The essential oil (EO) of Ocimum gratissimum inhibited Staphylococcus aureus at a concentration of 0.75 mg/ml. The minimal inhibitory concentrations (MICs) for Shigella flexineri, Salmonella enteritidis, Escherichia coli, Klebsiella sp., and Proteus mirabilis were at concentrations ranging from 3 to 12 >mg/ml. The endpoint was not reached for Pseudomonas aeruginosa (>=24 mg/ml). The MICs of the reference drugs used in this study were similar to those presented in other reports. The minimum bactericidal concentration of EO was within a twofold dilution of the MIC for this organism. The compound that showed antibacterial activity in the EO of O. gratissimum was identified as eugenol and structural findings were further supported by gas chromatography/mass spectra retention time data. The structure was supported by spectroscopic methods.

Cryptococcal infection had an increased incidence in last years due to the explosion of acquired immune deficiency syndrome epidemic and by using new and effective immunosuppressive agents. The currently antifungal therapies used such as amphotericin B, fluconazole, and itraconazole have certain limitations due to side effects and emergence of resistant strains. So, a permanent search to find new drugs for cryptococcosis treatment is essential. Ocimum gratissimum, plant known as alfavaca (Labiatae family), has been reported earlier with in vitro activity against some bacteria and dermatophytes. In our work, we study the in vitro activity of the ethanolic crude extract, ethyl acetate, hexane, and chloroformic fractions, essential oil, and eugenol of O. gratissimum using an agar dilution susceptibility method towards 25 isolates of Cryptococcus neoformans. All the extracts of O. gratissimum studied showed activity in vitro towards C. neoformans. Based on the minimal inhibitory concentration values the most significant results were obtained with chloroformic fraction and eugenol. It was observed that chloroformic fraction inhibited 23 isolates (92%) of C. neoformans at a concentration of 62.5 µg/ml and eugenol inhibited 4 isolates (16%) at a concentration of 0.9 µg/ml. This screening may be the basis for the study of O. gratissimum as a possible antifungal agent.

Aqueous extracts of the leaves of Ocimum gratissimum at 10, 25, 40 and 50% (w/v) concentrations induced the production of phytoalexins in soybean cotyledons and sorghum mesocotyls. The aqueous extracts also induced systemic resistance in cucumber to Colletotrichum lagenarium, reflected by reduction in disease incidence and an increase in chitinase production. Modes of action and the existence of possible elicitors of defense response in O. gratissimum leaf extracts are discussed.

We have investigated the antinociceptive effects of the essential oil of Ocimum gratissimum L. (Labiatae) (EOOG) in two classical models of pain in male Swiss mice (25-35 g), the writhing test and the formalin test. At doses of 30, 100 and 300 mg/kg (po), EOOG produced a dose-dependent inhibition (from 58.3 ± 4.4 to 40.7 ± 6.3, 36.4 ± 3.6 and 24.6 ± 3.6, respectively; N = 8-10, P<0.05) of acetic acid-induced writhing, causing up to a ~60% inhibition at the highest dose used, comparable to that obtained with indomethacin (10 mg/kg, po). At the same doses, EOOG predominantly inhibited the late (inflammatory) phase of the formalin-induced pain response (from 59.3 ± 8.3 to 40.4 ± 4.8, 23.2 ± 2.8 and 25.3 ± 5.5, respectively; N = 6, P<0.05), with a maximal reduction of ~60% of the control, although a significant reduction of the initial (neurogenic) phase was also observed at 300 mg/kg (from 62.5 ± 6.07 to 37 ± 5.9; P<0.05). On the basis of these data, we conclude that EOOG possesses interesting antinociceptive properties in the writhing and formalin tests. Due to the relatively low toxicity of EOOG, further detailed examination is strongly indicated for a better characterization of its pharmacological properties and its potential therapeutic value.

O isolamento e a identificação de antioxidantes naturais, extraÃdos de plantas, contribuem para a descoberta de novas fontes de compostos quÃmicos. A alfavaca (Ocimum gratissimum L.) é uma planta conhecida por seus aspectos medicinais e pelo uso na culinária. A atividade antioxidante do extrato bruto e do óleo essencial das folhas de alfavaca foi comprovada através do método do tiocianato férrico. A porcentagem de inibição da oxidação lipÃdica foi de 96,89% para o extrato bruto e de 92,44% para o óleo essencial, ambos na concentração de 0,02%. O extrato bruto foi purificado através da cromatografia em coluna com sÃlica-gel e fase móvel hexano:acetato de etila em diferentes proporções. Para a identificação, foram utilizadas análises espectrais (infravermelho, ressonância magnética de hidrogênio e carbono 13). A substância isolada foi o eugenol, que apresentou 86,56% de atividade antioxidante. Alguns constituintes do óleo essencial foram caracterizados por cromatografia de fase gasosa, sendo o eugenol o principal componente (53,90%). Desta forma, a alfavaca apresenta-se como uma nova fonte de eugenol e, conseqüentemente, de antioxidante natural.

A partir da formulação de quatro bebidas, duas alcoólicas e duas não alcoólicas, com e sem açúcar, respectivamente, e do extrato reconstituÃdo (alcoolatura/planta verde) de Ocimum gratisimum L. ("alfavacão", "alfavaca", "alfavaca-cravo") - Labiatae - (Lamiaceae) em diferentes concentrações (5, 15 e 30%), foi determinada, através de testes de suspensão em sistema de tubos múltiplos, a intensidade de atividade de inativação bacteriana (IINAB/bactericidia), sobre Salmonella Enteritidis (ATCC 11076), bem como a aceitabilidade/preferência sensorial por escala hedônica destes quatro produtos. Todas as formulações apresentaram atividade bactericida para Salmonella Enteritidis, diretamente proporcional à s concentrações do extrato e ao tempo de exposição da bactéria à s bebidas, destacando-se, neste sentido, a formulação não alcoólica com açúcar. Na análise sensorial, a preferência aumentou com o decréscimo da concentração de extrato de Ocimum gratissimum na formulação. A bebida não alcoólica com açúcar, na concentração de 5% de extrato, destacou-se na preferência/aceitabilidade.

To determine the repellent activity of Ocimum gratissimum volatile oil against Simulium damnosum (blackflies), a 12 month (January-December 2003) field study was conducted in three onchocerciasis endemic communities (Idomido, Obio camp, and Ikot Adaha) in Ini Local Government Area of Akwa Ibom State, Nigeria. The result revealed that topical application of 20% (v/v) concentration of the oil with liquid paraffin as a base, reduced the biting rate of S. damnosum by 90.2, 81.6, and 79.7%, in Idomido, Obiocamp, and Ikot Adaha respectively. The oil gave protection against the bite of S. damnosum for at least 3 h. A total of 710 adults S. damnosum were caught by individuals treated with Ocimum oil, as against 4296 caught by the control group. When the flies caught by the treated individuals were dissected none of them was infected with microfilariae of Onchocerca volvulus. Human-vector contact and onchocerciasis transmission could be reduced by the topical application of the volatile oil during the peak biting periods of the vector.

This work reports extraction yield and chemical characterization of the extracts obtained by high-pressure CO2 extraction of a cultivar of Ocimum basilicum L. The experiments were performed in the temperature range of 20 to 50 °C, from 100 to 250 atm of pressure. Chemical analyses were carried out by gas chromatography coupled to mass spectrometry, permitting to identify 23 compounds that were grouped into five chemical classes. Results showed that temperature and solvent density influenced positively the extraction yield. At 20 °C and 0.41 g cm-3 occurred a rise in the concentration of monoterpenes, oxygenated monoterpenes, sesquiterpenes and oxygenated sesquiterpenes.

Different concentrations of basil essential oil (Ocimum basilicum L.) (0.19; 0.38; 0.75; 1.87; 3.75 and 6.00 mg.g-1) were evaluated in relation to their antioxidant activity using the DPPH● radical methodology. From the IC50 obtained data, the concentrations of 0.19; 0.38; 0.75; 1.87; 3.75; 6.00 and 12.00 mg.mL-1 were applied directly to the product and these were sensorially evaluated by the test of control difference. The concentrations related to the highest acceptability (0.19; 0.38 and 0.75 mg.g-1) were tested for antioxidant activity in the internal part of Italian type salami - during the processing and after 30 days of storage, in terms of lipid and protein oxidation. The oxidation of lipids was determined using the method of TBARS. The method of carbonyl compounds was employed for proteins oxidation. Five different formulations of salami were elaborated: blank (without the use of antioxidant); control (using sodium eritorbate as antioxidant); and adding 0.19; 0.38 and 0.75 mg.g-1 of basil essential oil. The product was kept between 25 ºC and 18 ºC and UR between 95% and 70%, for 28 days. Analyses were carried out on the processing day and after 2, 7, 14, 21 and 28 days, and also following 30 days of storage. The basil essential oil in vitro presented an antioxidant activity of IC50 12 mg.mL-1. In the internal part of the Italian type salami the commercial antioxidant (control) and the formulation containing 0.75 mg.g-1 of basil essential oil presented antioxidant activity in relation to the lipids, but not to the proteins - during processing and storage.

Studies have been carried out in order to increase the stability of vegetable oils due to economic and health protection reasons. There is a growing interest in the addition of natural antioxidants; especially herbs and spices. For this reason, this study aimed at evaluating the antioxidant potential of the oleoresins of oregano, basil, and thyme, as well as their behavior when applied to soybean oil under various concentrations. Firstly, the antioxidant activity was determined by the β-carotene/linoleic acid system and by the quantification of total phenolic compounds. Next, different concentrations of oleoresins (500 to 3000 mg.kg-1) were added to the soybean oil, and its antioxidant potential was analyzed using the oxidative stability through a Rancimat equipment. The value of 3000 mg.kg-1 of thyme and oregano oleoresins was the concentration that presented the greatest oxidative stability to soybean oil making them a natural alternative to vegetable oil conservation.

A análise da atividade antibacteriana de óleos essenciais de ervas medicinais (Ocimum gratissimum, L., Cybopogum citratus (DC) Stapf. e Salvia officinalis, L.) foi verificada frente a 100 cepas de bactérias isoladas de indivÃduos da comunidade com diagnóstico de infecção urinária. Os microrganismos foram semeados em ágar Muller Hinton e os extratos aplicados com replicador de Steers e incubados a 37°C por 24 horas. Verificou-se que Salvia officinalis, L. apresentou ação inibitória superior à s outras ervas, tendo eficácia de 100% quando testadas em espécies de Klebsiella e Enterobacter, 96% em Escherichia coli, 83% contra Proteus mirabilis e 75% contra Morganella morganii.

Objetivou-se avaliar dois métodos de inoculação de Colletotrichum gloeosporioides em maracujá, testar a patogenicidade de diferentes isolados, o efeito fungitóxico e a composição quÃmica dos óleos essenciais das espécies medicinais alecrim-pimenta (Lippia sidoides Cham.), capim-santo [Cymbopogon citratus (D. C.) Stapf.], alfavaca-cravo (Ocimum gratissimum L.), no controle da antracnose [Colletotrichum gloeosporioides (Penz.)], associado ao estádio de maturação de frutos de maracujazeiro-amarelo. Avaliaram-se três experimentos, onde se testou a patogenicidade de seis isolados do fungo em delineamento inteiramente casualizado, com seis repetições, outro com o mesmo delineamento em esquema fatorial 2x2 (suspensão de conÃdios e disco de micélio) e frutos (verdes e maduros), com seis repetições. No tratamento com frutos, utilizou-se o delineamento inteiramente casualizado, em esquema fatorial 5x3+1, sendo cinco concentrações (0; 2; 4; 6 e 8µL mL-1) e três espécies medicinais, mais o tebuconazol, com cinco repetições. Fez-se a caracterização quÃmica dos óleos por cromatografia gasosa, com espectrometria de massas. Todos os isolados foram patogênicos. Os frutos maduros apresentaram maior diâmetro das lesões, quando inoculados com suspensão de conÃdios. O óleo de C. citratus proporcionou o menor diâmetro das lesões nos frutos, até a concentração de 6 µL mL-1. Na concentração de 8 µL mL-1, todos os óleos inibiram o desenvolvimento do fungo. O timol (30,24%), o citral (77,74%) e o eugenol (92,89%) foram componentes majoritários em L. sidoides, C. citratus e O. gratissimum, respectivamente.

Aedes aegypti L. is the major vector of dengue fever, an endemic disease in Brazil. In an effort to find effective and affordable ways to control this mosquito, the larvicidal activities of essential oils from nine plants widely found in the Northeast of Brazil were analyzed by measurement of their LC50. The essential oils were extracted by steam distillation and their chemical composition determined by GL-chromatography coupled to mass spectroscopy. The essential oils from Cymbopogon citratus and Lippia sidoides, reported in the literature to have larvicidal properties against A. aegypti, were used for activity comparison. The results show that Ocimum americanum and Ocimum gratissimum have LC50 of 67 ppm and 60 ppm respectively, compared to 63 ppm for L. sidoides and 69 ppm for C. citratus. These results suggest a potential utilization of the essential oil of these two Ocimum species for the control of A. aegypti.

Calycomyza hyptidis Spencer (Diptera, Agromyzidae): descriptions, redescriptions and first record in Ocimum basilicum (Lamiaceae) in Brazil. All phases of the leafminer Calycomyza hyptidis Spencer are for the first time described, including the larva, puparium and adult female. Illustrations are presented for male and female terminalia, mine, larva and pupa. The species is first recorded in leaves of Ocimum basilicum L. (Lamiaceae) in Brazil.

O manjericão (Ocimum basilicum L.) é uma hortaliça da famÃlia Lamiaceae, utilizada na culinária ou como matéria prima para a indústria de fármacos e óleos essenciais. Amostras de plantas de manjericão apresentando sintomas de murcha, seca de hastes e podridão de colo foram coletadas na área rural de Brazlândia (DF) durante a estação chuvosa de 2005. Outras duas amostras foram coletadas em plantas cultivadas em campo aberto e casas de vegetação na região de Ponte Alta (DF). Isolados de um fungo, identificado como Fusarium oxysporum, foram obtidos em todas as amostras. Testes de patogenicidade foram conduzidos com mudas das cultivares O. basilicum 'Dark Opal' e 'Italian Large Leaf', e de acessos das espécies O. americanum L. (manjericão de folha miúda), O. campechianum Mill. (alfavaca), Origanum manjorana L. (manjerona), Origanum vulgare L. (orégano), Mentha arvensis L. (menta), Coleus blumei Benth. (tapete), Leonorus sibiricus L. (rubim) e Leonotis nepetaefolia (L.) W.T. Aiton (cordão-de-frade). Todos os isolados fúngicos mostraram-se altamente virulentos sobre as duas cultivares de manjericão. Em O. campechianum e O. americanum os isolados causaram apenas suave escurecimento vascular e leve redução de crescimento, sendo avirulentos sobre acessos das espécies O. manjorana, O. vulgare, M. arvensis, C. blumei, L. sibiricus e L. nepetaefolia. Este conjunto de dados indicou que o agente causal da doença é o fungo F. oxysporum f. sp. basilici, constituindo-se no primeiro registro formal deste patógeno no Brasil. Os lotes de sementes utilizados nas áreas de ocorrência da doença foram submetidos a um teste de sanidade visando verificar a presença do patógeno. O fungo F. oxysporum f. sp. basilici foi detectado em quatro dos seis lotes e os isolados obtidos das sementes contaminadas mostraram similar sintomatologia e um idêntico perfil de virulência aos verificados em campo e casa de vegetação, sugerindo que as sementes representam o mais provável veÃculo de introdução e potencial disseminação deste patógeno no Brasil.
