Uma das práticas mais difundidas para a avaliação de ativos operacionais é o fluxo de caixa descontado, ou FCD. Contudo, uma das versões mais comumente aceitas de FCD, o custo médio ponderado de capital, ou WACC no original inglês, tornou-se obsoleta. Este artigo discute uma metodologia alternativa ao WACC, chamada de VPA (valor presente ajustado), ou APV (adjusted present value), em inglês. O artigo mostra que essa metodologia apresenta vantagens superiores quando comparada ao WACC, especificamente pelo fato de oferecer mais informações de relevância administrativa, a um índice menor de erro, ajudando os executivos a analisar não só quanto vale um ativo, mas também a origem de seu valor.

For the purpose of research a large quantity of anti-measles IgG working reference serum was needed. A pool of sera from five teenagers was prepared and named Alexandre Herculano (AH). In order to calibrate the AH serum, 18 EIA assays were performed testing in parallel AH and the 2nd International Standard 1990, Anti-Measles Antibody, 66/202 (IS) in a range of dilutions (from 1/50 to 1/25600). A method which compared parallel lines resulting from the graphic representation of the results of laboratory tests was used to estimate the power of AH relative to IS. A computer programme written by one of the authors was used to analyze the data and make potency estimates. Another method of analysis was used, comparing logistic curves relating serum concentrations with optical density by EIA. For that purpose an existing computer programme (WRANL) was used. The potency of AH relative to IS, by either method, was estimated to be 2.4. As IS has 5000 milli international units (mIU) of anti-measles IgG per millilitre (ml), we concluded that AH has 12000 mIU/ml.

The most practicable assay for measurement of measles IgG (mIgG) in large numbers of sera is an enzyme immunoassay (EIA). To assess how EIA results would agree with those by the gold standard method of plaque reduction neutralization (PRN) we compared the results from the two methods in 43 pairs of maternal and umbilical cord sera, and sera from the corresponding infants when aged 11 - 14 months. In maternal-cord sera, the differences between mean antibody levels by EIA or PRN were not statistically significant, though in individual sera, differences could be large. However, agreement was less good for infants sera, in which levels of mIgG were very low. The conclusions of a study of transplacental transport of mIgG would not be affected by the use of either technique. When studying waning immunity in infants, PRN should be the method of choice, while results from studies using EIA should be interpreted with caution.

Melioidosis is an emerging infection in Brazil and neighbouring South American countries. The wide range of clinical presentations include severe community-acquired pneumonia, septicaemia, central nervous system infection and less severe soft tissue infection. Diagnosis depends heavily on the clinical microbiology laboratory for culture. Burkholderia pseudomallei, the bacterial cause of melioidosis, is easily cultured from blood, sputum and other clinical samples. However, B. pseudomallei can be difficult to identify reliably, and can be confused with closely related bacteria, some of which may be dismissed as insignificant culture contaminants. Serological tests can help to support a diagnosis of melioidosis, but by themselves do not provide a definitive diagnosis. The use of a laboratory discovery pathway can help reduce the risk of missing atypical B. pseudomallei isolates. Recommended antibiotic treatment for severe infection is either intravenous Ceftazidime or Meropenem for several weeks, followed by up to 20 weeks oral treatment with a combination of trimethoprim-sulphamethoxazole and doxycycline. Consistent use of diagnostic microbiology to confirm the diagnosis, and rigorous treatment of severe infection with the correct antibiotics in two stages; acute and eradication, will contribute to a reduction in mortality from melioidosis.

DNA amplification techniques are being used increasingly in clinical laboratories to confirm the identity of medically important bacteria. A PCR-based identification method has been in use in our centre for 10 years for Burkholderia pseudomallei and was used to confirm the identity of bacteria isolated from cases of melioidosis in Ceará since 2003. This particular method has been used as a reference standard for less discriminatory methods. In this study we evaluated three PCR-based methods of B. pseudomallei identification and used DNA sequencing to resolve discrepancies between PCR-based results and phenotypic identification methods. The established semi-nested PCR protocol for B. pseudomallei 16-23s spacer region produced a consistent negative result for one of our 100 test isolates (BCC #99), but correctly identified all 71 other B. pseudomallei isolates tested. Anomalous sequence variation was detected at the inner, reverse primer binding site for this method. PCR methods were developed for detection of two other B. pseudomallei bacterial metabolic genes. The conventional lpxO PCR protocol had a sensitivity of 0.89 and a specificity of 1.00, while a real-time lpxO protocol performed even better with sensitivity and specificity of 1.00, and 1.00. This method identified all B. pseudomallei isolates including the PCR-negative discrepant isolate. The phaC PCR protocol detected the gene in all B. pseudomallei and all but three B. cepacia isolates, making this method unsuitable for PCR-based identification of B. pseudomallei. This experience with PCR-based B. pseudomallei identification methods indicates that single PCR targets should be used with caution for identification of these bacteria, and need to be interpreted alongside phenotypic and alternative molecular methods such as gene sequencing.

Interleukin 8 (CXCL8) is an autocrine chemokine specific for the chemoattraction and activation of granulocytes, NKT cells and T lymphocytes. Patients with tuberculosis and latent Mycobacterium tuberculosis infection were assessed for the spontaneous expression of CXCR1 (CD128) and CXCR2 on lymphocytes and monocytes. Compared with ex vivo profiles, increased spontaneous CXCR2 expression and normal CXCR1 expression were found on lymphocytes in two out of 59 individuals. Monocytes showed normal ex vivo profiles for both receptors. After stimulation with purified protein derivative, the in vitro levels of CXCL8 were below the median levels of all patients with prior tuberculosis. Spontaneous CXCR2 modulation did not cause notable variation in the in vitro levels of CXCL8.

Three new species of Erythroxylumfrom Amazoniaare described: Enythroxylum nelson-rosaePlowman, E. schunkeiPlowman and E. tucuruiendePlowman. An emended des-cription is given for E; ligustrinum DC., previously know from a single collection from French Guiana: its is here cited from Brazil for the first time. A new variety, E. ligustrinum var. carajasensePlowman, is also described. A provisional list of all Erythroxylum species occurring in Amazonia and peripheral areas is appended.

A síntese de lactonas sesquiterpênicas do tipo guaiano é de interesse devido as atividades biológicas associadas às estruturas complexas deste grupo de produtos naturais. Planejou-se a sua síntese a partir de monoterpenos abundantes isolados de óleos essenciais brasileiros. Assim p-ment-l-eno foi transformado em 2-bromo-6-isopropil-3-metil-ciclohept-2-enona por adição de dibromocarbeno, expansão de anel com rearranjo usado AgNO3, e finalmente oxidação com clorocromato de piridinio. Redução quimioseletiva do bromo com zinco em ácido acético e alquilação termodinâmica em α,β-insaturada. Tratamento com ácido sulfúrico concentrado efetuou hidrólise e desidratação a um furano anelado a um ciclohepteno de estrutura de um oxa-guaiano.

1) Inicialmente foi dado um breve resumo dos métodos básicos do melhoramento no milho os quais podem ser reunidos em dois grupos principais: o processo do milho híbrido, com as suas variantes, e os processos dos sintéticos. Estes últimos podem ainda ser subdivididos em duas categorias: os sintéticos simples e os sintéticos balançados. Na obtenção dos sintéticos simples toma-se inicialmente em consideração a capacidade combinatória das linhagens a serem misturadas, e se executa em cada geração de sintético uma seleção massal de conservação. Nos balançados devemos acrescentar uma forte seleção, na fase preparatória, contra todos os híbridos que dão segregações mendelianas fortes demais. 2) No curso de um breve resumo histórico ficou evidente que a idéia de se aproveitarem os sintéticos no melhoramento do milho, formulada pela primeira vez por Hayes e Garber (1919) deu resultados práticos apreciáveis. Assim Hayes, Rinke e Tsinang (1944) obtiveram produções de sintéticos que eram equivalentes de um híbrido duplo, Minhybrid 403. Lonnquist (1949) registrou produções de sintéticos idênticos ao híbrido duplo, US 13. Roberts, Wellhausen, Palácios e Guaves (1949) e Wellhausen (1950) relataram resultados bastante satisfatórios, obtidos no México. 3) Ficou demonstrado que as fórmulas de Sewall Wright (1932) e de Mangelsdorf (1939) não podem ser consideradas como explicações gerais do método, pois pela sua derivação pode-se mostrar facilmente que elas exigem certas premissas que nem sempre são justificáveis. 4) Para eliminar confusões na terminologia foi desenvolvido um esquema básico da constituição de sintéticos supondo que se parte de linhagens autofecundadas e que foram plantadas em conjunto para a reprodução de cruzamento livre. A geração que consiste das plantas autofecundadas, plantadas em mistura, é denominada SyO. A geração seguinte, a qual contém uma maior percentagem de híbridos simples e uma menor per-centagem de descendentes de cruzamentos dentro de mesma linhagem (descendentes consanguíneos) representa assim a geração Syl. A geração que segue depois de novo cruzamento livre, Sy2, será então composta de híbridos entre quatro linhagens (híbridos duplos"), entre três linhagens ("three way crosses"), entre duas linhagens ("híbridos simples") e descendentes de combinações consanguíneas, ("inbreds"). Porém se houver uma seleção em Sy1 que elimina todos os descendentes de combinações consanguíneas, sobrevivendo apenas híbridos simples, então a geração Sy2 será composta de híbridos entre plantas que não tem nenhuma das linhagens originais em comum, os que têm uma linhagem em comum e finalmente aqueles que têm duas linhagens em comum. 5) Empregando esta classificação das gerações, podemos verificar que a geração Sy1 de Lonnquist corresponde à geração Sy1 do esquema básico, a geração Sy1 deHayes et al corresponde à geração Sy2 do esquema básico é a geração Sy1 de Wellhausen et al corresponde aproximadamente à geração Sy3 do esquema básico. 6) Uma teoria mais correta dos sintéticos deve-se basear nas regras da genética em populações, as quais foram empregadas por Brieger para justificar o processo dos sintéticos balançados. Uma discussão mais detalhada desta teoria será assim dada numa outra publicação que se ocupara especialmente com ossintéticos balançados.

Fecal egg count scores were used to investigate the distribution and abundance of intestinal helminths in the population of a rural village. Prevalences of the major helminths were 41% with Ascaris lumbricoides 60% with Trichuris trichiura and 50% with Necator americanus. All three parasites showed a highly aggregated distribution among hosts. Age/prevalence and age/intensity profiles were typical for both A. lumbricoides and T. trichiura with the highest worm burdens in the 50-10 year old children. For hookworm both prevalence and intensity curves were convex in shape with maximum infection levels in the 30-40 year old age class. Infected females had higher burdens of T. trichiura than infected males in all age classes of the population; there were no other effects of host gender. Analysis of associations between parasites within hosts revealed strong correlations between A. lumbricoides and T. lumbricoides and T. trichiura. Individuals with heavy infections of A. lumbricoides and T. trichiura showed highly significant aggregation within households. Associations between a variety of household features and heavy infections with A. lumbricoides and T. trichiura are described.

Blood eosinophilia and tissue infiltration by eosinophils are frequently observed in allergic inflammation and parasitic infections. This selective accumulation of eosinophils suggested the existence of endogenous eosinophil-selective chemoattractants. We have recently discovered a novel eosinophil-selective chemoattractant which we called eotaxin in an animal model of allergic airways disease. Eotaxin is generated in both allergic and non-allergic bronchopulmonary inflammation. The early increase in eotaxin paralled eosinophil infiltration in the lung tissue in both models. An antibody to IL-5 suppressed lung eosinophilia, correlating with an inhibition of eosinophil release from bone marrow, without affecting eotaxin generation. This suggests that endogenous IL-5 is important for eosinophil migration but does not appear to be a stimulus for eotaxin production. Constitutive levels of eotaxin observed in guinea-pig lung may be responsible for the basal lung eosinophilia observed in this species. Allergen-induced eotaxin was present mainly in the epithelium and alveolar macrophages, as detected by immunostaining. In contrast there was no upregulation of eotaxin by the epithelial cells following the injection of Sephadex beads and the alveolar macrophage and mononuclear cells surrounding the granuloma were the predominant positive staining cells. Eotaxin and related chemokines acting through the CCR3 receptor may play a major role in eosinophil recruitment in allergic inflammation and parasitic diseases and thus offer an attractive target for therapeutic intervention.

Chemokines (chemoattractant cytokines) induce potent and selective chemotaxis of leukocyte subsets in vitro. Here, we review briefly the chemokines shown to induce eosinophil chemotaxis in vitro and describe a novel model for the study of the ability of chemokines to stimulate eosinophil migration in vivo. Eosinophils were purified from the blood of mice over-expressing the IL-5 gene and labelled with 111In. Only the C-C chemokines, eotaxin and MIP-1alpha, but not RANTES, MCP-1, MCP-3, MCP-4, MIP-1ß, KC and MIP-2, effectively induced the recruitment of 111In-eosinophils in mouse skin. We suggest that this mouse model will be useful in assessing the role of endogenously-generated chemokines in mediating eosinophil migration to sites of allergic inflammation in vivo.

Schistosomiasis is a major neglected tropical disease caused by trematodes from the genus Schistosoma. Because schistosomes exhibit a complex life cycle and numerous mechanisms for regulating gene expression, it is believed that spliced leader (SL) trans-splicing could play an important role in the biology of these parasites. The purpose of this study was to investigate the function of trans-splicing in Schistosoma mansoni through analysis of genes that may be regulated by this mechanism and via silencing SL-containing transcripts through RNA interference. Here, we report our analysis of SL transcript-enriched cDNA libraries from different S. mansoni life stages. Our results show that the trans-splicing mechanism is apparently not associated with specific genes, subcellular localisations or life stages. In cross-species comparisons, even though the sets of genes that are subject to SL trans-splicing regulation appear to differ between organisms, several commonly shared orthologues were observed. Knockdown of trans-spliced transcripts in sporocysts resulted in a systemic reduction of the expression levels of all tested trans-spliced transcripts; however, the only phenotypic effect observed was diminished larval size. Further studies involving the findings from this work will provide new insights into the role of trans-splicing in the biology of S. mansoni and other organisms. All Expressed Sequence Tags generated in this study were submitted to dbEST as five different libraries. O objetivo deste estudo é revisar a literatura relativa à criança com câncer e sua família, a fim de identificar temas que têm sido pesquisados e levantar indicadores de necessidades, subsidiando a sistematização da assistência de enfermagem. A coleta de dados sistemática foi realizada em bancos de dados informatizados, entre 1997 e 2002, utilizando-se as palavras-chave child, cancer, chronic illness/disease, family e nursing. Realizou-se, também, busca não-sistemática de publicações científicas. Os resultados foram apresentados em três temas: impacto do câncer infantil no sistema familiar; processo de adaptação e estratégias de enfrentamento utilizadas pelos pais diante da doença e o processo de perda e luto diante da morte da criança. A revisão demonstrou que a enfermagem está construindo um conhecimento específico sobre as necessidades individuais, culturais e regionais das famílias de crianças com câncer, para uma assistência de enfermagem que considere o cuidado de acordo com a singularidade de cada caso.
