Hepatitis C virus (HCV) is a major cause of liver disease throughout the world. The NS5A and E2 proteins of HCV genotype 1 were reported to inhibit the double-stranded (ds) RNA-dependent protein kinase (PKR), which is involved in the cellular antiviral response induced by interferon (IFN). The response to IFN therapy is quite different between genotypes, with response rates among patients infected with types 2 and 3 that are two-three-fold higher than in patients infected with type 1. Interestingly, a significant percentage of HCV genotype 3-infected patients do not respond to treatment at all. The aim of this paper was to analyse the sequences of fragments of the E2 and NS5A regions from 33 outpatients infected with genotype 3a, including patients that have responded (SVR) or not responded (NR) to treatment. HCV RNA was extracted and amplified with specific primers for the NS5A and E2 regions and the PCR products were then sequenced. The sequences obtained covered amino acids (aa) 636-708 in E2 and in NS5A [including the IFN sensitivity determining region (ISDR), PKR-binding domain and extended V3 region)]. In the E2 and NS5A regions, we did observe aa changes among patients, but these changes were not statistically significant between the SVR and NR groups. In conclusion, our results suggest that the ISDR domain is not predictive of treatment success in patients infected with HCV genotype 3a.

The objective of this study is to understand the structural flexibility and curvature of the E2 protein of human papillomavirus type 18 using molecular dynamics (6 ns). E2 is required for viral DNA replication and its disruption could be an anti-viral strategy. E2 is a dimer, with each monomer folding into a stable open-faced β-sandwich. We calculated the mobility of the E2 dimer and found that it was asymmetric. These different mobilities of E2 monomers suggest that drugs or vaccines could be targeted to the interface between the two monomers.

Determination of the ApoE allele distribution in five South American Amerindian tribes revealed absence of the ApoE2 allele, accompanied by high ApoE3 and low ApoE4 allele frequencies for most tribes, a distribution only previously reported for the Inuit Eskimo from Greenland.

Two recombinant baculoviruses were produced in order to obtain a bovine viral diarrhea virus (BVDV) immunogen: AcNPV/E2 expressing E2 glycoprotein, and AcNPV/E0E1E2 expressing the polyprotein region coding for the three structural proteins of BVDV (E0, E1, and E2). Mice were immunized with Sf9 cells infected with the recombinant baculoviruses in a water in oil formulation and the production of neutralizing antibodies was evaluated. Since E2 elicited higher neutralizing antibody titers than E0-E1-E2 polyprotein, it was selected to immunize cattle. Calves received two doses of recombinant E2 vaccine and were challenged with homologous BVDV 37 days later. The recombinant immunogen induced neutralizing titers which showed a mean value of 1.5 ± 0.27 on the day of challenge and reached a top value of 3.36 ± 0.36, 47 days later (84 days post-vaccination). On the other hand, sera from animals which received mock-infected Sf9 cells did not show neutralizing activity until 25 days post-challenge (62 days post-vaccination), suggesting that these antibodies were produced as a consequence of BVDV challenge. Even when no total protection was observed in cattle, in vitro viral neutralization assays revealed that the recombinant immunogen was able to induce neutralizing antibody synthesis against the homologous strain as well as against heterologous strains in a very efficient way.

Utilizando 5 partidas de S. mamsoni adultos, obtidos pela infecção de cobaios e camundongos, obtiveram-se três extratos antigênicos (E1, E2 e E3). Estes extratos, em contacto com soro de doentes esquistossomóticos, forneceram as linhas de precipitação da Fig. 1 pelo processo de imunoeletroosmoforese.

ABSTRACTINTRODUCTION:In this study, the molecular characteristics of group A rotavirus (RVA) were compared in samples obtained before and after RVA vaccine-introduction in Brazil.METHODS:Eighty samples were screened for the presence of RVA. Positive samples were molecularly analyzed.RESULTS:RVA positivity was 16.9%, with a predominance of G2P[4]. Periods: pre-vaccination: predominance of IId (G1), IId (G2) lineages, and I1 and E1 genotypes; post-vaccination: predominance of Ib (G1), IIa, and IIc (G2) lineages and I2 and E2 genotypes.CONCLUSIONS:Although changes in RVA-circulation pattern were observed in the post-vaccination period, it could not be attributed to vaccination process.

Esta pesquisa teve como objetivo caracterizar a reprodução e o perfil hormonal dos esteróides sexuais do pirarucu, Arapaima gigas, além de validar um aspecto do dimorfismo sexual secundário. O experimento foi realizado na Fazenda Santo Antônio II, em Presidente Figueredo-AM, no período de fevereiro de 2004 a fevereiro de 2005. Foram selecionados 24 exemplares e divididos em três grupos de oito peixes (grupo "A", "B" e "C" com pirarucus de mais de quatro anos de idade, com três anos e dois anos, respectivamente). Os pirarucus foram estocados em três tanques de 400m², um para cada grupo. Aspectos comportamentais, como o interesse por alimento, brigas, formação de casais e presença de ninhos, foram observados ao longo de todo o experimento. Amostras de sangue foram coletadas mensalmente, de todos os peixes, observou-se a presença de uma mancha alaranjada no macho como uma característica sexual secundária. Foram mensurados os níveis de testosterona (T), 17beta-estradiol (E2) e 17alfa-hidroxi-progesterona (17aOHP) através de radioimunoensaio (RIA) de fase sólida. Todos os machos dos grupos "A" e "B" puderam ser identificados pela mancha alaranjada na região inferior da cabeça, corroborada pela concentração de testosterona, comprovando o conhecimento empírico dos ribeirinhos. Os níveis de T e E2 nos peixes do grupo "A" e "B" tiveram maiores picos no início do período de chuva, enquanto que o 17aOHP teve oscilações constantes e pequenos picos no final do período chuvoso, os peixes do grupo "C" acompanharam essa tendência em concentrações menores.

OBJECTIVE: To establish the allelic and genotypic frequencies related to apolipoprotein E (ApoE) polymorphism and association of the genotypes with risk factors and cardiovascular morbidity in an elderly population with longevity. METHODS: We analyzed 70 elderly patients aged 80 years or more who were part of the Projeto Veranópolis. We used the gene amplification technique through the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and cleavage with the restriction enzyme Hha I to identify the ApoE genotypes. The most frequent genotypes were compared considering biological variables and cardiovascular risks and morbidity. RESULTS: The frequencies of the E2, E3, and E4 alleles were 0.05, 0.84, and 0.11, respectively, and of the genotypes were as follows: E3E3 (0.70), E3E4 (0.22), E2E3 (0.06), and E2E2 (0.02). Individuals with the E3E4 had a mean age greater than those with the E3E3. No association was observed between the genotypes and the variables analyzed, except for obesity, which was associated with the E3E3 genotype. Individuals with the E3E4 genotype had high levels of LDL-cholesterol and fibrinogen as compared with those with the E3E3 genotype. CONCLUSION: The results suggest that the E4E4 genotype may be associated with early mortality. A balance between the protective or neutral factors and the cardiovascular risk factors may occur among the individuals with different genotypes, attenuating the negative effects of the E4 allele.
Gravidade da lesão angiográfica coronariana e polimorfismo da APOE nas síndromes coronarianas agudas

FUNDAMENTO: Existem evidências de associação entre o polimorfismo da apolipoproteína E (APOE) e a doença coronariana, entretanto há controvérsias. OBJETIVO: Avaliar a associação entre o número de vasos coronarianos acometidos por obstrução significativa definida por angiografia, o polimorfismo da APOE e as variáveis clínicas. MÉTODOS: Estudo transversal multicêntrico que envolveu 207 pacientes (138 homens) com síndrome coronariana aguda (SCA) em Niterói (RJ - Brasil), os quais realizaram angiografia coronariana e determinação do genótipo para o polimorfismo APOE *2*3*4, pelo método de Restriction Fragment Length Polymorphism (RFLP). RESULTADOS: A frequência dos alelos APOE *2 foi de 6,8%, *3 foi de 82,5%, e *4 foi de 10,7%. Quanto ao número de vasos lesados, 27% dos pacientes apresentavam obstrução uniarterial, 33,8%, biarterial, e 39,1%, triarterial ou de tronco da coronária esquerda. O grau de lesão multivascular não se relacionou com a presença do alelo *4 (p = 0,78), mas com a idade > 55 anos (p = 0,025), o ex-tabagismo (p = 0,004) e a dislipidemia (p = 0,05) na análise multivariada e com doença arterial coronariana prévia (p = 0,05), diabete (p = 0,038) e síndrome metabólica (p = 0,021) na análise univariada. A prevalência de dislipidemia, diabete e hipertensão arterial sistêmica (HAS) foi elevada em relação a estudos semelhantes, com aumento progressivo da prevalência de HAS (p = 0,59) e de diabete (p = 0,06), de acordo com o número de vasos lesados. CONCLUSÃO: O polimorfismo da APOE não se associou ao número de vasos coronarianos com obstrução significativa em qualquer faixa etária. Por outro lado, a idade > 55 anos, o ex-tabagismo e a dislipidemia associaram-se à lesão multivascular.
Pressão arterial obtida pelos métodos oscilométrico e auscultatório antes e após exercício em idosos

FUNDAMENTO: Diferentes métodos de medida da pressão arterial (PA) têm sido utilizados em avaliações clínicas e científicas. Entretanto, os métodos empregados apresentam limitações e particularidades a serem consideradas. OBJETIVO: Avaliar se valores semelhantes de PA são obtidos em idosos hipertensos submetidos ao exercício resistido, ao usarem-se os métodos oscilométrico (Omron-HEM-431) e auscultatório (esfigmomanômetro de mercúrio). MÉTODOS: Dezesseis idosos hipertensos realizaram três sessões experimentais randomizadas com diferentes volumes: as sessões controle (C: 40 minutos), exercício 1 (E1: 20 minutos) e exercício 2 (E2: 40 minutos). A PA foi medida simultaneamente pelos dois métodos, a cada 5 minutos durante 20 minutos antes das sessões e durante 60 minutos após as mesmas. RESULTADOS: No período pré-intervenção houve boa concordância entre as medidas da pressão arterial sistólica (PAS) e diastólica (PAD) obtidas pelos dois métodos, havendo também elevada concordância geral após as sessões (Coeficiente de Lin = 0,82 e 0,81, respectivamente). Houve melhor concordância da PAD após a sessão controle do que após as sessões de exercício. A diferença entre as medidas obtidas entre os dois métodos foi maior para a PAD do que para a PAS após todas as sessões (p < 0.001). Independentemente do método, pode-se verificar a queda da PAS e da PAD que ocorreram nos primeiros 60 minutos após a realização dos exercícios. CONCLUSÃO: Os métodos auscultatório e oscilométrico foram concordantes antes e após as sessões controle e de exercícios, havendo, no entanto, maiores diferenças da PAD do que da PAS, sendo esta última muito semelhante entre métodos.

Background: When performing the Valsalva maneuver (VM), adults and preadolescents produce the same expiratory resistance values. Objective: To analyze heart rate (HR) in preadolescents performing VM, and propose a new method for selecting expiratory resistance. Method: The maximal expiratory pressure (MEP) was measured in 45 sedentary children aged 9-12 years who subsequently performed VM for 20 s using an expiratory pressure of 60%, 70%, or 80% of MEP. HR was measured before, during, and after VM. These procedures were repeated 30 days later, and the data collected in the sessions (E1, E2) were analyzed and compared in periods before, during (0-10 and 10-20 s), and after VM using nonparametric tests. Results: All 45 participants adequately performed VM in E1 and E2 at 60% of MEP. However, only 38 (84.4%) and 25 (55.5%) of the participants performed the maneuver at 70% and 80% of MEP, respectively. The HR delta measured during 0-10 s and 10-20 s significantly increased as the expiratory effort increased, indicating an effective cardiac autonomic response during VM. However, our findings suggest the VM should not be performed at these intensities. Conclusion: HR increased with all effort intensities tested during VM. However, 60% of MEP was the only level of expiratory resistance that all participants could use to perform VM. Therefore, 60% of MEP may be the optimal expiratory resistance that should be used in clinical practice.

Numa pesquisa efetuada em gânglios linfáticos de 3.674 suínos, aparentemente normais, e abatidos no Matadouro Municipal de Salvador e no Frigorífico Frimisa, isolaram-se 503 amostras de Salmonellas, sendo 461 ou 91,65% provenientes de gânglios crurais e pré-crurais e 42 culturas restantes, ou sejam 8,35% obtidas de outros gânglios. o estudo das 503 amostras, representando 13,69% de isolamentos no total dos suínos examinados, mostrou a existência de 52 sorotipos diferentes com dominância do grupo sorológico somático E1 em 48,70% das salmonellas seguindo-se o grupo B com 27,43%, C1 com 6,16%, L com 4,97%, C2 com 4,77% e D1 com 3,38%. Em menor percentagem registraram-se os grupos G1, N1, J, E2, e3 e E4.

The infection pattern in Swiss mice and Triatomine bugs (Rhodnius neglectus) of eleven clones and the original stock of a Trypanosoma cruzi isolate, derived from a naturally infected Didelphis marsupialis, were biochemically and biologically characterized. The clones and the original isolate were in the same zymodeme (Z1) except that two clones were found to be in zymodeme 2 when tested with G6PDH. Although infective, neither the original isolate nor the clones were highly virulent for the mice and lesions were only observed in mice infected with the original stock and one of the clones (F8). All clones and the original isolate infected bugs well while only the original isolate and clones E2 and F3 yielded high metacyclogenesis rates. An observed correlation between absence of lesions in the mammal host and high metacyclogenesis rates in the invertebrate host suggest a evolutionary trade off i.e. a fitness increase in one trait which is accompanied by a fitness reduction in a different one. Our results suggest that in a species as heterogeneous as T. cruzi, a cooperation effect among the subpopulations should be considered.

This study was designed to assess the effect of GB virus (GBV)-C on the immune response to human immunodeficiency virus (HIV) in chronically HIV-infected and HIV- hepatitis C virus (HCV)-co-infected patients undergoing antiretroviral therapy. A cohort of 159 HIV-seropositive patients, of whom 52 were HCV-co-infected, was included. Epidemiological data were collected and virological and immunological markers, including the production of interferon gamma (IFN-γ) and interleukin (IL)-2 by CD4, CD8 and Tγδ cells and the expression of the activation marker, CD38, were assessed. A total of 65 patients (40.8%) presented markers of GBV-C infection. The presence of GBV-C did not influence HIV and HCV replication or TCD4 and TCD8 cell counts. Immune responses, defined by IFN-γ and IL-2 production and CD38 expression did not differ among the groups. Our results suggest that neither GBV-C viremia nor the presence of E2 antibodies influence HIV and HCV viral replication or CD4 T cell counts in chronically infected patients. Furthermore, GBV-C did not influence cytokine production or CD38-driven immune activation among these patients. Although our results do not exclude a protective effect of GBV-C in early HIV disease, they demonstrate that this effect may not be present in chronically infected patients, who represent the majority of patients in outpatient clinics.

The hepatitis C virus (HCV) encodes approximately 10 different structural and non-structural proteins, including the envelope glycoprotein 2 (E2). HCV proteins, especially the envelope proteins, bind to cell receptors and can damage tissues. Endothelial inflammation is the most important determinant of fibrosis progression and, consequently, cirrhosis. The aim of this study was to evaluate and compare the inflammatory response of endothelial cells to two recombinant forms of the HCV E2 protein produced in different expression systems (Escherichia coli and Pichia pastoris). We observed the induction of cell death and the production of nitric oxide, hydrogen peroxide, interleukin-8 and vascular endothelial growth factor A in human umbilical vein endothelial cells (HUVECs) stimulated by the two recombinant E2 proteins. The E2-induced apoptosis of HUVECs was confirmed using the molecular marker PARP. The apoptosis rescue observed when the antioxidant N-acetylcysteine was used suggests that reactive oxygen species are involved in E2-induced apoptosis. We propose that these proteins are involved in the chronic inflammation caused by HCV.
