The frequency of viral markers for hepatitis B (HBV) and C (HCV), human immunodeficiency virus-1 (HIV-1) and human T-lymphotropic virus-1 (HTLV-1) was evaluated in 32 Brazilian ß-thalassemia multitransfused patients. Additionaly the serum concentrations of ferritin and alanine aspartate transaminase (ALAT) were determined. The results show a high prevalence of markers of infection by HBV (25.0%) and HCV (46.8%) and a low prevalence of markers for HIV-1 and HTLV-1. No correlations were demonstrated between the presence of the hepatitis markers and the number of units transfused or the serum concentrations of ferritin and ALAT.

One of the main difficulties in studying dengue virus infection in humans and in developing a vaccine is the absence of a suitable animal model which develops the full spectrum of dengue fever, dengue haemorrhagic fever, and dengue shock syndrome. It is our proposal to present morphological aspects of an animal model which shows many similarities with the dengue infection in humans. BALB/c mice were intraperitoneally infected with non-neuroadapted dengue virus serotype 2 (DENV-2). Histopathological and morphometrical analyses of liver tissue revealed focal alterations along the infection, reaching wide-ranging portal and centrolobular veins congestion and sinusoidal cell death. Additional ultrastructural observations demonstrated multifocal endothelial injury, platelet recruitment, and alterated hepatocytes. Dengue virus antigen was detected in hepatocytes and in the capillar endothelium of the central lobular vein area. Liver function tests showed high levels of aspartate transaminase and alanine transaminase enzyme activity. Lung tissue showed interstitial pneumonia and mononuclear cells, interseptal oedema, hyperplasia, and hypertrophy of the bronchiolar epithelial cells. DENV-2 led to a transient inflammatory process, but caused focal alterations of the blood-exchange barrier. Viremia was observed from 2nd to 11th day p.i. by isolation of DENV-2 in C6/36 mosquito cell line inoculated with the supernatant of macerated liver, lung, kidney, and cerebellum tissues of the infected mice.

Two species of wild potato Solanum commersonii, subspecies commersonii and malmeanum, and S. chacoense, subspecies muelleri occur in southern Brazil. Their rusticity and easy adaptation to extreme climatic conditions make them valuable for breeding programs. The objective of this work was to analyze the isoenzymatic variability of 113 clones of wild potato subspecies. They were collected and maintained at Embrapa-Centro de Pesquisa Agropecuária de Clima Temperado, at Pelotas, RS, Brazil. Enzymes involved in energetic (group I) or in peripherical (group II) metabolism constituted the material used. Polyacrylamide horizontal gel electrophoresis was used to analyze peroxidase, aspartate transaminase, phosphoglucomutase and isocitrate dehydrogenase isoenzymes. Solanum spp. has considerable genetic variability for isoenzymatic patterns. Cluster analysis classified the clones into 51 subgroups, based on electrophoretic variants of group I enzymes, and into 89, when group II enzyme variants were added. Genotypic differentiation of S. chacoense muelleri in relation to S. commersonii commersonii and S. commersonii malmeanum is evident when expressed through similarity and cluster analysis.

The objective of this work was to characterize 27 potato genotypes, using molecular markers. Polyacrylamide gel electrophoresis, RAPD techniques and isozymes of esterase, phosphoglucomutase and soluble proteins were analyzed in tubers, and isocitrate dehydrogenase, aspartate transaminase, phosphoglucomutase and peroxidase, in leaves. Eighteen primers were tested and four were chosen, kits OPX (01, 04 and 09) and OPY (07), to analyze RAPD markers in leaf extracts. Similarity and cluster analysis were conducted using Jaccard coefficient and the unweighted pair-group method using arithmetic average. Despite the differences detected in the analysis of proteins and isozymes in the tubers, as well as of isozymes in the leaves, the characterization of all genotypes through gel electrophoresis was not possible, while RAPD markers were efficient to characterize all the 27 genotypes.

The aim of the present study was to assess the influence of hyperbaric oxygenation (HBO) on rat liver regeneration before and after partial hepatectomy. Rats were sacrificed 54 h after 15% hepatectomy, liver and body weights were measured, and serum alanine transaminase (ALT) and aspartate transaminase (AST) activity and albumin levels were determined. The lipid peroxide level, as indicated by malondialdehyde production in the remnant liver was measured, and liver sections were analyzed by light microscopy. Five groups of 10 rats in each group were studied. The preHBO and pre-hyperbaric pressure (preHB) groups were treated before partial hepatectomy with 100% O2 and 21% O2, respectively, at 202,650 pascals, daily for 3 days (45 min/day). The control group was not treated before partial hepatectomy and recovered under normal ambient conditions after the procedure. Groups postHBO and postHB were treated after partial hepatectomy with HBO and HB, respectively, three times (45 min/day). The preHBO group presented a significant increase in the initiation of the regeneration process of the liver 54 h postoperatively. The liver/body weight ratio was 0.0618 ± 0.0084 in the preHBO compared to 0.0517 ± 0016 g/g in the control animals (P = 0.016). In addition, the preHBO group showed significant better liver function (evaluated by the lowest serum ALT and AST activities, P = 0.002 and P = 0.008, respectively) and showed a significant decrease in serum albumin levels compared to control (P < 0.001). Liver lipid peroxide concentration was lowest in the preHBO group (P < 0.001 vs control and postHBO group) and light microscopy revealed that the composition of liver lobules in the preHBO group was the closest to normal histological features. These results suggest that HBO pretreatment was beneficial for rat liver regeneration after partial hepatectomy.

Fazendo determinações dos níveis de transamino.se sérica (T-GP e T-GO) em Cebus apella, antes e após inoculação de Schistosoma manroni e após terapêutica pelo aminonitrotiazol, os autores puderam observar, antes da inoculação, os seguinces valores: T-GP = 28.3, T-GO = 24,7. Os desvios padrões foram respectivamente de 11,2 e 11,3 e os valores extremos variaram para T-GO 10 e 60 µF e entre 15 e 57 µF para T-GP. Após inoculação, em apenas 1 caso puderam observar, 81 dias após, um valor para T-GP de 70 µF que pode ser considerado como discretamente aumentado. Não encontraram alterações após terapêutica pelo Cl 32.644 Ba (Aminonitrotiazol).

Folhas de feijoeiro (Phaseolus vulgaris L.) normais e deficientes em potássio dos cultivares Rosinha Precoce e Goiano Precoce foram analisadas para determinação das atividades de transaminase glutâmico-oxaloacético (E.C. 2.6.1.1) e transcarbamilase de ornitina (E.C. 2.1.3.3). Em plantas deficientes em potássio houve aumento na atividade das duas enximas estudadas e nos dois cultivares. Esses resultados explicariam o acúmulo de citrulina e arginina em plantas deficientes em potássio devido a maior atividade da transcarbamilase de ornitina, que possivelmente seria um mecanismo de reduzir a toxidez por amônia.

Estudou-se a atividade da transaminase glutâmico oxaloacética (TGO) em diferentes tecidos (fígado, músculo, rim, pulmão, baço e soro sanguíneo) de ratos e de soro humano. verificou-se que a atividade da enzima proveniente de qualquer um destes tecidos é aumentada cerca de tr~es vezes quando a incubação se faz a 60ºC, ao invés de 37ºC. São feitas considerações acerca da importância deste fato.

The nucleus tractus solitarius (NTS) plays an important role in the control of autonomic reflex functions. Glutamate, acting on N-methyl-D-aspartate (NMDA) and non-NMDA ionotropic receptors, is the major neurotransmitter in this nucleus, and the relative contribution of each receptor to signal transmission is unclear. We have examined NMDA excitatory postsynaptic currents (NMDA-EPSCs) in the subpostremal NTS using the whole cell patch clamp technique on a transverse brainstem slice preparation. The NMDA-EPSCs were evoked by stimulation of the solitary tract over a range of membrane potentials. The NMDA-EPSCs, isolated pharmacologically, presented the characteristic outward rectification and were completely blocked by 50 µM DL-2-amino-5-phosphonopentanoic acid. The I-V relationship of the NMDA response shows that current, with a mean (± SEM) amplitude of -41.2 ± 5.5 pA, is present even at a holding potential of -60 mV, suggesting that the NMDA receptors are weakly blocked by extracellular Mg2+ at near resting membrane potentials. This weak block can also be inferred from the value of 0.67 ± 0.17 for parameter delta obtained from a fit of the Woodhull equation to the I-V relationship. The maximal inward current measured on the I-V relationship was at -38.7 ± 4.2 mV. The decay phase of the NMDA currents was fitted with one exponential function with a decay time constant of 239 ± 51 and 418 ± 80 ms at a holding potential of -60 and +50 mV, respectively, which became slower with depolarization (e-fold per 145 mV). The biophysical properties of the NMDA receptors observed in the present study suggest that these receptors in the NTS contain NR2C subunits and may contribute to the synaptic signal integration.

Growing consistent evidence indicates that hypofunction of N-methyl-D-aspartate (NMDA) transmission plays a pivotal role in the neuropathophysiology of schizophrenia. Hence, drugs which modulate NMDA neurotransmission are promising approaches to the treatment of schizophrenia. The aim of this article is to review clinical trials with novel compounds acting on the NMDA receptor (NMDA-R). This review also includes a discussion and translation of neuroscience into schizophrenia therapeutics. Although the precise mechanism of action of minocycline in the brain remains unclear, there is evidence that it blocks the neurotoxicity of NMDA antagonists and may exert a differential effect on NMDA signaling pathways. We, therefore, hypothesize that the effects of minocycline on the brain may be partially modulated by the NMDA-R or related mechanisms. Thus, we have included a review of minocycline neuroscience. The search was performed in the PubMed, Web of Science, SciELO, and Lilacs databases. The results of glycine and D-cycloserine trials were conflicting regarding effectiveness on the negative and cognitive symptoms of schizophrenia. D-serine and D-alanine showed a potential effect on negative symptoms and on cognitive deficits. Sarcosine data indicated a considerable improvement as adjunctive therapy. Finally, minocycline add-on treatment appears to be effective on a broad range of psychopathology in patients with schizophrenia. The differential modulation of NMDA-R neurosystems, in particular synaptic versus extrasynaptic NMDA-R activation and specific subtypes of NMDA-R, may be the key mediators of neurogenesis and neuroprotection. Thus, psychotropics modulating NMDA-R neurotransmission may represent future monotherapy or add-on treatment strategies in the treatment of schizophrenia.

O objetivo foi avaliar os padrões isoenzimáticos de Esterase (EST - EC 3.1.1.1), Fosfatase Ácida (ACP - EC 3.1.3.2), Malato Desidrogenase (MDH - EC 1.1.1.37), Álcool Deshidrogenase (ADH - EC 1.1.1.1) e Glutamato Oxalacetato Transaminase (GOT - EC 2.6.1.1), em sementes e plântulas de dois cultivares de cevada (MN 721 e Scarlett). As sementes foram fornecidas pela empresa Westermann, localizada no município de Piratini, Rio Grande do Sul, onde foi realizado o cultivo da cevada em 2007. Foram colhidas em três épocas com diferentes percentuais de umidade, secas a 13% de umidade e armazenadas em câmara fria. Os cinco sistemas isoenzimáticos analisados apresentaram variações na expressão, principalmente quando comparados entre sementes e plântulas. Concluiu-se que há variação no padrão de expressão das enzimas EST, ACP, MDH, ADH e GOT entre sementes e plântulas. A expressão das enzimas EST e GOT foi pouco e muito influenciada pela época de colheita, respectivamente.

O tratamento de sementes com a utilização de silício em sementes de boa qualidade constitui prática para o aumento da produtividade. O objetivo deste trabalho foi avaliar o efeito do recobrimento de sementes de arroz com duas fontes de silício, em seus atributos fisiológicos, enzimáticos e sanitários. Empregaram-se os cultivares de arroz Irga 424 e Puitá Inta CL e de duas fontes de silício: silicato de alumínio e casca de arroz carbonizada moída, consistindo nas doses de 0; 30; 60; 90 e 120 g 100 kg-1 (de cada produto aplicado) de sementes mais polímero e água, totalizando um volume de calda de 1 L 100 kg-1 de sementes. O delineamento experimental foi o inteiramente casualizado, com quatro repetições. A qualidade fisiológica das sementes foi avaliada no (LAS-FAEM\UFPel) pelos testes de germinação, primeira contagem de germinação, comprimento da parte aérea e raiz, teste de frio e emergência em campo. Para diferenciação isoenzimática, as isoenzimas analisadas foram: esterase, glutamato oxalacetato transaminase e peroxidase, para todos os tratamentos. A avaliação da qualidade sanitária das sementes foi realizada pelo método do papel de filtro ou "Blotter Test". Doses crescentes de casca de arroz carbonizada e de silicato de alumínio, até 120 g 100 kg-1 de sementes, incrementam o vigor de sementes de arroz, avaliados pelo comprimento de raiz e pela emergência a campo. As fontes casca de arroz carbonizada e caulim controlam a incidência de fungos de solo nas sementes de arroz.

Thirty-two patients bitten by venomous snakes sixteen by Bothrops spp. and sixteen by Crotalus durissus terrificus were studied. The group comprised thirty males and two females, aged eight to sixty-three years (mean 33±15). Bromsulphalein tests were increased in the majority of patients bitten by Crotalus durissus terrificus. The correlation coefficient of Spearman was positive between bromsulphalein tests and alanine aminotransferase levels, and between alanine aminotransferase and aspartate aminotransferase levels only in the Crotalus group. The only patient who died was bitten by Crotalus durissus terrificus and showed hydropic degeneration and mitochondrial injury in the liver. It was concluded that the hepatic damage might have been caused by at least two possible mechanisms: venom effect on liver mitochondria and cytokine effects on hepatocyte, specially interleukin-6.

A descriptive study was carried out in 104 patients with Plasmodium vivax malaria, from the region of Turbo (Antioquia, Colombia). Clinical features and levels of hemoglobin, glycemia, serum bilirubin, alanine-aminotransferase (ALT), aspartate-aminotransferase (AST), creatinine and complete blood cell profile were established. 65% of the studied individuals were men and their mean age was 23. Of all individuals 59% had lived in the region for > 1 year and 91% were resident in the rural area. 42% were farmers and 35% had a history of malaria. The mean parasitaemia was 5865 parasites/mm³. The evolution of the disease was short (average of 4.0 days). Fever, headache and chills were observed simultaneously in 91% of the cases while the most frequent signs were palmar pallor (46%), jaundice (15%), hepatomegaly (17%), and spleen enlargement (12%). Anemia was found in 39% of the women and in 51% of the men, 8% of individuals had thrombocytopaenia and 41% had hypoglycemia.
