The objective of this work was to optimize in vitro plant regeneration via organogenesis from tissues of adult 'Hamlin', 'Pêra', and 'Valência' sweet orange plants. Explants were grown in EME culture medium with different concentrations of 6-benzylaminopurine (BAP) and naphthaleneacetic acid (NAA), at 27ºC in the absence of light for 50 days, followed by a 16-hour photoperiod for 20 days. Regeneration was assessed 50 and 70 days after in vitro culture. Organogenesis in cultivars Hamlin and Valência was promoted by EME supplemented with BAP, while NAA showed no apparent effect.

The objective of this work was to evaluate the effects of lignin, hemicellulose, and cellulose concentrations in the decomposition process of cover plant residues with potential use in no-tillage with corn, for crop-livestock integrated system, in the Cerrado region. The experiment was carried out at Embrapa Cerrados, in Planaltina, DF, Brazil in a split plot experimental design. The plots were represented by the plant species and the subplots by harvesting times, with three replicates. The cover plants Urochloa ruziziensis, Canavalia brasiliensis, Cajanus cajan, Pennisetum glaucum, Mucuna aterrima, Raphanus sativus, Sorghum bicolor were evaluated together with spontaneous plants in the fallow. Cover plants with lower lignin concentrations and, consequently, higher residue decomposition such as C. brasiliensis and U. ruziziensis promoted higher corn yield. High concentrations of lignin inhibit plant residue decomposition and this is favorable for the soil cover. Lower concentrations of lignin result in accelerated plant decomposition, more efficient nutrient cycling, and higher corn yield.

The objective of this work was to evaluate the effect of different drip irrigation regimes and planting densities on the incidence of the leaf miner, Leucoptera coffeella, in arabica coffee plants for one year. The experiment was carried out in 2008, in a complete randomized block design, in a split-plot in time arrangement, with four replicates. The treatments consisted of four drip irrigation regimes - soil water balance, irrigations at 20 and 60 kPa soil tensions, and a nonirrigated treatment -, which were distributed at three plant densities: 2, 500, 5, 000, and 10, 000 plants per hectare. The evaluations were made on a monthly basis between January and December 2008. The highest pest occurrence period was from August to November, a season with low-air relative humidity preceded by a drought period. Irrigated coffee plants showed an incidence of intact mines 2.2 times lower than that of nonirrigated plants. Irrigation and increasing of plant density contribute to the reduction of coffee leaf miner occurrence.

The objective of this work was to evaluate the natural attack by Acromyrmex crassispinus in initial Pinus taeda plantations without control measures against ants, as well as the effect of defoliation in seedlings of P. taeda. Evaluations of the attack of leaf-cutting ants on P. taeda plantations were done monthly in the first six months, then 9 and 12 months after planting. The percentages of plants that were naturally attacked by ants were registered. The effect of defoliation was evaluated by artificial defoliation, simulating the natural patterns of attack by A. crassispinus on P. taeda seedlings. The natural attack of A. crassispinus was greater during the first months after planting, being more intense in the first 30 days. Artificial defoliation indicated that there were no significant losses in diameter and height in plants with less than 75% defoliation. However, there were significant losses in diameter and height in plants with 100% defoliation, independently of the cut of the apical meristem, and also plant death. The control of leaf-cutting ants in P. taeda plantings, in which A. crassispinus is the most frequent leaf-cutting ant, should be intense only at the beginning of planting, since the most severe attacks occur during this time.

The objective of this work was to evaluate the effects of UV-B radiation on the vegetative growth and on the gas exchange characteristics of passion fruit plants (Passiflora edulis) grown in greenhouse. The average unweighted UV-B radiation near the apex of the plants was 8 W m-2 for the UV-B treatment (high UV-B), and 0.8 W m-2 for the control plants (low UV-B). Plants were irradiated with UV-B for 7 hours per day, centered on solar noon, during 16 days. High UV-B radiation resulted in lower shoot dry matter accumulation per plant. The content of UV-B absorbing compounds and anthocyanins was increased in the plants exposed to high UV-B radiation, when compared with the control. UV-B radiation did not affect stomatal conductance or transpiration rate, but reduced photosynthesis and instantaneous water-use efficiency, and increased intercellular CO2 concentration. The accumulation of UV-B-absorbing compounds and anthocyanins did not effectively shield plants from supplementary UV-B radiation, since the growth and photosynthetic processes were significantly reduced.

The objective of this work was to obtain transgenic tomato plants expressing the PfCP-2.9 protein (a chimera of the antigens MSP1 and AMA1 of Plasmodium falciparum). Cotyledons of seven-day-old tomatoes, cultivar Summers, were transformed via Agrobacterium tumefaciens. Transgenic expression in the T0 plants was verified in the DNA extracted from fruits. PCR analysis was used to test the presence of the gene of interest in the T1 generation. Reverse transcriptase PCR provided evidence of gene expression at the RNA level, and Western blot analysis confirmed the presence of the protein of interest in the T1 plants. This is the first report of successful transformation with the expression of a malaria antigen (PfCP-2.9) in transgenic tomato plants from the T0 and T1 generations.

The objective of this work was to determine the sink-source relationships and their effects on the number and growth of runner tips of 'Camino Real' strawberry stock plants. Three types of sources were evaluated: one defoliation at 96 days after planting (DAP), two defoliations at 50 and 96 DAP, and mother plants without defoliation. Four types of sink were accessed: runner tips collected weekly and monthly, four stolons with rooted runner tips in pots, and four freely-grown stolons. A completely randomized experimental design was used in a split-plot arrangement, with four replicates. The source types were placed in the plots, and sink types in the subplots. The number of runner tips, the crown diameter, and the dry matter mass were determined. Number and growth of tips were higher on plants without defoliation, and decreased 44.7% on twice-defoliated mother plants. The two-defoliation management did not reduce runner tip dry matter mass only on plants with rooted stolons, which produced runner tips 50% heavier. Defoliation of mother plants bearing rooting stolons can be used to reduce their growth, without reducing the emission and growth of runner tips.

Polpas de mangas foram submetidas a um processo de secagem solar após pré-tratamento osmótico, com o intuito de obter-se produtos de umidade intermediária. Foram testados quatro pré-tratamentos osmóticos, ou seja, imersão em xaropes de sacarose com 45º Brix, 55º Brix, 65º Brix e imersão seqüenciada em xaropes de 45; 55 e 65º Brix. O processamento envolveu os seguintes passos: desidratação osmótica nas soluções de sacarose adicionadas de substâncias conservantes e secagem solar. Após a secagem, os tratamentos com 45 e 65º Brix foram selecionados através de análise sensorial para avaliação da estabilidade durante 180 dias de armazenamento à temperatura ambiente (28º C). Durante todo o processamento e/ou após a obtenção dos produtos finais, três tipos de avaliações foram realizadas: determinações físico-químicas (atividade de água, pH, acidez total, vitamina C, sólidos solúveis, umidade, açúcar total e açúcares redutores), análises microbiológicas e testes sensoriais. De acordo com os resultados obtidos, os dois produtos apresentaram boa estabilidade no que se refere às propriedades microbiológicas e sensoriais durante o armazenamento.

The lack of good quality planting material has limited the expansion and contributed to yield reduction of the Brazilian pineapple culture. Alternatives of 'Pérola' pineapple slips management were studied aiming at obtaining superior planting material within a shorter time period and making good use of healthy slips of low vigor, that are commonly discarded by growers. Two experiments were carried out at the Experimental Field of Embrapa Cassava & Fruits, Cruz das Almas, Bahia, Brazil, and another one in a commercial plantation in the region of Itaberaba, BA, using blocks or entirely randomized designs with at least four replications. In the first one, the development of slips of different initial sizes (6 to 20 cm long), when grown on mother plants after fruit harvest, was compared with that of slips grown in a nursery after their removal from the mother plants. It became clear that larger slips grow more vigorously and that the removal from the mother plant delays their growth. However, results from the second study showed that those slips grown in nursery had vegetative and agronomic performance similar to that of conventional slips and close to that of plantlets produced from plant stem sections. Independently from the type of planting material used, the larger ones presented a more vigorous growth and produced higher yields. In the third study, it was observed the influence of mineral fertilization, pest control and growth regulator application after fruit harvest on slip development. The treatments applied did not significantly accelerate slip growth. Slips reached 50 cm length and at least 300 g fresh weight within 90 days after fruit harvest, indicating that vigorous plants have enough nutritional storage material for slips development.

The experiment was carried out at the Embrapa Semi-Árido, Petrolina-PE, Brazil, in order to study the physiological responses of umbu plants propagated by seeds and by stem cuttings under water stress conditions, based on leaf water potential and gas exchange measurements. Data were collected in one-year plants established in pots containing 30 kg of a sandy soil and submitted to twenty-day progressive soil water deficit. The evaluations were based on leaf water potential and gas exchange data collection using psychrometric chambers and a portable infra-red gas analyzer, respectively. Plants propagated by seeds maintained a significantly higher water potential, stomatal conductance, transpiration and photosynthesis under decreasing soil water availability. However, plants propagated by stem cuttings were unable to maintain a favorable internal water balance, reflecting negatively on stomatal conductance and leaf gas exchange. This fact is probably because umbu plants propagated by stem cuttings are not prone to formation of root tubers which are reservoirs for water and solutes. Thus, the establishing of umbu plants propagated by stem cuttings must be avoided in areas subjected to soil water deficit.

The effects of 0, 30 and 60 mM NaCl and substrates (red peat, sand or 3:1:1 [w/w] mixture of peat, sand, or soil) on vegetative growth of lulo, an Andean fruit species, during 12 weeks were studied. The experiment was carried out by using 2000 cm³ of polypropylene plastic pots under greenhouse conditions. Plant height, number of leaves and nodes, leaf area, total plant dry matter (DM), and shoot/root ratio were evaluated. With the increase of salt concentration, the plant height, the number of leaves and nodes, the leaf areas and plant dry mass DM decreased, whereas shoot/root ratio increased. Sand grown lulo plants were most affected by salinity and presented total mortality at 60 mM NaCl. On the other hand, plants held either in peat or in substrate mixture developed larger height, greater leaf and node numbers, higher leaf area and dry matter content. Shoot/root ratio in control (soil) and sand-grown plants (30 mM NaCl) was lower.

O objetivo deste trabalho foi avaliar os efeitos da cobertura de videira 'Cabernet Sauvignon' com lona plástica translúcida sobre a disponibilidade de luz e água, a concentração foliar de clorofila e a fotossíntese. As plantas com cinco anos de idade foram conduzidas em sistema 'Y' sobre porta-enxerto Paulsen 1103. O experimento seguiu o delineamento em blocos ao acaso, com dois tratamentos (plantas sem e com cobertura plástica) e quatro repetições de 15 plantas (unidade experimental). A cobertura plástica reduziu o suprimento às plantas de radiações ultravioleta (UV), azul, verde, vermelho, vermelho distante e total (300-750 nm), bem como a disponibilidade hídrica nas camadas superficiais do solo (0-30 cm). As plantas cobertas apresentaram maior taxa fotossintética máxima e condutância estomática em relação às plantas descobertas. Os pontos de compensação e de saturação de luz, a eficiência quântica aparente, a respiração no escuro, a concentração foliar de clorofilas e o potencial hídrico foliar de base não foram influenciados pelo uso da cobertura plástica. A cobertura plástica reduziu a radiação e a disponibilidade hídrica nas camadas superficiais do solo, porém favoreceu a assimilação foliar de CO2.

A energia solar é a fonte primária para a fotossíntese e a transpiração vegetal para que uma cultura expresse seu potencial produtivo em um dado local. O método proposto neste estudo pretende facilitar o cálculo do volume de água (litros/planta/dia) necessário para uma irrigação localizada com o mínimo desperdício possível em pomares cítricos e de macieiras, utilizando-se de dados usualmente disponíveis, tais como área foliar, densidade de fluxo de radiação solar global, saldo de radiação e déficit de saturação de vapor médio diário do ar. Considerando-se que a irrigação localizada consome bem menos água do que o sistema de aspersão, e que a outorga de água para irrigação está cada vez mais limitada, tal estudo vem a ser certamente de grande importância para assegurar a autossustentabilidade da agricultura irrigada, especialmente em regiões áridas e semiáridas. Foram utilizados neste trabalho, para desenvolvimento da metodologia proposta, dados de fluxo de seiva medidos através do método de fluxo de calor, em pomar de lima-ácida-Tahiti com área foliar de 48 e 99 m², bem como em pomar de macieiras com área foliar aproximada de 5; 8; 9; 11; 16 e 21 m². Os resultados obtidos indicaram que a metodologia proposta, baseada na habilidade das plantas em converter energia solar fixada em água transpirada, mostrou-se viável para avaliar a lâmina de irrigação de plantas cítricas e macieiras nas localidades estudadas.

O trabalho teve por objetivo avaliar padrões de interceptação da radiação fotossinteticamente ativa (RFA) em vinhedos de 'Moscato Giallo', com e sem cobertura plástica sobre o dossel vegetativo. O experimento foi conduzido no município de Flores da Cunha, na "Serra Gaúcha" (29°06'S, 51°20'W, 541 m), na safra de 2005/2006. Consistiu de fileiras de plantas descobertas e cobertas com filme de polietileno de baixa densidade transparente com 160µm de espessura. Foram feitas medições contínuas de RFA em diferentes alturas do dossel, nos dois sistemas. O índice de área foliar (IAF) foi determinado por método não destrutivo, tendo-se ajustado modelos relacionando IAF com graus-dia acumulados. Também foram ajustados modelos relacionando a eficiência de interceptação de RFA com IAF, nos vinhedos cobertos e descobertos. A cobertura plástica promoveu aumento de 12% na eficiência de interceptação da radiação pelo vinhedo. O vinhedo coberto apresentou menor coeficiente de extinção de RFA que em céu aberto.

This work aimed to evaluate the influence of naphthaleneacetic acid (NAA) and gibberellic acid (GA3) plant regulators in in vitro etiolation and subsequent regeneration of the PE x SC-60 pineapple hybrid. Nodal segments of in vitro plants with approximately 5-7 cm height were incubated in basic MS culture medium supplemented with 0.0; 0.5 and 1.0 mg L-1 of naphthaleneacetic acid (NAA) in combination with gibberellic acid (GA3) in concentrations of 0.0; 0.5 and 1.0 mg L-1, and maintained at 27 ºC under dark condition. Evaluations were carried out at 90 and 180 days after incubation period. The best results for length of etiolated stems were obtained with 1.0 mg L-1 of NAA. In the experiment followed by the regeneration, stems with 3 cm from the etiolation treatment, were cultivated in proliferation medium and the number of regenerated plants per treatment was evaluated at 60 days of cultivation. The treatment that promoted the best etiolation of plants also promoted the worst regeneration rates, demonstrating the residual effect of the auxin used in the previous step in the regeneration of plants of the pineapple hybrid evaluated.
