We compared the in vitro growth of promastigotes from two Leishmania species in TC-100 and Schneider media. Leishmania (Leishmania) amazonensis replication rates were similar in both tissue culture media and reached maximum rates by 48 h. In contrast Leishmania (Viannia) braziliensis growth was significantly greater in TC-100 but maximum rates were achieved by 96 h. Folic acid appears to be the limiting factor and supplementation of Schneider media with this nutrient improved L. (V.) braziliensis replication rates and decreased the time of maximum replication to 48 h.

Antimalarial drugs including the antifolate, pyrimethamine-sulfadoxine (PS), can modulate the prevalence and intensities of gametocytaemia following treatment of acute malaria infections. They may also directly influence the transmission and spread of drug insensitivity. Little is known of the effects of co-trimoxazole (Co-T), another antifolate antimalarial, on gametocytes in children with acute malaria infections. We compared the effects of Co-T and PS on the prevalence and intensities of gametocytaemia and gametocyte sex ratios in 102 children aged 0.5-12 years presenting with acute and uncomplicated falciparum malaria. Compared to pre-treatment, both drugs significantly increased gametocyte carriage post-initiation of treatment. However, gametocyte carriage was significantly lower on day 14 in those treated with Co-T than PS. Significant increase in gametocytaemia with time occurred in PS - but not Co-T-treated children. Kaplan-Meier survival curve of the cumulative probability of remaining gametocyte-free in children who were agametocytaemic at enrolment showed that by day 7 of follow up, children treated with PS had a significantly higher propensity to have developed gametocytes than in Co-T-treated children (Log-rank statistic 5.35, df = 1, P = 0.02). Gametocyte sex ratio changes were similar following treatment with both drugs. PS and Co-T treatment of acute malaria infections in children from this endemic area is associated with significant increases in prevalence and intensities of gametocytaemia but these effects are more marked in those treated with PS than Co-T.

The present work provides information on Trypanosoma cruzi genotype circulating in endemic areas of Chagas disease in Panama. A total of 26 crude stocks of T. cruzi, isolated from the blood of persons with different clinical profiles of Chagas disease were collected and crio-conserved until used. Most of the stocks had been characterized by means of isoenzyme electrophoresis on cellulose acetate membranes. The clinical profiles of infected persons included 9 (34.6%) asymptomatic and 17 acute (65.4%) including 5 (19.2%) fatal cases, 2 under 5 years old and 3 adults. A multiplex-PCR assay based on the amplification of the non-transcribed spacer of the mini-exon gene was performed. All stocks of T. cruzi included in the study were found to correspond to Tc I group. This result supports the predominance of T. cruzi-I in the transmission cycles affecting the human population in the Republic of Panama.

The effects of pyrimethamine-sulphadoxine (PS), chloroquine plus chlorpheniramine, a H1 receptor antagonist that reverses chloroquine resistance in Plasmodium falciparum in vitro and in vivo (CQCP), and amodiaquine plus pyrimethamine-sulphadoxine (AQPS) on gametocyte production were evaluated in 157 children with acute, symptomatic, uncomplicated falciparum malaria who were treated with these drugs. PS was significantly less effective than CQCP or AQPS at clearing asexual parasitaemia or other symptoms of malaria. Gametocyte carriage on days 3, 7, and 14 were significantly higher in those treated with PS. The ratio of the density (per µl blood) of peripheral young gametocyte (PYG), that is, < stage III to peripheral mature gametocyte (PMG), that is, stage IV and V, an index of continuing generation of gametocytes, rose to 1 by day 7 of treatment in those treated with PS, but remained consistently below 1 in the other treatment groups. PYG-PMG density ratio increased significantly from day 0-14 in those treated with PS and CQCP (chi2 = 76, P = 0.000001 and chi2 = 42.2, P = 0.00001, respectively) but decreased significantly in those treated with AQPS (chi2 = 53.2, P = 0.000001). Both PS-sensitive and -resistant infections generated PYG (18 of 29 vs 13 of 20, chi2 = 0.04, P = 0.93) but PYG was present only in those with resistant response to CQCP. Combination of PS with amodiaquine (AQ), that is, (AQPS) resulted in less production of PYG, but in this setting, PYG was not indicative of response to AQPS. These data indicate that PS enhanced production or release of young gametocytes when used alone, but generated less young gametocytes when used in combination with AQ. PYG may be used as an indicator of response to CQCP but not PS or PS-based combination drugs.

High-fat diets induce weight gain and fatty liver in wild-type mice. Schistosomiasis mansoni infection also promotes hepatic injury. This study was designed to quantify hepatic alterations in schistosomiasis mansoni-infected mice fed a high fat-rich chow compared to mice fed a standard rodent chow, using stereology. Female SW mice fed each either high-fat diet (29% lipids) or standard chow (12% lipids) over 8 months, and then were infected with Schistosoma mansoni cercariae. Four experimental groups were studied: infected mice fed a high-fat diet (IHFC) or standard chow (ISC), uninfected mice fed a high-fat diet (HFC) or standard chow (SC). Mice were sacrificed during early infection (9 weeks from exposure). The following hepatic biometry and the stereology parameters were determined: volume density (hepatocytes [h], sinusoids [s], steatosis [st] and hepatic fibrosis [hf]); numerical density (hepatocyte nuclei - Nv[h]); absolute number of total hepatocyte N[h], normal hepatocyte N[nh], and binucleated hepatocyte N[bh], percentage of normal hepatocyte P[nh] and binucleated hepatocyte P[bh]. IHFC and HFC groups exhibited TC, HDL-C, LDL-C, and body mass significantly greater (p < 0.05) than control group. No significant differences were found regards liver volume (p = 0.07). Significant differences were observed regards P[nh] (p = 0.0045), P[bh] (p = 0.0045), Nv[h] (p = 0.0006), N[h] (p = 0.0125), N[bh] (p = 0.0164) and N[nh] (p = 0.0078). IHFC mice group presented 29% of binucleated hepatocytes compared to HFC group (19%), ISC group (17%) and SC (6%). Volume density was significantly different between groups: Vv[h] (p = 0.0052), Vv[s] (p = 0.0025), Vv[st] (p = 0.0004), and Vv[hf] (p = 0.0007). In conclusion, schistosomiasis mansoni infection with concurrent high-fat diet promotes intensive quantitative changes in hepatic structure, contributing to an increasing on hepatic regeneration.

Transmission of Trypanosoma cruziduring pregnancy is estimated to occur in less than 20% of infected mothers; however, the etiopathogenesis is not completely understood. The Centre for Studies on Chagas Disease provides confirmation of T. cruziinfection for individuals living in central Brazil. In this retrospective hospital-based study, all requests for diagnosis of T. cruziinfection in individuals less than 21 years old from 1994-2014 were searched. We end with 1,211 individuals and their respective infected mothers. Congenital transmission of infection was confirmed in 24 individuals (2%) in central Brazil, an area where the main T. cruzi lineage circulating in humans is TcII. This low prevalence of congenital Chagas disease is discussed in relation to recent findings in the south region of Brazil, where TcV is the main lineage and congenital transmission has a higher prevalence (approximately 5%), similar to frequencies reported in Argentina, Paraguay and Bolivia. This is the first report to show geographical differences in the rates of congenital transmission of T. cruziand the relationship between the prevalence of congenital transmission and the type of Tc prevalent in each region.

Lutzomyia columbiana es un flebotomíneo considerado como vector sospechoso de Leishmania mexicana y Leishmania braziliensis en Colombia. Este insecto pertenece al grupo verrucarum, que incluye algunos taxones isomórficos, lo que ha estimulado la búsqueda de marcadores moleculares que permitan, además de diferenciar las especies, estudiar sus relaciones de parentesco. En este artículo se describe por primera vez la estructura putativa del ARN de transferencia mitocondrial para serina que reconoce el codón UCN (ARNtSer) de Lu. columbiana. El ADN genómico fue extraído, amplificado y secuenciado a partir de seis especímenes colectados con cebo humano. La estructura secundaria del ARNtSer fue inferida con el programa tRNAscan-SE 1.21. El gen ARNts consistió de 67 pares de bases (pb), encontrándose un solo haplotipo en los seis individuos secuenciados. El ARNtSer de Lu. columbiana mostró 7 apareamientos intracatenarios en el brazo aceptor del aminoácido, 3 en el brazo dihidrouridina (DHU), 5 en el brazo del anticodón y 5 en el brazo ribotimidina-pseudouridina-citosina (TøC). El tamaño de las lupas correspondió a 5 nucleótidos en la DHU, 7 en la anticodón, 4 en la variable y 7 en la TøC. Lu. columbiana se distingue del resto de especies de Lutzomyia y Phlebotomus secuenciadas a la fecha por la presencia de una guanina en la posición nucleotídica 64, que produce un apareamiento no canónico tipo uracilo-guanina en el brazo aceptor. Se necesitan más estudios para confirmar la utilidad del ARNtSer como marcador molecular para la discriminación de especies de flebotomíneos.

Neste trabalho, foram comparadas as características dos solos de áreas de floresta e áreas de pastagens, considerando quatro diferentes idades de instalação, em três sítios, no estado de Rondônia. Os sítios foram escolhidos com o auxílio de imagens do sensor TM/Landsat, obtidas entre 1987 e 1997, sendo dois em Argissolo Vermelho-Amarelo (PVA) e um em Luvissolo Crômico (TC). Amostras de solo foram colhidas na camada de solo a 20 cm e submetidas à análise de fertilidade e granulométrica, obtendo-se os dados de pH, bases trocáveis, fósforo disponível, saturação por alumínio e por bases e carbono orgânico. Para o solo distrófico e de baixa fertilidade natural (PVA), a adição de cinza proveniente da queima da floresta e os restos vegetais nos primeiros anos de implantação de pastagem influenciaram bastante a dinâmica de solo. Para os dois sítios de PVA, o nível de saturação por bases manteve-se superior na área de pastagem, mesmo com mais de 10 anos de uso, ao encontrado na área de floresta. Já o alumínio mostrou baixo nível após a implantação de pastagem em todas as classes de idade estudadas. O fósforo disponível sofreu queda rápida após atingir o valor de pico e, após um período de mais de 10 anos de uso com pastagem, apresentou valor praticamente igual ao original, nos dois tipos de solo estudados. No caso do solo eutrófico (TC), o efeito da cinza, em geral, foi de menor intensidade em relação ao do solo distrófico (PVA), mostrando alta perda de nutrientes no tempo, decorrente de fatores associados ao baixo teor de argila do solo e ao relevo local mais acentuado.

A fitorremediação de solos e substratos contaminados por elementos tóxicos tem despertado crescente interesse entre pesquisadores e técnicos. Particularmente em relação ao As, o obstáculo ao emprego desta técnica é o pequeno número de espécies identificadas capazes de acumular este elemento. O objetivo deste trabalho foi avaliar o potencial de plantas de estilosante (Stylosanthes humilis HBK), amendoim (Arachis pintoi Krapov. & Gregory), aveia (Avena strigosa Schreb) e azevém (Lolium multiflorum Lam.) como espécies fitorremediadoras de áreas contaminadas por As. Amostras de Latossolo Vermelho-Amarelo foram incubadas por 15 dias com diferentes doses de As: 0; 50; 100; e 200 mg dm-3. Em seguida, realizaram-se a semeadura das quatro espécies e as respectivas adubações. Aos 65 dias após a semeadura, as plantas foram avaliadas quanto à altura, à matéria seca da parte aérea e raízes. Determinaram-se os teores de As nas folhas jovens, intermediárias e basais, no caule e nas raízes, bem como o conteúdo e o índice de translocação (IT) de As. Por meio de análises de regressão, foram estimados os teores críticos (TC) de As disponíveis no solo, que proporcionaram redução de 50 % da matéria seca. As espécies estudadas apresentaram comportamento diferenciado quanto à tolerância ao As, com destaque para azevém, amendoim e estilosante, que não apresentaram lesões foliares decorrentes de fitotoxidez por esse elemento. Os TC para as plantas de aveia e azevém foram significativamente superiores aos observados para as demais espécies, caracterizando-as como espécies tolerantes ao As. As plantas de amendoim e estilosante apresentaram maior capacidade de absorção e maior IT de As para a parte aérea. As plantas de amendoim apresentaram maiores teores nas folhas basais e raízes, mostrando potencial para serem utilizadas em programas de fitorremediação. As plantas de azevém, amendoim e estilosante podem ser utilizadas na fitoestabilização e, ou, na revegetação de áreas contaminadas por As, uma vez que apresentaram tolerância a esse elemento. Por se tratar de espécies forrageiras, quando utilizadas para esses fins, cuidados especiais são necessários, como o isolamento da área, para evitar a entrada do elemento na cadeia trófica.

A sustentabilidade do arroz (Oryza sativa L.) irrigado em solos de várzea está alicerçada na utilização da rotação e sucessão de culturas, fundamentais para o controle do arroz-vermelho e preto. Os reflexos sobre os atributos dos solos de várzea merecem estudos em especial sobre a compactação do solo. O objetivo deste trabalho foi identificar camadas compactadas em Planossolo submetido a diferentes sistemas de cultivo e preparo, avaliando-se a densidade do solo (Ds) pela Tomografia Computadorizada de Raios Gama (TC). A análise foi realizada em um experimento de longa duração, conduzido de 1985 a 2004, na Estação Experimental da Embrapa Clima Temperado, Capão do Leão, RS, num delineamento experimental em blocos ao acaso, com sete tratamentos, cada um com quatro repetições (T1 - um ano de arroz com preparo convencional do solo seguido de dois anos de pousio; T2 - cultivo contínuo de arroz com preparo convencional do solo; T4 - rotação de arroz e soja (Glycine max L.) com preparo convencional do solo; T5 - rotação de arroz, soja e milho (Zea maiz L.) em preparo convencional do solo; T6 - plantio direto de arroz no verão em sucessão do azevém (Lolium multiflorum L.) no inverno; T7 - rotação de arroz sob plantio direto e soja sob preparo convencional do solo; T8 - testemunha: solo sem cultivo). A Tomografia Computadorizada de Raios Gama permitiu detectar que o plantio direto de arroz no verão em sucessão do azevém no inverno não resultou na formação de camadas compactadas; a utilização de dois anos de pousio, no sistema de produção de arroz irrigado, não foi suficiente para evitar a formação de uma camada superficial compactada; e a rotação de arroz, soja e milho com preparo convencional do solo apresentou duas camadas compactadas (0,0 a 1,5 cm e 11 a 14 cm), indicando que essas podem limitar a produção agrícola nesse sistema de produção em Planossolos.

The occurrence of Umbric Ferralsols with thick umbric epipedons (> 100 cm thickness) in humid Tropical and Subtropical areas is a paradox since the processes of organic matter decomposition in these environments are very efficient. Nevertheless, this soil type has been reported in areas in the Southeast and South of Brazil, and at some places in the Northeast. Aspects of the genesis and paleoenvironmental significance of these Ferralsols still need a better understanding. The processes that made the umbric horizons so thick and dark and contributed to the preservation of organic carbon (OC) at considerable depths in these soils are of special interest. In this study, eight Ferralsols with a thick umbric horizon (UF) under different vegetation types were sampled (tropical rain forest, tropical seasonal forest and savanna woodland) and their macromorphological, physical, chemical and mineralogical properties studied to detect soil characteristics that could explain the preservation of high carbon amounts at considerable depths. The studied UF are clayey to very clayey, strongly acidic, dystrophic, and Al-saturated and charcoal fragments are often scattered in the soil matrix. Kaolinites are the main clay minerals in the A and B horizons, followed by abundant gibbsite and hydroxyl-interlayered vermiculite. The latter was only found in UFs derived from basalt rock in the South of the country. Total carbon (TC) ranged from 5 to 101 g kg-1 in the umbric epipedon. Dichromate-oxidizable organic carbon represented nearly 75 % of TC in the thick A horizons, while non-oxidizable C, which includes recalcitrant C (e.g., charcoal), contributed to the remaining 25 % of TC. Carbon contents were not related to most of the inorganic soil variables studied, except for oxalate-extractable Al, which individually explained 69 % (P < 0.001) of the variability of TC in the umbric epipedon. Clay content was not suited as predictor of TC or of the other studied C forms. Bulk density, exchangeable Al3+, Al saturation, ECEC and other parameters obtained by selective extraction were not suitable as predictors of TC and other C forms. Interactions between organic matter and poorly crystalline minerals, as indicated by oxalate-extractable Al, appear to be one of the possible organic matter protection mechanisms of these soils.

The application of organic residues to the soil can increase soluble organic carbon (SOC) and affect the pH and electrolytic conductivity (EC) of the soil. However, the magnitude of these changes depends on the type of residue and the applied dose. This study aimed to evaluate the effect of increasing C rates contained in organic residue on the pH, EC, water-extractable total carbon (WETC), water-extractable organic carbon (WEOC), and water-extractable inorganic carbon (WEIC) in soil treated with manure (chicken, swine, and quail), sawdust, coffee husk, and sewage sludge. The levels of total C (TC- KH2PO4), organic carbon (OC- KH2PO4), and inorganic C (IC- KH2PO4) extractable by a 0.1 mol L-1 KH2PO4 solution were also quantified in soil under the effect of increasing rates of chicken and quail manures. The following rates of organic residue C were applied to a dystrophic Red Latosol (Oxisol) sample: 0, 2,000, 5,000, 10,000, and 20,000 mg kg-1. The addition of organic residues to the soil increased pH, except in the case of sewage sludge, which acidified the soil. The acidity correction potential of chicken and quail manure was highest, dependent on the manure rate applied; regardless of the dose used, sawdust barely alters the soil pH. At all tested rates, the EC of the soil treated with swine manure, coffee husk, and sawdust remained below 2.0 dS m-1, which is a critical level for salinity-sensitive crops. However, the application of chicken or quail manure and sewage sludge at certain rates increased the EC to values above this threshold level. Highest levels of WETC, WEOC, and WEIC were obtained when chicken and quail manure and coffee husk were applied to the Oxisol. The quantities of SOC extracted by KH2PO4 were higher than the quantities extracted by water, demonstrating the ability of soil to adsorb C into its colloids.

O objetivo deste trabalho foi avaliar a fertilidade de sêmen suíno pelo teste de ligação de espermatozóides a um substrato sintético. A motilidade (MOT) e o porcentual de espermatozóides ligados (PEL) foram avaliados após 5, 24, 48 e 72 horas de armazenamento a 17ºC. O PEL foi determinado em soluções contendo 6,25 ou 12,5 milhões de espermatozóides/mL, com ou sem albumina sérica bovina (BSA), preparadas a partir de dois a cinco ejaculados de cada um dos quatro machos. Cinqüenta e oito leitoas foram inseminadas, uma vez, 24 horas após o início do estro. Houve correlação positiva (P = 0,0001; r = 0,33) entre a MOT e o PEL. O PEL foi maior com 12,5 milhões de espermatozóides/mL e na presença de BSA (P<0,05). Após 72 horas, o macho 3 apresentou PEL inferior ao dos outros três (P<0,05). As taxas de clivagem (TC) e de embriões morfologicamente normais não diferiram entre indivíduos, mas o macho 3 apresentou menos de 70,0% de TC no quartil superior, enquanto os outros tiveram mais de 75,0%. Os machos diferem quanto à capacidade de ligação de seus espermatozóides ao substrato sintético, a partir de 24 horas de armazenamento do sêmen. A ligação dos espermatozóides ao substrato sintético é maior com a inclusão de BSA e com o aumento da concentração espermática.

The objective of this work was to evaluate the effects of alkaline solution marinades on the characteristics of pork subjected to post-mortem pH decrease in pig muscle. The pH of carcasses was measured in a commercial slaughterhouse (n = 526), 45 min after slaughtering (pH45) and, then, the carcasses were divided into the groups with pH45<5.7 or pH45>5.7. Ten samples of the longissimus dorsi muscles of each group were collected and distributed in an entirely randomized design, in a 2x4 factorial arrangement, with two conditions (pH45<5.7 or pH45>5.7), and four marinade solutions: TC, no marinade; TM1, sodium bicarbonate and sodium chloride; TM2, sodium tripolyphosphate and sodium chloride; TM3, sodium bicarbonate, sodium tripolyphosphate and sodium chloride. There was no interaction between pH45 of the meat and the marinade treatments. Meat with pH45<5.7 showed higher values for lightness, and for purge loss (PL), exudate loss (EL), cooking loss (CL) and shear force (SF). Marinating increased the pH, reduced the lightness, EL, CL and SF, and improved tenderness, juiciness and flavor of meat. Marinades with solutions containing chloride, bicarbonate, and sodium tripolyphosphate are effective in the improvement of pork quality, making physical characteristics of marinated meat similar to those of fresh pork, as a consequence of accelerated postmortem glycolysis.
