The ocelot (Leopardus pardalis) is included in list of wild felid species protected by CITES and is part of conservation strategies that necessarily involve the use of assisted reproduction techniques, which requires practical and minimally invasive techniques of high reproducibility that permit the study of animal reproductive physiology. The objective of this study was to compare and validate two commercial assays: ImmuChem Double Antibody Corticosterone 125I RIA from ICN Biomedicals, Costa Mesa, CA, USA; and Coat-a-Count Cortisol 125I RIA from DPC, Los Angeles, CA, USA, for assessment of fecal glucocorticoid metabolites in ocelots submitted to ACTH (adrenocorticotropic hormone) challenge. Fecal samples were collected from five ocelots kept at the Brazilian Center of Neotropical Felines, Associação Mata Ciliar, São Paulo, Brazil, and one of the animals was chosen as a negative control. The experiment was conducted over a period of 9 days. On day 0, a total dose of 100 IU ACTH was administered intramuscularly. Immediately after collection the samples were stored at 20C in labeled plastic bags. The hormone metabolites were subsequently extracted and assayed using the two commercial kits. Previously it was performed a trial with the DPC kit to check the best extraction method for hormones metabolites. Data were analyzed with the SAS program for Windows V8 and reported as means ± SEM. The Schwarzenberger extraction method was slightly better when compared with the Wasser extraction method (103,334.56 ± 19,010.37ng/g of wet feces and 59,223.61 ± 12,725.36ng/g of wet feces respectively; P=0,0657). The ICN kit detected an increase in glucocorticoid metabolite concentrations in a more reliable manner. Metabolite concentrations (ng/g wet feces) on day 0 and day 1 were 66,956.28 ± 36,786.93 and 92,991.19 ± 28,555.63 for the DPC kit, and 205,483.32 ± 83,811.32 and 814,578.75 ± 292,150.47 for the ICN kit, respectively. The limit of detection for the ICN kit was 7.7 ng/mL for 100% B/Bo (25ng/mL for 88%B/Bo) and for the DPC kit it was 0.2ug/dL for 90.95% B/Bo (1ug/dL for 81.27% B/Bo). In conclusion it was confirmed that the Schwarzenberger extraction method and the ICN kit are superior for extracting and measuring fecal glucocorticoid metabolites in ocelot fecal samples.

Toxoplasmosis is caused by the obligate intracellular protozoan parasite Toxoplasma gondii and affects warm-blooded vertebrates, including pets and man. Dogs are epidemio-logically important since they act as sentinels for the infection in humans. The present study aimed to determine the presence of antibodies to T. gondii in 205 serum samples from dogs in Ubatuba, Sao Paulo state, Brazil, through indirect immunofluorescence reaction (IFAT), as well as the risk factors related to toxoplasmosis in the animals such as breed, age, sex, access to outdoors, homemade food ingestion, access to untreated water, and contact with rodents. Toxoplasmosis-positive samples accounted for 52/205 (25.4%), with titers ranging from 16 to 256. The serological results presented significant association (P<0.05) with homemade food ingestion (45/118; 38.1%; CI95% 29.9%-47.2%) (OR=7.0; CI95% 3.0-16.6), and with access to outdoors where those that do not have access to the street were prevalent (37/121; 30.6%; CI95% 23.1%-39.3%) (OR=0.5; CI95% 0.2-1.0). These results show that toxoplasmosis in this region is related to problems of sanitary education, mainly concerning the appropriate cooking of foods, since most positive animals did not show significant association with the presence of rodents or untreated water consumption but showed, instead association with ingestion of homemade food. Thus, toxoplasmosis is a public health problem in the studied region, and sanitary measures are needed to control the infection due to the strict relationship between man and dog and the presented risk factors

Senecio spp. poisoning is the main cause of cattle mortality in the central region of Rio Grande do Sul. This paper reports an outbreak of seneciosis in cattle with high prevalence of photosensitization, where 83 out of 162 cows (51.3%) presented this clinical sign. The outbreak occurred in September 2013, affecting adult cows that were held in a 205 hectare-pasture from April to October 2013 with abundant Senecio brasiliensis infestation. Main clinical signs were weight loss, excessive lacrimation or mucopurulent ocular discharge, nasal serous discharge, ventral diphteric glossitis, crusts in the nose, teats, dorsum of ears, and vulva. Liver biopsy was performed in all the cows under risk; the histopathological findings in the liver biopsies consisted of fibrosis, megalocytosis, and biliary ductal proliferation and were present in 73.4% of the biopsied animals. Six cows had increased serum activity of gamma glutamyl transferase. Three affected cows were necropsied. The main necropsy findings were a hard liver, distended gall bladder, edema of the mesentery and abomasum. Liver histological changes in the necropsied cows were similar to those of the biopsied livers. Spongiosis was detected in the brain of necropsied cows and is characteristic of hepatic encephalopathy.

As perdas de produtividade das culturas em decorrência da competição de plantas concorrentes geralmente aumentam quanto mais semelhantes forem suas características morfofisiológicas. O objetivo deste trabalho foi avaliar os efeitos da competição de cultivares de soja, simulando espécies daninhas dicotiledôneas, sobre cultivares de soja portadores de características diferenciais de planta. Para isso, conduziu-se um experimento em campo, em Eldorado do Sul-RS. Os fatores e tratamentos comparados foram: três condições de competição (ausência de plantas de soja concorrentes e presença de plantas concorrentes do cultivar de soja BRS 205 ou Cobb); e quatro cultivares reagentes de soja à competição (IAS 5, BR-16, CD 205 e Fepagro RS-10). Avaliaram-se variáveis relacionadas à produtividade dos cultivares e outras correlatas. Características dissimilares em plantas de cultivares de soja, como estatura e ciclo, resultaram em habilidade competitiva diferenciada com plantas concorrentes. O cultivar de soja CD 205, de estatura elevada e ciclo tardio, mostrou alta habilidade competitiva; IAS 5, de baixa estatura e ciclo precoce, apresentou baixa tolerância à competição, enquanto BR-16 e Fepagro RS-10 são intermediários. Cultivares com elevada habilidade competitiva, além de tolerarem a competição, preservando o potencial de produtividade, também suprimem a produção de grãos das plantas concorrentes.

Características morfofisiológicas diferenciais de plantas podem influenciar as relações de competição entre cultura e plantas daninhas. O objetivo do presente trabalho foi avaliar a influência das características de plantas de quatro cultivares de soja sob competição ou não com outros dois cultivares de soja, simulando espécies daninhas dicotiledôneas, durante a fase vegetativa de crescimento. Para isso, conduziram-se dois experimentos: um em vasos, visando fornecer subsídios quanto ao crescimento inicial das plantas de soja; e outro em campo, a fim de acompanhar características de crescimento das plantas. Em campo, foram comparados os seguintes fatores e tratamentos: três condições de competição (ausência de plantas concorrentes; presença de plantas dos cultivares de soja BRS 205 ou Cobb); e quatro cultivares reagentes de soja à competição (IAS 5, BR-16, CD 205 e Fepagro RS-10). Cultivares de soja apresentaram variações na velocidade de emergência e de crescimento inicial de plantas. A presença de plantas concorrentes, independentemente das características próprias, afetou a ramificação da soja. Os cultivares Fepagro RS-10 e BR-16 apresentaram rápido crescimento durante a fase inicial de desenvolvimento; CD 205, ao contrário, mostrou lento crescimento nesta fase. Cultivares de soja de ciclo precoce cobriram com mais rapidez o solo durante os primeiros 45 dias do que os tardios.

One of the very important components in the organic maize production costs refers to spending on weed control. In this research were assessed the effects of maize hybrids (AG 1051 and BR 205) in an intercropping with Gliricidia (Gliricidia sepium) and mechanical hoeing on weed control. The treatments assessed were: A - maize monocropping + two hoeings (20 and 40 days after sowing); B - maize with one hoeing at 20 days + intercropping with gliricidia sowed after hoeing; C - maize sowing intercropped with gliricidia at the time of maize sowing + hoeing at 40 days; D - maize sowing intercropped with gliricidia at the time of maize sowing without hoeing; E - monocropping maize without hoeing. In the intercroppings, gliricidia was sowed in broadcast seeding with 30 viable seeds m-2. Maize hybrids did not differ in their effects on weed growth and grain yield. Treatments A, B and C have reduced weed growth, compared to treatments D and E. The highest grain yield was obtained with treatment A and the lowest with treatment E. It was concluded that intercropping maize and gliricidia is not a good alternative for an integrated weed management in maize crops in the conditions assessed.

ABSTRACT The combination of crop residues or crop extracts is often more advantageous in controlling weeds, than the application of each residue or extract singly. This suggests that in intercropping with maize, the combination of tree species can be more advantageous than species isolated in weed control. The objective of this study was to evaluate the effects of intercropping with a combination of leguminous on the weed growth and corn yield. A randomized-block design with split plots (cultivars in plots) and five replicates was established. The cultivars BR 205 and AG 1041 were subject to the following treatments: two weedings (A), intercropping with sabiá (B), gliricidia (C), gliricidia + sabiá (D) and no weeding (E). In the B and C, 30 viable seeds m-2 of the leguminous were sown. In the D, 15 seeds of each species were sown m-2. The legumes were sown by random casting during corn planting. The sequence of the best treatments in reducing the growth of weeds is A > B = C = D = E. The sequence of the best treatments when are considered the yields of baby corn, green corn and grain is A > B > C > D > E. The cultivars do not differ in regards to the reduction in weed growth. In terms of corn yield cultivar BR 205 is the best.

O Município de Nova Lima, na Região Metropolitana de Belo Horizonte (MG), encontra-se no contato entre os domínios do Cerrado e da Mata Atlântica, que são dois hotspots de biodiversidade. Uma amostragem florística foi realizada em uma floresta semidecídua (20°00' S e 43°55' W) com o objetivo de avaliar sua relação com outras florestas semidecíduas e ombrófilas presentes em ambos os domínios a avaliar sua afinidade fitogeográfica. A região estudada encontra-se sob alta pressão antrópica devido à expansão urbana. A amostragem florística foi baseada na coleta de todos os indivíduos lenhosos presentes fora e dentro de 44 parcelas retangulares de 20 × 5 metros. A floresta estudada foi comparada com outras presentes no Distrito Federal e dos Estados de Minas Gerais, São Paulo, Espírito Santo, Rio de Janeiro e Goiás usando análises multivariadas. Foram encontradas 205 espécies representando 49 famílias botânicas. O registro de espécies com distribuição restritas a Mata Atlântica foi 15 vezes maior que do Cerrado. A flora arbórea da floresta estudada é, em boa medida, um subconjunto da flora das florestas ombrófilas, tendo espécies provavelmente mais eficientes em resistir e competir sob condições de seca mais prolongada. Assim, a floresta semidecídua estudada pode ser considerada como floresta atlântica sensu lato.

Five bovine milk protein polymorphisms were studied in Zebuine cattle raised in Brazil, through horizontal electrophoresis on starch gel containing urea and 2-mercaptoethanol, using basic and acidic buffer systems. Allelic frequencies for a-La, b-Lg, aS1-Cn, b-Cn and k-Cn loci were estimated in six Gyr herds (N = 283), six Guzerat herds (N = 205), one Nelore herd (N = 17) and one Sindi herd (N = 22), all from São Paulo or Minas Gerais State, Brazil. Genotypic frequencies observed for each locus and breed studied are in accordance with the assumption of genetic equilibrium, demonstrating absence of high inbreeding levels for the breeds tested. The FST value found indicated significant genetic differentiation among breeds; however, the Gyr and Guzerat herds showed significantly different gene frequencies. Genetic distance estimates among zebuine breeds studied and the Holstein breed, taken as a reference for a taurine breed, showed strong differences between these two racial groups

The use of hydroxyurea (HU) can improve the clinical course of sickle cell disease. However, several features of HU treatment remain unclear, including the predictability of drug response and determination of adequate doses, considering positive responses and minimal side effects. In order to identify adequate doses of HU for treatment of sickle cell disease, 10 patients, 8 with sickle cell anemia and 2 with Sß thalassemia (8SS, 2Sß), were studied for a period of 6 to 19 months in an open label dose escalation trial (10 to 20 mg kg-1 day-1). Hemoglobin (Hb), fetal hemoglobin (Hb F) and mean corpuscular volume (MCV) values and reticulocyte, neutrophil and platelet counts were performed every two weeks during the increase of the HU dose and every 4 weeks when the maximum HU dose was established. Reduction in the number of vasoocclusive episodes was also considered in order to evaluate the efficiency of the treatment. The final Hb and Hb F concentrations, and MCV values were significantly higher than the initial values, while the final reticulocyte and neutrophil counts were significantly lower. There was an improvement in the concentration of Hb (range: 0.7-2.0 g/dl) at 15 mg HU kg-1 day-1, but this concentration did not increase significantly when the HU dose was raised to 20 mg kg-1 day-1. The concentration of Hb F increased significantly (range: 1.0-18.1%) when 15 mg HU was used, and continued to increase when the dose was raised to 20 mg kg-1 day-1. The final MCV values increased 11-28 fl (femtoliters). However, reticulocyte (range: 51-205 x 109/l) and neutrophil counts (range: 9.5-1.3 x 109/l) obtained at this dose were significantly lower than those obtained with 15 mg kg-1 day-1. All patients reported a decrease in frequency or severity of vasoocclusive episodes. These results suggest that a hydroxyurea dose of 15 mg kg-1 day-1 seems to be adequate for treatment of sickle cell disease in view of the minimal side effects observed and the improvement in laboratory and clinical parameters.

The anthropometric status and metabolic control of 51 recently diagnosed Brazilian schoolchildren with type 1 diabetes (DM1), during the first 5 years of the disease, were compared with those of normal children (60 girls and 132 boys) belonging to the same environmental condition and pubertal stage. Metabolic control was evaluated on the basis of fasting plasma glucose (FPG) and HbA1c levels. The criteria of the National Center for Health Statistics were used for anthropometric evaluation. FPG (205 ± 51 mg/dl for girls vs 200 ± 34 mg/dl for boys) and % above upper normal limit of median HbA1c (1.8% for girls vs 2.5% for boys with diabetes) were not significantly different during follow-up. The Z-score of the last height evaluation was lower in the girls' group (-0.14 vs -0.53, P<0.05). By forward stepwise analysis, the Z-score of the initial height was statistically significant as a determinant factor for height at the end of the study in both girls and boys with DM1. The Z-score of weight at last evaluation was not different from that at diagnosis in either sex. However, analysis according to pubertal stage showed a tendency to a weight increase in the girls. The weight recovery and height loss in girls with DM1 follows the trend of the normal Brazilian population.

Osteoporosis is a common manifestation of Cushing's syndrome, but the mechanisms responsible for this abnormality have not been defined. With the objective of analyzing parathyroid hormone (PTH) secretion in chronic hypercortisolism (CH), we evaluated 11 healthy subjects and 8 patients with CH, 6 with Cushing's disease and 2 with adrenal adenoma. These volunteers were submitted to tests of PTH stimulation through hypocalcemia (EDTA), PTH suppression through hypercalcemia (iv and oral calcium), and evaluation of bone mineral density (BMD) by DEXA. During the test of PTH stimulation, the calcium and magnesium concentrations of the normal and CH groups were similar. Patients with CH showed an increased PTH response to the hypocalcemic stimulus compared to controls. PTH values were significantly higher in the CH group at 70 (17.5 ± 3.5 vs 10.2 ± 1.3 pmol/l, P = 0.04), and 120 min (26.1 ± 5.9 vs 11.3 ± 1.9 pmol/l, P = 0.008) of EDTA infusion. The area under the curve for PTH during EDTA infusion was also significantly higher in patients with CH than in normal subjects (1867 ± 453 and 805 ± 148 pmol l-1 2 h-1, P = 0.02). During the test of PTH suppression, calcium, magnesium and PTH levels of the patients with hypercortisolism and controls were similar. BMD was decreased in patients with hypercortisolism in the spine (0.977 ± 0.052 vs 1.205 ± 0.038 g/cm² in controls, P<0.01). In conclusion, our results show that subjects with CH present decreased bone mass mainly in trabecular bone. The use of dynamic tests permitted the detection of increased PTH secretion in response to a hypocalcemic stimulus in CH patients that may probably be involved in the occurrence of osteoporosis in this state.

The influence of drug concentrations on the development of persistent posttransplant hyperlipidemia was investigated in 82 patients who received cyclosporin A (CsA) and prednisone plus sirolimus (SRL) (52) or azathioprine (AZA) (30) during the first year after transplantation. Blood levels of CsA and SRL, daily doses of AZA and prednisone, and cholesterol, triglyceride, and glucose concentrations were determined during each visit (pretransplant and 30, 60, 90, 120, 180, and 360 days posttransplant). Persistent hyperlipidemia was defined as one-year average steady-state cholesterol (CavCHOL) or triglyceride (CavTG) concentrations above 240 and 200 mg/dL, respectively. Mean cholesterol and triglyceride concentrations increased after transplantation (P < 0.01) and were higher in patients receiving SRL compared to AZA (P < 0.001). Patients receiving SRL showed a significantly higher number of cholesterol (>229 or >274 mg/dL) and triglyceride (>198 or >282 mg/dL) determinations in the upper interquartile ranges. CsA and SRL interquartile ranges correlated with cholesterol concentrations (P = 0.001) whereas only SRL interquartile ranges correlated with triglyceride concentrations (P < 0.0001). Only pretransplant cholesterol concentration >205 mg/dL was independently associated with development of persistent hypercholesterolemia (CavCHOL >240 mg/dL, relative risk (RR) = 20, CI 3.8-104.6, P = 0.0004) whereas pretransplant triglyceride concentration >150 mg/dL (RR = 7.2, CI 1.6-32.4, P = 0.01) or >211 mg/dL (RR = 19.8, CI 3.6-107.9, P = 0.0006) and use of SRL (RR = 3, CI 1.0-8.8, P = 0.0049) were independently associated with development of persistent hypertriglyceridemia (CavTG >200 mg/dL). Persistent hypercholesterolemia was more frequent among patients with higher pretransplant cholesterol concentrations and was dependent on both CsA and SRL concentrations. Persistent hypertriglyceridemia was more frequent among patients with higher pretransplant triglyceride concentrations and was dependent on SRL concentrations.

Serum hormone levels were compared between captive and free-living maned wolves and seasonal variations of sex hormones were studied. Blood samples were collected from 16 male and 26 female adult animals from Brazilian zoos, and from 30 male and 24 female free-living adults to determine serum progesterone and testosterone by radioimmunoassay. Serum testosterone concentrations varied (P < 0.05) across seasons for 16 captive males, being higher in autumn (2184.7 ± 355.1 pg/mL) than in summer (1080.7 ± 205.4 pg/mL), winter (1270.1 ± 276.6 pg/mL) and spring (963.9 ± 248.1 pg/mL), although they did not differ between summer, winter and spring. Testosterone concentration of 30 free-living males differed (P < 0.05) between autumn (824.1 ± 512.2 pg/mL), winter (14.4 ± 8.0 pg/mL) and spring (151.9 ± 90.5 pg/mL). Comparison between captive and free-living animals showed no difference in autumn (P > 0.05). Sixteen captive males showed higher testosterone concentration during winter and spring compared with 30 free-living animals (P < 0.05). Progesterone concentration varied among seasons in 26 captive females (P < 0.05), being higher in autumn (15.3 ± 3.1 ng/mL) than in summer (6.6 ± 1.5 ng/mL), winter (5.3 ± 3.1 ng/mL) and spring (4.3 ± 0.7 ng/mL). Progesterone concentration of 24 free-living females varied between autumn (17.1 ± 6.0 ng/mL) and winter (1.7 ± 0.3 ng/mL) (P < 0.05), but we could not obtain data for spring or summer. No difference in progesterone levels was observed between captive and free-living females in autumn and winter.

Foi estudado o efeito da temperatura do ar (205-215°C) e do tempo de cozimento (3,5 - 4,1 minutos), sobre a temperatura interna e o rendimento durante o cozimento do filé de frango empanado usando a metodologia de superfície de resposta, obtendo-se modelos estatísticos adequados. Verificou-se que as melhores condições de processo foram o emprego de temperaturas entre 206 e 209°C e tempos entre 2,5 e 3,6 minutos.
