The polypeptides of 46 and 58kDa were recognized in different T. cruzi strains (Y, WSL and Colombiana) by serum of all chagasic patients studied. These polypeptides were isolated from T. cruzi Y strain and used in ELISA. The sensitivity and specificity were 97.6% [CI 95%: 86-100%] and 100% [CI 95%: 89.3-100%], respectively when Tc 46 was used. When Tc 58 was used the sensitivity and specificity were 100% [CI 95%: 89.6-100%] and 90.2% [CI 95%: 75.9-96.8%], respectively.

INTRODUCTION: Studies strongly indicate Dientamoeba fragilis as one of the causes of diarrhea in human immunodeficiency virus (HIV) patients. METHODS: The objective of the present study was to evaluate the prevalence of D. fragilis associated with the causes of diarrhea in 82 HIV/ AIDS patients hospitalized at the Instituto de Infectologia Emílio Ribas from September 2006 to November 2008. RESULTS: In total, 105 samples were collected from 82 patients. Unprotected sex was the most frequent cause of HIV infection (46.3%), followed by the use of injectable or non-injectable drugs (14.6%). Patients presented with viral loads of 49-750,000 copies/ mL (average: 73,849 ± 124,850 copies/mL) and CD4 counts ranging of 2-1,306 cells/mm³ (average: 159 ± 250 cells/mm³). On an average, the odds of obtaining a positive result by using the other techniques (Hoffman, Pons and Janer or Lutz; Ritchie) were 2.7 times higher than the chance of obtaining a positive result by using the simplified iron hematoxylin method. Significant differences were found between the methods (p = 0.003). CONCLUSIONS: The other techniques can detect a significantly greater amount of parasites than the simplified iron hematoxylin method, especially with respect to Isospora belli, Cryptosporidium sp., Schistosoma mansoni, and Strongyloides stercoralis, which were not detected using hematoxylin. Endolimax nana and D. fragilis were detected more frequently on using hematoxylin, and the only parasite not found by the other methods was D. fragilis.

OBJETIVO: Avaliar como a pancreatite aguda é vista e tratada pelo cirurgião brasileiro. MÉTODO: Trata-se de um questionário prospectivo enviado aos membros do Colégio Brasileiro de Cirurgiões (CBC). Dois mil questionários foram enviados obtendo-se 618 respostas (30,9%). O questionário constituía de perguntas relacionadas à experiência do cirurgião e ao tratamento da PA. RESULTADOS: Cento e oitente e dois entrevistados (33,6%) disseram tratar até cinco casos por ano e 147 (27,2%) tratam de seis a 10 casos por ano. Dentre os critérios utilizados para a definição de PA, o mais citado foi a avaliação clínica por 306 (57,4%) entrevistados, seguido dos critérios de Ranson por 294 (55,2%) e TC por 262 (49,2%). Com relação ao uso da TC, 275 (51,5%) entrevistados responderam que realizam a TC em todos os doentes com PA. O suporte nutricional parenteral foi o método de escolha indicado por 248 (46,6%) entrevistados. A infecção da coleção e/ou necrose pancreática é a principal indicação operatória com 447 (83,6%) respostas. O melhor período para operar um doente com PA grave foi considerado até quatorze dias por 278 (54,2%) entrevistados. Com relação aos antibióticos, 371 (68,6%) entrevistados disseram que utilizam antibióticos no tratamento da PA grave. CONCLUSÃO: Estes resultados demonstram uma ampla variação no tratamento da PA no Brasil. A pouca experiência dos cirurgiões e os temas controversos são os fatores principais responsável para esta falta de padronização. A realização de um Consenso nacional baseado nos novos conceitos e na experiência mundial é crucial para ajustar estas condutas.

Male sex determination in humans is controlled by the SRY gene, which encodes a transcriptional regulator containing a conserved high mobility group box domain (HMG-box) required for DNA binding. Mutations in the SRY HMG-box affect protein function, causing sex reversal phenotypes. In the present study, we describe a 19-year-old female presenting 46,XY karyotype with hypogonadism and primary amenorrhea that led to the diagnosis of 46,XY complete gonadal dysgenesis. The novel p.E89K missense mutation in the SRY HMG-box was identified as a de novo mutation. Electrophoretic mobility shift assays showed that p.E89K almost completely abolished SRY DNA-binding activity, suggesting that it is the cause of SRY function impairment. In addition, we report the occurrence of the p.G95R mutation in a 46,XY female with complete gonadal dysgenesis. According to the three-dimensional structure of the human SRY HMG-box, the substitution of the conserved glutamic acid residue by the basic lysine at position 89 introduces an extra positive charge adjacent to and between the positively charged residues R86 and K92, important for stabilizing the HMG-box helix 2 with DNA. Thus, we propose that an electrostatic repulsion caused by the proximity of these positive charges could destabilize the tip of helix 2, abrogating DNA interaction.

This study aimed to verify the influence of pH and temperature on the lysis of yeast using experimental design. In this study, the enzymatic extract containing β-1,3-glucanase and chitinase, obtained from the micro-organism Moniliophthora perniciosa, was used. The experiment showed that the best conditions for lysis of Pseudozyma sp. (CCMB 306) and Pseudozyma sp. (CCMB 300) by lytic enzyme were pH 4.9 at 37 ºC and pH 3.9 at 26.7 ºC, respectively. The lytic enzyme may be used for obtaining various biotechnology products from yeast.
