Plum (Prunus salicina Lindl. cv. Harry Pickstone), a China indigenous fruit, is widely produced and consumed in countries such as Japan and Brazil. The practice of thinning is common in horticulture and the fruits removed are discarded as waste. Like the great majority of vegetables, these thinning discards also contain essential oils which have not been investigated until the present time. The extraction of the plum thinning discards volatile oil, through the hydrodistillation method, produced a yield of 0.06% (m/m) and a total of 21 components were identified, with 11 of them being responsible for 72,9% of the total oil composition. The major compounds determined through GC and GC-MS were Z-α-bisabolene (13.7%), n-hexadecanoic acid (12.7%), phytol (12.7%), and β-caryophyllene (10.4%).

Flooded rice cultivation promotes anaerobic conditions, favoring the formation of short chain organic acids such as acetic acid, which may be toxic to the crop. The objective of this study was to evaluate the effect of acetic acid on rice seeds coated with rice husk ash. The experiment was arranged in a 2 x 5 x 5 factorial randomized design, with two cultivars (IRGA 424 and BRS Querência), five doses of coating material (0, 2, 3,4 e 5 g kg-1 seed) and five concentrations of acetic acid (0, 3, 6, 9 and 12 mM), with 4 replications, totaling 50 treatments. The variables first count of germination, germination, shoot and root length, dry weight of shoots and roots were recorded. The results showed that coating rice seeds with rice husk ash up to 5 g kg-1 seed does not influence the performance of rice seeds of cultivars IRGA 424 and BRS Querência when exposed to concentrations of 12 mM acetic acid. The presence of acetic acid in the substrates used for seed germination reduced the vigor and viability of seeds of cultivars IRGA 424 and BRS Querência, as well as seedling development, affecting mainly the roots of BRS Querência.

Eight triterpenes, maniladiol, breine, ursa-9(11):12-dien-3beta-ol, oleana-9(11):12-dien-3beta-ol, 3alpha-hydroxy-tirucalla-8,24-dien-21-oic acid, 3alpha-hydroxy-tirucalla-7,24-dien-21-oic, alpha and beta amyrines were isolated as binary mixtures obtained from the chloroform extract of the oil-resin of Protium heptaphyllum March. The identification of the compounds was based mainly in 13C NMR data and mass spectra. The diene and the tetracyclic acid triterpenes were not reported before in the literature as constituents of the studied resin.

Several polyunsaturated fatty acids (PUFA) belonging to the ômega 6 series, such as cis-6,9,12 gamma-linolenic acid, as well as those of the ômega 3 series, such as cis-5,8,11,14,17-eicosapentaenoic acid and cis-4,7,10,13,16,19-docosahexaenoic acid are of considerable interest due to their nutritional and therapeutic properties. Methods used for the concentration of PUFA from natural sources include urea adduct formation, solvent winterization, supercritical fluid extraction and lipase-catalyzed reaction. Lipases are known to have little reactivity on PUFA and these acids can be enriched by selective hydrolysis, direct esterification of glycerol with PUFA and interesterification. Since lipase reactions are advantageous with respect to fatty acid, positional specificities and mild incubation condition, these enzymes are considered to be suitable for the production of PUFA concentrates for medical purposes.

The phytochemical investigation of the stem bark and leaves of G. elliptica provided a mixture of the norisoprenoids blumenol B and 6-epiblumenol B along with the triterpenes friedelin, as the major constituent, friedelanol, ursa-9(11),12-dien-3-ol, a-amyrin, β-amyrin, morentenol, epifriedelanol, as well as the sesquiterpenes trans-caryophyllene, α-humulene, ethyl hydnocarpate and other fatty acid esters. The identification of the compounds was performed on basis of spectrometric methods such as GC-MS, IR, MS and 1D and 2D NMR. Stem bark extracts showed significant leishmanicidal activity against promastigote forms of Leishmania braziliensis, with the best results for the chloroform extract.

A new trachylobane diterpene ent-trachyloban-18,19-diol (1) was isolated from root bark of Croton floribundus, along with known diterpenes ent-trachyloban-19-oic acid (2), 15b-hydroxy-ent-trachyloban-19-oic acid (3), ent-trachyloban-19-ol (4), ent-kaur-16-en-19-oic acid (5), ent-kaur-16-ene-6a,19-diol (6) and ent-16a-hydroxykaur-11-en-19-oic acid (7). ent-trachyloban-18,19-diol (1) was submitted to derivatization reactions affording four new compounds (8-11). Cytotoxic activity of diterpenes 1, 3, 4, 7-11 against three human cancer cell lines was evaluated. No compounds showed cytotoxic potential with IC50 values greater than 25 mg/mL. Compound 6 was evaluated against five human cancer cell lines, showing moderate effect against three cancer cell lines, MDA-MB-435, HCT-8 and HCT-116, with IC50 values of 14.32, 13.47 and 12.1 mg/mL, respectively.

The phytochemical study of hexane extract from leaves of Salacia crassifolia resulted in the isolation of 3β-palmitoxy-urs-12-ene, 3-oxofriedelane, 3β-hydroxyfriedelane, 3-oxo-28-hydroxyfriedelane, 3-oxo-29-hydroxyfriedelane, 28,29-dihydroxyfriedelan-3-one, 3,4-seco-friedelan-3-oic acid, 3β-hydroxy-olean-9(11):12-diene and the mixture of α-amirin and β-amirin. β-sitosterol, the polymer gutta-percha, squalene and eicosanoic acid were also isolated. The chemical structures of these constituents were established by IR, 1H and 13C NMR spectral data. Crude extracts and the triterpenes were tested against Entamoeba histolytica, Giardia lamblia and Trichomonas vaginalis and no activity was observed under the in vitro assay conditions. The hexane, chloroform, ethyl acetate and ethanol crude extracts, and the constituent 3,4-seco-friedelan-3-oic acid and 28,29-dihydroxyfriedelan-3-one showed in vitro antimicrobial activity against Salmonella typhimurium, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes, Streptococcus sanguinis and Candida albicans.

Objetivo: determinar a viabilidade da identificação ultra-sonográfica precoce do sexo fetal. Métodos: foram estudados prospectivamente 592 fetos por meio de exame ultra-sonográfico entre 11 e 14 semanas na Clínica Obstétrica da Faculdade de Medicina da Universidade de São Paulo. O tubérculo genital foi determinado como horizontal ou vertical através do plano sagital e correlacionado com o sexo ao nascimento ou ao resultado do cariótipo quando este foi realizado. Resultados: o sexo fetal determinado pela avaliação ultra-sonográfica coincidiu com o sexo real em 84% dos casos. O sucesso da identificação aumenta com o progredir da gestação, sendo de 72%, 85% e 89% com 11, 12 e 13 semanas, respectivamente. A taxa de acerto também aumenta com o treinamento dos operadores, sendo de 83,5% no início e de 93,6% no final do estudo. Conclusão: por meio da determinação ultra-sonográfica do tubérculo genital podemos predizer o sexo fetal na maior parte dos casos com potencial aplicabilidade na redução das indicações de procedimentos invasivos para detecção de doenças ligadas ao sexo.

Cancer cachexia induces host protein wastage but the mechanisms are poorly understood. Branched-chain amino acids play a regulatory role in the modulation of both protein synthesis and degradation in host tissues. Leucine, an important amino acid in skeletal muscle, is higher oxidized in tumor-bearing animals. A leucine-supplemented diet was used to analyze the effects of Walker 256 tumor growth on body composition in young weanling Wistar rats divided into two main dietary groups: normal diet (N, 18% protein) and leucine-rich diet (L, 15% protein plus 3% leucine), which were further subdivided into control (N or L) or tumor-bearing (W or LW) subgroups. After 12 days, the animals were sacrificed and their carcass analyzed. The tumor-bearing groups showed a decrease in body weight and fat content. Lean carcass mass was lower in the W and LW groups (W = 19.9 ± 0.6, LW = 23.1 ± 1.0 g vs N = 29.4 ± 1.3, L = 28.1 ± 1.9 g, P < 0.05). Tumor weight was similar in both tumor-bearing groups fed either diet. Western blot analysis showed that myosin protein content in gastrocnemius muscle was reduced in tumor-bearing animals (W = 0.234 ± 0.033 vs LW = 0.598 ± 0.036, N = 0.623 ± 0.062, L = 0.697 ± 0.065 arbitrary intensity, P < 0.05). Despite accelerated tumor growth, LW animals exhibited a smaller reduction in lean carcass mass and muscle myosin maintenance, suggesting that excess leucine in the diet could counteract, at least in part, the high host protein wasting in weanling tumor-bearing rats.

The objective of the present study was to determine the effects of retinoic acid on the growth of the mouse mammary cells HC11 and HC11ras, which are a model for in vitro breast cancer progression. The expression of the two classes (RARs and RXRs) of retinoic acid receptor mRNAs was determined by Northern blot analysis. Receptor functional integrity was determined by testing whether RAR ß mRNA could be induced by retinoic acid. The effects of a 72-h exposure to 50 µM 13-cis retinoic acid on HC11 and HC11ras cell proliferation and HC11 cell differentiation were investigated by flow cytometric cell cycle analysis, and by determination of ß-casein mRNA expression, respectively. The possibility that retinoic acid would induce the expression of the vitamin D receptor and synergize with vitamin D, a known inhibitor of HC11 cell growth, was also investigated. HC11 cells expressed higher mRNA levels of both RAR a and RAR g when compared to HC11ras cells. In contrast, RAR ß, as well as RXR a, ß and g expression was low in both HC11 and HC11ras cells. In addition, RAR ß mRNA was induced by retinoic acid treatment in both cells. In spite of these observations, no effects were seen on cell proliferation or differentiation upon exposure to retinoic acid. Neither vitamin D receptor induction nor synergy with vitamin D on growth inhibition was observed. We conclude that the RAR expression profile could be related to the transformed state in HC11ras cells and that the retinoic acid resistance observed merits further investigation.

A prevalência dos anticorpos protetores contra os três tipos de poliovírus e os níveis de imunidade para os grupos etários de 7, 8, 9, 10, 11, 12, 13, 14 e 15 e mais anos de idade foram determinados, por meio de um inquérito soro-epidemiológico, na população escolar do 19 grau da rede da Prefeitura Municipal de São Paulo. O estudo se baseou numa amostra representativa desta população infantil - exames de 1.489 crianças - e os níveis de imunidade foram determinados em 13 Regionais Administrativas da Prefeitura, bem como nas três zonas geográficas da capital consideradas - "Centro", "Intermediária" e "Periférica". Os resultados mostraram que apenas cerca da metade das crianças apresentaram anticorpos contra todos os três tipos de poliovírus nos diferentes grupos etários examinados, exceto no grupo de 13 anos, em que a percentagem de triplo-imunes alcançou 68,0%. Conseqüentemente, o estado imunitário da população ainda não alcança os níveis desejáveis de 75% de triplo-imunes e a principal causa desta situação foi a prevalência relativamente baixa dos anticorpos contra o poliovírus do tipo 3, em quase todos os grupos etários examinados. O estudo mostrou também que o estado imunitário da população escolar é semelhante nas três zonas geográficas da capital, havendo, entretanto, diferenças consideráveis nas 13 Regionais Administrativas da Prefeitura, onde as percentagens de crianças triplo-imunes variaram de 38,6% a 66,9%. Doses de reforço de vacina oral são recomendadas às crianças em idade escolar.

OBJETIVO: Avaliar como as características demográficas, sociais e comportamentais se associam a diferentes tipos de atividade física. MÉTODOS: Avaliaram-se 37.692 indivíduos de amostra representativa da população portuguesa, no âmbito do Inquérito Nacional de Saúde, 1998-99. A maioria era constituída por mulheres (53,1%) e idade 20 anos. A avaliação da atividade física diária foi baseada em questionário e classificada como: total, de lazer e exercício. Cada tipo foi dicotomizado em baixa intensidade (atividades leves/moderadas) e alta intensidade (atividades pesadas/muito pesadas). Calcularam-se odds ratios (OR) e respectivos intervalos de confiança de 95% por regressão logística não condicional. RESULTADOS: Em ambos os sexos, verificou-se associação inversa significativa entre idade e diferentes tipos de atividade física, e entre a obesidade e a atividade de lazer e exercício. A escolaridade (<4; 5-11; 12 anos) associou-se positivamente com a atividade física de lazer (OR 1; 1,58; 2,39 nas mulheres e OR 1; 1,44; 2,08 nos homens) e com o exercício (OR 1; 3,50; 9,77 nas mulheres e OR 1; 3,42; 7,61 nos homens) e de forma inversa com a AF total (OR 1; 0,65; 0,20 nas mulheres e 1; 0,47; 0,09 nos homens). Independentemente da idade, os solteiros eram frequentemente mais ativos. A atividade de lazer associou-se negativamente ao consumo de bebidas alcoólicas para ambos os sexos e nos homens, ao consumo de tabaco. CONCLUSÕES: Os jovens, normoponderais, solteiros, não-bebedores e os homens não-fumadores apresentaram maior probabilidade de serem fisicamente ativos. Em ambos os sexos, observou-se um efeito diferencial da escolaridade segundo os tipos de atividade física.

OBJETIVO: Analisar a associação entre a conduta conservadora em lesão intraepitelial cervical de alto grau com o índice de recidiva da neoplasia e faixa etária. MÉTODOS: Estudo transversal e retrospectivo realizado com 509 mulheres (15-76 anos) atendidas no período de 1996 a 2006, com colpocitologia oncótica alterada, em um serviço público de referência em Maringá, PR. Os dados foram coletados dos prontuários médicos e estudadas as variáveis diagnóstico definitivo, tipos de tratamento, ocorrência da lesão e recidivas, analisados por meio de testes de associação de qui-quadrado de Pearson e teste exato de Fisher. RESULTADOS: A lesão intraepitelial cervical de alto grau ocorreu em 168 casos; destes, 31 mulheres foram submetidas à amputação cônica, 104 a cirurgias de alta frequência, nove histerectomizadas e 24 receberam conduta conservadora. Dentre as mulheres com lesão de alto grau e tratadas de forma conservadora, oito (33,3%) recidivaram, enquanto dentre as submetidas à conduta não conservadora dez (6,9%) recidivaram, sendo essa diferença estatisticamente significante (p = 0,0009), RP = 4,8 (IC95% 2,11;10,93). Para aquelas que fizeram o seguimento clínico-citológico, três (30,0%) e, dentre as cauterizadas, cinco (35,7%) recidivaram no prazo de três anos, sem diferença significante (p = 0,5611). A recidiva abaixo e acima de 30 anos ocorreu, respectivamente, em sete (13,8%) e 11 (12,2%) mulheres (p = 0,9955). CONCLUSÕES: A idade da mulher não influencia o prognóstico de recidiva. O tratamento conservador deve ser indicado como conduta de exceção, dada a alta taxa de recidiva, e o seguimento deve ser rigoroso, com acompanhamento citológico e colposcópico de até três anos, período em que ocorre a maioria das recidivas.

From January 1984 to May 1994, 17 of 239 children under 15 years old stung by Tityus serrulatus (15.1%) or Tityus bahiensis (84.9%) presented severe envenoming. Of these 17 patients (1-11 years old; median=2 yr) 14 were stung by T.serrulatus and three by T.bahiensis. All of them received scorpion antivenom i.v. at times ranging from 45 min. to 5 h after the accident (median=2h). On admission, the main clinical manifestations and laboratory and electrocardiographic changes were: vomiting (17), diaphoresis (15), tachycardia (14), prostration (10), tachypnea (8), arterial hypertension (7), arterial hypotension (5), tremors (5), hypothermia (4), hyperglycemia (17), leukocytosis (16/16), hypokalemia (13/17), increased CK-MB enzyme activity (>6% of the total CK, 11/12), hyperamylasemia (11/14), sinusal tachycardia (16/17) and a myocardial infarction-like pattern (11/17). Six patients stung by T.serrulatus had depressed left ventricular systolic function assessed by means of echocardiography. Of these, five presented pulmonary edema and four had shock. A child aged two-years old presented severe respiratory failure and died 65 h after being stung by T.serrulatus. Severe envenomations caused by T.serrulatus were 26.2 times more frequent than those caused by T.bahiensis (p<0.001).

Aim: The objective was to describe an outbreak of bloodstream infections by Burkholderia cepacia complex (Bcc) in bone marrow transplant and hematology outpatients.Methods: On February 15, 2008 a Bcc outbreak was suspected. 24 cases were identified. Demographic and clinical data were evaluated. Environment and healthcare workers' (HCW) hands were cultured. Species were determined and typed. Reinforcement of hand hygiene, central venous catheter (CVC) care, infusion therapy, and maintenance of laminar flow cabinet were undertaken. 16 different HCWs had cared for the CVCs. Multi-dose heparin and saline were prepared on counter common to both units.Findings: 14 patients had B. multivorans(one patient had also B. cenopacia), six non-multivorans Bcc and one did not belong to Bcc. Clone A B. multivorans occurred in 12 patients (from Hematology); in 10 their CVC had been used on February 11/12. Environmental and HCW cultures were negative. All patients were treated with meropenem, and ceftazidime lock-therapy. Eight patients (30%) were hospitalized. No deaths occurred. After control measures (multidose vial for single patient; CVC lock with ceftazidime; cleaning of laminar flow cabinet; hand hygiene improvement; use of cabinet to store prepared medication), no new cases occurred.Conclusions: This polyclonal outbreak may be explained by a common source containing multiple species of Bcc, maybe the laminar flow cabinet common to both units. There may have been contamination by B. multivorans (clone A) of multi-dose vials.
