur and liver. Pb mobilization from the aorta was only effective with the L-arginine/DMSA treatment. Blood Pb concentrations in Pb-treated groups were not different from those of the Pb/water group. Pb increased blood pressure starting from the 5th week. L-arginine and DMSA treatments (4th week) and the combination of L-arginine/DMSA (3rd and 4th weeks) decreased blood pressure levels of intoxicated rats. These levels did not reach those of nonintoxicated rats. Treatment with L-arginine/DMSA was more effective than the isolated treatments in mobilizing Pb from tissues and in reducing the blood pressure of intoxicated rats.

The microenvironment of the tumor plays an important role in facilitating cancer progression and activating dormant cancer cells. Most tumors are infiltrated with inflammatory cells which secrete cytokines such as tumor necrosis factor-a (TNF-a). To evaluate the role of TNF-a in the development of cancer we studied its effects on cell migration with a migration assay. The migrating cell number in TNF-a -treated group is about 2-fold of that of the control group. Accordingly, the expression of E-cadherin was decreased and the expression of vimentin was increased upon TNF-a treatment. These results showed that TNF-a can promote epithelial-mesenchymal transition (EMT) of MCF-7 cells. Further, we found that the expression of Snail, an important transcription factor in EMT, was increased in this process, which is inhibited by the nuclear factor kappa B (NFkB) inhibitor aspirin while not affected by the reactive oxygen species (ROS) scavenger N-acetyl cysteine. Consistently, specific inhibition of NFkB by the mutant IkBa also blocked the TNF-a-induced upregulation of Snail promoter activity. Thus, the activation of NFkB, which causes an increase in the expression of the transcription factor Snail is essential in the TNF-a-induced EMT. ROS caused by TNF-a seemed to play a minor role in the TNF-a-induced EMT of MCF-7 cells, though ROS per se can promote EMT. These findings suggest that different mechanisms might be responsible for TNF-a - and ROS-induced EMT, indicating the need for different strategies for the prevention of tumor metastasis induced by different stimuli.

This paper describes the antioxidant effects of thirteen phenolic compounds isolated from plants of the genus Lychnophora. Two assays were performed to evaluate these effects: a cellular test that measured the luminol-enhanced chemiluminescence produced by neutrophils stimulated with opsonized zymosan and a cell-free test involving horseradish peroxidase-H2O2-luminol. In both assays, the antioxidant activity of the phenolic compounds was dependent on their concentration and chemical structure. Our results suggest that the ability of phenolic compounds from Lychnophora species to scavenge and inhibit the generation of ROS may be a mechanism underlying the anti-inflammatory activity of extracts from Lychnophora spp.

The richness, diversity, abundance and prevalence of mite species associated with sigmodontine rodents of different species in Entre Ríos province, Argentina are studied. Five of the six species of mites were reported for the first time in the study area. The richness and diversity of mites was higher on Oligoryzomys flavescens and O. delticola than on Akodon azarae. Androlaelaps rotundus was dominant and exhibited higher values of mean abundance and prevalence on A. azarae, Mysolaelaps microspinosus on O. flavescens and Gigantolaelaps mattogrossensis on O. delticola.

Carcasses represent a trophic and reproductive resource or shelter for arthropods, which are a representative component of the decomposition process. Four experiments, one per season, were conducted in a semi-rural area of Bahía Blanca, Argentina, to study the trophic roles of cadaveric beetles, evaluating the abundance, composition and dominance during all decomposition stages and seasons. Species of necrophagous, necrophilous and omnivorous habits were found. Abundance, composition and dominance of beetles in relation to their trophic roles changed according to seasons and decomposition stages. Guilds and patterns of succession were established in relation to those periods. Trophic roles could be an indicator of beetle associations with decomposition stages and seasons.

Trinta amostras de solos foram selecionadas com o objetivo de caracterizar o Fator Capacidade de Fósforo (FCP) e sua relação com propriedades de solos de Entre Ríos, Argentina. Foram estimadas isotermas de adsorção e realizado um teste rápido de adsorção de P, denominado P-remanescente. As equações de Freundlich e Langmuir descreveram adequadamente o fenômeno de adsorção de P. A capacidade máxima de adsorção variou de 46 a 716 mg kg-1 de P no solo, apresentando os solos analisados de muito baixo a médio FCP. A capacidade de adsorção de P aumentou com a quantidade de argila e não foi afetada pelo teor de carbono orgânico do solo. O P-remanescente permitiu caracterizar os solos em relação ao fator capacidade e estimar sua capacidade máxima de adsorção de P.

Blood-derived products are commonly administered to horses and humans to treat many musculoskeletal diseases, due to their potential antioxidant and anti-inflammatory effects. Nevertheless, antioxidant effects have never been shown upon horse synovial fluid cells in vitro. If proved, this could give a new perspective to justify the clinical application of blood-derived products. The aim of the present study was to investigate the antioxidant effects of two blood-derived products - plasma (unconditioned blood product - UBP) and a commercial blood preparation (conditioned blood product - CBP)¹ - upon stimulated equine synovial fluid cells. Healthy tarsocrural joints (60) were tapped to obtain synovial fluid cells; these cells were pooled, processed, stimulated with lipopolysaccharide (LPS) or phorbol 12-myristate 13-acetate (PMA), and evaluated by flow cytometry for the production of reactive oxygen species (ROS). Upon addition of any blood-derived product here used - UBP and CBP - there was a significant decrease in the oxidative burst of synovial fluid cells (P<0.05). There was no difference between UBP and CBP effects. In conclusion, treatment of stimulated equine synovial cells with either UBP or CBP efficiently restored their redox equilibrium.

Recent studies have reported that exogenous gangliosides, the sialic acid-containing glycosphingolipids, are able to modulate many cellular functions. We examined the effect of micelles of mono- and trisialoganglioside GM1 and GT1b on the production of reactive oxygen species by stimulated human polymorphonuclear neutrophils using different spectroscopic methods. The results indicated that exogenous gangliosides did not influence extracellular superoxide anion (O2.-) generation by polymorphonuclear neutrophils activated by receptor-dependent formyl-methionyl-leucyl-phenylalanine. However, when neutrophils were stimulated by receptor-bypassing phorbol 12-myristate 13-acetate (PMA), gangliosides above their critical micellar concentrations prolonged the lag time preceding the production in a concentration-dependent way, without affecting total extracellular O2.- generation detected by superoxide dismutase-inhibitable cytochrome c reduction. The effect of ganglioside GT1b (100 µM) on the increase in lag time was shown to be significant by means of both superoxide dismutase-inhibitable cytochrome c reduction assay and electron paramagnetic resonance spectroscopy (P < 0.0001 and P < 0.005, respectively). The observed phenomena can be attributed to the ability of ganglioside micelles attached to the cell surface to slow down PMA uptake, thus increasing the diffusion barrier and consequently delaying membrane events responsible for PMA-stimulated O2.- production.

Pancreatic cancer is the fourth leading cause of cancer death. Gemcitabine is widely used as a chemotherapeutic agent for the treatment of pancreatic cancer, but the prognosis is still poor. Berberine, an isoquinoline alkaloid extracted from a variety of natural herbs, possesses a variety of pharmacological properties including anticancer effects. In this study, we investigated the anticancer effects of berberine and compared its use with that of gemcitabine in the pancreatic cancer cell lines PANC-1 and MIA-PaCa2. Berberine inhibited cell growth in a dose-dependent manner by inducing cell cycle arrest and apoptosis. After berberine treatment, the G1 phase of PANC-1 cells increased by 10% compared to control cells, and the G1 phase of MIA-PaCa2 cells was increased by 2%. Whereas gemcitabine exerts antiproliferation effects through S-phase arrest, our results showed that berberine inhibited proliferation by inducing G1-phase arrest. Berberine-induced apoptosis of PANC-1 and MIA-PaCa2 cells increased by 7 and 2% compared to control cells, respectively. Notably, berberine had a greater apoptotic effect in PANC-1 cells than gemcitabine. Upon treatment of PANC-1 and MIA-PaCa2 with berberine at a half-maximal inhibitory concentration (IC50), apoptosis was induced by a mechanism that involved the production of reactive oxygen species (ROS) rather than caspase 3/7 activation. Our findings showed that berberine had anti-cancer effects and may be an effective drug for pancreatic cancer chemotherapy.

Os fatores prognósticos histológicos podem colaborar para determinar a evolução desta neoplasia. OBJETIVO: Correlacionar a expressão imunoistoquímica do p53 e Ki-67 com idade, grau histológico, comprometimento de linfonodos e estadiamento patológico de pacientes com carcinomas epidermóides de laringe. MÉTODO: Foram avaliados trinta casos consecutivos de carcinomas epidermóides de laringe, submetidos à técnica de imunoistoquímica para verificar a expressão dos anticorpos p53 e Ki-67. RESULTADO: A idade média foi de 56,2 anos e a imunoexpressão dos marcadores foi mais observada no grupo com mais de 50 anos de idade, especialmente a do anticorpo Ki-67 (p=0,032). Não houve relação do p53 e Ki-67 com o comprometimento de linfonodos. O Ki-67 se expressou em 70% nos casos de alto grau histológico e 80% nos de baixo, enquanto o p53 em 70% apenas nos de alto grau. O estadiamento patológico mostrou que no grupo dos carcinomas avançados a expressão do p53 foi de 61,5%, enquanto o Ki-67 mostrou positividade para os casos precoces (100%) e avançados (73,1%). CONCLUSÃO: Não houve diferenças significantes entre a imunoexpressão do p53 e do Ki-67 em carcinomas epidermóides de laringe, exceto no grupo de pacientes com mais de 50 anos, onde a expressão do Ki-67 foi significativamente maior.

Camu camu es una fruta nativa de la Amazonía, que llama la atención por el alto contenido de vitamina C (6,116 mg/ 100 g de pulpa), está en proceso de domesticación, por lo cual se está investigando un método de propagación vegetativa que permita avanzar en el proceso de mejoramiento genético. El objetivo de este trabajo fue evaluar la capacidad rizogénica de plantas matrices promisorias de camu camu "Myrciaria dubia (Kunth) McVaugh" según el aumento del número de hojas, mediante la técnica de estacas herbáceas en cámaras de subirrigación. El ensayo fue conducido mediante un Diseño de Bloques Completamente al Azar (DBCA) con arreglo factorial 9Ax3B, con 3 repeticiones y 15 estacas por unidad experimental. El factor A, estuvo constituido por nueve plantas matrices y el factor B: pares de hojas con 3 niveles: 1; 2 y 3 pares. El enraizamiento fue evaluado después de 90 días. Se observó que existió interacción estadística significativa para las variables: porcentaje de enraizamiento, longitud y número de raíces. Para las variables porcentaje de callo y porcentaje de mortalidad se encontró efecto de la planta matriz y pares de hojas. Los resultados muestran que el porcentaje de enraizamiento estuvo influenciado por efectos intrínsecos adherentes a la variabilidad genotípica de las plantas matrices, presentando un alto grado de dispersión, que osciló entre 91,11 % y 0,00 %, mostrando una alta variabilidad y marcada influencia de la planta matriz sobre el proceso de rizogénesis, influyendo de manera altamente significativa en el enraizamiento. Con respecto al área foliar, estacas con 2 y 3 pares de hojas, independiente de la planta matriz, presentaron mayor capacidad de enraizamiento. Con base en estos resultados se concluye que el efecto de la variabilidad genotípica y el área foliar influyen de manera altamente significativa en el proceso de rizogénesis de estacas herbáceas de camu camu.

O artigo apresenta a metodologia dos Grupos de Promoção à Saúde, concebida como um instrumento capaz de contribuir com o desenvolvimento da autonomia e condições de vida e saúde. O método é identificado como uma intervenção coletiva e interdisciplinar de saúde, constituída por um processo grupal e orienta para o desenvolvimento de atitudes e comportamentos direcionados à transformação contínua do nível de saúde e condições de vida dos seus participantes. Os Grupos são identificados no contexto do Sistema Único de Saúde como uma prática que contribui com a superação do modelo biomédico. Desenvolvem ações em que a saúde é tomada em sua positividade, ao mesmo tempo em que podem servir ao atendimento da demanda de assistência a idosos nos sistemas de saúde.

OBJETIVO: Analisar o modelo de atuação de coordenadores de grupos de promoção da saúde em unidades básicas de saúde vinculadas à formação de profissionais. PROCEDIMENTOS METODOLÓGICOS: Estudo qualitativo realizado no município de Florianópolis, SC, em 2001. Foram analisados quatro grupos, totalizando 24 sessões em unidades básicas de saúde. Procedeu-se à observação participante para iniciar o trabalho de campo. Os relatos foram analisados por meio da técnica de análise do discurso do tipo enunciativo-pragmático. ANÁLISE DOS RESULTADOS: As formas de atuação dos coordenadores congruentes ao modelo preventivista foram: opressiva, pedagógica bancária, biologicista e higienista, prescritiva de condutas e culpabilizadora, infantilizadora, redutora das problemáticas coletivas, desfavorecedora do setting grupal, além de utilização de solilóquio. As formas de atuação consonantes ao Modelo da Nova Promoção foram: facilitadora da livre expressão e autonomia, comunicadora empática, construcionista, acolhedora, utilizou a escuta ativa, além de promover a superação da violência e alienação. CONCLUSÕES: Os coordenadores atuaram primordialmente por meio do modelo preventivista e sem utilizar recursos técnicos e teóricos alusivos à metodologia grupal na área da saúde. As atuações identificadas nos modelos preventivista e a nova promoção da saúde desvelam características que se fundamentam, respectivamente, nas éticas da - opressão/subordinação e; - na cooperação/aceitação dos usuários como livres e responsáveis por suas escolhas e conseqüências.

The prevalence of thermotolerant Campylobacter in diarrhoeic and healthy children as well as in dogs, hens, ducks and pigeons was determined in Southern Chile. Campylobacter were found in 34.5% of the faecal samples examined. The isolation rate of Campylobacter in diarrhoeic and healthy children was 16.3% and 6.4% respectively. Despite C.jejuni was always more frequent than C. coli, the latter was isolated with a high frequency (29%) from patients with diarrhoea. C.jejuni and C. coli biotypes I and II were found in healthy and diarrhoeic children and were predominant in all the animals species studied. This may point out towards the possible origin of strains infecting children.

Neutrophils, eosinophils and macrophages are cells that interact with invading parasites and naive hosts have been shown to have anti-parasitic activity. The initial reaction of these leukocytes is the generation of reactive oxygen species (ROS) to play in parasite expulsion. The present work was carried out to study the effect of total extract, scolex and membrane fractions from Cysticercus cellulosae on respiratory burst by pig neutrophils. Hydrogen peroxide (H2O2) production by neutrophils incubated with metacestode fractions from C. cellulosae showed an increase of: 190% (total extract), 120% (scolex) and 44% (membrane). High antioxidant catalatic activity (33%, 28%, 28% by total extract, scolex and membrane, respectively) was observed in neutrophils incubated with metacestode fractions, which could be an attempt at self-protection. Scolex and membrane fractions increased the phagocytic capacity of neutrophils (44% and 28%, respectively). On the other hand, total cysticerci did not alter the phagocytosis, possibly due to modifications in membrane function, caused by high ROS production from neutrophils in the presence of total cysticerci. Total fraction from C. cellulosae is toxic for neutrophils as shown by the decrease in phagocytic capacity, probably caused by high levels of ROS formation. The difference in toxicity of total extract, scolex and membrane fractions on neutrophils can be explained by the presence of an antigenic effect of the vesicular fluid in the total extract of C. cellulosae.
