The hormone 1,25-dihydroxyvitamin D3 (1,25-(OH)2D3), the active form of vitamin D3, is an important regulator of calcium homeostasis, exerts antiproliferative effects on various cell systems and can induce differentiation in some kinds of hematopoietic cells. These effects are triggered by its receptor, vitamin D receptor (VDR), a phosphoprotein member of the nuclear receptor superfamily, which functions as a transcriptional factor. VDR binds as a heterodimer with retinoid X receptor (R X R) to hexameric repeats, characterized as vitamin D-responsive elements present in the regulatory region of target genes such as osteocalcin, osteopontin, calbindin-D28K, calbindin-D9K, p21WAF1/CIP1, TGF-ß2 and vitamin D 24-hydroxylase. Many factors such as glucocorticoids, estrogens, retinoids, proliferation rate and cell transformation can modulate VDR levels. VDR is expressed in mammary tissue and breast cancer cells, which are potential targets to hormone action. Besides having antiproliferative properties, vitamin D might also reduce the invasiveness of cancer cells and act as an anti-angiogenesis agent. All of these antitumoral features suggest that the properties of vitamin D could be explored for chemopreventive and therapeutic purposes in cancer. However, hypercalcemia is an undesirable side effect associated with pharmacological doses of 1,25-(OH)2D3. Some promising 1,25-(OH)2D3 analogs have been developed, which are less hypercalcemic in spite of being potent antiproliferative agents. They represent a new field of investigation.

Our objective was to evaluate the concentrations of serum 25-hydroxyvitamin D [25(OH)D], serum calcium, serum phosphorus, alkaline phosphatase, and parathormone (PTH) in patients with polyarticular juvenile idiopathic arthritis (JIA) and to associate them with disease duration and activity, bone mineral density and use of medications. In a cross-sectional and controlled study, 30 patients with polyarticular JIA were evaluated and compared to 30 healthy individuals matched for age and gender. Clinical status, anthropometry, laboratory markers in both patients and controls, and bone mineral density, only in the patients, were measured. Of the 30 patients included in the study, 23 (76.7%) were female and 16 (53.3%) non-Caucasian; mean age was 14 years (range = 4 to 20 years). Mean disease duration was 5 years (range = 1 to 12 years). The mean concentrations of serum albumin-corrected calcium (9.04 ± 0.41 mg/dL) and alkaline phosphatase (153.3 ± 100.1 IU) were significantly lower in patients with JIA than in controls (P < 0.0001 and P = 0.001, respectively). No differences in 25(OH)D, PTH or serum phosphorus were observed between JIA and control subjects. Regarding 25(OH)D concentration, 8 patients (26.7%) and 5 controls (16.7%) had 25(OH)D concentrations compatible with deficiency (lower than 20 ng/mL) and 14 patients (46.7%) and 18 controls (60%) had concentrations compatible with insufficiency (20-32 ng/mL). These values were not associated with disease activity, use of medications or bone mineral density. We observed a high frequency of 25(OH)D insufficiency and deficiency in the study sample. The compromised bone metabolism emphasizes the importance of follow-up of JIA patients.

Este trabalho teve como objetivo avaliar o efeito de indutores de florescimento em três cultivares de abacaxizeiro em Rio Branco-Acre. O delineamento experimental foi em blocos ao acaso, em esquema fatorial 3 x 2 x 2, com quatro repetições, sendo combinadas três cultivares de abacaxi (RBR-1, SNG-2 e SNG-3), dois indutores de florescimento (carbureto de cálcio- CaC2 e etefon-ácido 2-cloroetilfosfônico) e duas épocas de aplicação (aos 10 e 12 meses do plantio). As plantas induzidas com etefon aos 10 meses apresentaram maior percentagem de florescimento (96,25%) quando comparadas com as induzidas com CaC2 (62,92%). A percentagem de florescimento foi significativamente superior nas plantas induzidas aos 12 meses com CaC2 (85,42%) quando comparadas com as induzidas aos 10 meses (62,92%).

Low bone remodeling and relatively low serum parathyroid hormone (PTH) levels characterize adynamic bone disease (ABD). The impact of renal transplantation (RT) on the course of ABD is unknown. We studied prospectively 13 patients with biopsy-proven ABD after RT. Bone histomorphometry and bone mineral density (BMD) measurements were performed in the 1st and 12th months after RT. Serum PTH, 25-hydroxyvitamin D, 1,25-dihydroxyvitamin D, and osteocalcin were measured regularly throughout the study. Serum PTH levels were slightly elevated at transplantation, normalized at the end of the third month and remained stable thereafter. Bone biopsies performed in the first month after RT revealed low bone turnover in all patients, with positive bone aluminum staining in 5. In the 12th month, second biopsies were performed on 12 patients. Bone histomorphometric dynamic parameters improved in 9 and were completely normalized in 6, whereas no bone mineralization was detected in 3 of these 12 patients. At 12 months post-RT, no bone aluminum was detected in any patient. We also found a decrease in lumbar BMD and an increase in femoral BMD. Patients suffering from ABD, even those with a reduction in PTH levels, may present partial or complete recovery of bone turnover after successful renal transplantation. However, it is not possible to positively identify the mechanisms responsible for the improvement. Identifying these mechanisms should lead to a better understanding of the physiopathology of ABD and to the development of more effective treatments.

The range of 25-hydroxyvitamin D (25OHD) concentration was determined in a young healthy population based on bone metabolism parameters and environmental and behavioral aspects. We studied 121 healthy young volunteers (49 men, 72 women) living in São Paulo (23º 34' south latitude) belonging to three occupational categories: indoor workers (N = 28), medical school students (N = 44), and resident physicians (N = 49). Fasting morning blood samples were collected once from each volunteer from August 2002 to February 2004, and 25OHD, total calcium, albumin, alkaline phosphatase, phosphorus, creatinine, intact parathyroid hormone, osteocalcin, and type I collagen carboxyterminal telopeptide were measured. Data are reported as means ± SD. Mean subject age was 24.7 ± 2.68 years and mean 25OHD level for the entire group was 78.7 ± 33.1 nM. 25OHD levels were lower (P < 0.05) among resident physicians (67.1 ± 27.0 nM) than among students (81.5 ± 35.8 nM) and workers (94.0 ± 32.6 nM), with the last two categories displaying no difference. Parathyroid hormone was higher (P < 0.05) and osteocalcin was lower (P < 0.05) among resident physicians compared to non-physicians. Solar exposure and frequency of beach outings showed a positive association with 25OHD (P < 0.001), and summer samples presented higher results than winter ones (97.8 ± 33.5 and 62.9 ± 23.5 nM, respectively). To define normal levels, parameters such as occupational activity, seasonality and habits related to solar exposure should be taken into account. Based on these data, we considered concentrations above 74.5 nM to be desired optimal 25OHD levels, which were obtained during the summer for 75% of the non-physicians.

We evaluated the concentrations of 25-hydroxyvitamin D [25(OH)D] in children and adolescents with juvenile systemic lupus erythematosus (JSLE) and associated them with disease duration and activity, use of medication (chloroquine and glucocorticoids), vitamin D intake, calcium and alkaline phosphatase levels, and bone mineral density. Thirty patients with JSLE were evaluated and compared to 30 healthy individuals, who were age and gender matched. Assessment was performed of clinical status, disease activity, anthropometry, laboratory markers, and bone mineral density. The 30 patients included 25 (83.3%) females and 16 (53.3%) Caucasians, with a mean age of 13.7 years. The mean age at diagnosis was 10.5 years and mean disease duration was 3.4 years. Mean levels of calcium, albumin, and alkaline phosphatase were significantly lower in patients with JSLE compared with controls (P<0.001, P=0.006, and P<0.001, respectively). Twenty-nine patients (97%) and 23 controls (77%) had 25(OH)D concentrations lower than 32 ng/mL, with significant differences between them (P<0.001). Fifteen patients (50%) had vitamin D levels <20 ng/mL and 14 had vitamin D levels between 20 and 32 ng/mL. However, these values were not associated with greater disease activity, higher levels of parathormone, medication intake, or bone mineral density. Vitamin D concentrations were similar with regard to ethnic group, body mass index, height for age, and pubertal stage. Significantly more frequently than in controls, we observed insufficient serum concentrations of 25(OH)D in patients with JSLE; however, we did not observe any association with disease activity, higher levels of parathormone, lower levels of alkaline phosphatase, use of medications, or bone mineral density alterations.

Chromoblastomycosis (CR) is a subcutaneous chronic mycosis characterized by a granulomatous inflammatory response. However, little is known regarding the pattern of leukocyte subsets in CR and the pathways involved in their recruitment. The objective of this study was to assess the cellular subsets, chemokine, chemokine receptors and enzymes in CR. The inflammatory infiltrate was characterized by immunohistochemistry using antibodies against macrophages (CD68), Langerhans'cells (S100), lymphocytes (CD3, CD4, CD8, CD45RO, CD20 and CD56) and neutrophils (CD15). The expression of MIP-1alpha (Macrophage inflammatory protein-1alpha), chemokine receptors (CXCR3 and CCR1) and enzymes (superoxide dismutase-SOD and nitric oxide synthase-iNOS) was also evaluated by the same method. We observed an increase in all populations evaluated when compared with the controls. Numbers of CD15+ and CD56+ were significantly lower than CD3+, CD4+, CD20+ and CD68+ cells. Statistical analysis revealed an association of fungi numbers with CD3, CD45RO and iNOS-positive cells. Furthermore, MIP-1alpha expression was associated with CD45RO, CD68, iNOS and CXCR3. Our results suggest a possible role of MIP-1alpha and fungi persistence in the cell infiltration in CR sites.

As espécies de plantas aquáticas podem causar inúmeros inconvenientes ao uso múltiplo da água quando elas se desenvolvem desordenadamente. Assim, o objetivo deste trabalho foi avaliar a eficiência de diferentes herbicidas no controle químico de plantas de Alternanthera philoxeroides, Enhydra anagallis e Pycreus decumbens em caixa-d'água. Quando as plantas atingiram o seu pleno desenvolvimento (antes do florescimento), foram aplicados, nas espécies Alternanthera philoxeroides e Enhydra anagallis, os herbicidas: 2,4-D amina (U-46 D FLUID 720) a 2.880 g e.a. ha-1; diquat (REWARD 240) a 480 g i.a. ha-1; imazapyr (ARSENAL 250) a 500 e 750 g e.a. ha-1; glyphosate (RODEO 480) a 3.360 g e.a. ha-1 com e sem o surfatante Aterbane BR (0,5% v v-1); glyphosate + diquat (3.360 + 480 g i./e.a. ha-1 ); glyphosate + 2,4-D (3.360 + 2.880 g e.a. ha-1); e diquat + 2,4-D (480 + 2.880 g i./e.a. ha-1), além de uma testemunha sem aplicação de herbicida. Para a espécie Pycreus decumbens foram aplicados: 2,4-D amina (U-46 D FLUID 720) a 2.880 g e.a. ha-1; diquat (REWARD 240) a 480 g i.a. ha-1; propanil (STAM 480) a 2.880 g i.a. ha-1; glyphosate (RODEO 480) a 3.360 g e.a. ha-1 mais o surfatante Aterbane BR (0,5% v v-1); glyphosate + propanil (3.360 + 2.880 g i./e.a. ha-1); glyphosate + diquat (3.360 + 480 g i./e.a. ha-1); glyphosate + 2,4-D (3.360 + 2.880 g e.a. ha-1); propanil + 2,4-D (2.880 + 2.880 g i./e.a. ha-1); e diquat + 2,4-D (480 + 2.880 g i./e.a. ha-1), além de uma testemunha sem aplicação de herbicida. Os herbicidas foram aplicados com um pulverizador estacionário, pressurizado a ar comprimido e equipado com um reservatório de 2 litros, pontas Teejet XR11002VS, com um consumo de calda de 200 L ha-1. As avaliações de controle das plantas daninhas foram visuais, por meio de uma escala de percentual de notas, além de se avaliar a massa seca das plantas. Verificou-se que o controle químico apresenta-se como uma boa alternativa de manejo para as espécies A. philoxeroides, E. anagallis e P. decumbens, e a mistura de herbicidas pode aumentar a eficiência de controle. E. anagallis apresentou alta sensibilidade à ação dos herbicidas; entretanto, as espécies A. philoxeroides e P. decumbens evidenciaram alta capacidade de regeneração, principalmente quando se utilizaram herbicidas de ação de contato.
Effect of D-alpha-tocopherol on tubular nephron acidification by rats with induced diabetes mellitus

The objective of the present study was to determine if treatment of diabetic rats with D-alpha-tocopherol could prevent the changes in glomerular and tubular function commonly observed in this disease. Sixty male Wistar rats divided into four groups were studied: control (C), control treated with D-alpha-tocopherol (C + T), diabetic (D), and diabetic treated with D-alpha-tocopherol (D + T). Treatment with D-alpha-tocopherol (40 mg/kg every other day, ip) was started three days after diabetes induction with streptozotocin (60 mg/kg, ip). Renal function studies and microperfusion measurements were performed 30 days after diabetes induction and the kidneys were removed for morphometric analyses. Data are reported as means ± SEM. Glomerular filtration rate increased in D rats but decreased in D + T rats (C: 6.43 ± 0.21; D: 7.74 ± 0.45; D + T: 3.86 ± 0.18 ml min-1 kg-1). Alterations of tubular acidification observed in bicarbonate absorption flux (JHCO3) and in acidification half-time (t/2) in group D were reversed in group D + T (JHCO3, C: 2.30 ± 0.10; D: 3.28 ± 0.22; D + T: 1.87 ± 0.08 nmol cm-2 s-1; t/2, C: 4.75 ± 0.20; D: 3.52 ± 0.15; D + T: 5.92 ± 0.19 s). Glomerular area was significantly increased in D, while D + T rats exhibited values similar to C, suggesting that the vitamin prevented the hypertrophic effect of hyperglycemia (C: 8334.21 ± 112.05; D: 10,217.55 ± 100.66; D + T: 8478.21 ± 119.81µm²). These results suggest that D-alpha-tocopherol is able to protect rats, at least in part, from the harmful effects of diabetes on renal function.

Graft rejection is the major cause of failure of HLA mismatched bone marrow transplantation because of residual host immunity. we have proposed to use a monoclonal murine antibody specific for the LFA-1 molecule (25-3) to prevent graft failure in HLA mismatched bone marrow transplantation (BMT). The rationale for this approach is three fold: LFA-1 deficient patients (3/3) do not reject HLA mismatched BMT; anti LFA-1 blocka in vitro the induction of T cell responses and T/ non T cytotoxic functions; LFA-1 is not expressed by other cells than leucocytes. We have accordingly treated twenty two patients with inherited diseases and 8 with leikemia. The bone marrow was T cells depled by E rosetting of Campath antibody. The antibody was given at days -3, -1, +1, +3, +5 at dose of .1 mg/kg/d for the first 9 and then .2mg/kg/d from day -3 to +6. Engraftment occured in 23/30 patients as shown by at least HLA typing. Hematological recovery was rapid, GVH was limited. Side effects of antibody infusion included fever and possibly an increased incidence of early bacteral infection (sepsis, 1 death). Immunological reconstitution occured slowly leading in six cases to EBV-induced B cell poliferation (1 death and in two others to transient auto immune hemolytic anemia. There has been only one secondary graft rejection. Sisteen patients are alive 3 to 26 months post transplant with functional grafts. Although the number of patients treated is still low the absence of late rejection so far, gives hope for long term maintenance of the graft using anti LFA-1. Since the antibody is an IgG 1 unable to bind human complement, and since it is known to inhibit phagocytosis, there is a good suggestion that 25-3 act through functional blocking of host T and non T luymphocytes at both induction and effector levels.

Na execução de uma análise química de solo são empregados diversos procedimentos que, mesmo seguindo-se o protocolo preconizado pelo método de análise utilizado, estão sujeitos a variações nos resultados analíticos, causadas por manipulação das amostras ou dos materiais utilizados. Objetivou-se neste estudo avaliar a influência do grau de moagem da amostra, do tipo de frasco e do volume vazio no frasco na execução dos métodos Mehlich-1 e Mehlich-3 para determinar o P no solo. Para tanto, foram conduzidos três experimentos. No primeiro experimento, as amostras de solo foram moídas e tamisadas em peneiras com aberturas de 2,000; 1,700; 0,850; 0,600; e 0,300 mm. No segundo, foram utilizados dois modelos de frasco (erlenmeyer e snap-cap), ambos com volume de 50 mL. Já no terceiro, foi alterado o volume vazio no frasco, mantendo-se a relação solo:solução extratora utilizando-se as quantidades dentro do frasco de 1:10; 1,5:15; 2,5:25; 3:30; e 4:40 cm³ cm-3. O grau de moagem das amostras não influenciou a capacidade de extração do Mehlich-1; entretanto, a capacidade extrativa do Mehlich-3 foi influenciada, principalmente em solos argilosos. Tanto para o Mehlich-1 quanto para o Mehlich-3, os teores de P extraído foram significativamente mais elevados com o uso de frasco tipo snap-cap em relação ao erlenmeyer. O volume vazio no frasco alterou os teores de P extraído para o Mehlich-1 e Mehlich-3 em 100 e 64 % das amostras, respectivamente. Deve-se padronizar a intensidade da moagem das amostras de solo para extração do P pela solução de Mehlich-3. Um modelo único de frasco deve ser adotado pelos laboratórios de rotina para análise do P, independentemente do método de extração, mantendo-se sempre constante no frasco o volume da amostra (cm³) para o volume de solução extratora (cm³).

In this paper we report the synthesis of biologically active compounds through a [3+4] cycloaddition reaction to produce the main frame structure, followed by several conventional transformations. The 1,2alpha,4alpha,5-tetramethyl-8-oxabicyclo[3.2.1]oct-6-en-3-one (11) obtained from a [3+4] cycloaddition reaction was converted into 1,2alpha,4alpha,5-tetramethyl-6,7-exo-isopropylidenedioxi-8 -oxabicyclo[3.2.1]octan-3-one (13) in 46% yield. This was further converted into the alcohols 1,2alpha,4alpha,5-tetramethyl-6,7-exo-isopropylidenedioxi-8-oxabicyclo[3.2.1]octan-3 alpha-ol (14), 1,2alpha,4alpha,5-tetramethyl-6,7-exo-isopropylidenedioxi-8 -oxabicyclo[3.2.1]octan-3beta-ol (15), 1,2alpha,4alpha,5-tetramethyl-3-butyl-6,7-exo-isopropylidenedioxi-8-oxabicyclo[3.2.1]octan-3 alpha-ol (17), 1,2alpha,4alpha,5-tetramethyl-3-hexyl-6,7-exo-isopropylidenedioxi-8-oxabicyclo[3.2.1]octan-3 alpha-ol (18) and 1,2alpha,4alpha,5-tetramethyl-3-decyl-6,7-exo-isopropylidenedioxi-8-oxabicyclo[3.2.1]octan-3 alpha-ol (19). Dehydration of 17, 18 and 19 with thionyl chloride in pyridine resulted in the alkenes 20, 21 and 22 in ca. 82% - 89% yields from starting alcohols. The herbicidal activity of the compounds synthesized was evaluated at a concentration of 100 µg g-1. The most active compound was 21 causing 42,7% inhibition against Cucumis sativus L.

O presente trabalho teve como objetivo determinar os efeitos de subdoses do herbicida 2,4-D na cultura do fumo. Os experimentos foram conduzidos a campo durante o ano de 2003, no município de Santa Isabel do Ivaí - PR, utilizando-se de fumo tipo Virgínia. No primeiro experimento, foram aplicadas, no estádio de pré-florescimento, doses de 0,84; 1,68; 3,36; 6,72; 13,44 e 26,88 g e.a. ha-1 de 2,4-D, equivalentes à deriva de 0,125; 0,25; 0,5; 1,0; 2,0 e 4,0% de produtos comerciais aplicados a 1,0 L ha-1 (670 g e.a. ha-1). Para o segundo experimento, os tratamentos foram constituídos pela combinação em esquema fatorial de duas doses (6,72 e 13,44 g e.a. ha-1, equivalentes a 1,0 e 2,0% de deriva) e três épocas de aplicação após o transplante para o campo. Nenhum sintoma visual marcante de fitotoxicidade foi observado dentro da faixa de doses avaliada. Visualmente, alguns dos possíveis efeitos atribuídos à fitotoxicidade ocasionada pela deriva de 2,4-D na cultura do fumo podem estar associados à utilização de antibrotantes. Nenhuma das doses aplicadas no primeiro experimento afetou a produtividade da cultura, nem o aspecto e qualidade das folhas, tendo o fumo tolerado até 4,0% de deriva avaliada. Resultados obtidos no segundo experimento confirmaram esses dados, constatando-se que níveis de deriva de até 2,0% (13,44 g e.a. ha-1), aplicados em três fases após o transplante das mudas para o campo, não causaram qualquer efeito negativo sobre a produtividade da cultura.

Em geral, a eficiência de herbicidas inibidores da ACCase é reduzida quando da aplicação em conjunto com herbicidas latifolicidas. Dois experimentos - em casa de vegetação e campo - objetivaram determinar a existência de antagonismo nas associações de clodinafop-propargyl a metsulfuron-methyl ou 2,4-D, para controle de azevém anual. Em casa de vegetação, o delineamento foi o completamente casualizado, com 21 tratamentos compostos pela aplicação de clodinafop (0, 3, 6, 12, 24, 48 e 96 g ha-1) isolado ou em mistura com metsulfuron-methyl (2 e 4 g ha-1) ou 2,4-D (470 g ha-1). Em campo, o delineamento foi de blocos casualizados, com 12 tratamentos, arranjados em fatorial 6 x 2, composto pelos níveis de clodinafop-propargyl (0, 3, 6, 12, 24, 48 e 96 g ha¹) isolado ou em mistura com metsulfuron-methyl (2 g ha-1). A avaliação visual de controle, em casa de vegetação, revelou I50 das misturas de clodinafop-propargyl + metsulfuron-methyl (2 e 4 g ha-1) ou clodinafop-propargyl + 2,4-D (470 g ha-1), respectivamente, 33, 84 e 151% superiores ao de clodinafop-propargyl isolado. Já para matéria verde de azevém, o I50 das misturas supracitadas foi, respectivamente, 119, 244 e 72% superior ao de clodinafop-propargyl isolado. Em campo, ocorreu redução da matéria verde de azevém com a elevação dos níveis de clodinafop-propargyl isolado, mas não houve variação da matéria verde com a elevação dos níveis de clodinafop-propargyl, em associação a metsulfuron-methyl. Os resultados evidenciam a existência de antagonismo entre clodinafop-propargyl e os herbicidas metsulfuron-methyl e 2,4-D. São feitas considerações sobre as possíveis vantagens da aplicação dos herbicidas em mais de uma operação de controle, em comparação à associação entre graminicidas e latifolicidas.

Human herpesvirus-8 (HHV-8) appears to be transmitted mainly by sexual contact. However, several studies suggest that in developing countries the infection may be acquired early in life by routes other than sexual transmission. The present study estimated the seroprevalence of HHV-8 in Brazilian children born to HIV-1-infected mothers. The serum samples were collected in a cross-sectional cohort study from 99 children born to HIV-infected mothers (median age 3.27 years; range 1.5-13.8 years) attending the outpatient clinic of the Federal University of São Paulo. IgG antibodies to HHV-8 latency-associated nuclear antigen and lytic phase antigens were detected by immunofluorescence assays. The samples tested were collected from children aged 12 months or older to exclude the possibility of cross-placental antibody transport. The total prevalence of anti-lytic antibodies in this population (5/99; 5%) reveals that HHV-8 infection can occur during childhood. Children aged 1.5 to 2 years had a seroprevalence of 2% (1/50) and children aged 3.25 to 13.8 years had a seroprevalence of 8% (4/49). This difference was not statistically significant, probably because of the small size of the sample, but it suggests that HHV-8 infection occurs more commonly late in infancy. Further prospective studies are necessary to evaluate the timing and risk factors for primary HHV-8 infection in the pediatric population.
