O presente trabalho objetivou avaliar a eficácia do tratamento Ovsynch associado à inseminação artificial em tempo prefixado em vacas Bos taurus e Bos indicus. Foram utilizados rebanhos das raças Holandesa, Caracu, Nelore e Mantiqueira. Também foi incluído um rebanho de vacas Gir, com problemas de fertilidade. Cada rebanho foi dividido em três grupos. O grupo 1 recebeu o tratamento Ovsynch e foi inseminado em tempo prefixado. O grupo 2 foi inseminado no cio induzido com cloprostenol. O grupo 3 foi inseminado no cio natural. As taxas de concepção e de prenhez foram determinadas por ultra-sonografia. Não existiu interação significativa das variáveis reprodutivas analisadas para rebanho, idade, período pós-parto, número de parição e presença do bezerro. A taxa de concepção não diferiu (P>0,05) entre os grupos, ao passo que a taxa de prenhez foi superior (P<0,05) nos grupos 1 e 2 em comparação ao grupo 3. No rebanho Gir, o tratamento Ovsynch não alterou a taxa de concepção nem a de prenhez. Independentemente da raça, os tratamentos Ovsynch e cloprostenol não afetam a taxa de concepção, mas melhoram a taxa de prenhez. O tratamento Ovsynch não aumenta a fertilidade de vacas com problemas reprodutivos inespecíficos.

Abstract: The objective of this work was to estimate allelic frequencies of the polymorphisms IGF2/MboII (G > T) of the insulin-like growth factor 2 (IGF2) gene, DQ499531.1:g.134A > T of the pro-melanin-concentrating hormone (PMCH) gene, and DQ667048.1:g.3290G > T of the RARrelated orphan receptor C (RORC) gene in beef cattle of different genetic groups, and to evaluate the associations between these polymorphisms and traits related to carcass composition and meat quality. Data on carcass and meat quality of 499 animals was used: of 313 Nellore (Bos indicus) and of 186 Nellore crossed with different taurine (Bos taurus) breeds. For the IGF2/MboII polymorphism, the frequencies found for the G allele were 0.231 and 0.631 for Nellore and crossed breeds, respectively. For the DQ499531.1:g.134A > T polymorphism, the allelic frequencies of A were 0.850 for Nellore and 0.905 for crossed breeds. For the DQ667048.1:g.3290G > T polymorphism, the allelic frequencies of G were 0.797 and 0.460 for Nellore and crossed breeds, respectively. The evaluated single nucleotide polymorphisms (SNPs) are not significantly associated with carcass and meat traits (rib eye area, back fat thickness, shear force, total lipids, and myofibrillar fragmentation index), suggesting little utility of the analyzed polymorphisms of the IGF2, PMHC, and RORC genes as selection markers in the studied cattle populations.

Eighteen young steers were inoculated with Toxoplasma gondii and randomly distributed into three groups of six animals each: GI, 2.5x10(5) "P" strain oocysts, GII, 5.0x10(6) "RH" strain tachyzoites, and GIII (Control). Clinical, serological and parasitemia exams were realized. Parasite investigation by bioassay and PCR was realized on semen and fragments of skeletal musculature, lymph nodes, brain, retina, spleen, liver, lung, testicle, epididymis and seminal vesicle. Blood and semen samples were collected on days -2, -1, 1, 3, 5, 7, 14 and weekly thereafter, up to postinfection day (PID) 84. The inoculated steers (GI and GII) presented hyperthermia from PID 3 to 16. Antibodies against T. gondii were detected through the indirect fluorescence antibody test (IFAT) on PID 5 (1:16) in both inoculated groups (oocysts and tachyzoites), reaching peaks of 1:4096 on PID 7. Parasitemia outbursts occurred in all infected bovines, principally from PID 7 to 28, independent of the strain and inoculate used. Bioassays revealed the presence of parasites in semen samples of animals infected with oocysts (GI) and tachyzoites (GII) on several experimental days between PID 7 and 84. Tissue parasitism by T. gondii was diagnosed by bioassay and the PCR technique in several organ and tissue fragments. These findings suggest the possibility of sexual transmission of T. gondii in the bovine species.

The episodes of diarrhea caused by neonatal bovine rotavirus group A (BoRVA) constitute one of the major health problems in the calf rearing worldwide. The main G (VP7) and P (VP4) genotypes of BoRVA strains involved in the etiology of diarrhea in calves are G6P[1], G10P[11], G6P[5], and G8P[1]. However, less frequently, other G and P genotypes have been described in BoRVA strains identified in diarrheic fecal samples of calves. This study describes the identification and molecular characterization of an emerging genotype (G6P[11]) in BoRVA strains involved in the etiology of a diarrhea outbreak in beef calves in a cattle herd of high production in extensive management system. The diarrhea outbreak, which showed high morbidity (60%) and lethality (7%) rates, occurred in calves (n= 384) Nelore (Bos indicus) up to 30-day-old from the State of Mato Grosso do Sul, Brazil. BoRVA was identified in 80% (16/20) of the fecal samples analyzed by polyacrylamide gel electrophoresis (PAGE) technique. In all PAGE-positive fecal samples were amplified products with 1,062-bp and 876-bp in the RT-PCR assays for VP7 (G type) and VP4 (VP8*) (P type) of BoRVA, respectively. The nucleotide sequence analysis of VP7 and VP4 genes of four wild-type BoRVA strains showed G6-III P[11]-III genotype/lineage. The G6P[11] genotype has been described in RVA strains of human and animal hosts, however, in calves this genotype was only identified in some cross-sectional studies and not as a single cause of diarrhea outbreaks in calves with high morbidity and lethality rates as described in this study. The monitoring of the G and P genotypes of BoRVA strains involved in diarrhea outbreaks in calves is important for both animal and public health by allowing the identification of the most frequent genotypes, the characterization of novel genotypes and to identify reassortments with genotypes described in animal and human hosts. The results of this study show the importance of the monitoring of the genotypes of BoRVA strains involved in episodes of bovine neonatal diarrhea as for characterization of frequency of occurrence and pathogenic potential of uncommon genotypes as for monitoring of the emergency of different BoRVA genotypes not included in commercial vaccines.

Amostras de contrafilé (músculo L. dorsi ) provenientes de 26 bovinos, sendo 14 Polled Hereford (HH), sete 3/4Hereford 1/4Nelore (3/4H1/4N) e cinco 5/8Hereford 3/8Nelore (5/8H3/8N), machos castrados, abatidos aos dois anos de idade, foram coletadas 24 h após o abate e analisadas quanto à atividade de calpastatina e textura, tanto no 1o dia post mortem quanto após um período de maturação de 10 dias a 2o C. A atividade de calpastatina foi determinada pelo ensaio de inibição da m-calpaína e a textura através da força de cisalhamento (Warner-Bratzler). A carne de novilhos 5/8H3/8N apresentou, no 1o dia, maiores (p<0,05) atividade de calpastatina e força de cisalhamento do que a carne de novilhos HH e 3/4H1/4N. Não foram detectadas diferenças (p>0,05) entre os grupos HH e 3/4H1/4N para as mesmas características. Após 10 dias, houve uma diferença na atividade de calpastatina, porém não significativa (p>0,05), entre o grupo 5/8H3/8N (1,57U/g) e os demais (HH=1,23U/g; 3/4H1/4N=1,35U/g), e diferença significativa entre os grupos HH e 5/8H3/8N para força de cisalhamento (3,67 e 5,00kg, respectivamente). Conclui-se que a atividade de calpastatina determinada 24 h post mortem pode ser útil para a previsão da textura da carne, maturada ou não, em programas de melhoramento genético, e que a participação crescente do genótipo Bos indicus nos rebanhos da Região Sul, a par das conhecidas vantagens zootécnicas, poderá resultar em carne de pior textura.

The sweat glands are important in thermoregulation of cattle in a warm environment as they help dissipate heat through evaporation. Studies on gland histology are important to define its secretion potential and the capacity of perspiration and heat removal. The objective of this study was to determine, by histomorphometry, glandular epithelium height, the depth of the gland, length of the glandular portion and number of glands per cm2 of the sweat glands of the three age groups of Nellore cattle. Thirty females were used in this study. They were equally divided into calves, heifers and cows. Histological sections were obtained and analyzed by digital images in Trinocular BX40 Olympus microscope coupled to an Oly - 200 camera, connected to a computer. The images were obtained with microscope with 2x, 4x, 10x and 40x magnification objectives. The measurements were performed using HL Image 97 program. The height of glandular epithelium, depth of the glands, length and density of the glandular portion per cm2 , were all analyzed. The calves showed greater height of the glandular epithelium than heifers (P = 0.0024), and cows (P = 0.0191). The depth of the gland was not influenced by age. Cows had higher length of secretory portion than heifers (P = 0.0379) and calves (P = 0.0077). Heifers had a greater number of sweat glands per cm2 of skin than cows (P = 0.023). In cattle, the height of glandular epithelium and the density decreases as animals get older. On the other hand, the length of the secretor portion increases but with no changes in the depth of the sweat glands

The objective of this work was to estimate the allele polymorphism frequencies of genes in Nellore cattle and associate them with meat quality and carcass traits. Six hundred males were genotyped for the following polymorphisms: DGAT1 (VNTR with 18 nucleotides at the promoter region); ANK1, a new polymorphism, identified and mapped here at the gene regulatory region NW_001494427.3; TCAP (AY428575.1:g.346G>A); and MYOG (NW_001501985:g.511G>C). In the association study, phenotype data of hot carcass weight, ribeye area, backfat thickness, percentage of intramuscular fat, shear force, myofibrillar fragmentation index, meat color (L*, a*, b*), and cooking losses were used. Allele B from the ANK1 gene was associated with greater redness (a*). Alleles 5R, 6R, and 7R from the DGAT1 VNTR gene were associated with increased intramuscular fat, reduced cooking losses and increased ribeye area, respectively. The single nucleotide polymorphism (SNP) of the TCAP gene was not polymorphic, and MYOG alleles were not associated with any of the evaluated characteristics. These results indicate that ANK1 and DGAT1 genes can be used in the selection of Nellore cattle for carcass and meat quality.
Hematological profile of beef cattle with divergent residual feed intake, following feed deprivation

The objective of this work was to characterize the hematological profiles of steers and bulls, according to residual feed intake (RFI), after feed deprivation. Twenty‑month‑old Nellore steers and bulls were fed feedlot diets for 70 and 56 days, respectively. RFI was calculated as the difference between actual feed intake and expected feed intake. More and less efficient steers and bulls, according to RFI, were subjected to 24 hours of food deprivation. Blood was sampled prior to and following the withdrawal period. Hematological analyses included total and differential white blood cell count, red blood cell count and morphology, and plasma glucose concentration. Variation in RFI did not influence plasma glucose concentration or blood cell profiles. Glucose concentrations in bulls decreased from 114 to 97 mg dL‑1, but remained unchanged in steers, and the neutrophil:lymphocyte ratio increased from 0.39 to 0.57 following deprivation. Hematological profiles do not differ between more and less efficient steers and bulls, according to RFI.

The objective of this work was to evaluate the effects of single-nucleotide polymorphisms (SNPs) in the genes IGF1 (AF_017143.1:g.198C>T), MSTN (AF_320998.1:g.433C>A), MYOD1 (NC_007313:g.1274A>G) and MYF5 (NC_007303:g.1911A>G) on carcass and meat traits in Nelore (Bos indicus) and Nelore x B. taurus. A total of 300 animals were genotyped and phenotyped for rib eye area (REA), backfat thickness (BT), intramuscular fat (IF), shear force (SF) and myofibrillar fragmentation index (MFI). The effects of allele substitution for each SNP were estimated by regression of the evaluated phenotypes on the number of copies of a particular allele using the general linear model. The polymorphism at IGF1 was non-informative in Nelore animals. In crossbred animals, the IGF1 C allele was associated with greater REA. However, this relation was not significant after Bonferroni correction for multiple testing. The A allele of the MSTN polymorphism was absent in Nelore cattle and was only found in two crossbred animals. The polymorphisms of MYOD1 and MYF5 were little informative in Nelore animals with G allele frequency of 0.097 and A allele frequency of 0.031, respectively. These markers show no association with the analyzed traits in the total sample of evaluated animals.

Several characteristics are important in a traceability system of animal products, such as age at slaughter, breed composition, besides information of the productive chain. In general, the certification agent records information about the animals and the system which it came from, although cannot guarantee that the slaughtering, meat processing and distribution are error proof. Besides, there is a differential price, at least at the international market, based on sex and breed composition of the animals. Genetic markers allow identification of characteristics controlled in the beef cattle traceability program, as sex and breed composition, in order to correctly identify and appraise the final product for the consumer. The hypothesis of this study was that the majority beef samples retailed in the local market originate from female with a great participation of zebu breeds. Therefore, the objective of this work was to characterize retail beef samples with DNA markers that identify cattle sex and breed composition. Within 10 beef shops localized in Pirassununga, SP, Brazil, 61 samples were collected, all were genotyped as harboring Bos taurus mitochondrial DNA and 18 were positive for the Y chromosome amplification (male). For the marker sat1711b-Msp I the frequency of the allele A was 0.278 and for the marker Lhr-Hha I the frequency of the allele T was 0.417. The results of sat1711b-Msp I and Lhr-Hha I allelic frequencies are suggestive that the proportion of indicus genome compared with the taurine genome in the market meat is smaller than the observed in the Nellore breed. The procedure described in this study identified sex and subspecies characteristics of beef meat samples, with potential application in meat products certification in special as an auxiliary tool in beef cattle traceability programs.

Este estudo avaliou o efeito do tamanho do folículo na capacidade dos ovócitos de sofrerem maturação nuclear e citoplasmática. Ovários de vacas Nelore (Bos indicus) foram coletados logo após o abate, e os complexos cúmulus-ovócitos foram aspirados de folículos de diferentes categorias, e classificados de acordo com seus diâmetros em: 1-2 mm, 3-5 mm, 6-8 mm e > ou = 9 mm. A medida do diâmetro e a fixação dos ovócitos para avaliação do estádio nuclear antes da maturação foram feitas logo após a aspiração. Ovócitos morfologicamente viáveis foram maturados, fecundados e cultivados in vitro. Os ovócitos obtidos de folículos de 1-2 mm apresentaram menor diâmetro (P<0,01) do que os dos demais grupos. Antes da maturação, 89,8%, 90,1%, 85,7% e 100% dos ovócitos provenientes de folículos de 1-2, 3-5, 6-8 e > ou = 9 mm de diâmetro, respectivamente, apresentavam vesícula germinativa. O tamanho do folículo não influenciou (P<0,05) a taxa de maturação nuclear. A porcentagem de ovócitos que chegaram a metáfase II foi de 88,8%, 87,8%, 92,9% e 100% no que se refere aos ovócitos de folículos de 1-2, 3-5, 6-8 e > ou = 9 mm, respectivamente. As taxas de penetração e clivagem foram semelhantes (P>0,05) nos grupos de 1-2 mm (93,4% e 81,9%), 3-5 mm (90,7% e 79,6%), 6-8 mm (91,3% e 77,8%) e > ou = 9 mm (92,0% e 78,3%). Da mesma forma, não houve diferenças (P>0,05) entre as categorias de folículos no que se refere às taxas de polispermia, descondensação da cabeça do espermatozóide e formação de pro-núcleos. Os resultados deste estudo demonstram que na espécie B. indicus, folículos com diâmetro de 1 mm até > ou = 9 mm não influenciaram a capacidade dos ovócitos de reiniciar e completar a meiose e de clivar após maturação e fecundação in vitro.

A determinação da absorção real de fósforo (P) em bovinos deve levar em consideração a fração endógena mínina do mineral, que se perde nas fezes. De modo geral esses cálculos são feitos utilizando-se tabelas cujos valores foram obtidos em outros países, com outras raças de animais e em condições bem diferentes das condições brasileiras. O trabalho teve como objetivo determinar a perda endógena de P nas fezes e estimar a exigência mínima de P em novilhos da raça Nelore (Bos indicus). Foram utilizados 18 novilhos castrados, com peso médio de 190,82±27,53 kg e idade aproximada de 12 meses, divididos em três grupos de seis animais, e mantidos em gaiolas metabólicas individuais. Os animais receberam dieta básica constituída de feno de Brachiaria decumbens e uma mistura de concentrados durante os 30 dias de período experimental. Os tratamentos consistiram de diferentes quantidades de fosfato bicálcico em níveis de 0,12, 0,24 e 0,36% de P, com base na dieta total. Foram aplicadas injeções de 32P nos animais para a determinação da perda endógena fecal de P. A perda endógena mínima fecal de P para uma ingestão zero do mineral, calculada por interpolação, foi de 5,72 mg/kg de peso vivo e para um balanço zero, o requerimento mínimo foi de 8,84 mg/kg de peso vivo por dia.

The aim of this study was to investigate the follicular dynamics during estrous cycle in Gir breed (Bos indicus) cows. Follicular growth and atresia during estrous cycle were evaluated using a portable ultrasound device. Luteal activity was evaluated by serum progesterone levels. Cycles with two (6.67%), three (60.00%), four (26.67%) and five (6.67%) follicular waves were observed. There was no difference (P>0.05) in dominant or subordinate follicles growth or atresia rates among follicular waves. The maximum diameter of the ovulatory follicle was higher than the diameter of the other dominant follicles in cycles with four waves, and higher than the diameter of the second dominant follicle in cycles with three waves (P<0.05). There was no difference (P>0.05) in estrous cycle length (21.11±1.76 and 22.25±1.71 days) or progesterone levels during diestrous (4.48±1.45 and 5.08±1.40 ng/mL) between cycles with three or four waves. Follicular dynamics in Gir cattle is characterized by a higher incidence of cycles with three or four waves, associated with a low persistence of the dominant follicle.

Avaliou-se a digestibilidade dos nutrientes de uma ração completa fornecida a bovinos de diferentes grupos genéticos. A digestibilidade foi estimada pela Lignina em Detergente Ácido, em delineamento de blocos ao acaso, com repetição no tempo. Foram efetuadas análises químicas da ração e das fezes, a cada 28 dias. De cada grupo genético foram colhidas fezes de 16 animais, no mínimo, a cada 28 dias. Observaram-se diferenças no aproveitamento dos nutrientes, destacando-se o Guzerá em todas as variáveis, com exceção da proteína bruta e da fibra em detergente ácido. Por outro lado, o Caracu mostrou-se menos eficiente na digestibilidade dos nutrientes, com exceção da proteína bruta e da fibra em detergente ácido.

Avaliou-se o efeito de concentrações espermáticas e períodos de incubação na fecundação in vitro com sêmen de touros Gir (Bos indicus). Oócitos maturados in vitro foram fecundados com sêmen de dois touros, nas concentrações de 1, 2 ou 4x10(6) espermatozóides/mL, por 12 ou 18 horas, e avaliada a taxa de fecundação e polispermia. Posteriormente, foi avaliada a taxa de clivagem com 72 horas e de blastocisto no oitavo dia pósfecundação. Um segundo experimento verificou o efeito de 1, 2 e 4x10(6) espermatozóides/mL por 18 horas, utilizando sêmen de três touros separadamente, bem como o efeito de 12 e 18 horas de incubação, na concentração de 1 a 2x10(6) espermatozóides/mL. Observou-se maior (P<0,05) polispermia na concentração de 4x10(6) espermatozóides/mL e que a associação dessa concentração com 18 horas aumentou a taxa de clivagem (P<0,05). No segundo experimento, o aumento do período de incubação melhorou (P<0,05) a taxa de clivagem para um dos touros e, quando comparados entre si, os animais produziram taxa de clivagem e de blastocistos diferentes (P<0,05) em uma mesma concentração ou período de incubação. Os resultados revelaram que a polispermia é o principal efeito do aumento da concentração espermática e que a eficiência entre touros na fecundação in vitro é dependente da concentração e período de incubação.
