Infecções experimentais de descendentes de Biomphalaria tenagophila de Itajubá, MG (Brasil) foram realizadas utilizando-se a conhecida cepa "LE" de Schistosoma mansoni, isolada em Belo Horizonte. Como controle, utilizou-se B. glabrata de Belo Horizonte, MG. Em conseqüência, assinalou-se, pela primeira vez, a infecção de B. tenagophila de Itajubá, à cepa de S. mansoni adaptada à B. glabrata de Belo Horizonte. Isto é, ao final do experimento, 5 (3,3%) entre os 149 exemplares sobreviventes, infectaram-se. Face ao atual surto industrial da região, com grande aporte de migrantes, foi chamada a atenção para a necessidade de medidas profiláticas, em decorrência da possibilidade de instalação de foco da doença no Sul de Minas Gerais.

"Híbridos" de Biomphalaria tenagophila provenientes dos cruzamentos de linhagens albinas de Belo Horizonte (BH) ou de Joinvile (SC), com melânicos de Cabo Frio (CF), do Taim (Ta) ou de Curitiba (PR), submetidos à infecção pela cepa do Schistosoma mansoni de Belo Horizonte (=LE), apresentaram os seguintes resultados: em F1, os "híbridos" TaSC, PRSC e CFTa exibiram taxas de 4,5%, 12,5% e 11,2% de suscetibilidade; em F2, todos os "híbridos" foram negativos e em F3, um exemplar albino, filho de (CFBH) ². TaBH se infectou com a LE. Dentre os controles, a B. glabrata apresentou taxas de 66,7 a 93,6% de suscetibilidade à LE e a B. tenagophila de Joinvile exibiu taxas de infecção de 17,1 e 33,3% pela cepa SJ; e os "híbridos" BHTa e BHCF, taxas de 6,0 a 53,8% também pela cepa SJ. Houve grande influência da linhagem materna nas taxas de suscetibilidade. Devido ao fato de descendentes do cruzamento de linhagens refratárias a LE (CF, Ta e BH), terem se infectado, é recomendado o uso de "híbridos" para a detecção de gens de suscetibilidade em tais linhagens. São ainda discutidas, a necessidade do uso de maior número de miracídios nos testes de infecção e a falta de relação entre a freqüência de contatos parasitas-hospedeiros e as taxas de infectividade. Considerando que estas dependem de características genéticas preexistentes na população, a cepa LE seria uma variedade genética (ou raça) distinta da cepa SJ, dotada de pouca aptidão em infectar as diversas populações de B. tenagophila, exceto a de Joinvile (SC).

In the experimental schistosomiasis mansoni glucocorticoids cause a reduction in the worm burden when administered in the week of infection or, the longest, at the next week. In order to determinate the probable(s) site(s) of reduction of the worm burden, mice were infected with cercariae of LE strain of S. mansoni and dexamethasone was administered daily (50 mg/kg, subcutaneously) starting 1 hour before infection until the eighth day. Mice were sacrificed daily starting on the third day after infection until the ninth day, and schistosomula from lungs were collected. Six weeks after infection, the remaining mice were sacrificed and perfused for adult worm recovery. Analysis of the results showed that the non-treated mice presented larger numbers of lung larvae than the treated ones, and this difference was also found later in the worm burden in the portal system. This difference may reflect the early death of larvae in treated animals, before or after reaching the lungs.

Mice infected with about 90 cercariae of Schistosoma mansoni (LE strain) were treated during five consecutive days with dexamethasone (50 mg/Kg, subcutaneously), starting on the 42th day of infection. Groups of five mice were then daily sacrificed from the first day after onset of treatment until the first day after. The perfusion of the portal system was performed and a piece of the intestine was processed for qualitative and quantitative oograms. This treatment carries to larger numbers of eggs in the tissues of treated mice, when compared with untreated groups. No changes were observed in the kinetics of oviposition, as all stages of viable eggs were observed in the tissues of treated and control mice. These data reinforce the hypothesis of a partial blockade of the egg excretion in immunossupressed mice.

The analysis of 58 patients with chronic hepatitis C without cirrhosis and treated with interferon-alpha demonstrated that hepatitis C viral (HCV) load does not correlate with the histological evolution of the disease (p = 0.6559 for architectural alterations and p = 0.6271 for the histological activity index). Therefore, the use of viral RNA quantification as an evolutive predictor or determinant of the severity of hepatitis C is incorrect and of relative value. A review of the literature provided fundamental and interdependent HCV (genotype, heterogeneity and mutants, specific proteins), host (sex, age, weight, etc) and treatment variables (dosage, time of treatment, type of interferon) within the broader context of viral kinetics, interferon-mediated immunological response (in addition to natural immunity against HCV) and the role of interferon as a modulator of fibrogenesis. Therefore, viral load implies much more than numbers and the correct interpretation of these data should consider a broader context depending on multiple factors that are more complex than the simple value obtained upon quantification.

Uma auxiliar de laboratório infectou-se acidentalmente, com cercárias de Schistosoma mansoni, cepa LE, mantida rotineiramente em nossos laboratórios. Decorridos 5 meses, o exame parasitológico de fezes revelou 108 ovos/g . A pacientefoi tratada com oxamniquine, porém a infecção continuou ativa (6 ovos/g). Foi então obtido o isolado SSF mantido no modelo Biomphalaria glabrata - camundongo albino. Os resultados obtidos no estudo comparativo, entre o isolado SSF e a cepa LE, que lhe deu origem, mostraram que a duração do período pré-patente e o índice de infectividade em camundongos, bem como a resposta aos agentes esquistossomicidas (hycanthone, oxamniquine epraziquantel) não apresentaram diferenças estatisticamente significativas. Por outro lado, o número de miracídios obtidos dos intestinos e fígados dos camundongos infectados foi o dobro com a cepa LE, quando comparados com aquele do isolado SSF. Também a variação do peso dos animais foi bastante diferente. Concluiu-se que apenas uma passagem pelo hospedeiro humano não mudou substancialmente as características da cepa estudada.

Data on Schistosoma mansoni-Entamoeba histolytica coinfection are scarce in the literature. In the present study, hamsters that had been infected for 70 days with Schistosoma mansoni (LE strain) were inoculated via the portal vein with two strains of trophozoites of Entamoeba histolytica: ICB-EGG (highly virulent) and ICB-RPS (non-virulent). The most evident result of coinfection was increased morbidity and mortality, in comparison with either of the infections alone. Histologically, there were no evident signs of interaction between these two infections. The morphological findings of schistosomal granuloma and amoebic abscesses in the liver were similar to those seen in the respective single-infection controls. However, there was severe wasting of the animals with both infections and greater numbers of amoebic lesions in their livers. The results obtained indicated that schistosomiasis aggravates the course of amoebiasis in hamsters.
