This review describes the use of catalytic asymmetric aldol reactions of silyl enol ethers and silyl (thio)ketene acetals with aldehydes (the Mukaiyama aldol reaction) in order to illustrate its synthetic utility. A variety of Lewis acid and basic reagents were employed for catalytic aldol reactions with high diastereo- and enantioselectivities. The origins of the selectivity of these reactions are discussed and some representative examples of their application in the synthesis of natural products are presented. New developments in chiral heterobimettalic lanthanoid catalysis and enantioselective aldol reactions in aqueous media are also included.

Chalcone (1) and its fluorinated derivatives 2-4, as well as their cyclic analogues 5-10, were synthesized through an aldol condensation reaction between the corresponding ketone and aldehyde. These compounds were characterized by IR, EIMS and ¹H and 13C NMR spectral data. Modern NMR techniques allowed us to conclude that the compounds obtained show E configuration. These techniques were also employed to investigate the equilibrium involving the s-cis and s-trans conformations of 1-4, with this equilibrium being dependent on the fluorine substitution on both aromatic rings, A or B. IR studies indicated that the yield of the s-cis conformation in the fluorinated derivatives is 57.4±1.4; 88.1±0.4 and 66.4±0.7%, for 2, 3 and 4, respectively, based on previous ¹H NMR calculations for chalcone. Theoretical calculations, using the MMX method, were employed to justify the variation of chemical shifts for the fluorinated derivatives and cyclic analogues. These chemical shifts are consequence of the anisotropic effect showed by the carbonyl group on these compounds.

High levels of substrate-based 1,5-stereoinduction are obtained in the boron-mediated aldol reactions of beta-oxygenated methyl ketones with achiral and chiral aldehydes. Remote induction from the boron enolates gives the 1,5-anti adducts, with the enolate pi-facial selectivity critically dependent upon the nature of the beta-alkoxy protecting group. This 1,5-anti aldol methodology has been strategically employed in the total synthesis of several natural products. At present, the origin of the high level of 1,5-anti induction obtained with the boron enolates is unclear, although a model based on a hydrogen bonding between the alkoxy oxygen and the formyl hydrogen has been recently proposed.

Dibenzalacetone and other aldol condensation products are known sunscreens commonly used in cosmetics. This type of compounds can easily be prepared in an Organic Chemistry Lab by reaction of aldehydes with ketones in basic medium. These compounds can be incorporated in poly(methyl methacrylate) and used as UV light absorbers, for example in sunglasses. This project has the advantage of using inexpensive reagents which are readily available in Chemistry Laboratories. This experiment can also be a base starting point for discussions of organic, polymer and photochemistry topics.

This work describes an overview of our synthesis of pteridic acids A and B and discloses some interesting results related to the lithium enolate-mediated aldol reaction used as key step to set up the C5-C15 fragment of these natural products. This first example, as far we know, of an aldol reaction between a chiral enolate of a (Z) enone and a chiral aldehyde has driven us to a series of experiments showing the remarkable relation between enolization selectivity and reaction conditions.
