Este estudo teve como objetivo averiguar o conhecimento dos alunos da 5a série das escolas públicas estaduais de Manaus sobre a floresta amazônica e seus recursos. As entrevistas foram realizadas em 14 escolas, com o auxílio de um questionário padrão contendo perguntas abertas. De cada escola foram entrevistados 10 alunos, perfazendo um total de 140 no final da pesquisa. Os resultados mostraram a necessidade de enfatizar nos currículos escolares, assuntos sobre a região amazônica.

The composite membranes prepared via incorporation of 12.5% of molecular sieves 3A, 4A, 5A and 13X into chitosan/poly(vinyl alcohol) (1:1). The composite membranes were immersed in the cross-linker sulfuric acid in order to acquire high proton conductivity for applications in electrolytes in PEMCF and DMF. The influence of the molecular sieves on the structural, optical, thermal, mechanical, morphologic and protonic conductivity properties and water/methanol swelling degree of membranes were investigated.

ABSTRACT This paper studied the effect of adding an enzyme (ellulose) on anaerobic digestion of maize silage. We compared materials at chopping lengths of 8 mm (MSL), 4mm (MSS) and natural size (Ms) under a mesophilic and discontinuous operation (batch process). Hence, we found the process to be significantly influenced by particle size. Moreover, the ellulose addition did not significantly impact biogas production after a 35-day digestion period. Ms and MSS displayed an improved response to all variables when compared with MSL and MSL+C, with significant differences. Studies on the refractory fraction at infinite time (R0) have demonstrated that the lowest values correspond to Ms and MSS (0.122 and 0.155, respectively). The Kinetic approach and the Ultimate Biodegradability test are useful tools to evaluate the effect of the addition of an enzyme to the anaerobic process.

Invasive bacteria can induce their own uptake and specify their intracellular localization; hence it is commonly assumed that proximate modulation of host cell transcription is not required for infection. However, bacteria can also modulate, directly or indirectly, the transcription of many host cell genes, whose role in the infection may be difficult to determine by global gene expression. Is the host cell nucleus proximately required for intracellular infection and, if so, for which pathogens and at what stages of infection? Enucleated cells were previously infected with Toxoplasma gondii, Chlamydia psittaci, C. trachomatis, or Rickettsia prowazekii. We enucleated L929 mouse fibroblasts by centrifugation in the presence of cytochalasin B, and compared the infection with Shigella flexneri M90T 5a of nucleated and enucleated cells. Percent infection and bacterial loads were estimated with a gentamicin suppression assay in cultures fixed and stained at different times after infection. Enucleation reduced by about half the percent of infected cells, a finding that may reflect the reduced endocytic ability of L929 cytoplasts. However, average numbers of bacteria and frequency distributions of bacterial numbers per cell at different times were similar in enucleated and nucleated cells. Bacteria with actin-rich tails were detected in both cytoplasts and nucleated cells. Lastly, cytoplasts were similarly infected 2 and 24 h after enucleation, suggesting that short-lived mRNAs were not involved in the infection. Productive S. flexneri infection could thus take place in cells unable to modulate gene transcription, RNA processing, or nucleus-dependent signaling cascades.

O vírus rábico RV194-2, uma variante avirulenta da cepa CVS (Challenge Vírus Standard), produz uma infecção inaparente quando inoculado intracerebralmente em camundongos adultos. Sugerindo que a resposta imunológica do hospedeiro permite a eliminação do vírus do sistema nervoso central. Por esta razão foram estudadas a indução de interferon e a resposta imune humoral em camundongos BALB/c inoculados com vírus RV194-2. Durante a infecção, estes camundongos apresentaram elevados níveis de interferon no plasma e no cérebro com altos títulos de anticorpos neutralizantes anti-rábicos. A 2-5A sintetase. um marcador da ação dos interferons,foi também analisada no cérebro destes animais. Sua atividade, aumentou, paralelamente, á produção de interferon, demonstrando que este interferon é bioquímicamente ativo. O vírus RV194-2 também induziu, 45 dias após sua inoculação, proteção aos animais quando desafiados com a cepa virulenta CVS. Estes resultados demonstram que a cepa RV194-2possui um alto nível imunogênico.

Avaliações preliminares sobre uma pintura inseticida à base de diazinon, clorpirifós e piriproxifen em formulação micro-encapsulada (Inesfly 5A IGR ®) mostrou efetiva e persistente atividade contra Triatoma infestans intra e peridomiciliar, numa região altamente infestada do Chaco Boliviano. Ressaltam, além disso, a boa manuseabilidade do produto e o bom aspecto deixado pela pintura em casas e anexos tratados, bem como uma excelente aceitação pela população e autoridades sanitárias locais, o que estimula novas investigações e o emprego do produto em maior escala e contra outros vetores da doença de Chagas.

In order to study the action of herbicides - sodium salt, amine salt and ester of 2,4-D, TCA and 2,4,5-T a preliminary experiment for pre-emergence weed control was corried out, and the corresponding results are given in table I and II. The corn used in the experiments was of the flint type 1A 3531. The loam soil on which the experiment has been carried out is called "terra roxa". All treatments were highly significant when compared with the check plots, except the 2B one in the control of broad leaf weeds, and 4B in the control of grass weeds. Among these treatments there are no significant differences. But we note the following: (table I). a) treatments of higher concentrations were superior to lower ones. b) the treatments which gave the best control for broad leaf weeds were in the following decreasing order: 1A, 5A and 3A. For grass weeds, they were 5A, 1A and 3A. c) the amine 2,4-D (600 grs. per hectare) supplied very good control when we get into consideration that on the acid basis, it was in very low concentration. d) TCA in high concentration affected the germination, growth and yield, in the lower one it did not show good control of weeds, especially of grasses. It is not suitable for pre-emergence control in corn. e) 2,4,5-T was not better than the 2,4-D products. As it is much more expensive than the others, economically its use in pre-emergence weed control in corn is not praticable. f) all the products used controled grass weeds as well as broad leaf ones; this show the superiority of the pre-emergence treatment method over that of post-emergence. g) Even a dose as strong as the treatment 1A (3.400g. of 2,4-D acid per hectare) did not damage corn production (table II). h) the superiority noted in the production of all the treatments with the exception of 2A, which damaged the plants, we atribute to the lack of competion between corn and weeds; all chek-plots suffered this competition, because they were not Probably, there was, also, hormonial effect of 2,4-D on the corn plant. Not withstanding the fact that the present experiment has been successful, we think that new researches are necessary, especially with the purpose of studying factors as climate and soil which in other countries, interferred with the success of the pre-emergence weed control.

Em 20 portadores de Lepra, internados na Colônia de São Roque, Paraná, 13 dos quais eram da forma lepromatosa e os 7 restantes da forma nervosa pura ou tuberculoide, o autor estudou a reação intradérmica de antigenos bacilares (Leprolinas Souza-Araujo 1, 1a, 3, 5, 5a), comparativamente com as reações produzidas, ao mesmo tempo, com antígeno lepromatoso (Emulsão de lepromas, lepromina). Observou o seguinte: 1.° - Que a lepromina, comportou-se como habitualmente, isto é, negativa nos lepromatosos e ositiva nos nervosos e tuberculoides e sempre mais intensa ao fim da quarta semana. 2.° - Que as leprolinas provocam reações, que o autor não conclui serem positivas ou negativas no sentido imunológico da expressão em virtude de estar estudando-as neste sentido, nos leprotomatosos e tuberculoides, mas, sempre mais intensas entre a 1.ª e 2.ª semanas, diminuindo de intensidade na 3.ª e desaparecendo ou quase desaparecendo ao fim da quarta semana: nódulos, nódulos-eritema, ulcerações. 3.° - Que nestas reações houve destruição de tecidos, pois a presença tardia de cicatrizes, mesmo quando não houve ulceração, mostra êste fato. 4.° - Que as inoculações das lepromina, não trouxeram nenhum inconveniente a saúde dos paciente; embora um deles tenha apresentado nos primeiros dias leve reação geral; outro, intensa fusão de suas lesões nodulars de lepra e, dois outros, a chamada Reação leprótica.

By employing 4-methylumbelliferyl-beta-D-NN',N"-triacetylchitotriose substrate in a semi quantitative assay, chitinolytic activity in viable spores of Encephalitozoon cuniculi and E. intestinalis was detected and dependence on reaction time, spore concentration, concentration of substrate and temperature were demonstrated. It was possible to block the chitinolytic activity by chitin hydrolysate. By incubation at 80°C for 10 min or at 55°C for 20 min the spores were loosing the chitinolytic activity. Incubation of the spores in trypsin reduced the chitinolytic activity. Cellulase activity could not be detected.

The Bolivian Chaco is part of the endemic region of Chagas disease and an area where pyrethroid resistant Triatoma infestans (Hemiptera: Reduviidae) populations has been reported. The World Health Organization identified these resistant populations as an important focus for research. The objective of this study was to evaluate the residual effect of a micro-encapsulated formulation containing organophosphate active ingredients and a juvenile hormone analogue (Inesfly 5A IGR) on the mortality of T. infestans. Studies took place in rural houses of the Bolivian Chaco that were treated up to 34 months before and evaluated the susceptibility to pyrethroids of the offspring of field collected insects. Thirty houses were randomly selected within three communities to carry out wall bio-assays with T. infestans nymphs. Mortality was recorded 24, 48 and 72 h after wall contact. Eggs laid by females collected in the area were used to obtain first-instar nymphs and carry out pyrethroid susceptibility tests. The wall bio-assays showed that the micro-encapsulated insecticide eliminates T. infestans populations and produces detectable mortality of insects exposed to walls treated 34 months prior to the tests. The discriminant dose of deltamethrin (0.01 mg/mL) showed 65% nymph survival, whereas at the highest tested dose (1.0 mg/mL) 14% of the nymphs survived. These results show that Inesfly 5A IGR is an appropriate tool for the elimination of intradomestic and peridomestic populations of T. infestans resistant to pyrethroids.

Enzymatic activity is an important property for soil quality evaluation. Two sequences of experiments were carried out in order to evaluate the enzymatic activity in a soil (Rhodic Eutrudox) amended with cattle manure, earthworm casts, or sewage sludges from the municipalities of Barueri and Franca. The activity of commercial enzymes was measured by microcalorimetry in the same soil samples after sterilization. In the first experiment, the enzyme activities of cellulase, protease, and urease were determined in the soil samples during a three month period. In the second sequence of experiments, the thermal effect of the commercial enzymes cellulase, protease, and urease on sterilized soil samples under the same tretaments was monitored for a period of 46 days. The experimental design was randomized and arranged as factorial scheme in five treatments x seven samplings with five replications. The treatment effects were statistically evaluated by one-way analysis of variance. Tukey´s test was used to compare means at p < 0.05. The presence of different sources of organic residues increased the enzymatic activity in the sampling period. Cattle manure induced the highest enzymatic activity, followed by municipal sewage sludge, whereas earthworm casts induced the lowest activity, but differed from control treatment. The thermal effect on the enzyme activity of commercial cellulase, protease, and urease showed a variety of time peaks. These values probably oscillated due to soil physical-chemical factors affecting the enzyme activity on the residues.

Integrated crop-livestock systems (ICLs) are a viable strategy for the recovery and maintenance of soil characteristics. In the present study, an ICL experiment was conducted by the Instituto Agronômico do Paraná in the municipality of Xambre, Parana (PR), Brazil, to evaluate the effects of various grazing intensities. The objective of the present study was to quantify the levels of microbial biomass carbon (MBC) and soil enzymatic activity in an ICL of soybean (summer) and Brachiaria ruziziensis (winter), with B. ruziziensis subjected to various grazing intensities. Treatments consisted of varying pasture heights and grazing intensities (GI): 10, 20, 30, and 40 cm (GI-10, GI-20, GI-30, and GI-40, respectively) and a no grazing (NG) control. The microbial characteristics analysed were MBC, microbial respiration (MR), metabolic quotient (qCO2), the activities of acid phosphatase, β-glucosidase, arylsuphatase, and cellulase, and fluorescein diacetate (FDA) hydrolysis. Following the second grazing cycle, the GI-20 treatment (20-cm - moderate) grazing intensity) contained the highest MBC concentrations and lowest qCO2 concentrations. Following the second soybean cycle, the treatment with the highest grazing intensity (GI-10) contained the lowest MBC concentration. Soil MBC concentrations in the pasture were favoured by the introduction of animals to the system. High grazing intensity (10-cm pasture height) during the pasture cycle may cause a decrease in soil MBC and have a negative effect on the microbial biomass during the succeeding crop. Of all the enzymes analyzed, only arylsuphatase and cellulase activities were altered by ICL management, with differences between the moderate grazing intensity (GI-20) and no grazing (NG) treatments.

A procedure is described to regenerate plants from protoplasts of Brazilian citrus cultivars, after isolation, fusion and culture. Protoplasts were isolated from embryogenic cell suspension cultures and from leaf mesophyll of seedlings germinated in vitro. The enzyme solution for protoplast isolation was composed of mannitol (0.7 M), CaCl2 (24.5 mM), NaH2PO4 (0.92 mM), MES (6.15 mM), cellulase (Onozuka RS - Yakult, 1%), macerase (Onozuka R10 - Yakult, 1%) and pectolyase Y-23 (Seishin, 0.2%). Protoplast culture in liquid medium after chemical fusion lead to the formation of callus colonies further adapted to solid medium. Somatic embryo formation occurred spontaneously after two subcultures, on modified MT medium supplemented with 500 mg/L of malt extract. Well defined embryos were germinated in modified MT medium with addition of GA3 (2.0 muM) and malt extract (500 mg/L). Plant regeneration was also achieved by adventitious shoots obtained through direct organogenesis of not well defined embryos in modified MT medium with addition of malt extract (500 mg/L), BAP (1.32 muM), NAA (1.07 muM) and coconut water (10 mL/L). Plantlets were transferred to root medium. Rooted plants were transferred to a greenhouse for further adaptation and development.

The objective of this work was to identify growth-promoting bacteria isolated from Agaricus blazei and to evaluate their effect on mushroom mycelial growth and productivity. A total of 56 A. blazei-associated bacterial isolates were obtained from casing soil and identified by 16S rRNA gene sequencing. Bacteria were evaluated as to phosphate-solubilization ability, nitrogen-fixation capability, and secretion of cellulase. Superior isolates were tested for their to effect on A. blazei productivity, micelial growth, and on the contents of the polysaccharide-protein complex and of N, P, K, Ca, and Mg. Bacterial isolates were identified as actinobacteria (60%), firmicutes (20%), and proteobacteria (20%). Among them, ten isolates had phosphate-solubilization ability, eight showed nitrogen-fixation capability, and 12 isolates promoted A. blazei mycelium growth. Bacterial inoculation reduces time till harvest in up to 26 days, increases fresh mushroom yield up to 215%, and increases total polysaccharide-protein complex content twofold when compared to the non-inoculated control. The actinobacteria group is the predominant A. blazei-associated phylum.

O objetivo deste trabalho foi validar 19 marcadores microssatélites para resistência do trigo à giberela, em uma população não estruturada. Foram utilizados marcadores moleculares descritos na literatura como flanqueando QTLs de resistência à giberela em trigo, nos cromossomos 3B, 5A e 6B. Foram avaliadas 96 linhagens e cultivares de trigo quanto à severidade da infecção por giberela, em dois anos de avaliação. As linhagens e as cultivares foram genotipadas com 19 marcadores microssatélites. Os dados obtidos foram analisados pelo teste de Tukey e pelas análises de correlação, regressão linear simples e regressão múltipla; também foi estimada a eficiência de seleção dos marcadores moleculares. A severidade da doença variou de 1,95 a 41,3%, na média dos dois anos. Foram validados os QTLs nos três cromossomos avaliados. Os marcadores Xgwm389, Xgwm533, Xbarc180, Xbarc24, Wmc397, Xbarc101 e Wmc398 foram associados significativamente à resistência do trigo à giberela, tendo sido identificados alelos de resistência e de suscetibilidade. Os marcadores Wmc397, Xbarc101 (cromossomo 6B) e Xbarc180 (cromossomo 5A) têm potencial para uso na seleção assistida por marcadores moleculares, para resistência do trigo à giberela.
