Para se detectar diferenças imuno-antigênicas entre 8 amostras de P. brasiliensis isoladas de diferentes áreas endêmicas (Botucatu: Pb 1, 2 e 3; São Paulo: Pb: 18, 192 e 265; Venezuela: Pb 9 e 73), esutdaram-se: 1. A reatividade antigênica de cada amostra nas reações de imunofluorescência indireta (II) e de imunodifusão dupla em gel de agar (ID) contra painel de 20 soros controles positivos para paracoccidioidomicose; 2. A capacidade de induzir resposta imune humoral (medida por imunodifusão) e celular (medida pelo teste de coxim plantar) em camundongos imunizados com an-tígenos de cada amostra. Observamos: 1. As amostras Pb 265 e Pb 9 mostraram-se mais reativas na II; 2. Os antígenos das amostras Pb 192 e Pb 73 foram significativamente mais reativas na ID; 3. Estes dados demonstram diferenças de antigenicidade entre estas amostras; 4. A amostra Pb 18 mostrou baixo poder indutor de resposta imune celular e alta capacidade de indução de resposta imune humoral em camundongos imunizados, revelando dissociação de sua imunogenicidade. Estas diferenças podem indicar a existência de cepas distintas do fungo ou refletir modificações do parasita no hospedeiro ou du rante seu cultivo.

The experimental model of paracoccidioidomycosis induced in mice by the intravenous injection of yeast-forms of P. brasiliensis (Bt2 strain; 1 x 10(6) viable fungi/animal) was used to evaluate sequentially 2, 4, 8, 16 and 20 weeks after inoculation: 1. The presence of immunoglobulins and C3 in the pulmonary granuloma-ta, by direct immunofluorescence; 2. The humoral (immunodiffusion test) and the cellular (footpad sweeling test) immune response; 3. The histopathology of lesions. The cell-immune response was positive since week 2, showing a transitory depression at week 16. Specific antibodies were first detected at week 4 and peaked at week 16. At histology, epithelioid granulomas with numerous fungi and polymorphonuclear agreggates were seen. The lungs showed progressive involvement up to week 16, with little decrease at week 20. From week 2 on, there were deposits of IgG and C3 around fungal walls within the granulomas and IgG stained cells among the mononuclear cell peripheral halo. Interstitital immunoglobulins and C3 deposits in the granulomas were not letected. IgG and C3 seen to play an early an important role in. the host defenses against P. brasiliensis by possibly cooperating in the killing of parasites and blocking the antigenic diffusion.

PURPOSE: To study quantitatively C cells in the thyroids of non-isogenic rats to determine the possible effects of pinealectomy on the number of these cells, and consequently on the synthesis and secretion of calcitonin. METHODS: Twenty male rats of an outbred strain (200-300 g) were used in the present study. One group of 10 animals was pinealectomized 50 days prior to sacrifice. Thyroid tissue was stained for calcitonin (Dako Corporation) at a 1:1500 dilution. The number of C cells observed was expressed as number of cells/cm². Data were analyzed statistically by Mann-Whitney test. RESULTS: The number of C cells in pinealectomized and normal animals ranged from 489 to 2084 per cm² and 227 to 1584 per cm², respectively, a difference that was statistically significant (P <0.05). CONCLUSIONS: These results showed consistent differences in the number of C cells after pinealectomy when compared to controls. We believe that pinealectomy increases the number of C cells in the rat thyroid.

FUNDAMENTO: A insuficiência cardíaca (IC) cursa com frequentes descompensações e admissões ao serviço de emergência. Vacinação contra Influenza (INF) e Pneumococo (PNM) são recomendadas nas diretrizes, entretanto, as infecções respiratórias são a terceira causa de hospitalização na IC. OBJETIVO: Avaliar a frequência da vacinação contra INF e PNM em pacientes com IC na rede pública. MÉTODOS: Em estudo observacional realizado em Teresópolis, região serrana fluminense, foram utilizadas três estratégias: (I) estudo das requisições para vacina contra INF e/ou PNM na Secretaria Municipal de Saúde, entre 2004 e 2006; (II) inquérito direto a 61 pacientes com IC atendidos na atenção básica sobre sua situação vacinal contra INF e PNM; (III) inquérito direto sobre situação vacinal contra INF e PNM a 81 pacientes com IC crônica descompensada atendidos na única emergência aberta à rede pública. RESULTADOS: Na estratégia I, a vacinação contra INF e/ou PNM foi de 15,3% daqueles com indicações por doenças cardiovasculares e respiratórias. A mediana do tempo entre a indicação e a vacinação foi de 32 dias. Na estratégia II, o percentual de vacinados contra INF, com idade > 60 anos, foi de 23,1%, e de 24,6% contra PMN em todas as idades. Na estratégia III, o percentual de pacientes vacinados contra INF foi de 35,8% e contra PNM foi de 2,5%. CONCLUSÃO: A taxa de vacinação contra INF e PNM em pacientes com IC é muito baixa e ainda menor naqueles descompensados atendidos em serviço de emergência.

Este estudo teve como objetivo caracterizar e comparar a alimentação de 13 espécies de peixes em três zonas (zona de influência fluvial, zona de transição e zona lacustre) do reservatório de Itupararanga. Foram analisados 436 estômagos, sendo encontrados 17 recursos alimentares, todos de origem autóctone. As dietas predominantes das espécies foram: peixe, inseto, material vegetal e detritos na zona de influência fluvial, peixe, sedimento e material vegetal na zona de transição e inseto, escama e peixe na zona lacustre. Através do Índice Alimentar, as espécies foram categorizadas nas guildas: piscívora, insetívora, detritívora e carcinófaga. O Cluster indicou separação da guilda insetívora em espécies generalistas (insetos diversos) e especialista (Chironomidae) o que aumenta para cinco guildas. Na zona fluvial foram verificadas quatro guildas; na zona de transição três guildas e na zona lacustre duas guildas. Observou-se ainda uma dominância de espécies piscívoras que pode estar relacionada à idade do reservatório. Este estudo ampliou o conhecimento dos hábitos alimentares dos peixes em reservatórios antigos e suas diferenças, comparando as zonas existentes.

Valores determinados em laboratório, de vazão, raio de alcance e do perfil radial de aplicação de água do aspersor canhão PLONA-RL400, foram utilizados em simulações digitais da uniformidade de aplicação de água desse aspersor operando, na ausência de ventos, com diferentes ângulos de giro e espaçamentos entre carreadores, em sistemas autopropelidos de irrigação. Os valores simulados de uniformidade de aplicação de água foram apresentados em três grupos distintos, cada um dos quais representando condições operacionais (bocal e pressão) que determinam a ocorrência da mesma forma geométrica (I, II ou III) do perfil radial adimensional de aplicação de água do aspersor avaliado. Para os perfis do tipo I, II e III, observou-se que espaçamentos de carreadores menores que 50% do diâmetro molhado ou situados entre 80 e 90% do diâmetro molhado proporcionaram os maiores valores de uniformidade. Em todas as formas geométricas do perfil, os melhores valores de uniformidade de aplicação de água foram obtidos com ângulos de giro do aspersor entre 180 e 210º.

Apolipoprotein E (apoE - e2, e3, e4 alleles) plays a role in the regulation of lipid metabolism, with the e4 considered to be a risk factor for coronary artery disease (CAD). We aimed to evaluate the apoE polymorphisms in Brazilians with CAD and their influence on the lipid profile and other risk factors (hypertension, diabetes mellitus, smoking). Two hundred individuals were examined: 100 patients with atherosclerosis confirmed by coronary angiography and 100 controls. Blood samples were drawn to determine apoE polymorphisms and lipid profile. As expected, the e3 allele was prevalent in the CAD (0.87) and non-CAD groups (0.81; P = 0.099), followed by the e4 allele (0.09 and 0.14, respectively; P = 0.158). The e3/3 (76 and 78%) and e3/4 (16 and 23%) were the most common genotypes for patients and controls, respectively. The lipid profile was altered in patients compared to controls (P < 0.05), independently of the e4 allele. However, in the controls this allele was prevalent in individuals with elevated LDL-cholesterol levels only (odds ratio = 2.531; 95% CI = 1.028-6.232). The frequency of risk factors was higher in the CAD group (P < 0.05), but their association with the lipid profile was not demonstrable in e4 carriers. In conclusion, the e4 allele is not associated with CAD or lipid profile in patients with atherosclerosis. However, its frequency in the non-CAD group is associated with increased levels of LDL-cholesterol, suggesting an independent effect of the e4 allele on lipid profile when the low frequency of other risk factors in this group is taken into account.

The objective of the present study was to determine the prevalence of electrolyte disturbances in AIDS patients developing acute kidney injury in the hospital setting, as well as to determine whether such disturbances constitute a risk factor for nephrotoxic and ischemic injury. A prospective, observational cohort study was carried out. Hospitalized AIDS patients were evaluated for age; gender; coinfection with hepatitis; diabetes mellitus; hypertension; time since HIV seroconversion; CD4 count; HIV viral load; proteinuria; serum levels of creatinine, urea, sodium, potassium and magnesium; antiretroviral use; nephrotoxic drug use; sepsis; intensive care unit (ICU) admission, and the need for dialysis. Each of these characteristics was correlated with the development of acute kidney injury, with recovery of renal function and with survival. Fifty-four patients developed acute kidney injury: 72% were males, 59% had been HIV-infected for >5 years, 72% had CD4 counts <200 cells/mm³, 87% developed electrolyte disturbances, 33% recovered renal function, and 56% survived. ICU admission, dialysis, sepsis and hypomagnesemia were all significantly associated with nonrecovery of renal function and with mortality. Nonrecovery of renal function was significantly associated with hypomagnesemia, as was mortality in the multivariate analysis. The risks for nonrecovery of renal function and for death were 6.94 and 6.92 times greater, respectively, for patients with hypomagnesemia. In hospitalized AIDS patients, hypomagnesemia is a risk factor for nonrecovery of renal function and for in-hospital mortality. To determine whether hypomagnesemia is a determinant or simply a marker of critical illness, further studies involving magnesium supplementation in AIDS patients are warranted.

The hypothalamus is a forebrain structure critically involved in the organization of defensive responses to aversive stimuli. Gamma-aminobutyric acid (GABA)ergic dysfunction in dorsomedial and posterior hypothalamic nuclei is implicated in the origin of panic-like defensive behavior, as well as in pain modulation. The present study was conducted to test the difference between these two hypothalamic nuclei regarding defensive and antinociceptive mechanisms. Thus, the GABA A antagonist bicuculline (40 ng/0.2 µL) or saline (0.9% NaCl) was microinjected into the dorsomedial or posterior hypothalamus in independent groups. Innate fear-induced responses characterized by defensive attention, defensive immobility and elaborate escape behavior were evoked by hypothalamic blockade of GABA A receptors. Fear-induced defensive behavior organized by the posterior hypothalamus was more intense than that organized by dorsomedial hypothalamic nuclei. Escape behavior elicited by GABA A receptor blockade in both the dorsomedial and posterior hypothalamus was followed by an increase in nociceptive threshold. Interestingly, there was no difference in the intensity or in the duration of fear-induced antinociception shown by each hypothalamic division presently investigated. The present study showed that GABAergic dysfunction in nuclei of both the dorsomedial and posterior hypothalamus elicit panic attack-like defensive responses followed by fear-induced antinociception, although the innate fear-induced behavior originates differently in the posterior hypothalamus in comparison to the activity of medial hypothalamic subdivisions.

This study investigated the influence of cueing on the performance of untrained and trained complex motor responses. Healthy adults responded to a visual target by performing four sequential movements (complex response) or a single movement (simple response) of their middle finger. A visual cue preceded the target by an interval of 300, 1000, or 2000 ms. In Experiment 1, the complex and simple responses were not previously trained. During the testing session, the complex response pattern varied on a trial-by-trial basis following the indication provided by the visual cue. In Experiment 2, the complex response and the simple response were extensively trained beforehand. During the testing session, the trained complex response pattern was performed in all trials. The latency of the untrained and trained complex responses decreased from the short to the medium and long cue-target intervals. The latency of the complex response was longer than that of the simple response, except in the case of the trained responses and the long cue-target interval. These results suggest that the preparation of untrained complex responses cannot be completed in advance, this being possible, however, for trained complex responses when enough time is available. The duration of the 1st submovement, 1st pause and 2nd submovement of the untrained and the trained complex responses increased from the short to the long cue-target interval, suggesting that there is an increase of online programming of the response possibly related to the degree of certainty about the moment of target appearance.

A fim de elaborar um doce de manga em massa com excedentes de produção, foi iniciado o estudo de parâmetros físico-químicos da manga, variedade "coquinho" e do doce obtido dessa manga, empregando-se diversas formulações. Primeiramente, foi determinada a composição da manga madura e o efeito da variação do teor de polpa, de sólidos totais e de ácido cítrico na consistência do gel obtido. Foram testadas formulações variando quatro parâmetros: teores de pectina e de ácido cítrico adicionados, proporção entre polpa e açúcar na mistura inicial e concentração de sólidos no produto final. A textura foi avaliada usando um texturômetro. O teor de pectina requerido foi da ordem de 1% sobre o peso do produto final. A adição de ácido cítrico deve ser mantida na faixa entre 0,6 e 1,0%. Acima de 1% a textura do produto ficou prejudicada pelo efeito de hidrólise da pectina. Abaixo de 0,6%, a inversão do açúcar pode ser insuficiente para estabilizar a textura, acarretando a formação de cristais durante o armazenamento que comprometem a qualidade do doce. O incremento do teor de sólidos solúveis aumenta a dureza do produto, e um tempo de cozimento elevado pode acarretar uma hidrólise excessiva da pectina, resultando na não formação do gel.
