Swine are susceptible to different mycobacteria species, being Mycobacterium bovis an agent of tuberculosis, with most significant zoonotic risks, while M. avium determines a granulomatous lymphadenitis with low zoonotic risk. Currently performed intradermal tests present some important limitations, such as the lack of ability to detect anergic animals or to differentiate among mycobacterial species. In order to improve the TB diagnosis, serological assays have been developed, with encouraging results. The purpose of this study was to evaluate the performance of a MPB70-ELISA in 82 piglets divided into four groups: sensitized by inactivated M. bovis, M. avium, inoculated with oil adjuvant, or with saline solution. The test was able to discriminate between an animal sensitized by M. bovis and animals of the three other groups, including M. avium-sensitized animals; for this reason, we suggest that MPB70-ELISA could be used as a complementary tool for discriminating the agent of the mycobacteriosis, and therefore to diagnose tuberculosis in a swine herd.

A herd infected naturally with tuberculosis was investigated by different diagnostic methods. Ninety days after a screening test that identified 21 cows as skin test positive, a Comparative Intradermal Tuberculin Test (CITT) was performed in those 21 cows and in 29 other randomly selected skin test negative cows. Milk samples and nasal swabs were collected prior to the CITT for bacteriological culture and PCR, while blood samples were collected for IFN release and antibody responses to MPB70 and MPB83, at three time points post tuberculin injection. Animals positive by CITT were slaughtered and disease confirmation undertaken. Based on the Kappa test, IFN was comparable to the standard tests (culture, PCR and CITT) at all three sampling points. Results from both antibody ELISAs were similar but were not comparable to the standard tests. T-test analysis of the CITT, IFN and ELISAs demonstrated that their performances were not correlated. There is increasing recognition that individually, available diagnostic tests do not detect all infected cattle. Therefore, a comprehensive strategy for the diagnosis of bovine TB should include test results for the detection of both cellular and humoral immune responses where there may be animals at different stages of infection.

O desempenho do ensaio por enzimas imuno-adsorvidas (ELISA), mediante o emprego de conjuntos de reativos produzidos em escala comercial, para determinação de aflatoxina, foi avaliado em condições experimentais, através de análises repetidas, em amostras de leite em pó reconstituído contaminadas com concentrações conhecidas da fração M1 da toxina. Para os níveis de 0,10; 0,20; 0,50; e, 1,00 ng/ml, os percentuais de recuperação foram: 83,0%; 87,5%; 103,0%; e, 111,8%, respectivamente. O desvio-padrão relativo, para as referidas concentrações, foi, respectivamente, 65,5%; 31,8%; 10,9% e 13,6% (n=10, para cada nível de contaminação). Os resultados obtidos demonstram que o método é apropriado para pesquisas e levantamentos sobre a ocorrência de aflatoxina M1 em leite, sobretudo nas faixas de concentração entre 0,20 - 1,00 ng/ml.

One of the main difficulties related to the detection of the Hepatitis Delta Virus (HDV) antigen and antibody has been the source of the needed HD antigen since HDV containing human and animal livers are very difficult to obtain and since yield is low. This fact prompted us to try to use the serum of patients in the acute phase of HDV infection as a source of HDAg and turn to enzyme immunoassays (EIA) instead of RIA for the sake of easiness and economy in the amount of HDAg needed. The antigen for EIA was obtained from patients during the acute phase of HDV infection and the antibody from patients who have been carriers for many years. For the detection of the antigen, a sandwich type method was employed, whereas for the antibody a competition assay was developed. In order to assess the relative specificity and sensibility of the test, the antibody assay was compared to a commercial RIA (C. RIA, Abbott) and to a non-commercial RIA (NC RIA). Forty-two sera were tested by the two methods and only in two cases discrepant results were obtained. Its is concluded that: 1) sera from patients in the acute and chronic phases of HDV infection can be used as source of both antigen and antibody, for immunoassays; 2) EIA and RIA have comparable relative specificity and sensibility and 3) EIA is easier to perform, cheaper, non-hazardous, has a longer shelf-life and saves scarce HDAg.

Foi avaliado o teste imunoenzimático (ELISA), para detecção de anticorpos da classe IgM na leptospirose humana. Nas amostras de soros analisadas, o teste ELISA demonstrou ser mais sensível, específico e precoce, quando comparado ao teste de soroaglutinação microscópica. A análise dos resultados obtidos nesta avaliação demonstra que o teste ELISA permite detectar níveis baixos de anticorpos circulantes, e também anticorpos não aglutinantes. não detectáveis através do teste de soroaglutinação microscópica.

Comunicamos nuestra experiencia con la aplicación del inmunoensayo enzimático al diagnóstico serológico de Plasmodium vivax con antígeno homólogo. Este se obtuvo a partir de una muestra de sangre de un paciente y luego de lisar los glóbulos rojos con detergente (NP-40). En un "pool" conformado con 11 sueros de pacientes con malaria por P. vivax se encontraron anticuerpos de las clases IgM, IgG e IgA. Se introdujo además la proteína A como sonda secundaria para determinar la respuesta anti-P. vivax en una muestra de 30 sueros de pacientes con un primer episodio palúdico. Se encontró un 93% de correlación con el diagnóstico parasitológico y la prueba resultó específica y reproducible.

Realizou-se estudo comparativo de diferentes sorotipos de Leptospira interrogans utilizados na preparação de antígenos empregados no teste ELISA, para a detecção de anticorpos da classe IgM, em amostras de soro na fase precoce e tardia da leptospirose humana. Foram utilizados dez sorotipos, escolhidos entre os que apresentaram maior reatividade na soroaglutinação microscópica (SAM), na cidade de São Paulo. Os cinco sorotipos que apresentaram melhores resultados individualmente no teste ELISA-IgM (canicola, hebdomadis, icterohaemorrhagiae, cynopteri e brasiliensis), foram também estudados em mistura antigênica. Os antígenos não tratados apresentaram maior reatividade do que os antígenos tratados com Triton X - 100 (4%) à temperatura de 50ºC, durante 4 horas. O teste ELISA-IgM empregando os sorotipos não tratados, isoladamente, e em mistura antigênica, mostrou-se altamente sensível, podendo ser empregado como teste de triagem para o diagnóstico precoce da leptospirose humana. Outra aplicação do teste é permitir a detecção do início de situações epidêmicas ou de surtos, possibilitando acionar medidas de vigilância epidemiológica.

A dot enzyme-linked immunosorbent assay (DOT-ELISA) was developed to detect specific antibodies in cerebrospinal fluid (CSF) for human neurocysticercosis immunodiagnosis, with Cysticercus cellulosae antigen dotted on a new solid-phase. This was represented by sheets of a synthetic polyester fabric impregnated with a polymerized resin (N-methylol-acrylamide). A very stable preparation was thus obtained, the antigen being covalently bound by cross-linking with free N-methylol groups on the resin. Since robust, no special care was necessary for handling the solid-phase. The test could be performed at room-temperature. From 30 CSF samples assayed, 14 were positive, from a group of 15 cases of neurocysticercosis, with titers from 1 to 128; 15 other samples, from normals or other neurological diseases, were all negative. Test characteristics seem to indicate it as adequate for epidemiological surveys. A more detailed study on sensitivity, specificity, reproducibility and the use in serum samples is being conducted.

Le test ELISA-TÉTANOS (Biosys, France) a été utilisé pour le titrage des anti-corps tétaniques (sensibilité = 0,0025 UI/ml) en sérums humains de donneurs de sang, 566 hommes et 108 femmes, âges de 18 à 58 ans, moyenne de 29 ans, provenant de São Paulo, SP, Brésil. L'OMS, acceptant seulement la séroneutralisation sur souris (NT), la méthode de référence, pour les études sur la protection contre le tétanos, préconise le titre de 0,01 UI/ml comme minimum protecteur. BOURLEAUD & HUET ont proposé la limite de 0,06 UI/ml quand s'emploie le test ELISA, en attendant à une certaine discordance inévitable entre les méthodes. Parmi les 674 sérums étudiés, 178 (26,41%) n'ont pas présenté d'anticorps (< 0,0025 UI/ml); 413 (61,28%) ont présenté des résultats égaux ou supérieurs à 0,01 UI/ml et en 232 (34,42%) les titres ont été égaux ou supérieurs à 0,06 UI/ml. Le pourcentage d'individus protégés a été inversement proportionnel à 1'âge: environ 50% dans le groupe le plus jeune (hommes de 18 à 35 ans et femmes de 18 à 23 ans) contre environ 10% dans le groupe de plus de 42 ans ont présenté des titres sûrement protecteurs (> 0,06 UI/ml).

An enzyme-linked immunosorbent assay (ELISA) for measles antibodies was compared with Plaque Neutralization (PRN), Haemagglutination inhibition (HI) and Fluorescent antibody (IFA) tests in 181 sera from vaccinated children and umbilical cord. Of 179 positive samples by the sensitive PRN, only two, with titers of 8, were negative by ELISA (copositivity of 98.9%). IFA and HI presented, respectively, copo-sitivities of 93.3% and 82.7%. The ELISA presented a high sensitivity as well as a good reproducibility and represents an alternative for the time consuming PRN for detection of low measles antibodies.

Para evaluar la utilidad del método de recolección de muestras de sangre en papel de filtro para la detección de anticuerpos anti-dengue mediante un ELISA de Inhibición recientemente desarrollado en nuestro laboratorio, se realizó una toma simultánea de muestras de sangre en papel de filtro y de suero, de donantes de Banco de Sangre. Ambas muestras fueron conservadas a -20ºC y probadas a los 15 dias, 3 y 6 meses respectivamente. A las muestras que resultaron positivos se les amplió el rango de dilución para determinar título. Al realizar la comparación entre ambas muestras, sangre en papel de filtro y suero, encontramos que no existían diferencias de detección significativas, tanto para los casos positivos como los negativos. No obstante se observó en ambas muestras y de forma general una disminución del titulo de anticuerpos (una dilución) al transcurrir el tiempo máximo establecido en nuestro estudio (6 meses).

Diagnostic performance indexes of sensitivity, specificity, positive predictive value and efficiency were determined for dot-ELISA and IgG-ELISA tests in 340 leishmaniasis sera. Sensitivity of the dot-ELISA was significantly lower than IgG-ELISA's; the two tests had indexes of specificity and positive predictive value of the same magnitude. Seventy-eight sera gave a negative dot-ELISA test result and a positive IgG-ELISA test result. When sera were classified according to different criteria as how to interpret this diversity, the kappa statistic did not corroborate the classification indicating that the two tests display a substantial strength of agreement. The results presented indicate that performance indexes accrued in a survey where variables arc well known may be extrapolated to other population studies if the disease presents itself as highly prevalent (due to a selection bias or not) and may be expected to discriminate a disease status among test positives.

The dot-enzyme-linked immunosorbent assay (dot-ELISA) was standardized using somatic (S) and excretory-secretory (ES) antigens of Toxocara-canis for the detection of specific antibodies in 22 serum samples from children aged 1 to 15 years, with clinical signs of toxocariasis. Fourteen serum samples from apparently normal individuals and 28 sera from patients with other pathologies were used as controls. All samples were used before and after absorption with Ascaris suum extract. When the results were evaluated in comparison with ELISA, the two tests were found to have similar sensitivity, but dot-ELISA was found to be more specific in the presence of the two antigens studied. Dot-ELISA proved to be effective for the diagnosis of human toxocariasis, presenting advantages in terms of yield, stability, time and ease of execution and low cost.

En una zona endémica de la República Argentina se llevó a cabo un ensayo de campo de la prueba inmunoenzimática ELISA para la detección de antígenos (cAg) y complejos inmunes circulantes (CIC) en sueros de pacientes chagásicos crónicos. Del total de 215 muestras de sangre analizadas, 51 fueron positivas para ELISA-CIC y 45 lo fueron para ELISA-cAg. De los 74 (34,32% de la población) sujetos considerados infectados con dos reacciones serológicas positivas, 49 (66,21%) presentaron CIC en suero, en tanto que en 43 (58,11%) de ellos se encontró cAg por ELISA. Solo en 2 casos serológicamente no reactivos, se detectaron inespecíficamente CIC y cAg. Dentro del grupo considerado no infectado, se observó reactividad inespecífica de bajo título por una de las pruebas serológicas en 16 (11,35%) de 141 individuos. Estos sueros arrojaron resultados consistentemente negativos por ELISA-CIC y cAg demostrando la utilidad de estos métodos de diagnóstico antigénico en casos de serología conflictiva. La determinación de fracciones antigénicas circulantes por ELISA en individuos chagásicos crónicos permite evidenciar la infección por T. cruzi de manera más directa que midiendo la respuesta inmune humoral en el huésped, presentando además mayor sensibilidad que el diagnóstico parasitológico clásico

An Enzyme-linked immunosorbent assay ELISA was evaluated for the detection of IgA antibodies in the human leptospirosis. The assay proved to be sensitive and specific when compared with the ELISA-IgM, in the examinated serum samples. The results found suggest that IgA antibodies became positive later in leptospirosis, and will can be an evolutive indicator in the development of the disease
