It is well known that the culture media used in the presumptive diagnosis of suspiciuous colonies from plates inoculated with stools for isolation of enteric organisms do not always correctly indicate the major groups of enterobacteria. In an effort to obtain a medium affording more exact indications, several media (1-9) have been tested. Modifications of some of these media have also been tested with the result that a satisfactory modification of Monteverde's medium was finaly selected. This proved to be most satisfactory, affording, as a result of only one inoculation, a complete series of basic indications. The modification involves changes in the formula, in the method of preparation and in the manner of storage. The formulae are: A. Thymol blue indicator: NaOH 0.1/N .............. 34.4 ml; Thymol blue .............. 1.6 g; Water .................... 65.6 ml. B. Andrade's indicator. C. Urea and sugar solution: Urea ..................... 20 g; Lactose ................... 30 g; Sucrose ................... 30 g; Water .................... 100 ml. The mixture (C.) should be warmed slightly in order to dissolve the ingredients rapidly. Sterilise by filtration (Seitz). Keep stock in refrigeratior. The modification of Monteverde's medium is prepared in two parts. Semi-solid part - Peptone (Difco) 2.0 g; NaCl 0.5 g; Agar 0.5 g; Water 100.0 ml. Boil to dissolve the ingredients. Adjust pH with NaOH to 7.3-7.4. Boil again for precipitation. Filter through cotton. Ad indicators "A" 0.3 ml and "B" 1.0 ml. Sterilise in autoclave 115ºC, 15 minutes in amounts not higher than 200 ml. Just before using, add solution "C" asseptically in amounts of 10 ml to 200 ml of the melted semi-solid medium, maintained at 48-50ºC. Solid part - Peptone (Difco) 1.5 g; Trypticase (BBL) 0.5 g; Agar 2.0 g; Water 100,00 ml. Boil to dissolve the ingredients. Adjust pH with NaOH to 7.3-7.4. Boils again. Filter through cotton. Add indicators "A" 0.3 ml and "B" 1.0 ml; ferrous ammonium sulfate 0.02 g; sodiun thiosulfate 0.02 g. Sterilise in autoclave 115ºC, 15 minutes in amounts not higher than 200 ml. Just before using, add solution "C" asseptically in amounts of 10 ml to 200 ml of the melted solid medium, maintained at 48-50ºC. Final medium - The semi-solid part is dispensed first (tubes about 12 x 120 mm) in 2.5 ml amounts and left to harden at room temperature, in vertical position. The solid part is dispensed over the hardened semi-solid one in amounts from 2.0 ml to 2.5 ml and left to harden in slant position, affording a butt of 12 to 15 mm. The tubes of medium should be subjected to a sterility test in the incubator, overnight. Tubes showing spontaneous gas bubbles (air) should then be discarded. The medium should be stored in the incubator (37ºC), for not more than 2 to 4 days. Storage of the tubes in the ice-box produces the absorption of air which is released as bubbles when the tubes are incubated at 37ºC after inoculation. This fact confirmed the observation of ARCHAMBAULT & McCRADY (10) who worked with liquid media and the aplication of their observation was found to be essential to the proper working conditions of this double-layer medium. Inoculation - The inoculation is made by means of a long straight needle, as is usually done on the triple sugar, but the needel should penetrate only to about half of the height of the semi-solid column. Indol detection - After inoculation, a strip of sterelized filter papaer previously moistened with Ehrlich's reagent, is suspended above the surface of the medium, being held between the cotton plug and the tube. Indications given - In addition to providing a mass of organisms on the slant for serological invetigations, the medium gives the following indications: 1. Acid from lactose and/or sucrose (red, of yellowsh with strains which reduce the indicators). 2. Gas from lactose and/or sucrose (bubbles). 3. H[2]S production, observed on the solid part (black). 4. Motility observed on the semi-solid part (tubidity). 5. Urease production, observed on solid and semi-solid parts (blue). 6. Indol production, observed on the strip of filter paper (red or purplish). Indol production is not observed with indol positive strains which rapidly acidify the surface o the slant, and the use of oxalic acid has proved to give less sensitive reaction (11). Reading of results - In most cases overnight incubation is enough; sometimes the reactions appear within only a few hours of incubation, affording a definitive orientation of the diagnosis. With some cultures it is necessary to observe the medium during 48 hours of incubation. A description showing typical differential reaction follows: Salmonella: Color of the medium unchanged, with blackening of the solid part when H[2]S is positive. The slant tends to alkalinity (greenish of bluish). Gas always absent. Indol negative. Motility positive or negative. Shigella: Color of the medium unchanged at the beginning of incubation period, but acquiring a red color when the strain is late lactose/sucrose positive. Slant tending to alkalinity (greenish or purplish). Indol positive or negative. Motility, gas and H[2]S always negative. Proteus: Color of the medium generally changes entirely to blue or sometimes to green (urease positive delayed), with blackening of solid part when H[2]S is positive. Motility positive of negative. Indol positive. Gas positive or negative. The strains which attack rapidly sucrose may give a yellow-greenish color to the medium. Sometimes the intense blue color of the medium renders difficult the reading of the H[2]S production. Escherichiae and Klebsiellae: Color of the medium red or yellow (acid) with great and rapid production of gas. Motility positive or negative. Indol generally impossible to observe. Paracoli: Those lactose of sucrose positive give the same reaction as Esherichia. Those lactose or sucrose negatives give the same reactions as Salmonellae. Sometimes indol positive and H[2]S negative. Pseudomonas: Color of the medium unchanged. The slant tends to alkalinity. It is impossible to observe motility because there is no growth in the bottom. Alkaligenes: Color of the medium unchanged. The slant tends to alkalinity. The medium does not alter the antigenic properties of the strains and with the mass of organisms on the slant we can make the serologic diagnosis. It is admitted that this medium is somewhat more laborious to prepare than others used for similar purposes. Nevertheless it can give informations generally obtained by two or three other media. Its use represents much saving in time, labor and material, and we suggest it for routine laboratory work in which a quick presumptive preliminary grouping of enteric organisms is needed.

A razão nitrogênio ureico/creatinina (N u/c) foi medida em indivÃduos de ambos os sexos nos grupos de 3 a 10 anos, 11 a 15 anos e acima de 15 anos de idade, em uma amostra das populações das comunidades de Vila de Icapara, Pontal do Ribeira e diaristas de Iguape, localizadas no litoral sul de São Paulo, no Vale do Ribeira. Foi calculada a adequação do consumo de proteÃnas entre as famÃlias, através dos resultados de um inquérito alimentar feito pelo método das pesagens. Partiu-se da hipótese de que haveria maior proporção de indivÃduos com a razão N u/c maior ou igual a 5 nas famÃlias que apresentaram consumo proteico maior ou igual a 80,0% de adequação; através do teste chi2; 0,05, obteve-se uma associação entre a adequação do consumo de proteÃnas nas famÃlias e a razão N u/c nos indivÃduos das respectivas famÃlias.

OBJECTIVE: To assess the association between pre-gestational obesity and weight gain with cesarean delivery and labor complications. METHODS: A total of 4,486 women 20-28 weeks pregnant attending general prenatal care clinics of the national health system in Brazil from 1991 to 1995 were enrolled and followed up through birth. Body mass index categories based on prepregnancy weight and total weight gain were calculated. Associations between body mass index categories and labor complications were adjusted through logistic regression analysis. RESULTS: Obesity was present in 308 (6.9%) patients. Cesarean delivery was performed in 164 (53.2%) obese, 407 (43.1%) pre-obese, 1,045 (35.1%) normal weight and 64 (24.5%) underweight women. The relative risk for cesarean delivery in obese women was 1.8 (95% CI: 1.5-2.0) compared to normal weight women. Greater weight gain was particularly associated with cesarean among the obese (RR 4th vs 2nd weight gain quartile 2.2; 95% CI: 1.4-3.2). Increased weight at the beginning of pregnancy was associated with a significantly higher adjusted risk of meconium with vaginal delivery and perinatal death and infection in women submitted to cesarean section. Similarly, greater weight gain during pregnancy increased the risk for meconium and hemorrhage in women submitted to vaginal delivery and for prematurity with cesarean. CONCLUSIONS: Pre-gestational obesity and greater weight gain independently increase the risk of cesarean delivery, as well as of several adverse outcomes with vaginal delivery. These findings provide further evidence of the negative effects of prepregnancy obesity and greater gestational weight gain on pregnancy outcomes.

OBJECTIVE: To examine the relationship between social contextual factors and child and adolescent labor. METHODS: Population-based cohort study carried out with 2,512 families living in 23 subareas of a large urban city in Brazil from 2000 to 2002. A random one-stage cluster sampling was used to select families. Data were obtained through individual household interviews using questionnaires. The annual cumulative incidence of child and adolescent labor was estimated for each district. New child and adolescent labor cases were those who had their first job over the two-year follow-up. The annual cumulative incidence of child and adolescent labor was the response variable and predictors were contextual factors such as lack of social support, social deprivation, unstructured family, perceived violence, poor school quality, poor environment conditions, and poor public services. Pearson's correlation and multiple linear regression were used to assess the associations. RESULTS: There were selected 943 families corresponding to 1,326 non-working children and adolescents aged 8 to 17 years. Lack of social support, social deprivation, perceived violence were all positively and individually associated with the annual cumulative incidence of child and adolescent labor. In the multiple linear regression model, however, only lack of social support and perceived violence in the neighborhood were positively associated to child and adolescent labor. No effect was found for poor school quality, poor environment conditions, poor public services or unstructured family. CONCLUSIONS: Poverty reduction programs can reduce the contextual factors associated with child and adolescent labor. Violence reduction programs and strengthening social support at the community level may contribute to reduce CAL.

OBJECTIVE To analyze the effects of acupressure at the SP6 point on labor duration and cesarean section rates in parturients served in a public maternity hospital.METHODS This controlled, randomized, double-blind, pragmatic clinical trial involved 156 participants with gestational age ≥ 37 weeks, cervical dilation ≥ 4 cm, and ≥ 2 contractions in 10 min. The women were randomly divided into an acupressure, placebo, or control group at a university hospital in an inland city in the state of Sao Paulo, Brazil, in 2013. Acupressure was applied to the SP6 point during contractions for 20 min.RESULTS The average labor duration was significantly different between the SP6 acupressure group [221.5 min (SD = 162.4)] versus placebo [397.9 min (SD = 265.6)] and versus control [381.9 min (SD = 358.3)] (p = 0.0047); however, the groups were similar regarding the cesarean section rates (p = 0.2526) and Apgar scores in the first minute (p = 0.9542) and the fifth minute (p = 0.7218) of life of the neonate.CONCLUSIONS The SP6 acupressure point proved to be a complementary measure to induce labor and may shorten the labor duration without causing adverse effects to the mother or the newborn. However, it did not affect the cesarean section rate.

A radiometric assay system has been used to study oxidation patterns of (U-14C) L-amino acids by drug-susceptible and drug-resistant mycobacteria. Drug-susceptible M. tuberculosis (H37Rv TMC 102 and Erdman) along with the drug-resistant organism M. tuberculosis (H37 Rv TMC 303), M. bovis, M. avium, M. intracellulare, M. kansasii and M. chelonei were used. The organisms were inoculated into a sterile reaction system with liquid 7H9 medium and one of the (U-14C) L-amino acids. Each organism displayed a different pattern of amino acid oxidation, but these patterns were not distinctive enough for identification of the organism. Complex amino acids such as proline, phenylalanine and tyrosine were of no use in identification of mycobacteria, since virtually all organisms failed to oxidize them. There was no combination of substrates able to separate susceptible from resistant organisms.

Os autores estudaram mensalmente e por um perÃodo de seis meses 62 pacientes através de exames clÃnico-laboratoriais e que receberam transfusão de sangue no Hospital das ClÃnicas da F.M.U.F.Pe. Constataram em 3 deles sinais clÃnicos-laboratoriais e histopatológicos de hepatite por vÃrus (hepatite por sóro homólogo). Após revisão da literatuta recomendam que os estudos prossigam a fim de que os riscos de transmissão da hepatite pós-transfusional sejam eliminados.

Os Autores realizando um levantamento coproparasitológico no Instituto de Medicina Tropical da U.F.Pe., com relação a incidência de Hymenolepis nana e Taenia sp no perÃodo de 1968 a 1970, encontram Ãndice de infestação muito baixo, em tôrno de 0,05% a 2,7% para Taenia sp e 0,36% para Hymenolepis nana.

Anti-U is a rare red blood cell alloantibody that has been found exclusively in blacks. It can cause hemolytic disease of the newborn and hemolytic transfusion reactions. We describe the case of a female newborn presenting a strongly positive direct antiglobulin test due to an IgG antibody in cord blood. Anti-U was recovered from cord blood using acid eluate technique. Her mother presented positive screening of antibodies with anti-U identified at delivery. It was of IgG1 and IgG3 subclasses and showed a titer of 32. Monocyte monolayer assay showed moderate interaction of Fc receptors with maternal serum with a positive result (3.1%). The newborn was treated only with 48 hours of phototherapy for mild hemolytic disease. She recovered well and was discharged on the 4th day of life. We conclude that whenever an antibody against a high frequency erythrocyte antigen is identified in brown and black pregnant women, anti-U must be investigated.

As plantas de Uncaria guianensis e U. tomentosa são lianas trepadeiras, arbustos trepadores ou rasteiros ascendentes que ocorrem em amplas áreas da Amazônia brasileira e paÃses da América Central e do Sul. Possuem excepcional importância medicinal, com sua casca, lenho e folhas utilizadas pelos povos amazônicos e pela indústria farmacêutica. Face a necessidade da identificação das plantas em condições de campo e de laboratório, o presente trabalho descreveu a estrutura anatômica do lenho de Uncaria guianensis e U. tomentosa, ocorrentes no Estado do Acre, Brasil. Os resultados mostraram que o lenho das espécies é caracterÃstico da famÃlia Rubiaceae, a exceção dos vasos arredondados, grandes e solitários, parenquima axial difuso e largura dos raios que são próprios das lianas.

O presente trabalho estudou o efeito da salinidade na sobrevivência e na duração do desenvolvimento larval do caranguejo-uçá, Ucides cordatus (do estuário do Rio Caeté, Norte do Brasil), até a fase de megalopa em sete tratamentos de salinidade (0, 5, 10, 15, 20, 25 e 30). A salinidade afetou significativamente a sobrevivência das larvas zoea, no entanto não afetou a duração do desenvolvimento larval (20,77 ± 1,56 dias). Nas salinidades 0, 5 e 10 houve total mortalidade das larvas zoea. Somente a partir da salinidade 15 observou-se um desenvolvimento completo até a fase de megalopa. A taxa de sobrevivência foi maior em salinidade 30 (72%) e menor em 15 (16%). A reduzida taxa de sobrevivência das larvas zoea de U. cordatus, em salinidades baixas, indica a necessidade de dispersão larval do estuário para as águas costeiras onde as condições de salinidade para o desenvolvimento larval são mais favoráveis. Caso contrário se não houvesse a dispersão, a reduzida salinidade das águas estuarinas no perÃodo chuvoso, causaria uma elevada mortalidade, afetando desta forma o recrutamento, a manutenção e o crescimento da população de U. cordatus nos manguezais.

Em decorrência dos estudos na subtribo Ecliptinae da Amazônia brasileira, é descrita uma nova espécie, Acmella marajoensis G.A.R. Silva & J.U.M. Santos. Até o presente momento, a espécie é considerada como endêmica da Ilha do Marajó. São apresentados diagnose em latim, descrição detalhada, comentários taxonômicos e ilustrações.

Três rações, com nÃvel alto, médio e baixo de energia - tratamento T1 , T2 T3 - foram ministradas em confinamento, à vontade, a bovinos recém-desmamados, machos inteiros, castrados e fêmeas, durante dois perÃodos de 17 semanas. Os machos inteiros exibiram a maior habilidade de ganho de peso, e as novilhas a menor habilidade. O tratamento T1 proporcionou os melhores ganhos e conversões, e o T2 os inferiores. Verificou-se pequena superioridade no teor protéico de T3, ao que se atribuiu sua vantagem sobre T2 no 1º perÃodo, e aparente crescimento compensatório no 2º perÃodo. O teor de uréia foi superior nos machos inteiros, e semelhante nos outros animais, superior em T3 e semelhante em T1 e T2, bem como aumentou consistentemente durante o experimento. Aparentemente a uréia refletiu o fornecimento adequado de proteÃna e, mais ainda, de energia. O teor de proteÃna-iodada não mostrou diferença significativa, mas apenas tendência de se elevar com o decorrer do tempo. Não houve correlação entre os dois componentes do sangue. Concluiu-se que a eficiência de utilização alimentar foi: superior nos machos inteiros, proporcional à quantidade de energia ingerida (no caso do 1º perÃodo); favorecida pelo teor maior de proteÃna de T3 em relação a T2 no 19 perÃodo, e talvez por ganho compensatório no 2º perÃodo; refletida pelos nÃveis de uréia no plasma, mas não correlacionado com proteÃna- iodada.

Neste estudo, foi analisada a composição dos Heteroptera do curso médio da bacia do rio JacuÃ, RS, Brasil, previamente à construção da U.H.E. Dona Francisca, bem como alguns fatores abióticos que poderiam afetar a distribuição e a abundância destes organismos. Nos ambientes lênticos, amostragens quantitativas foram realizadas utilizando-se peneiras, em seis localidades (janeiro 2000). Nos ambientes lóticos, as coletas foram feitas através de amostrador de Surber (de maio a outubro de 2000), privilegiando-se a captura de espécies tipicamente bentônicas. Em cada estação, pH, oxigênio dissolvido, precipitação mensal, temperatura do ar e da água e profundidade foram medidos. Nos rios e riachos, a velocidade da corrente também foi registrada. Quinze espécies foram registradas nos ambientes lênticos, sendo Belostoma sp. e Notonecta sp. as dominantes (70%). A abundância, a riqueza e os Ãndices de diversidade foram mais altos nos locais com maiores dimensões. Nos ambientes lóticos, foram assinaladas duas espécies de Naucoridae, Ambrysus teutonius La Rivers, 1951 e Cryphocricus vianai De Carlo, 1951, sendo a primeira dominante (65%). A abundância foi maior nos pontos com maior sombreamento, presença de árvores e/ou com detritos vegetais, ou com a macrófita Podostemum sp. (Podostemaceae) no fundo, e mais baixa em locais com curso semi-regulado. Possivelmente, fatores como temperatura e precipitação acumulada estejam relacionados com a abundância mensal dos Naucoridae. Os resultados deste estudo servirão de subsÃdio para futuros estudos de impacto ambiental após o enchimento do reservatório da U.H.E. Dona Francisca.
