Os fungos ectomicorrízicos são capazes de tolerar concentrações de metais pesados tóxicas às plantas hospedeiras, apesar de serem adversamente influenciados pelo excesso de alguns metais. Avaliou-se o crescimento de um isolado de Pisolithus tinctorius e outro de Suillus bovinus em meio de cultura líquido com doses crescentes de sais de Zn, Cu ou Cd adicionados individualmente em frascos de 125 mL que continham 50 mL de meio Mellin-Norkrans modificado (MNM), em pH 4,8. Os fungos cresceram por 20 dias em câmara de crescimento a 28ºC. O crescimento dos fungos foi inibido com a elevação das concentrações dos metais, porém de forma diferenciada. As concentrações suficientes para inibir 50% do crescimento foram de 2,71 x 10-3 mol L-1 de Zn, 1,18 x 10-3 mol L-1 de Cu e 12,2 x 10-6 mol L-1 de Cd, para o P. tinctorius, e de 2,15 x 10-3 mol L-1 de Zn, 0,12 x 10-3 mol L-1 de Cu e 7,2 x 10-6 mol L-1 de Cd, para o S. bovinus. O efeito inibitório dos metais sobre o crescimento dos fungos seguiu a seguinte ordem decrescente: Cd > Cu > Zn. O isolado de S. bovinus apresentou tolerância ao Zn similar à observada para o P. tinctorius, mas foi menos tolerante que este em relação aos outros dois metais. O crescimento de P. tinctorius foi favorecido por pequena dose de Cu. A produção de pigmentos extracelulares nestes isolados foi estimulada por todos os metais estudados. O P. tinctorius, o mais tolerante, produziu mais pigmentos extracelulares por grama de micélio, o que sugere a relação positiva entre a capacidade de produção de pigmentos e a tolerância aos metais.

Alterações no pH da rizosfera de plantas fixadoras de N2 parecem exercer papel fundamental na absorção de micronutrientes que têm sua disponibilidade dependente de alterações da acidez do solo. Estudaram-se variações na absorção de B, Cu, Fe, Mn e Zn durante o ciclo de crescimento e desenvolvimento da soja, induzidas pela fixação biológica de N2 e pelo pH inicial de amostras de dois solos (um LV argiloso e outro arenoso), em um ensaio conduzido em casa de vegetação. Essas amostras foram incubadas com doses de CaCO3 + MgCO3 (4:1) para elevar o pH (H2O) a valores de 5,2, 5,6, 6,2 e 6,6 no solo argiloso e 5,3, 5,6, 5,9 e 6,3 no solo arenoso. Após 60 dias de incubação, essas amostras receberam 450 mg dm-3 de P e 120 mg dm-3 de K no solo. Sementes de soja (Glycine max (L) Merrill), variedade Paranaíba, inoculadas com Bradyrhizobium japonicum, estirpes SEMIA 587 e SEMIA 5019, foram colocadas para germinar. Foram cultivadas quatro plantas por vaso (2,2 dm³) e colhidas aos 16, 20, 24, 28, 32, 36, 40, 46 e 54 dias após a emergência. Determinaram-se o pH da rizosfera (pHr), o pH do solo entre raízes - não rizosférico (pHnr), os teores de B, Cu, Fe, Mn e de Zn na parte aérea e raiz, o N apenas na parte aérea, o número de nódulos e o peso da matéria seca de parte aérea, raiz e nódulos. Observou-se que as mudanças ocorridas no pHr e pHnr foram dependentes do pH inicial dos solos (pHs) e da fixação biológica de N2. O acúmulo de B e de Fe na parte aérea não foi alterado pelos valores de pHr, modificados em função do pHs, exceto para o Fe no solo argiloso. Todavia, aumentos significativos no acúmulo destes nutrientes na parte aérea ocorreram com o aparecimento dos nódulos, a partir de 24 dias após a emergência. Para Cu, Mn e Zn, as diferenças apareceram sobretudo quanto ao pHs. O conteúdo de micronutrientes na planta revelou-se sensível a mudanças no pH rizosférico, principalmente após a nodulação.

Optimization of the main parameters of SWASV using boron-doped diamond electrode was described for the simultaneous determination of Zn, Cd, Pb and Cu free in coconut water. The values of electroanalytical parameters studied were optimized with the factorial design and center composite design. The optimized parameters for the preconcentration of metals were -1.50 V for potential, and 240 s for deposition time. For SWV, the optimized value was 11.56 mV for step potential. In addition, frequency and pulse height were defined at 100 Hz and 55 mV, respectively. Furthermore, the concentration of the supporting electrolyte (acetate buffer, pH 4.7) was optimized in 0.206 mol L-1. The optimized procedure was applied in two samples of coconut water: natural and processed. The limits of detection (LOD) obtained for Zn, Cd, Pb and Cu were 7.2; 4.4; 3.3 and 1.5 µg L-1, respectively. The concentrations of Cd and Pb were not detected. On the other hand, the values found for the concentrations of Zn and Cu were: < LOD (29 µg L-1) and (6.8 ± 0.9) µg L-1 for the natural sample; and (85.8 ± 4.2) µg L-1 and (7.7 ± 0.6) µg L-1 for the processed sample, respectively.

Knowledge of the radiochemical purity of radiopharmaceuticals is mandatory and can be evaluated by several methods and techniques. Planar chromatography is the technique normally employed in nuclear medicine since it is simple, rapid and usually of low cost. There is no standard system for the chromatographic technique, but price, separation efficiency and short time for execution must be considered. We have studied an alternative system using common chromatographic stationary phase and alcohol or alcohol:chloroform mixtures as the mobile phase, using the lipophilic radiopharmaceutical [99mTc(MIBI)6]+ as a model. Whatman 1 modified phase paper and absolute ethanol, Whatman 1 paper and methanol:chloroform (25:75), Whatman 3MM paper and ethanol:chloroform (25:75), and the more expensive ITLC-SG and 1-propanol:chloroform (10:90) were suitable systems for the direct determination of radiochemical purity of [99mTc(MIBI)6]+ since impurities such as99mTc-reduced-hydrolyzed (RH),99mTcO4- and [99mTc(cysteine)2]-complex were completely separated from the radiopharmaceutical, which moved toward the front of chromatographic systems while impurities were retained at the origin. The time required for analysis was 4 to 15 min, which is appropriate for nuclear medicine routines.

ABSTRACT OBJECTIVE To estimate the prevalences of tobacco use, tobacco experimentation, and frequent smoking among Brazilian adolescents. METHODS We evaluated participants of the cross-sectional, nation-wide, school-based Study of Cardiovascular Risks in Adolescents (ERICA), which included 12- to 17-year-old adolescents from municipalities of over 100 thousand inhabitants. The study sample had a clustered, stratified design and was representative of the whole country, its geographical regions, and all 27 state capitals. The information was obtained with self-administered questionnaires. Tobacco experimentation was defined as having tried cigarettes at least once in life. Adolescents who had smoked on at least one day over the previous 30 days were considered current cigarette smokers. Having smoked cigarettes for at least seven consecutive days was an indicator for regular consumption of tobacco. Considering the complex sampling design, prevalences and 95% confidence intervals were estimated according to sociodemographic and socio-environmental characteristics. RESULTS We evaluated 74,589 adolescents. Among these, 18.5% (95%CI 17.7-19.4) had smoked at least once in life, 5.7% (95%CI 5.3-6.2) smoked at the time of the research, and 2.5% (95%CI 2.2-2.8) smoked often. Adolescents aged 15 to 17 years had higher prevalences for all indicators than those aged 12 to 14 years. The prevalences did not differ significantly between sexes. The highest prevalences were found in the South region and the lowest ones, in the Northeast region. Regardless of sex, the prevalences were found to be higher for adolescents who had had paid jobs, who lived with only one parent, and who reported having been in contact with smokers either inside or outside their homes. Female public school adolescents were found to smoke more than the ones from private schools. CONCLUSIONS Tobacco use among adolescents is still a challenge. Intending to reduce the prevalence of tobacco use among young people, especially the ones under socioeconomic vulnerability conditions, Brazil must consolidate and increase effective public health care measures.

Cerebrospinal fluid (CSF) samples from 2083 patients with acquired immunodeficiency syndrome (AIDS) and neurological complications were bacteriologically examined during a period of 7 years (1984-1990). The percentage of patients who had at least one bacterial agent cultured from the CSF was 6.2%. Mycobacterium tuberculosis was the most frequently isolated agent (4.3%), followed by Mycobacterium avium complex or MAC (0.7%), Pseudomonas spp (0.5%), Enterobacter spp (0.4%), and Staphylococcus aureus (0.3%). Among 130 culture positive patients, 89 (68.5%) had M. tuberculosis and 15 (11.6%) had MAC. The frequency of bacterial isolations increased from 1988 (5.2%) to 1990 (7.2%), partly due to the increase in MAC isolations. Bacterial agents were more frequently isolated from patients in the age group 21-30 years and from women (p<0.05).

Trypanosoma cruzi trypomastigotes excrete-secrete a complex mixture of antigenic molecules. This antigenic mixture denominated trypomastigote excreted-secreted antigens contains a 150-160 kDa band that shows excellent performance in Chagas' disease diagnosis by immunoblotting. The present study partially characterized by two-dimensional gel electrophoresis the immunoreactivity against the 150-160kDa protein using sera samples from chagasic patients in different phases of the disease. Trypomastigote excreted-secreted antigen preparations were subjected to high-resolution two-dimensional (2D) gel electrophoresis followed by immunoblotting with sera from chagasic and non-chagasic patients. The 150-160kDa protein presented four isoforms with isoelectric focusing ranging from 6.2 to 6.7. The four isoforms were recognized by IgM from acute phase and IgG from chronic phase sera of chagasic patients. The 150-160kDa isoform with IF of approximately 6.4 became the immunodominant spot with the progression of the disease. No cross-reactivity was observed with non-chagasic or patients infected with Leishmania sp. In this study we provide basic knowledge that supports the validation of trypomastigote excreted-secreted antigens for serological diagnosis of Chagas' disease.

INTRODUCTION: Human T cell lymphotropic virus types 1 and 2 (HTLV-1/2) are endemic in Brazil and are screened for in transfusion services since 1993. This study evaluated the evolution of the prevalence of HTLV-1 and 2 in blood donors of the Hemominas Foundation from 1993 to 2007, and its geographical distribution in State of Minas Gerais, Brazil. METHODS: The Hemominas Foundation is a centralized blood center in Minas Gerais, Brazil. The sources of data were the Hemominas Foundation Technical Bulletin and files from the centralized serological laboratory. Donors were tested in the period using enzyme linked immuno sorbent assays (ELISA), followed by Western blot, when repeatedly reactive. The data were analyzed by EPIINFO 6.2 and TABWIN 3.5 softwares. RESULTS: The average seroprevalence in the period 1993-2007 was 0.1%. A steady decline occurred from 0.4% in 1993 to below 0.1% in 2002 and later, with a transient peak of 0.5% in 1994. HTLV reactivity distribution was asymmetrical in the state, with regions of higher prevalence, interspersed with low prevalence areas. Comparison of positive and negative donors verified that increasing age was proportional to virus positivity. Odds ratio for age ranged from 1.43 (30 to 39 years-old) to 3.09 (50 to 65 years-old). Women had a greater chance of being positive (OR-1.64), as previously described. CONCLUSIONS: Possible explanations for HTLV-1/2 prevalence decline are the exclusion of positive donors from the donor pool, an increase in repeat donors and ELISA test improvement, with reduction in the number of false positive results.

Na presente nota prévia os autores apresentam os primeiros dados acerca da extração e exportação de nutrientes através do cajueiro a partir de 1 a 14 anos de idade. As plantas foram colhidas de uma plantação localizada no Município de Terezinha, BA, situadas em um latossolo de baixa fertilidade, exceto em K. As plantas foram pesadas e analisadas para N, P, K, Ca, Mg, S, B, Cu, Fe, Mn e Zn. Constataram os autores que o cajueiro cresce continuamente até aos 14 anos. Uma árvore aos 10 anos de idade contém: N- 1,5 kg; P- 0,2 kg; K- 1,1 kg; Ca-0,2 kg;Mg-0,3 kg; S- 0,1 kg; B- 4,0 g;Cu-1,4 g;Fe- 2,0 g;Mn- 7,8 g;Zn- 2,6 g. Aquilataram os autores que a exportação de nutrientes através de 1 kg de frutos é a seguinte: N- 13,9 g; P- 1,3 g; K- 6,2 g; Ca- 0,3 g; Mg- 1,3 g; S- 0,5 g; B-18,5 mg; Cu-16,2 mg; Fe- 36,9 mg; Mn-13,2 mg. Finalmente observaram que a concentração de N, Mg, S, Cu, Fe e Zn não varia nas folhas em função da idade da planta; sendo que a concentração em P, K, Ca, B e Mn varia com a idade da planta.

De uma bem conduzida plantação de rúcula (Euruca sativa var. Cultivada) foram coletadas plantas aos 20, 27, 34 e 41 dias de idade. As plantas foram lavadas e divididas em folhas e raízes. Postas a secar a 80°C e analisadas para N, P, K, Ca, Mg, S, B, Cu, Fe , Mn e Zn pelos métodos convencionais de laboratório. Observou-se que o crescimento, expresso em produção de matéria seca, é crescente até os 34 dias, estabilizando-se após este período. A análise química mostrou que a concentração de K e Ca é elevada atingindo 2,19% para este último elemento aos 41 dias de idade da planta. Para os micronutrientes chama atenção a elevada concentração de zinco nas folhas atingindo 229 ppm aos 41 dias de idade. Uma área cultivada de 10 m² extrai pelas folhas , que é parte exportada as seguintes quantidades de elementos: 20 g de N, 1,86 g de P, 22,1 g de K, 7,56 g de Ca , 1,77 g de Mg, 1,99 g de S, 17,7 mg de B, 7,8 mg de Cu , 475,6 mg de Fe, 14,3 mg de Mn e 37,7 mg de Zn.

The serology of human Trypanosoma cruziinfection in the Rio Negro microregion is very complex because of the large numbers of false-positive cases that result from low antibody titres and cross-reactions with other infections. In the present study, we collected 4,880 blood samples on filter paper; of these, indirect immunofluorescence (IIF) was strongly reactive in 221 (4.5%), which were considered to be positive (IIF strongly reactive; high intensity of fluorescence) and weakly reactive in 302 (6.2%), which were considered to be doubtful (IIF weakly reactive; low intensity of fluorescence). The confirmatory test on the serum using at least two of three techniques (IIF, conventional ELISA and recombinant ELISA) on 137 samples that were positive in the screening test only confirmed 33 cases (24.1%). Of the 178 samples that were considered doubtful in the screening test, only 10 (5.6%) were considered to be positive in the confirmatory test. Finally, we recommend that the serological diagnosis of T. cruziinfection in the Amazon region be made using at least two different techniques, for example immunofluorescence and ELISA and confirmed by Western blot analysis when possible.

Cp'Mn(CO)3 (Cp'=h5-C5H4-CH 3) reacts with P(C6H5)2H in THF to give Cp'(CO)2MnPPh2H (Ph = Phenyl) (1), by substitution of one CO ligand. The reaction of 1 with CH3COCl and CH3S(O)2Cl in the presence of triethylamine occurs under electrophilic substitution on the diphenylphosphan ligand to yield the acetyl- and sulfonylphosphane complexes of manganese(I) Cp'(CO)2MnPPh2COCH3 (2) and Cp'(CO)2MnPPh2S(O)2 CH3 (3). The complex stabilisation of these molecules, wich are hitherto unknown in the free state, is only accomplished by blocking the free electron pair on phosphorus by coordination. The new complexes 1, 2 and 3 were analysed by IR, ¹H-NMR and 31P-MNR spectroscopy and their similar structures are discusssed.

The sequential extraction procedure proposed by Tessier and total digestion were applied for the analysis of sediment samples from Tubarão River. The recoveries were between 93.5 and 102.5% for Cu and 99.2 and 111% for Pb. The precision was tipically better than 6% for Cu and 3% for Pb. Comparison of the values obtained for the total digest with the sum of the extracted fractions showed that there were no significant losses in the extraction steps so that the method can be used for the monitoring availability and mobility of these analytes. According to ten points of sampling, was possible to determined the average of labiles phases (fractions 1 and 4) for Pb and Cu: 33.70% and 18.18%, respectively; and the inert phases (fraction 5- residual): 66.30% for Pb and 81.82% for Cu.

A method for determining copper by solid phase spectrophotometry (SPS) was optimized using the Doehlert design. Copper(II) was sorbed on a styrene-divinylbenzene anion-exchange resin as a Cu(II)-1-(2-pyridylazo)-2-naphthol (PAN) complex, at pH 7.0. Resin phase absorbances at 560 and 800 nm were measured directly. The detection limit was found to be 2.5 µg L-1. The relative standard deviation on ten replicate determinations of 10 µg Cu(II) in 1000 mL samples was 1.1%. The linear range of the determination was 5.0-100 µg L-1. The method was applied successfully to the determination of Cu(II) in natural water and vegetable samples.

A flow system based on the sandwich technique is proposed for the sequential determination of ascorbic acid, dipyrone, acetylcysteine, captopril and paracetamol. The procedure is based on the reduction of Cu(II) by the analytes followed by the spectrophotometric measurement of the complex of Cu(I) with 2,2'-biquinoline 4,4'-dicarboxylic acid. Linear responses were achieved in the µmol L-1 range, with coefficients of variation better than 1.7%. Sampling rate was estimated as 60 determinations per hour, consuming 230 µg of BQA and generating 2.5 mL of waste per determination. Results for commercial samples agreed with those obtained by procedures recommended by the American and European pharmacopeias at the 95% confidence level.
