Three isolates over 5 years from a patient with persistent relapsing mucosal leishmaniasis due to Leishmania (Viannia) braziliensis and 7 clones from one of these isolates were studied by zymodemes and scrodemes analysis. Results showed evidences of clonal phenotypic variation. Eight isoenzymes markers demonstrated clear differences on Cellulose Acetate (CA) and thin starch gel electrophoresis. Also a panel of specific monoclonal antibodies showed such differences. Our observations provide additional evidence that Leishmania (Viannia) braziliensis is composed by subpopulations of parasites with peculiar biochemical and antigenic characteristics.

Salmonella Infantis has been the second most common serovar in Argentina in the last two years, being isolated mostly from paediatric hospitalised patients. In order to determine the clonal relationship among Salmonella Infantis strains, we examined 15 isolates from paediatric patient faeces in Argentina (12 geographically related and 3 geographically non-related) by using antimicrobial susceptibility, plasmid profiling, repetitive extragenic palindromic (REP) PCR, enterobacterial repetitive intergenic consensus (ERIC) PCR, and low-frequency restriction analysis of chromosomal DNA by pulsed field gel electrophoresis (PFGE). Four Spanish strains were included as controls of clonal diversity in molecular techniques. Antibiotype and plasmid profile was not useful as epidemiological tools. PFGE and REP-PCR were able to discriminate between Argentinean and Spanish isolates of Salmonella Infantis allowing to detect genetically related strains in three different cities. This finding indicates that a possible spread of a clone of this serovar in the North-eastern Region of Argentina has taken place in 1998.

In the present study were evaluated the DNA macrorestriction profile and SCCmec types for nine multi-resistant MRSA selected. Also antimicrobial susceptibility testing by disk diffusion method was evaluated for 68 MRSA isolates against 12 antimicrobial agents. The isolates were recovered from blood culture collected from hospitalized patients in three hospitals of Porto Alegre, Brazil. PFGE and PCR for mecA and SCCmec I, II, III, IV types genes were done on selected nine isolates with susceptibility only to vancomycin, teicoplanin and linezolid. Two clone profiles, with five subtypes, were demonstrated among multi-resistant MRSA analyzed. Eight isolates showed harbor SCCmec type III and one isolate was not typeable. The knowledge of SCCmec type, clone and antimicrobial profiles among S. aureus is essential mainly to prevention and control of dissemination of the antimicrobial resistance.

Este estudo avalia a evolução da sensibilidade in vitro do Plasmodium falciparum em uma área de prospecção de ouro no Estado do Amapá no período de 1983 a 1990. Foram efetuados 75 testes para cloroquina e quinino, 74 para amodiaquina e 76 para mefloquina. Os resultados revelaram 81% de resistência à cloroquina e 27% para a amodiaquina, enquanto que para quinino e mefloquina não foram evidenciadas cepas resistentes. Contudo, para estas duas últimas drogas identificou-se uma crescente perda da sensibilidade ao longo do tempo. Aparentemente observa-se uma associação entre resistência à cloroquina e a diminuição da sensibilidade ao quinino.

Atypical P. vivax cases reported in Manaus municipality led us to detect a genetic isolate of P. vivax. Variable regions of SSUrRNA were examined from the initial time of infection and in the two recrudescences/relapses from a patient exhibiting chloroquine and primaquine resistance. A unique isolate, found at all stages of infection, suggests the presence of a clonal expansion.

The clonal structure of the Colombian strain of Trypanosoma cruzi, biodeme Type III and zymodeme 1, was analyzed in order to characterize its populations and to establish its homogeneity or heterogeneity. Seven isolated clones presented the basic characteristics of Biodeme Type III, with the same patterns of parasitemic curves, tissue tropism to skeletal muscle and myocardium, high pathogenicity with extensive necrotic-inflammatory lesions from the 20th to 30th day of infection. The parental strain and its clones C1, C3, C4 and C6, determined the higher levels of parasitemia, 20 to 30 days of infection, with high mortality rate up to 30 days (79 to 100%); clones C2, C5 and C7 presented lower levels of parasitemia, with low mortality rates (7.6 to 23%). Isoenzymic patterns, characteristic of zymodeme 1, (Z1) were similar for the parental strain and its seven clones. Results point to a phenotypic homogeneity of the clones isolated from the Colombian strain and suggest the predominance of a principal clone, responsible for the biological behavior of the parental strain and clones.

Côr das águas de Sepetiba; as pròpriamente marítimas, pela Escala de Forel e as outras côres aparentes totais feitas por comparação com escalas de SÉGUY e com o Dicionário de Côres de MAERZ & PAUL, 1950. Côr Forel nº6 mostrava no litoral cirrípedes como a Tetraclita squamosa, nas rochas; Águas com a cianofícea planctônica Trichodesmium erythraeum tinham côr de "fôlha de bananeira" ou alpine green. O verde esmeralda mais intenso no local 7, com plancto maciço da diatomácea Coscinodiscus; a parte central da Baía com 150 [quilometros quadrados], cheia de larvas de camarão Penaeus schmidti, águas de cõr de ferrugem nas superfícies das águas; os estuários côr caramelo, isabellinus e bistre. Êste levantamento preliminar serve para comparações futuras, pois a Baía de Sepetiba ainda não é poluída. Futuramente haverá um pôrto de minérios com 3.000.000 de toneladas anuais, cais do pôrto, siderúrgicas e outras indústrias que a poluirão. Várias cõres de águas poluídas da Baía de Guanabara, como águas negras, águas côr de asfalto e outras escurecidas não foram encontradas em Sepetiba. Também não foram encntradas "águas vermelhas" com plancto predominante de dinoflagelados do que já vimos na Baía de Guanabara seguidos de mortandadas de peixes. A fig. 3 mostra as curvas de transparência, e a relação entre côr e transparência é dada para as côres totais aparentes. Foi marcado o coeficiente de extinção da luz K, pelo cálculo a partir do Disco de Secchi. Havendo poluições todos êsses dados que apresentamos deverão ser alterados.

Relata-se uma atividade prospectiva para doença de Chagas em uma ampla região de projetos de colonização ao norte de Minas Gerais. Pesquisa de triatomíneos e tripanossomos e inquéritos clínico e sorológico na população revelaram presença marcante de triatomíneos na área, com mais de 60% dos domicílios infestados por Triatoma infestans ou T. sordida, com índices globais de positividade para Trypanosoma (Schizotrypanum) cruzi ao nível de 60% dos insetos capturados. Definem-se duas situações distintas segundo a domiciliação destas espécies, estando a de T. infestans seguramente ligada aos padrões precários de habitação das populações mais pobres, especialmente em Lagedinho e Gado Bravo, com altos índices de densidade domiciliar desta espécie. T. sordida predomina aparentemente ao Sul (Janaúba), sugerindo-se que a espécie esteja ainda em fase predominantemente silvestre, com focos esporádicos de domiciliação. A análise da população revela índices relativamente altos de infecção (globalmente em torno de 10%) demostrável sorologicamente, indicando-se transmissão recente através de exames positivos em pessoas de baixa idade. A aparente escassez de formas clínicas graves sugre, em princípio, que a doença de Chagas esteja, no momento, se implantando na região como doença humana, reproduzindo ali o que deve ter ocorrido em vastas extensões do país nos últimos 200 anos. Paralelamente ao interesse do estudo ecológico da interação homem-vetor-tripanossomo, a situação observada parece constituir importante modelo para situações concretas e atuais, como seja a conquista da Amazônia, inclusive e principalmente em relação às medidas de prevenção cabíveis em benefício de populações pioneiras.

To investigate the clonal diversity of Staphylococcus aureus strains isolated at João Pessoa, State of Paraíba, Brazil, digested genomic DNA were studied by pulsed-field gel electrophoresis (PFGE) in nine methicillin-resistant strains (MRSA) and three methicillin-sensitive strains (MSSA), selected among 67 isolates based on their antimicrobial susceptibility and epidemiology. The isolates were obtained between April and November 1992 from the Hospital of the Federal University of Paraíba, located in João Pessoa. Two MRSA isolates from the Oswaldo Cruz Hospital, São Paulo, Brazil, including an epidemic strain previously detected from different hospitals at the country were used as control. Five different patterns, were demonstrated by MRSA isolated in João Pessoa and these patterns were described in several epidemiologically unrelated hospitals in São Paulo. Our results suggest the interstate dissemination of a MRSA clone in João Pessoa which is similar to that described in other cities of Brazil.

After isolating three clones of Trypanasoma cruzi (Bolivia), we first characterized them according to parasitaemia, pleomorphism and virulence, and then histopathologically. The study's interest lies on the hypothesis that clonal evolution of T. cruzi has a major impact on biologically relevant properties of this parasite. Data obtained from the studies of parasitaemia, pleomorphism and virulence showed no differences between the groups studied. As a final point, the histopathological study shows us a muscular tissue tropism both in clones and in their mother strain (Bolivia). In this paper, we conclude that Bolivia strain and clones isolated from it, pertaining to the same major clone share similar biological properties.

Previous studies showed that two groups of Trypanosoma cruzi clonal genotypes named clonet 20 and clonet 39 were predominant in Triatoma infestans, the unique vector of Chagas disease in Bolivia. These groups of clones correspond to distinct genetic clusters. These clonets were detected in T. infestans and Rhodnius pictipes fecal samples before isolation and after culture by kDNA PCR (polymerase chain rreaction) and hybridization of the amplified products with clonet specific kDNA probes named 20 and 39 as previously reported. Forty eight T. infestans and three R. pictipes infected insects captured at random in different Bolivian departments were proceeded. As previously reported the direct identification of the two major clonets in fecal samples allowed the detection of abundant mixed infections: 41% in the original sample, however after culture, only 6% of mixed infections were detected. Among the 21 parasite stocks isolated from digestive tracts where mixed infections were initially detected (clonet 20 + 39) clonet 20 alone was detected in 81% of them. This result clearly showed that the culture step selected clonet 20 parasites over those belonging to clonet 39. The taxonomic status of the isolated stocks was also confirmed by isoenzyme typing, and correlation was observed between clustering topology and hybridization patterns with the probes 20 and 39.

Staphylococcus aureus is an important agent of healthcare-associated and community-acquired infections. A major characteristic of this microorganism is the ability to develop resistance to antimicrobial agents. Several molecular techniques have been applied for the characterization of S. aureus in epidemiological studies. In the present review, we discuss the application of molecular techniques for typing S. aureus strains and describe the nomenclature and evolution of epidemic clones of this important pathogen.
