The adherence of Pseudomonas fluorescens cells to nine food-processing contact surfaces was evaluated using the plate-count method. The surfaces include marble, granite, stainless steel, polyvinyl chloride, polyurethane, and silicone-coated cloth, which have been used only in a few studies concerning bacterial adherence. The number of cells adhered to the surfaces increased with contact time reaching 5.0-6.1 log CDM.cm-2 after 10 hours, which can be considered a well established adherence process. The number of adhered cells doubled in 29.5 minutes and 23.5 minutes on stainless steel and thin polyvinyl chloride-coated cloth, respectively. For the other surfaces, this value was 9.8 minutes on average. Marble, granite, thick polyvinyl-coated cloth, double-faced rugous polyurethane, and silicone-coated cloth were not different (p < 0.05) in their ability to adhere cells (CFU/cm²) after 2 and 10 hours. The surfaces that had higher percentage of similarity in the adhesion level and higher log CFU/cm² of adhered cells were double-faced rugous polyurethane, silicone-coated cloth, and granite. The surfaces showed very different microtopography characteristics when viewed using scanning electron microscopy. This experiment showed the importance of using appropriate materials for food contact during processing, which will affect the cleaning and sanitation procedures.

The effectiveness of cleaning and sanitizing procedures in controlling Staphylococcus aureus, Salmonella Enteritidis, and Pseudomonasfluorescens adhered to granite and stainless steel was evaluated. There was no significant difference (p > 0.05) in the adherence of pure cultures of these microorganisms to stainless steel. The numbers of P. fluorescens and S. Enteritidis adhered to granite were greater (p < 0.05) than the numbers of S. aureus. Additionally, the adherence of P. fluorescens was similar to the adherence of S. Enteritidis on granite surface. In a mixed culture with P. fluorescens, S aureus adhered less (p < 0.05) to stainless steel surfaces (1.31 log CFU.cm-2) than when in a pure culture (6.10 log CFU.cm-2). These results suggest that P. fluorescens inhibited the adherence of S. aureus. However, this inhibition was not observed in the adherence process for granite. There was a significant difference (p < 0.05) between the number of adhered cells before and after pre-washing for S. aureus on stainless steel and granite surfaces, and after washing with detergent for all microorganisms and surfaces. The efficiency of the cleaning plus sanitizing procedures was not significantly different (p > 0.05) between the surfaces. However, a significant difference was observed (p < 0.05) between the sanitizer solutions. Sodium hypochlorite and peracetic acid were more bactericidal (p < 0.05) than a quaternary ammonium compound. With regard to microorganisms, S. aureus was the least resistant to the sanitizers. These results show the importance of good cleaning and sanitization procedures to prevent bacterial adherence and biofilm formation.

The biofilm formation of Pseudomonas aeruginosa and Pseudomonas fluorescens on AISI 304 stainless steel in the presence of reconstituted skim milk under different temperatures was conducted, and the potential of three chemical sanitizers in removing the mono-species biofilms formed was compared. Pseudomonas aeruginosa cultivated in skim milk at 28 °C presented better growth rate (10.4 log CFU.mL-1) when compared with 3.7 and 4.2 log CFU.mL-1 for P. aeruginosa and P. fluorescens cultivated at 7 °C, respectively. Pseudomonas aeruginosa formed biofilm when cultivated at 28 °C. However, only the adhesion of P. aeruginosa and P. fluorescens was observed when incubated at 7 °C. The sodium dichloroisocyanurate was the most efficient sanitizer in the reduction of the adhered P. aeruginosa cells at 7 and 28 °C and those on the biofilm, respectively. The hydrogen peroxide was more effective in the reduction of adhered cells of P. fluorescens at 7 °C.

Para o aumento da produtividade agrícola no País, são necessários estudos para melhor aproveitamento da adubação mineral, a fim de uma produção de grãos com melhor qualidade nutricional. Este trabalho teve como objetivo avaliar o teor e o acúmulo de proteína, de cinzas e de nutrientes em grãos de milho provenientes de plantas submetidas à inoculação com Pseudomonas fluorescens e cultivadas em diferentes níveis de adubação NPK no solo. Foram testados seis tratamentos, em delineamento experimental de blocos casualizados, no esquema fatorial 3 x 2, sendo três níveis de adubação química com NPK (0,125 e 250 kg ha-1) e dois níveis de inoculante à base de P. fluorescens (com e sem), com quatro repetições, sendo instalado em Latossolo Vermelho eutroférrico, utilizando o cultivar de milho híbrido 30F35. Determinaram-se os teores e acúmulo de N, P, K, Ca, Mg, Zn, Cu, Fe, Mn, proteína e cinzas dos grãos. Os dados foram submetidos à análise de variância e as médias, comparadas pelo teste de Tukey (p < 0,05). A aplicação de P. fluorescens via inoculação incrementou os teores de P e K dos grãos de milho, independentemente dos níveis de adubação. Os teores dos nutrientes, de proteína e de cinzas dos grãos de milho não foram influenciados pelos níveis de adubação mineral, com exceção do Cu, que aumentou com a elevação dos níveis de adubação.

Oviposition attractants could be used for monitoring as well as controlling mosquitoes by attracting them to lay eggs at chosen sites. In the present study, culture filtrates of seven bacterial species were tested for their attractancy against gravid females of Culex quinquefasciatus. When their oviposition active indices (OAI) were studied, the culture filtrates of Bacillus cereus and Pseudomonas fluorescens exhibited oviposition attractancy (OAI = >0.3) at 100 ppm and the OAI were respectively 0.70 and 0.47. Culture filtrates of B. thuringiensis var. israelensis (wild type), B. t. var. israelensis (mutant) and B. sphaericus showed attractancy at 2000 ppm with OAI of respectively 0.71, 0.59 and 0.68. However, the OAI of B. megaterium as well as Azospirillum brasilense was 0.13 (at 2000 ppm), which was less than 0.3 required to be considered them as attractants. When the oviposition attractancy of the bacterial culture filtrates were compared with that of a known oviposition attractant, p-cresol (at 10 ppm), the culture filtrates of B. t. var. israelensis (wild type) and B. cereus were found to be more active than p-cresol, respectively with 64.2 and 54.3% oviposition.

Embora estudos recentes relatem a utilização de RCPC (Rizobactérias Promotoras de Crescimento de Plantas) no Brasil, raríssimos trabalhos avaliam a presença natural dessas espécies bacterianas no solo. O objetivo deste trabalho foi avaliar a ocorrência de RPCP em duas amostras de solo sob diferentes tipos de manejo, através da construção e do seqüenciamento de bibliotecas de DNA metagenômico. Utilizaram-se oligonucleotídeos específicos para amplificação da região hipervariável do espaço intergênico dos genes ribossomais 16S-23S de DNA extraído de diferentes solos, sob Eucalyptus sp. e sob mata. Os fragmentos obtidos foram inseridos em vetor e clonados. As bibliotecas geraram 495 clones, que foram seqüenciados e identificados através de comparações realizadas pelo software Blast. O solo sob Eucalyptus sp. apresentou maior número de RPCP do que sob mata. Os filos Actinobacteria e Proteobacteria eram maiores no solo sob Eucalyptus sp., estando o filo Firmicutes ausente no solo sob mata. Somente oito espécies diferentes de RPCP foram detectadas: Bacillus subtilis, Bacillus megaterium, Bradyrhizobium japonicum, Bradyrhizobium elkanii, Bradyrhizobium sp., Frankia sp., Pseudomonas fluorescens e Pseudomonas gladioli. O trabalho forneceu valiosos dados sobre a presença de RPCP em solos com espécies florestais e sua possível utilização em reflorestamentos, assim como para o melhor conhecimento desses microrganismos nos solos do Brasil.

O objetivo deste trabalho foi avaliar o efeito de rizobactérias, no crescimento de plântulas de pepino e no controle de tombamento, causado por Pythium aphanidermatum. Foram realizados em laboratório ensaios de: degradação de 1-aminociclopropano-1-carboxilato (ACC); colonização das raízes de plântulas de pepino; e pareamento de culturas. A identificação dos melhores isolados foi feita pela determinação das seqüências do gene 16S rDNA. Trinta e sete isolados, dos 165 testados, aumentaram a massa de matéria seca das plantas de pepino em até 63%. Desses, somente um isolado (N13 - Pseudomonas fluorescens) reduziu o tombamento de plântulas em 25%; 21 isolados inibiram o crescimento micelial de P. aphanidermatum, colonizaram o sistema radicular das plantas de pepino e cresceram em presença de ACC como única fonte de nitrogênio. Dos dez isolados que apresentaram resultados satisfatórios, cinco foram identificados como pertencentes aos gêneros Bacillus, quatro Pseudomonas e um Stenotrophomonas. Dos 165 isolados de rizobactérias testados, sete possuem potencial para promover o crescimento de plantas de pepino e um para controlar o tombamento causado por P. aphanidermatum.

Isolados de Pseudomonas veronii (DFs513), Bacillus spp. (DFs093 e DFs348), Bacillus cereus (DFs769), Rodhococcus fascians (DFs843 e DFs912) e Pseudomonas fluorescens (DFs831 e DFs842), selecionados para o controle de Xanthomonas axonopdis pv. phaseoli, bem como a combinação de alguns destes isolados bacterianos, foram avaliados quanto a possível influência sobre a transmissão de Colletotrichum lindemuthianum para plântulas de feijão, a partir de sementes naturalmente infectadas e/ou infestadas. Sementes de dois lotes foram microbiolizadas com suspensões dos biocontroladores, sendo que no primeiro ensaio, em rolo de papel, realizou-se a semeadura com oito repetições de 25 sementes, incubadas a 20 ± 2°C. Os percentuais de germinação das sementes e incidência do patógeno foram avaliados. Em um segundo ensaio avaliou-se a transmissão do patógeno para a planta em ensaio conduzido em substrato esterilizado, incubadas por 10 dias. Realizaram-se contagens diárias das plântulas emergidas, incidência do patógeno, massa seca das folhas e raízes. Para a antibiose in vitro, um isolado de C. lindemuthianum foi confrontado com os isolados biocontroladores. Todos os tratamentos proporcionam aumentos de massa foliar e radicular, tanto considerando o número total quanto por planta. Os isolados, DFs831, DFs842, DFs843 e DFs912 produzem antibióticos em testes in vitro. Todos os tratamentos possibilitaram a redução da transmissão de C. lindemuthianum para plantas em pelo menos um dos ensaios. Porém, o isolado DFs912 (Rhodococcus fascians) destaca-se, por apresentar em todos os experimentos reduções da transmissão que variam de 40 a 67%.

This cross-sectional study, performed in an oncology hospital in Goiania, aimed to characterize the prevalence of oral colonization and antimicrobial susceptibility of Pseudomonas spp. isolated from the saliva of healthcare workers. Microorganisms were subjected to biochemical tests, susceptibility profile, and phenotypic detection. Of 76 participants colonized with Gram negative bacilli, 12 (15.8%) harbored Pseudomonas spp. Of all isolates, P. aeruginosa (75.0%), P. stutzeri (16.7%), and P. fluorescens (8.3%), were resistant to cefoxitin, and therefore likely to be AmpC producers. The results are clinically relevant and emphasize the importance of surveillance to minimize bacterial dissemination and multiresistance.

Ecthyma gangrenosum (EG) due to Pseudomonas aeruginosa is a rare and invasive infection that can be associated with agammaglobulinemia. The cornerstone of the treatment is based on prompt recognition with appropriate antibiotic coverage and intravenous immunoglobulin. The authors report a case of EG emphasizing the clinical and therapeutic aspects of this condition.

Acanthamoeba is a “Trojan horse” of the microbial world. The aim of this study was to identify the presence of Pseudomonas as an amoeba-resistant microorganism in 12 isolates of Acanthamoeba. All isolates showed the genus Pseudomonas spp. as amoeba-resistant microorganisms. Thus, one can see that the Acanthamoeba isolates studied are hosts of Pseudomonas.

The enzymatic modification of aminoglycosides by aminoglycoside-acetyltransferases (AAC), aminoglycoside-adenyltransferases (AAD), and aminoglycoside-phosphotransferases (APH), is the most common resistance mechanism in P. aeruginosa and these enzymes can be coded on mobile genetic elements that contribute to their dispersion. One hundred and thirty seven P. aeruginosa isolates from the University Hospital, Cumana, Venezuela (HUAPA) were evaluated. Antimicrobial susceptibility was determined by the disk diffusion method and theaac, aadB and aph genes were detected by PCR. Most of the P. aeruginosa isolates (33/137) were identified from the Intensive Care Unit (ICU), mainly from discharges (96/137). The frequency of resistant P. aeruginosaisolates was found to be higher for the aminoglycosides tobramycin and amikacin (30.7 and 29.9%, respectively). Phenotype VI, resistant to these antibiotics, was the most frequent (14/49), followed by phenotype I, resistant to all the aminoglycosides tested (12/49). The aac(6´)-Ib,aphA1 and aadB genes were the most frequently detected, and the simultaneous presence of several resistance genes in the same isolate was demonstrated. Aminoglycoside resistance in isolates ofP. aeruginosa at the HUAPA is partly due to the presence of the aac(6´)-Ib, aphA1 andaadB genes, but the high rates of antimicrobial resistance suggest the existence of several mechanisms acting together. This is the first report of aminoglycoside resistance genes in Venezuela and one of the few in Latin America.
