Specimens of Fissurella crassa (Archaeogastropoda) from Ilo, southern Perú, are infected with the adult stage of the digenetic trematode Proctoeces lintoni (Fellodistomidae). The histopatological analysis of the male and female gonads show a strong effect of the parasite on the structure and function of these organs. P. lintoni live unencysted in the gonads, and the main mechanical damage is originated by the action of a well developed acetabulum. Chemical actions of parasitic secretions may also be involved. The infected gonads show altered structure and the gametogenic processes is aborted. There is no evidence of hemocytic response, but leucocite infiltration is evident at least in male infected gonads. An increased content of polysaccarides is evident in infected gonads.

We evaluated the potential antitubercular activity of triterpenes obtained from leaves and bark of Byrsonima crassa. From chloroform extracts of the leaves, by bioassay-guided fractionation, we obtained mixtures of known triterpenes: α-amyrin, β-amyrin and their acetates, lupeol, oleanolic acid, ursolic acid and α-amyrinone. Tested against Mycobacterium tuberculosis, the triterpenes exhibited minimum inhibitory concentrations (MICs) of 31.25 - 312.25 µg/mL. β-amyrin and friedelin, isolated from the chloroform extract of bark, showed MICs of 312.25 and 125 µg/mL respectively. This is the first report of the identification and determination of the activity of B. crassa triterpenes against M. tuberculosis.

Six bromotyrosine-derived compounds were isolated from the Caribbean marine sponge Aiolochroia crassa: 3-bromo-5-hydroxy-O-methyltyrosine (1), 3-bromo-N,N,N-trimethyltyrosinium (2), 3-bromo-N,N,N,O-tetramethyltyrosinium (3), 3,5-dibromo-N,N,N-trimethyltyrosinium (4), 3,5-dibromo-N,N,N,O-tetramethyltyrosinium (5), and aeroplysinin-1 (6). Structural determination was performed using NMR, MS and comparison with literature data. All isolated compounds were screened for their in vitro activity against Leishmania panamensis, Plasmodium falciparum and Trypanosoma cruzi. Compound 4 showed selective antiparasitic activity against Leishmania and Plasmodium parasites. This is the first report of compounds 1, 4 and 5 in the sponge A. crassa and the first biological activity reports for compounds 2-4. This work shows that bromotyrosines are potential antiparasitic agents.

A constitutive alkaline phosphatase was purified to apparent homogeneity as determined by polyacrylamide gel electrophoresis from mycelia of the wild strain 74A of the mold Neurospora crassa, after growth on acetate and in the presence of saturating amounts of inorganic phosphate (Pi) for 72 h at 30ºC. The molecular mass was 58 kDa and 56 kDa as determined by exclusion chromatography and SDS-PAGE, respectively. This monomeric enzyme shows an apparent optimum pH ranging from 9.5 to 10.5 and Michaelis kinetics for the hydrolysis of p-nitrophenyl phosphate (the Km and Hill coefficient values were 0.35 mM and 1.01, respectively), alpha-naphthyl phosphate (the Km and Hill coefficient values were 0.44 mM and 0.97, respectively), ß-glycerol phosphate (the Km and Hill coefficient values were 2.46 mM and 1.01, respectively) and L-histidinol phosphate (the Km and Hill coefficient values were 0.47 mM and 0.94, respectively) at pH 8.9. The purified enzyme is activated by Mg2+, Zn2+ and Tris-HCl buffer, and is inhibited by Be2+, histidine and EDTA. Also, 0.3 M Tris-HCl buffer protected the purified enzyme against heat inactivation at 70ºC(half-life of 19.0 min, k = 0.036 min-1) as compared to 0.3 M CHES (half-life of 2.3 min, k = 0.392 min-1) in the same experiment.

Molecular characterization of Paracoccidioides brasiliensis variant strains that had been preserved under mineral oil for decades was carried out by random amplified polymorphic DNA analysis (RAPD). On P. brasiliensis variants in the transitional phase and strains with typical morphology, RAPD produced reproducible polymorphic amplification products that differentiated them. A dendrogram based on the generated RAPD patterns placed the 14 P. brasiliensis strains into five groups with similarity coefficients of 72%. A high correlation between the genotypic and phenotypic characteristics of the strains was observed. A 750 bp-RAPD fragment found only in the wild-type phenotype strains was cloned and sequenced. Genetic similarity analysis using BLASTx suggested that this RAPD marker represents a putative domain of a hypothetical flavin-binding monooxygenase (FMO)-like protein of Neurospora crassa.

Revision of the Neotropical species of Empididae (Diptera) described by Mario Bezzi. IX. The species described in Atrichopleura Bezzi and the description of a new species. Four Neotropical species of Empididae described by Bezzi in Atrichopleura Bezzi, 1909 are redescribed: A. crassa (lectotype, pres. desig.), A. hirtipes (lectotype, pres. desig.), A. nitida (lectotype, pres. desig.), A. schnusei (lectotype, pres. desig.). Atrichopleura bezzii sp. nov. is described from among syntypes of A. schnusei.

Este trabalho objetivou investigar a di¢mica populacional de 12 espécies lenhosas de Cerrado sentido restrito. O estudo foi executado nas áreas de reserva legal (fragmentos 1 e 2) do Projeto de Colonização Gerais de Balsas, no sul do Maranhão, no período de 1995 a 2002. A maioria das espécies estudadas apresentou distribuição de freqüência dos indivíduos nas classes de diâmetro em forma de J invertido, característica de populações auto-regenerativas. As populações das espécies Byrsonima coccolobifolia, Sclerolobium paniculatum e Vochysia rufa, no fragmento 1, e B. coccolobifolia, Byrsonima crassa, Davilla elliptica e Qualea parviflora, no fragmento 2, destacaram-se por apresentar altas taxas de recrutamento que compensaram as elevadas taxas de mortalidade. Os maiores valores de incremento em diâmetro foram registrados nas espécies B. crassa, Q. parviflora, S. paniculatum e V. rufa, em ambos os fragmentos. As espécies que apresentaram alto recrutamento e alto incremento em diâmetro provavelmente permanecerão ocupando posição de destaque na estrutura da comunidade. No entanto, as populações das espécies Connarus suberosus, D. elliptica, Hirtella ciliata e Erythroxylum deciduum, no fragmento 1, e Salvertia convallariaeodora, no fragmento 2, £o mostraram altas taxas de recrutamento e podem ter sua sobrevivência comprometida no futuro, caso as tendências detectadas neste estudo permaneçam.

A composição vegetal, estrutura, diversidade, estádio sucessional e distribuição de espécies do cerrado do campus da UEG com 6 ha, foram inventariados usando 30 parcelas com 100 m² cada. A partir de dbs superior ou igual a 5 cm foram amostrados 515 indivíduos, representados por 20 famílias, 28 gêneros e 46 espécies. As famílias de maior riqueza foram Leguminosae, Vochysiaceae e Malpighiaceae. As espécies Qualea grandiflora, Byrsonima crassa, Erytroxylum tortuosum, Qualea parviflora e Miconia ferruginata apresentaram os maiores VIs. A diversidade da área (H¢ = 1,353) é menor que valores descritos para Cerrado, o que pode ser conseqüência tanto da abundância de espécies como Q. grandiflora e B. crasssa quanto de interferências antrópicas no local, incluindo queimadas. O valor do índice sucessional (IS = 2,3) indica uma comunidade em estágio intermediário de sucessão, já que seu valor oscila de 1 a 3 . A comunidade ajustou-se apenas ao modelo log-normal, o que, de acordo com a literatura, pode ser influência da proporção da abundância de espécies dominantes, intermediárias e raras.

In the State Park Parque Estadual das Fontes do Ipiranga - PEFI, located in the city of São Paulo, samples of submerged mixed leaf litter were collected monthly from 10 sites with different levels of eutrophication, from October 2003 to April 2005. Some abiotic factors, such as pH, temperature, conductivity and dissolved oxygen were simultaneously measured in 20 cm deep water at each site. The leaf litter samples were washed in laboratory, cut into 1 cm² pieces and incubated in Petri dishes containing distilled sterile water for five to seven days at 15 °C to 20 °C. Twenty-four aquatic Hyphomycete species were identified, with predominance of Anguillospora crassa Ingold, Lunulospora curvula Ingold, Tetrachaetum elegans Ingold and Camposporium pellucidum (Grove) Hughes. First reports for South America are: Anguillospora filiformis Greathead, Dendrospora erecta Ingold and Pyramidospora casuarinae Nilsson. These species, as well as Tetracladium setigerum (Grove) Ingold, Tricladium splendens Ingold and Varicosporium elodeae Kegel, are reported for the first time in Brazil. According to the multivariate analysis, the occurrence of the aquatic Hyphomycetes was mainly influenced by the trophic level of the aquatic environments.

O presente trabalho refere-se ao estudo taxo´mico dos gêneros Chaetomorpha e Rhizoclonium ocorrentes no litoral da Bahia. As espécies foram delimitadas com base no hábito, prese§a ou ausência de rizóides laterais, tipo de célula basal, forma e dimensões celulares. A prese§a de inclusões cristalinas nas células também foi verificada. Foram identificadas nove espécies, sete para o gênero Chaetomorpha (C. aerea (Dillwyn) Kütz., C. antennina (Bory) Kütz., C. brachygona Harv., C. clavata Kütz., C. crassa (C. Agardh) Kütz., C. minima Collins & Herv. e C. nodosa Kütz.) e duas para o gênero Rhizoclonium (R. africanum Kütz. and R. riparium (Roth) Kütz. ex Harv.). Chaetomorpha crassa, C. minima e C. nodosa são novas adições para o litoral nordeste do Brasil e C. aerea, C. clavata e R. africanum são adições para o litoral da Bahia. Três tipos de cristais foram encontrados: 1. cristais de sílica, em forma de finas agulhas agrupadas, presentes em C. aerea, C. antennina, C. brachygona, C. clavata e C. crassa; 2. cristais de oxalato de cálcio, em forma de octaedros, encontrados em C. antennina e C. clavata e, 3. cristais de carbonato de cálcio, em forma de cones formando agregados globosos, observados em C. clavata. Talos férteis de C. antennina, C. clavata, C. nodosa, R. africanum e R. riparium são ilustrados pela primeira vez para o Brasil. Descrições detalhadas das estruturas vegetativas e reprodutivas das espécies e comparações com táxons afins são apresentadas.

A floristic study of Coscinodiscophyceae and Fragilariophyceae in the Lago dos Tigres hydrologic system is presented. Phytoplankton samples were collected with a plankton net (25 µm mesh) at 11 sampling stations, three in lotic areas and eight in lacustrine zones. Eleven species of Coscinodiscophyceae and eight of Fragilariophyceae were identified; descriptions, comments, their geographic distributions for the midwestern region of Brazil, type of environment (lentic/lotic), and illustrations are provided. Among the taxa found, 13 were recorded for the first time in midwestern Brazil: Aulacoseira alpigena, A. distans, A. granulata var. angustissima, A. granulata var. valida, A. muzzanensis, Discostella stelligera var. stelligera, Fragilaria capucina var. fragilarioides, F. delicatissima var. delicatissima, F. javanica, Pleurosira laevis, Staurosira acutirostrata, Staurosirella crassa, and Ulnaria ulna.

Preference for specific protein substrates together with differential sensitivity to activators and inhibitors has allowed classification of serine/threonine protein phosphatases (PPs) into four major types designated types 1, 2A, 2B and 2C (PP1, PP2A, PP2B and PP2C, respectively). Comparison of sequences within their catalytic domains has indicated that PP1, PP2A and PP2B are members of the same gene family named PPP. On the other hand, the type 2C enzyme does not share sequence homology with the PPP members and thus represents another gene family, known as PPM. In this report we briefly summarize some of our studies about the role of serine/threonine phosphatases in growth and differentiation of three different eukaryotic models: Blastocladiella emersonii, Neurospora crassa and Dictyostelium discoideum. Our observations suggest that PP2C is the major phosphatase responsible for dephosphorylation of amidotransferase, an enzyme that controls cell wall synthesis during Blastocladiella emersonii zoospore germination. We also report the existence of a novel acid- and thermo-stable protein purified from Neurospora crassa mycelia, which specifically inhibits the PP1 activity of this fungus and mammals. Finally, we comment on our recent results demonstrating that Dictyostelium discoideum expresses a gene that codes for PP1, although this activity has never been demonstrated biochemically in this organism.

Leber's hereditary optic neuropathy (LHON) is a maternally inherited form of retinal ganglion cell degeneration leading to optic atrophy in young adults. Several mutations in different genes can cause LHON (heterogeneity). The ND6 gene is one of the mitochondrial genes that encodes subunit 6 of complex I of the respiratory chain. This gene is a hot spot gene. Fourteen Persian LHON patients were analyzed with single-strand conformational polymorphism and DNA sequencing techniques. None of these patients had four primary mutations, G3460A, G11788A, T14484C, and G14459A, related to this disease. We identified twelve nucleotide substitutions, G13702C, T13879C, T14110C, C14167T, G14199T, A14233G, G14272C, A14290G, G14365C, G14368C, T14766C, and T14798C. Eleven of twelve nucleotide substitutions had already been reported as polymorphism. One of the nucleotide substitutions (A14290G) has not been reported. The A14290G nucleotide substitution does not change its amino acid (glutamic acid). We looked for base conservation using DNA star software (MEGALIGN program) as a criterion for pathogenic or nonpathogenic nucleotide substitution in A14290G. The results of ND6 gene alignment in humans and in other species (mouse, cow, elegans worm, and Neurospora crassa mold) revealed that the 14290th base was not conserved. Fifty normal controls were also investigated for this polymorphism in the Iranian population and two had A14290G polymorphism (4%). This study provides evidence that the mtDNA A14290G allele is a new nonpathogenic polymorphism. We suggest follow-up studies regarding this polymorphism in different populations.
