Foi estimada a área queimada, a biomassa vegetal total acima e abaixo do solo, a formação de carvão, a eficiência de queimada e a concentração de carbono de diferentes paisagens naturais e agroecossistemas que foram atingidos pelos incêndios ocorridos durante a passagem do “El Niño” em 1997/98 no Estado de Roraima, extremo norte da Amazônia Brasileira. O objetivo foi o de calcular a emissão bruta de gases do efeito estufa liberados por combustão das diversas classes de biomassa que compõem cada tipo fitofisionômico atingido. A área total efetivamente queimada foi estimada entre 38.144-40.678 km2, sendo 11.394-13.928 km2 de florestas primárias (intactas, em pé) e, o restante, de savanas (22.583 km2), campinas / campinaranas (1.388 km2) e ambientes florestais já transformados como pastagens, área agrícolas e florestas secundárias (2.780 km2). O total de carbono afetado pelos incêndios foi de 42,558 milhões de toneladas, sendo que 19,73 milhões foram liberados por combustão, 22,33 milhões seguiram para a classe de decomposição e 0,52 milhões foram depositados nos sistemas na forma de carvão (estoque de longo prazo). A emissão bruta de gases do efeito estufa, em milhões de toneladas do gás, considerando apenas o emitido por combustão foi de 17,3 de CO2, 0,21-0,35 de CH4, 1,99-3,68 de CO, 0,001-0,003 de N,O, 0,06-0,09 de NOx e 0,25 de hidrocarbonetos não-metânicos (HCNM). O total de carbono equivalente a CO2 emitido por combustão, quando considerado o potencial de aquecimento global de cada gás em um horizonte de tempo de 100 anos utilizado pelo IPCC, foi de 6,1-7,0 milhões de toneladas.

Fascioliasis is an emerging/re-emerging vector-borne disease with the widest known distribution. Approximately 17 million people are infected around the world, being the Andean region the most affected area. There is an important necessity to develop sensitive and specific diagnostic tools to treat patients early and to avoid complications. In this paper we evaluated the immune response of infected humans against two antigenic preparations: the total soluble extract (FhTSE) and the adult worm vomit (FhAWV) in order to identify antigenic fractions specific for Fasciola hepatica. Both preparations were processed by SDS-PAGE and Western blot with human sera with fascioliasis (F), other parasitosis and healthy individuals. In the immunoblot of FhTSE, sera F recognised 16 bands with MW between eight and 110 kDa, from which those of 8, 9, 10, 38, 45 and 57 kDa were specific. In the preparation FhAWV, sera F recognised nine bands with MW from eight to 85 kDa, from which those of 8, 12, 15 and 24 kDa were specific. Some bands of cross-reaction were evident with sera from patients with other parasitoses, more frequent with the FhTSE. Bands within the MW mentioned, particularly that of eight kDa, have been shown to be specific by others, and deserve additional characterisation for their potential use in immunodiagnosis.

A proven case of human infection caused by Angiostrongylus costaricensis is reported for the first time in Venezuela. The patient was a 57-year-old female surgically operated because of signs of peritonitis with a palpable mass at the lower right quadrant of the abdomen. WBC count reported 16,600 cells/mm³, with 46% eosinophils. The tumoral aspect of ileocolic area and peritoneal lymph nodes prompted the resection of a large area of the terminal ileum, cecum, part of the ascending colon and a small part of the jejunum, where a small lesion was found. The pathology showed thickened areas of the intestinal wall with areas of hemorrhage and a perforation of the cecum. Histology showed intense eosinophil infiltration of the whole intestinal wall, granulomas with giant cells and eosinophils. Some of the granuloma surrounded round or oval eggs with content characterized by a large empty area, cells or embryo in the center, and sometimes nematode larvae. A cross section of an adult nematode worm was observed inside a branch of mesenteric artery. The intestinal affected area, the characteristics of the lesions, the presence of eggs in the submucosa with nematode larvae inside, and the observation of a nematode inside a mesenteric artery, makes sufficient criteria for the diagnosis of an infection by Angiostrongylus costaricensis.

A case of meningitis due to Staphylococcus warneri in a patient with a hyperinfection with Strongyloides stercoralis possibly associated with rituximab treatment for mantel cell lymphoma is reported for the first time in the literature. The patient was a 59-year-old woman, with a 3-year history of an apparently well controlled lymphoma after treatment with chemotherapy-immunotherapy and then immunotherapy alone, and diagnosis of strongyloidiasis. Meningitis was diagnosed by cerebrospinal fluid culture and tested with an automated plate system. The patient was successfully treated with vancomycin; although fever and productive cough persisted. Severe gastrointestinal symptoms and pneumonia developed three weeks later. Hyperinfection syndrome by S. stercoralis was diagnosed, with abundant larvae in feces and expectoration.

The first report to our knowledge, of hyperinfection by Strongyloides stercoralis (HS) and hypereosinophilia, associated to immune suppression by Rituximab (the only drug received for the last one year and 10 months), in a patient with mantle-cell lymphoma (MCL), is presented. The patient has a 3-year history of MCL, and developed two accesses of HS during 2008, including meningitis, pneumonia and presence of larvae of S. stercoralis in the lungs. We had a unique chance to look at cytotoxicity of filariform larvae in the expectoration after Ivermectin treatment, showing immobilization and death of larvae, associated with eosinophils attached to the cuticle of the parasite.

The causative agent of cholera, Vibrio cholerae, can enter into a viable but non-culturable (VBNC) state in response to unfavorable conditions. The aim of this study was to evaluate the in situ survival of V. cholerae in an aquatic environment of the Southern Caribbean Sea, and its induction and resuscitation from the VBNC state. V. cholerae non-O1, non-O139 was inoculated into diffusion chambers placed at the Cuare Wildlife Refuge, Venezuela, and monitored for plate, total and viable cells counts. At 119 days of exposure to the environment, the colony count was < 10 CFU/mL and a portion of the bacterial population entered the VBNC state. Additionally, the viability decreased two orders of magnitude and morphological changes occurred from rod to coccoid cells. Among the aquatic environmental variables, the salinity had negative correlation with the colony counts in the dry season. Resuscitation studies showed significant recovery of cell cultivability with spent media addition (p < 0.05). These results suggest that V. cholerae can persist in the VBNC state in this Caribbean environment and revert to a cultivable form under favorable conditions. The VBNC state might represent a critical step in cholera transmission in susceptible areas.

Ochocientos veinte reden nacidosfRN) de ≤2500g de la Matemidad Percy Bolandfueron examinados entre 1988 y 1989por los diferentes métodos de diagnóstico de la enfermedad de Chagas (Patologia de placenta, serologia, parasitologia y clínica) con el propósito de determinar su eflcacia y costo. El examen histopatologico permitió detectar 87 casos de infección placentaria. De este total, se observaron 43 (49%) casos RN positivos al examen parasitologico de la sangre del cordon. Este número aumento con la repetición de la prueba durante el primer mes de vida del nino, alcanzando el mismo nivel que la histopatologia. Con el examen serológico se detectaron 2 casos positivos. El signo clínico de alia especificidad de la enfermedad de Chagas en RN es la hepato-esplecnomegalia. Se discuten ventajas y desventajas en cuanto a costo y factibilidad de dos estrategias de detección de la enfermedad de Chagas congénito, la primera basada en la histopatologia y la otra en la parasitologia. Se concluye que los programas de detección de la enfermedad de Chagas no pueden ser uniformes, yaquese deben tomar en cuenta aspectos deprevalencia de la enfermedad, existencia del vector y disponibilidad de técnicas de laboratorio.

The most severe drought in tropical South America during the 20th century occurred in 1926. This extreme El Nino year is further documented anecdotally, in an update of the river stage observations at Manaus, and in annual rainfall records. The annual rainfall anomaly is an east-west dipole over tropical South America, with drought to the west over the Amazon basin whose discharge is documented at Manaus, and with a surplus to the east and including the Nordeste region of Brazil. Speculations about a role for aerosol in aggravating the drought are discussed.

Schistosomiasis mansoni endemic zone of Venezuela is located in the valleys of the north central mountain region, with an extension of 15,000 km2 and inhabited by 5.1 million persons. The disease was discovered in 1906, but an organized Control Program was not established until 1943. Its basic activity has been the control of the snail vector, but prevention of man-water contact, prevention of snail infection, treatment of infected people and sanitary instruction, have also been carried out. Prevalence has diminished from 14.7% (1943-60) to 0.9% (1981-84). At present few active foci still persist, but a low transmission rate and low morbidity makes it difficult to know the exact number of infected people, which has been estimulated to be about 50,000.

The aim of this study was to evaluate an enzyme-linked immunoassay with recombinant rhoptry protein 2 (ELISA-rROP2) for its ability to detectToxoplasma gondii ROP2-specific IgG in samples from pregnant women. The study included 236 samples that were divided into groups according to serological screening profiles for toxoplasmosis: unexposed (n = 65), probable acute infection (n = 48), possible acute infection (n = 58) and exposed to the parasite (n = 65). When an indirect immunofluorescence assay forT. gondii-specific IgG was considered as a reference test, the ELISA-rROP2 had a sensitivity of 61.8%, specificity of 62.8%, predictive positive value of 76.6% and predictive negative value of 45.4% (p = 0.0002). The ELISA-rROP2 reacted with 62.5% of the samples from pregnant women with probable acute infection and 40% of the samples from pregnant women with previous exposure (p = 0.0180). Seropositivity was observed in 50/57 (87.7%) pregnant women with possible infection. The results underscored that T. gondii rROP2 is recognised by specific IgG antibodies in both the acute and chronic phases of toxoplasmosis acquired during pregnancy. However, the sensitivity of the ELISA-rROP2 was higher in the pregnant women with probable and possible acute infections and IgM reactivity.

Este trabalho foi realizado com o objetivo de avaliar o potencial de multiplicação in vitro de sete cultivares de amoreira-preta: Brazos, Cherokee, Comanche, Ébano, Guarani, Tupy e Xavante. Utilizou-se protocolo da Embrapa Clima Temperado empregado em laboratórios comerciais. A desinfestação dos explantes foi realizada em soluções à base de álcool e hipoclorito de sódio; a cultura dos meristemas em meio semi-sólido MS com 1 mg L-1 BAP, 0,01 mg L-1 ANA e 0,1 mg L-1 AG3; a multiplicação em MS com 0,8 mg L-1 BAP; e o enraizamento em ½MS com 0,5 mg L-1 ANA, sempre a 25±4ºC, 20 µE m-2 s-1, e fotoperíodo de 16 horas. Partiu-se de 60 meristemas de cada cultivar, avaliando-se a taxa de multiplicação e os níveis de contaminação, vitrificação e oxidação durante as fases de estabelecimento (60 dias), multiplicação (seis subcultivos de 40 dias) e enraizamento in vitro (30 dias). O número estimado de plantas obtidas por meristema foi de 16.335 para 'Brazos', 24.211 para 'Cherokee', 19.778 para 'Comanche', 106.550 para 'Ébano', 14.275 para 'Guarani', 34.022 para 'Tupy' e 24.651 para 'Xavante'. A quantificação dessa variabilidade de resposta in vitro é importante para o planejamento da produção de mudas de cada cultivar nos laboratórios de micropropagação.

Objetivou-se avaliar o potencial de multiplicação in vitro de quatro das principais cultivares de framboeseira utilizadas no Brasil: Autumn Bliss, Batum, Dorman Red e Heritage. Utilizou-se protocolo empregado em laboratórios comerciais. A desinfestação dos explantes foi realizada em soluções à base de álcool e hipoclorito de sódio; a cultura dos meristemas em meio semissólido MS com 1 mg L-1 BAP, 0,01 mg L-1 ANA e 0,1 mg L-1 AG3; a multiplicação em MS com 0,8 mg L-1 BAP e 15 mg L-1 sulfato de ferro; e o enraizamento em ½MS com 0,1 mg L-1 ANA, sempre a 25±4ºC, 20 µE m-2 s-1 e fotoperíodo de 16 horas. Partiu-se de 60 meristemas de cada cultivar, avaliando-se a taxa de multiplicação e os níveis de contaminação, vitrificação e oxidação durante as fases de estabelecimento (60 dias), multiplicação (seis subcultivos de 40 dias) e enraizamento in vitro (30 dias). As cultivares de framboeseira apresentaram pronunciada variabilidade genética quanto ao potencial de multiplicação. O número estimado de mudas obtidas por meristema no sistema de micropropagação descrito foi de 56.664 para 'Autumn Bliss', 6.692 para 'Heritage', 1.942 para 'Batum' e 696 para 'Dorman Red'. A quantificação dessa variabilidade de resposta in vitro é importante para o planejamento da produção de mudas nos laboratórios de micropropagação.
