The kinetics of growth of Leishmania performed in vitro after internalization of the promastigote form in the cell and the occurrence of the transformation of the parasite into the amastigote form have been described by several authors. They used explants of macrophages in hamster spleen cell culture or in a human macrophage lineage cell, the U937. Using microscopy, the description of morphologic inter-relationship and the analysis of the production of specific molecules, it has been possible to define some of the peculiarities of the biology of the parasite. The present study shows the growth cycle of Leishmania chagasi during the observation of kinetic analysis undertaken with a McCoy cell lineage that lasted for a period of 144 hours. During the process, the morphologic transformation was revealed by indirect immunofluorescence (IF) and the molecules liberated in the extra cellular medium were observed by SDS-PAGE at 24-hour intervals during the whole 144-hour period. It was observed that in the first 72 hours the promastigote form of L. chagasi adhered to the cell membranes and assumed a rounded (amastigote-like) form. At 96 hours the infected cells showed morphologic alterations; at 120 hours the cells had liberated soluble fluorescent antigens into the extra cellular medium. At 144 hours, new elongated forms of the parasites, similar to promastigotes, were observed. In the SDS-PAGE, specific molecular weight proteins were observed at each point of the kinetic analysis showing that the McCoy cell imitates the macrophage and may be considered a useful model for the study of the infection of the Leishmania/cell binomial.

The production of interferon gamma (IFNgamma) guarantees effective T cell-mediated immunity against Mycobacterium tuberculosis infection. In the present study, we simply compare the in vitro immune responses to Mycobacterium antigens in terms of IFNg production in a total of 10 healthy Brazilian volunteers. Whole blood and mononuclear cells were cultivated in parallel with PPD, Ag85B, and M. bovis hsp65, and five-days supernatants were harvested for cytokine detection by ELISA. The inter-assay result was that the overall profile of agreement in response to antigens was highly correlated (r² = 0.9266; p = 0.0102). Potential analysis is in current progress to dictate the usefulness of this method to access the immune responses also in tuberculosis patients and its contacts.

Chromatin proteins play a role in the organization and functions of DNA. Covalent modifications of nuclear proteins modulate their interactions with DNA sequences and are probably one of the multiple factors involved in the process of switch on/off transcriptionally active regions of DNA. Histones and high mobility group proteins (HMG) are subject to many covalent modifications that may modulate their capacity to bind to DNA. We investigated the changes induced in the phosphorylation pattern of cultured Wistar rat Sertoli cell histones and high mobility group protein subfamilies exposed to 7 µM retinol for up to 48 h. In each experiment, 6 h before the end of the retinol treatment each culture flask received 370 KBq/ml [32P]-phosphate. The histone and HMGs were isolated as previously described [Moreira et al. Medical Science Research (1994) 22: 783-784]. The total protein obtained by either method was quantified and electrophoresed as described by Spiker [Analytical Biochemistry (1980) 108: 263-265]. The gels were stained with Coomassie brilliant blue R-250 and the stained bands were cut and dissolved in 0.5 ml 30% H2O2 at 60oC for 12 h. The vials were chilled and 5.0 ml scintillation liquid was added. The radioactivity in each vial was determined with a liquid scintillation counter. Retinol treatment significantly changed the pattern of each subfamily of histone and high mobility group proteins.

In this study, we investigated the potential role of high-mobility group box 1 (HMGB1) in severe acute pancreatitis (SAP) and the effects of growth hormone (G) and somatostatin (S) in SAP rats. The rats were randomly divided into 6 groups of 20 each: sham-operated, SAP, SAP+saline, SAP+G, SAP+S and SAP+G+S. Ileum and pancreas tissues of rats in each group were evaluated histologically. HMGB1 mRNA expression was measured by reverse transcription-PCR. Levels of circulating TNF-α, IL-1, IL-6, and endotoxin were also measured. In the SAP group, interstitial congestion and edema, inflammatory cell infiltration, and interstitial hemorrhage occurred in ileum and pancreas tissues. The levels of HMGB1, TNF-α, IL-1, IL-6 and endotoxin were significantly up-regulated in the SAP group compared with those in the sham-operated group, and the 7-day survival rate was 0%. In the SAP+G and SAP+S groups, the inflammatory response of the morphological structures was alleviated, the levels of HMGB1, TNF-α, IL-1, IL-6, and endotoxin were significantly decreased compared with those in the SAP group, and the survival rate was increased. Moreover, in the SAP+G+S group, all histological scores were significantly improved and the survival rate was significantly higher compared with the SAP group. In conclusion, HMGB1 might participate in pancreas and ileum injury in SAP. Growth hormone and somatostatin might play a therapeutic role in the inflammatory response of SAP.

This paper surveys the literature on fiscal competition. We consider tax and expenditure competition in a more general set up where different jurisdictions within a federation may compete in the provision of public goods in order to attract some residents (Tiebout, 1956) and expel others (Brueckner, 1999); and/or for business. We address the vast literature on welfare gains or losses of these types of competition. Then, we discuss the empirical evidence, focusing on estimates of the sensitiveness of production factors to tax differentials and on the importance of the strategic interdependence among jurisdictions. We combine econometric studies with some case studies. Last we discuss the design of mechanisms to cope with fiscal competition, especially under a more global environment where factors become more mobile.

Specifically, it analyses the main aspects of diplomatic relations between the Holy See and Russia, China, Islamic countries and Israel, and provides perspectives on the inter-civilization diplomacy of the Holy See during the current pontificate.

Tem sido observada, em milho, a redução da absorção de nitrogênio (N), por causa da deficiência de fósforo (P), mas o efeito dessa combinação na seleção de genitores ainda precisa de estudos. Os objetivos deste trabalho foram avaliar os efeitos das inter-relações de P e N na capacidade de combinação e seleção de genitores de milho. As 28 combinações híbridas (CH), obtidas de um dialelo completo entre oito genótipos de milho e três testemunhas, foram avaliadas em quatro ambientes, obtidos pelas combinações de alta e baixa disponibilidade de P e N. Avaliaram-se o rendimento de grãos (RG) e as características secundárias: altura de plantas (AP) e espigas (AE), índice de colheita (IC), peso volumétrico (PV), prolificidade (PRL) e massa seca da parte aérea (MSPA). O efeito do estresse de N no RG foi similar em baixo e alto P. O genótipo P3041 apresentou alto RG em todos os ambientes. As correlações genéticas das características secundárias com o RG foram afetadas pelos ambientes. Em alto P, a capacidade geral de combinação (CGC) foi significativa para RG apenas em alto N. Em baixo P, a CGC apresentou significância em ambos os níveis de N. Os genótipos AG1051 e P3041 tiveram estimativas positivas de CGC em todos os ambientes. Conclui-se que, para o rendimento de grãos, a CGC somente não é importante em alto P e baixo N e a CEC não é importante em estresse de P ou N.

A cultura da cana-de-açúcar desempenha, sobretudo no aspecto relacionado às fontes energéticas alternativas, importantíssimo papel. No ano de 2009, no município de Suzanápolis (SP), cujas coordenadas são 20º 28' 10'' S e 50º 49' 20'' W, no cerrado brasileiro foi analisada a produtividade da cana soca (2º ciclo) em razão da resistência à penetração, umidade gravimétrica e matéria orgânica de um Argissolo Vermelho eutrófico. O objetivo foi selecionar, entre os atributos pesquisados, aquele com a melhor correlação, a linear e a espacial, para explicar a variabilidade da produtividade da cana-de-açúcar. Foi instalada a malha geoestatística para a coleta de dados do solo e da planta, com 120 pontos amostrais, numa área de 14,53 ha. A matéria orgânica correlaciona-se linear e negativamente com a resistência à penetração, indicando que manejos do solo que visem seu aumento no perfil melhoram suas condições físicas e, por conseguinte, o desenvolvimento e a produtividade da cana-de-açúcar. A umidade gravimétrica (UG) e o teor de matéria orgânica do solo (MO) correlacionam-se direta, linear (UG2 e MO1) e espacialmente (UG1 e MO1) com a produtividade da cana-de-açúcar, demonstrando serem os melhores atributos, dentre os pesquisados, para estimar e aumentar a produtividade agrícola da cana-de-açúcar.

A autora, após sintetizar algumas das vertentes de estudo e conceitos atuais na temática da inter-relação trabalho-saúde mental, apresenta um estudo de caso individual. Trata-se do caso de um mecânico industrial do setor siderúrgico que desenvolveu um quadro psicopatol6gico. A evolução de manifestações de fadiga crônica e desencadeamento de uma crise mental aguda são examinadas em suas correlações ao hist6rico de vida e trabalho e a dinâmicas referidas às condições e à organização do trabalho no emprego atual.

The aim of this paper is to analyze the determining factors for the pricing of handsets sold with service plans, using the hedonic price method. This was undertaken by building a database comprising 48 handset models, under nine different service plans, over a period of 53 weeks in 2008, and resulted in 27 different attributes and a total number of nearly 300,000 data registers. The results suggest that the value of monthly subscriptions and calling minutes are important to explain the prices of handsets. Furthermore, both the physical volume and number of megapixels of a camera had an effect on the prices. The bigger the handset, the cheaper it becomes, and the more megapixels a camera phone has, the more expensive it becomes. Additionally, it was found that in 2008 Brazilian phone companies were subsidizing enabled data connection handsets.

Foram estudados os registros civis desde as suas origens, salientando aspectos legislativos do Brasil, chamando a atenção para algumas implicações que a legislação a eles referente trazem às estatísticas vitais e suas repercussões em planejamento de saúde e epidemiologia. Relativamente ao nascimento foram abordados alguns tópicos, com ênfase no problema do registro por local de ocorrência, fato que influe enormemente em programações de saúde materno-infantil. Foram salientados também alguns aspectos do sub-registro de nascimento, citando dados para São Paulo. Quanto aos óbitos foram analisados o registro por local de ocorrência, o prazo para registro, o problema das causas de morte e o relativo ao sub-registro. Com referência aos nascidos mortos, foi apresentada a conceituação de nascido vivo e nascido morto da Organização Mundial da Saúde e indicado o fato de sua não aplicação prática repercutir nos campos do Direito e da Estatística Vital. Foram feitas algumas recomendações no sentido de um maior entrosamento entre os vários profissionais - legisladores, médicos, oficiais de cartório, estaticistas de saúde - que, de uma maneira ou de outra, têm relação com os eventos vitais.

A controlled trial was performed with the purpose of investigating which factors could be considered of significant risk for the development of basal cell carcinoma. A total of 259 cases of basal cell carcinoma diagnosed from July 1991 to July 1992 were compared with 518 controls matched for age and sex. All subjects in both groups were white. Protocol data were submitted to statistical analysis by the chi-square test and by multiple conditional logistic regression analysis and the following conclusions were reached: 1) light skin color (types I and II of the Fitzpatrick classification), odds ratio of 2.8; outdoor work under constant sunlight, odds ratio of 5.0; the presence of actinic lesions due to exposure to the sun, odds ratio of 4.9, are risk factors perse. 2) Type III skin in the Fitzpatrick classification only represents a risk factor when the patient reports a history of intense sunburns, but not in the absence of such a history. 3) Sunburns per se do not represent a risk factor althorig the point made in item 2 of these conclusions is valid. 4) Other suspected risk factors whose significance was not confirmed by multiple conditioned logistic regression analysis were: residence in rural areas, light eyes and blond hair color, extent of the awareness of the "sun x skin cancer" relationship, familial occurrence of skin cancer, excessive exposure to the sun, and freckles appearing in childhood.
