The objective of this study was to evaluate the effect of Ascophyllum nodosum brown seaweed meal (FAM) on the health of Nile tilapia submitted to inoculation with Aeromonas hydrophila. The experiment was conducted for a period of 40 days using 120 Nile tilapia fingerlings, with age of 40 days, distributed in 20 tanks. A diet including Ascophyllum nodosum seaweed meal at 20g.kg-1 and a control diet (without FAM) were provided which constituted the treatments. Thirty days after beginning the experiment, A. hydrophila was inoculated by bacterial inoculum diluted in sterile saline solution at a concentration of 10(6) CFU ml-1. Except for the width, which was greater for the treatment with the provision of FAM (P<0.05), there was no influence on the performance parameters of the fingerlings, but the occurrence of lesions in animals inoculated with A. hydrophila and fed with FAM was lower and they also exhibited a decline in the lesions in a shorter period of time than the control group. FAM prevents hepatopancreatic congestion in infected animals. Ascophyllum nodosum brown seaweed meal reduced the number of lesions in fish in a shorter time when compared to the control group.

Em morangueiros, a mancha das folhas, causada por Mycosphaerella fragariae, é uma das doenças mais comuns na cultura, o que torna importantes os estudos de controle alternativo e de cultivares, sobre o progresso da epidemia. Em busca de práticas alternativas para seu controle, avaliou-se o efeito de cultivares de morangueiro e do extrato da alga Ascophyllum nodosum, na expressão dos sintomas de mycosphaerella, além de avaliar o ajuste de modelos de crescimento para o progresso temporal da incidência e severidade da doença. Utilizou-se o extrato de algas a 29%, na dose de 2 L ha-1, aplicado no solo, na folha e em ambos. As cultivares Dover, Toyonoka, Albion, Camarosa, Ventana, Campinas, Tudla e Camino Real foram avaliadas, quinzenalmente, quanto a incidência e severidade da doença. Utilizou-se o delineamento inteiramente casualizado, com parcelas subdivididas e três repetições. Com os dados de incidência e severidade, calculou-se a área abaixo da curva de progresso da doença e utilizou-se a análise de variância para avaliar o efeito de cultivares e do extrato de algas. Aos dados do progresso temporal da incidência e da severidade, em cada cultivar, foram ajustados os modelos monomolecular, logístico e de Gompertz. O extrato de algas não apresentou efeito ou interação com cultivares, no controle da mycosphaerella. Houve efeito de cultivar com menor intensidade da doença, em Albion e Ventana, devido, principalmente, ao atraso na epidemia, pela redução do inóculo inicial efetivo. Aos dados de progresso temporal da incidência e da severidade ajustaram-se, respectivamente, os modelos logístico e monomolecular, para todas as cultivares.

O uso de fertilizantes organominerais contendo extratos de algas é recente no Brasil. Em vista disso, este estudo teve como objetivo avaliar o efeito da aplicação foliar de fertilizante organomineral contendo extrato da alga Ascophyllum nodosum (Activo®) no desenvolvimento e nos teores de nutrientes em cenoura. O experimento foi conduzido na área de olericultura da Universidade Federal do Paraná, no município de Pinhais, PR, no ano de 2009, em sistema orgânico, utilizando cultivar do tipo Nantes. O delineamento experimental foi inteiramente casualizado, e os tratamentos consistiram na aplicação de fertilizante organomineral à base de extrato de algas. Foi utilizado um esquema fatorial com duas doses (0,2 e 0,4%) e duas épocas de aplicação (25 e 45 dias após o plantio), e um tratamento testemunha. As aplicações aumentaram a altura da parte aérea, o diâmetro radicular e o número de folhas, sendo esta variável também influenciada pela concentração. A produtividade aumentou com a aplicação do fertilizante organomineral, independentemente da concentração e da época de aplicação. Os teores nutricionais na parte aérea e nas raízes também foram alterados em decorrência das aplicações, sendo observada diminuição dos teores foliares de Cu e dos teores radiculares de N, Ca, Mg, Cu e Zn, possivelmente em função de efeito de diluição, pelo incremento da biomassa. Conclui-se que a aplicação do fertilizante organomineral contendo extrato de algas foi benéfica ao crescimento, desenvolvimento e produção das plantas de cenoura, o que resultou em diminuição na concentração de N, Ca, Mg, Cu e Zn na raiz e de Cu na parte aérea.

The results of 32 cases studied lead us to the conclusion that erythema nodosum's investigation routine is very important, once in our retrospective study, the percentage of cases of unknown etiology was 69.4%, and in this prospective study it is 21.8%. In 10 cases (31.2%), more than one causing agent was suspected. Infections (bacterial, helminthic, fungal, by protozoa) were diagnosed in 26 cases, streptococcal infection having predominated (12 cases). Drugs-dipirone, aspirin, anovulatory - were suspected as causing agents in 13 cases. The association of erythema nodosum and histoplasmosis capsulata is described for the first time in Brazil. We consider erythema nodosum to be a complex syndrome which should be regarded as a manifestation of underlying diseases. The fact that all 32 subjects were women, 26 of them during menacme, suggests that particular hormonal media may favor the action of various processes (infections and drugs), precipitating erythema nodosum's clinical picture.

Recent data suggest that the clinical course of reactional states in leprosy is closely related to the cytokine profile released locally or systemically by the patients. In the present study, patients with erythema nodosum leprosum (ENL) were grouped according to the intensity of their clinical symptoms. Clinical and immunological aspects of ENL and the impact of these parameters on bacterial load were assessed in conjunction with patients' in vitro immune response to mycobacterial antigens. In 10 out of the 17 patients tested, BI (bacterial index) was reduced by at least 1 log from leprosy diagnosis to the onset of their first reactional episode (ENL), as compared to an expected 0.3 log reduction in the unreactional group for the same MDT (multidrug therapy) period. However, no difference in the rate of BI reduction was noted at the end of MDT among ENL and unreactional lepromatous patients. Accordingly, although TNF-alpha (tumor necrosis factor) levels were enhanced in the sera of 70.6% of the ENL patients tested, no relationship was noted between circulating TNF-alpha levels and the decrease in BI detected at the onset of the reactional episode. Evaluation of bacterial viability of M. leprae isolated from the reactional lesions showed no growth in the mouse footpads. Only 20% of the patients demonstrated specific immune response to M. leprae during ENL. Moreover, high levels of soluble IL-2R (interleukin-2 receptor) were present in 78% of the patients. Circulating anti-neural (anti-ceramide and anti-galactocerebroside antibodies) and anti-mycobacterial antibodies were detected in ENL patients' sera as well, which were not related to the clinical course of disease. Our data suggest that bacterial killing is enhanced during reactions. Emergence of specific immune response to M. leprae and the effective role of TNF-alpha in mediating fragmentation of bacteria still need to be clarified.

We evaluated the antiviral activity of the marine alga, Ulva fasciata, collected from Rasa beach and Forno beach, Búzios, Rio de Janeiro, Brazil on the replication of human metapneumovirus (HMPV). The algae extracts were prepared using three different methodologies to compare the activity of different groups of chemical composites obtained through these different methodologies. Four out of the six extracts inhibited nearly 100% of viral replication. The results demonstrated that the majority of the extracts (five out of six) possess virucidal activity and therefore have the ability to interact with the extracellular viral particles and prevent the infection. On the other hand, only two extracts (from Forno beach, obtained by maceration and maceration of the decoction) were able to interact with cell receptors, hindering the viral entry. Finally, only the extract of algae collected at Forno beach, obtained by maceration presented intracellular activity. To our knowledge, this is a pioneer study on antiviral activity of marine algae against HMPV. It is also the first on antiviral activity against HMPV ever done in Brazil. The study also shows the effect of different environment factors and different chemical procedures used to obtain the extract on its biological properties.

Com iodo-131 e o emprego do método cromatográfico, foi possível isolar duas substâncias orgânicas lipídicas emuma alga marinha. Os quatro métodos químicos de análise estrutural, foram aplicados e nos deram como resultado, ser uma delas, o éster metílico do ácido oléico e a outra o estér metílico do ácido alfa-iodo palmítico, substância esta até então desconhecida na literatura científica, como existente em algas marinhas.

A partir de teleógina de Amblyomma nodosum Neumann, 1899, removida de Tamandua tetradactyla (Linnaeus, 1758) foram obtidas posturas que originaram larvas, em condições de laboratório. Um total de 219 larvas foi examinado em microscopia de luz, com objetiva de imersão e microscopia de confocal com varredura a laser. Buscou-se analisar a variação do número das cerdas internas dos lados direito e esquerdo, no fundo da cápsula do Órgão de Haller, com intuito de utilizá-las como um parâmetro quetotáxico na diagnose específica. A média do número de cerdas foi 5,06±0, 71, com valores limites da amplitude de 4-11.

Acid base properties of mixed species of the microalgae Spirulina were studied by potentiometric titration in medium of 0.01 and 0.10 mols L-1 NaNO3 at 25.0±0.10 C using modified Gran functions or nonlinear regression techniques for data fitting. The discrete site distribution model was used, permitting the characterization of five classes of ionizable sites in both ionic media. This fact suggests that the chemical heterogeneity of the ionizable sites on the cell surface plays a major role on the acid-base properties of the suspension in comparison to electrostatic effects due to charge-charge interactions. The total of ionizable sites were 1.75±0.10 and 1.86±0.20 mmolsg-1 in ionic media of 0.01 and 0.10 mols L-1 NaNO3, respectively. A major contribution of carboxylic groups was observed with an average 34 and 22% of ionizable sites being titrated with conditional pcKa of 4.0 and 5.4, respectively. The remaining 44% of ionizable sites were divided in three classes with averaged conditional pcKa of 6.9, 8.7 and 10.12, which may be assigned respectively to imidazolic, aminic, and phenolic functionalities.

Chemical investigation of the dichloromethane/methanol extract of the marine alga Bostrychia tenella has led to the isolation of two aromatic compounds, the new sulfate metabolite potassium 4-(hydroxymethyl)-benzenosulfonate (1) and the compound 1-methoxyphenethyl alcohol (2), described previously as a synthetic product. Their structures were determined by spectroscopic methods including NMR, MS, IR and by comparison with literature data.

This work describes a simple and economical experiment for the extraction and purification of chlorophyll a from Spirulina maxima. Extraction and purification of natural compounds can be considered one of the most illustrative experiments that can be performed in Organic Chemistry courses. Particularly, the chromatography of dyes and pigments allows students to have a better comprehension of the chromatography separations. These compounds represent an important class of organic pigments applied in pharmaceutical, cosmetic, detergent compositions, and various other fields and can be extracted from plants and algae. To extract, separate and purify chlorophyll a from associated pigments such as xanthophylls, carotenes, and pheophytins, very costly processes are reported. The present approach is perfectly adequate for use in Chemistry experiments for undergraduate students.

A method based on microwave-induced combustion (MIC) was applied for the decomposition of different types of edible seaweed (Nori, Hijiki and Wakame) for subsequent determination of bromine and iodine by inductively coupled plasma mass spectrometry (ICP-MS). Decomposition of 500 mg of each sample was achieved in less than 30 min. A single and diluted solution (150 mmol L-1 (NH4)2CO3) was used for the absorption of both analytes and a reflux step of 5 min was applied to improve analyte recoveries. Accuracy was evaluated using certified reference materials and agreement was between 103 and 108% for both elements.

A lectin present in the marine red alga Pterocladiella capillacea was purified and characterised by extraction of soluble proteins (crude extract) in 20 mM Tris-HCl buffer, pH 7.5. Among the analysed erythrocytes (human blood group A, B and O and the animals ox, goat, chicken and rabbit) the lectin agglutinated specifically rabbit erythrocytes. The hemagglutinating activity assay showed that the lectin was not dependent on divalent cations and was shown to be inhibited by the glycoproteins avidin and mucin. The purification procedure was conduced by precipitation of the crude extract with 80% saturation ammonium sulfate (F0/80) followed by affinity chromatography on guar-gum column. The lectin of P. capillacea was purified 14.5 fold and had a recovery of 27.4% of the original total specific activity present in the crude extract. The absence of carbohydrate suggested that the lectin is not a glycoprotein. The molecular mass of P. capillacea lectin, determined by gel filtration, was 5.8 kDa. SDS-PAGE in the presence of ß-mercaptoethanol gave one band, indicating that the native lectin is a monomeric protein. The activation energy of denaturation process (D G') was calculated to be 106.87 kJ . mol-1 at 70 ºC.
