Random amplified polymorphic DNA markers (RAPD) were used to estimate the variability of 14 genotypes of Brazilian wheat (Triticum aestivum L.), using a set of 50 random 10mer primers. A total of 256 reproducibly scorable DNA amplification products were obtained from 48 of the primers, 83% of which were polymorphic. Genetic distances among genotypes were calculated and a dendrogram and a principal coordinates analysis showing the genetic relationships among them were obtained. Despite the low variability found (average genetic distance of 27%), two groups of genotypes could be identified, which probably reflect how they were formed. Studies such as this one may be important in the planning and development of future improvement programs for this plant species.

Seed from the sensitive wheat (Triticum aestivumL.) cultivar Anahuac was treated to gamma-ray irradiation and eleven Al3+ tolerant mutants selected. The objective was to compare these mutants to the original Anahuac and to the tolerant wheat cultivars IAC-24 and IAC-60 from 1994 to 1996 in acid (Capão Bonito) and limed (Monte Alegre do Sul) soil field trials, in the State of São Paulo, Brazil. Grain yield and agronomic characteristics were analyzed. All the mutant lines yielded higher than the sensitive Anahuac cultivar in the acid soils of Capão Bonito. Under limed soil conditions, 10 mutants had a similar yield to the original sensitive cultivar and one a lower yield. The majority of the mutants were similar in yield to the tolerant cultivars IAC-24 and IAC-60 under both conditions. Some of the mutants showed altered agronomic characteristics, but these alterations did not generally influence the grain yield. The results indicated that tolerant lines with good characteristics may be obtained from a susceptible cultivar by mutation induction, thus allowing cropping under conditions where Al3 + is a limiting factor.

The use of winter legumes in southern Brazil is hindered by the slow growth of these species during establishment exposing soil surface to erosion. Introduction of these species along with spring wheat (Triticum aestivum L.) was studied as a means of increasing ground cover during their initial establishment period, without reducing wheat grain yield. Two experiments were conducted in nearby areas, one in each year. Birdsfoot trefoil (Lotus corniculatus L.), red clover (Trifolium pratense L.) cultivar Quiñequelli, white clover (T. repens L.), and arrowleaf clover (T. vesiculosum Savi) did not reduce cereal yield in either year. Wheat yield was reduced by intercropped red clover cultivar Kenland and by subclover (T. subterraneum L.) in the first year. No grain yield differences due to intercropping with any legume were detected in the second year, when rainfall was below normal. Intercropping with wheat showed to be a practical alternative to enhance ground cover at establishing forage legumes.

El objetivo del trabajo fue estudiar en la generación segregante del híbrido entre una cultivar de Lycopersicon esculentum homocigota para el gen nor y la accesión LA1385 de L. esculentum var. cerasiforme, la recombinación de caracteres productivos y de calidad de fruto a través de las metodologías de análisis multivariado. En las generaciones F1 y F2 y en los progenitores se evaluaron caracteres vegetativos y productivos (longitud de entrenudos, perímetro del tallo en las partes basal, media y apical, número de flores por inflorescencia, número de inflorescencias por planta y días a cosecha) y de calidad de fruto (peso, forma, sólidos solubles, acidez, firmeza, color y vida poscosecha). Se utilizaron las correlaciones canónicas entre los caracteres vegetativos y productivos y los de calidad de fruto y también un análisis de agrupamiento para las características del fruto, incluyendo los progenitores, la F1 y la F2. Estos análisis permitieron demostrar que las características productivas y de calidad de fruto recombinaron en la generación segregante. La vida poscosecha fue la característica de los frutos más importante para discriminar grupos en la F2. Para tres niveles de agrupamiento cada grupo de individuos F2 se comportó como alguno de los progenitores o la F1.

O objetivo deste trabalho foi avaliar a associação dos polimorfismos dos genes bGH, IGF-1 e PIT-1 com características de peso e ganho de peso numa população F2 de bovinos (Gir x Holandês), pela técnica de PCR-RFLP. As freqüências alélicas A e B, do gene PIT-1, e dos genótipos AA, AB e BB, nas populações parentais foram semelhantes entre si, mas diferentes das freqüências nas populações cruzadas F1 e F2, para esse gene. Quanto ao gene bGH, os animais da raça Holandesa apresentaram freqüência de 100% para o alelo E e os animais da raça Gir, 92% para o alelo F, resultando em alta freqüência de indivíduos heterozigotos nas populações F1 e F2. Quanto ao gene IGF-1, todos os animais da raça Holandesa eram heterozigotos (AB) e, nos animais Gir, a maioria dos indivíduos foi de homozigotos (AA), o que resultou em alta freqüência do alelo A nas populações F1 e F2. Foram encontradas associações significativas do alelo A do gene PIT-1 com as características de peso aos 60, 205, 365 dias e ganho de peso do nascimento aos 60 dias. Em bGH, observou-se efeito significativo do alelo E para peso aos 365 dias e ganho de peso do nascimento aos 60 dias, enquanto o efeito do alelo A do IGF-1 foi significativo somente para peso ao nascimento. Os alelos identificados podem ser usados como marcadores no melhoramento animal.

The objective of this study was to assess the impact of genetic breeding on grain yield, and to identify the physiological traits associated to the increment in yield and their related growth processes, for wheat cultivars grown in Southern Brazil, in the past five decades. Seven wheat cultivars released between 1940 and 1992, were compared for physiological aspects associated with grain yield. Grain yield, biological yield, biomass partitioning, harvest index and grain yield components were also determined. The number of grains per square meter was more affected by plant breeding and was better correlated with grain yield (r = 0.94, p<0.01) than with grain weight (r = -0.39ns). The higher number of grains per square meter was better correlated with the number of grains per spike in the modern cultivars than in the older ones. The genetic gain in grain yield was 44.9 kg ha-1 per year, reflecting important efforts of the breeding programs carried out in Southern Brazil. Grain yield changes, during the period of study, were better associated with biomass production (r = 0.78, p<0.01) than with harvest index (r = 0.65, p<0.01).

The objective of this work was to determine the genetic variability available for triticale (X Triticosecale Wittmack) crop improvement in Brazil. Forty-two wheat genomic microsatellites were used to estimate the molecular diversity of 54 genotypes, which constitute the base of one of the major triticale breeding programs in the country. Average heterozygosity was 0.06 and average and effective number of alleles per locus were 2.13 and 1.61, respectively, with average allelic frequency of 0.34. The set of genomic wheat microsatellites used clustered the genotypes into seven groups, even when the germplasm was originated primarily from only two triticale breeding programs, a fact reflected on the average polymorphic information content value estimated for the germplasm (0.36). The 71.42% transferability achieved for the tested microsatellites indicates the possibility of exploiting these transferable markers in further triticale genetic and breeding studies, even those mapped on the D genome of wheat, when analyzing hexaploid triticales.

O objetivo deste trabalho foi validar marcadores moleculares previamente associados a genes que conferem resistência à ferrugem-da-folha, em genótipos brasileiros de trigo. Cinco marcadores STS e SCAR, identificados como associados aos alelos de resistência dos genes Lr1, Lr9, Lr10 e Lr24, foram avaliados por PCR, em 25 genótipos de trigo com conhecida presença ou ausência desses alelos. O marcador STS, associado ao alelo de resistência do gene Lr1, não foi eficiente em identificar genótipos brasileiros que possuem este alelo de resistência. Os marcadores STS e SCAR, associados a Lr9, Lr10 e Lr24, foram eficientes na identificação de plantas que possuem o alelo de resistência desses genes, e podem ser utilizados na seleção por marcadores da resistência à ferrugem-da-folha, em genótipos brasileiros de trigo.

O objetivo deste estudo foi desenvolver testes de alelismo, inclusive cruzamentos entre um grupo de genótipos resistentes à ferrugem, do banco de germoplasma de soja da Embrapa, e as duas fontes de resistência com os genes Rpp2 e Rpp4. A ferrugem-asiática-da-soja, causada pelo fungo Phakopsora pachyrhizi, tem gerado perdas significativas na produtividade da cultura e preocupado os agricultores brasileiros. Até recentemente, existiam quatro genes de resistência descritos na literatura, mas somente Rpp2 e Rpp4 continuam efetivos no Brasil. Vinte e seis fontes com resistência a P. pachyrhizi foram cruzadas com PI 230970 e PI 459025 (fontes dos genes Rpp2 e Rpp4, respectivamente), e as plantas da geração F2 foram infectadas com uma suspensão com 2,5x10(4) esporos por mililitro e analisadas em casa de vegetação após 20 dias, para verificação da presença de lesões de resistência (RB) ou de suscetibilidade (TAN). Foram aplicados testes de qui-quadrado para investigar as hipóteses de segregação independente ou de alelos de resistência nos locos Rpp2 e Rpp4. Genes de resistência provenientes de PI 197182, PI 230971 e PI 417125 não segregaram em cruzamentos com a PI 230970, o que indica que esses genótipos apresentam um único gene de resistência presente no loco Rpp2. Cruzamentos com os outros 23 genótipos resultaram em populações segregantes, o que indica que estas possuem genes que não pertencem aos locos Rpp2 e Rpp4.
Morphological, pedigree, and molecular distances and their association with hybrid wheat performance

The objectives of this work were to estimate the genetic distance among wheat genotypes using morphological, pedigree, molecular, and combined morphological and molecular measures, to determine the correlations between these measures, and to evaluate the combining ability of the genotypes. Three generations and two planting designs were studied. Six wheat genotypes were crossed using a diallel design. The F1, F2 and F3generations were evaluated in the field, in the crop seasons of 2003, 2004 and 2005, under spaced plant and full-row planting designs. The estimated general and specific combining abilities of tested hybrids were influenced both by the generation and the planting design. The correlation coefficients among the distance measures and between these measures and genotype performances of different generations for the two planting designs were low to moderate. In order to obtain a more precise estimate of the genetic distance among cultivars and its association with the hybrid performance, more than one generation, planting design, and genetic distance estimation technique should be employed.

O objetivo deste trabalho foi avaliar a expressão, por RT-qPCR, dos genes de nodulação nodC e nodW e do gene nopP da estirpe CPAC 15, que provavelmente atuam na infecção das raízes da soja. Foram realizados dois experimentos. No primeiro, a expressão dos genes foi avaliada nas células após a incubação com genisteína por 15 min, 1, 4 e 8 horas. Os resultados revelaram que os três genes apresentaram maior expressão imediatamente após o contato com o indutor (15 min). No segundo experimento, a bactéria foi cultivada na presença de indutores (genisteína ou exsudatos de sementes de soja) por 48 horas. A expressão dos três genes foi maior na presença de genisteína, com valores de expressão para nodC, nodW e nopP superiores ao controle. Os resultados obtidos confirmam a funcionalidade dos três genes na estirpe CPAC 15, com ênfase para o nopP, cuja funcionalidade em Bradyrhizobium japonicum foi descrita pela primeira vez.

The objective of this work was to evaluate the effect of moisture and temperature on the development of Sclerotium rolfsii on soybean, corn, and wheat straw. Wheat straw produced the lowest number of sclerotia. Intermediate soil moisture level (70% of field capacity), and temperatures ranging between 25-30ºC favored sclerotia development. No sclerotia were formed at temperatures between 30-35ºC, on any type of straw.

The objective of this study was to identify gliadin band patterns and the extent of genetic diversity in durum wheat genotypes from Northwestern Iran and the Republic of Azerbaijan. Gliadins from 46 landraces and four cultivars were evaluated through acid PAGE analyses. Sixty-six polymorphic bands and 81 patterns were identified. Twenty-four different motility bands and 22 patterns were found in the ω gliadin region with 14 polymorph bands and 20 patterns for α and γ gliadins, and 14 bands and 19 different patterns for β gliadins. The combination of these patterns generated 38 and 39 combinations for Gli-1 and Gli-2 loci, respectively. The genetic diversity index (H) was higher for α gliadins (0.924), followed by ω and γ gliadins (0.899 and 0.878, respectively), and for β gliadin patterns (0.866). Extensive polymorphism (H = 0.875) was observed in four gliadin pattern regions, with higher genetic diversity in the Iranian landraces than in the Azerbaijani ones. Each genotype had special identifying patterns in the gliadin acid PAGE analysis, and cluster analysis based on Jaccard's similarity coefficients formed six groups. Gliadin has a simple, repeatable and economic analysis, and can be used in genetic studies

El objetivo de este trabajo fue comparar la variabilidad, diversidad y distancias genéticas entre cerdos criollos, Pelón Mexicano (CPM) y Cuinos (CC), con Yorkshire, cuanto a los genes candidatos CAST, DECR1, HAL, HFABP4, LEP, LIPE, MCR4, MYOG, RN y CHX, a través de analysis por PCR-RFLP. Se evaluaron 180 cerdos: 59 CPM, 65 CC y 56 Yorkshire. Se analizaron las frecuencias génicas y genotípicas, heterocigosidad, distancias genéticas y árboles filogenéticos entre grupos raciales. Para CAST, DECR1, HFABP4, LEP, MCR4 y CHX las frecuencias génicas y genotípicas fueron diferentes al comparar las tres razas. En LIPE, los CC fueron iguales a los Yorkshire; en cuanto a MYOG, los CPM fueron iguales a los Yorkshire. No hubo diferencias entre poblaciones criollas y Yorkshire en las frecuencias génicas y genotípicas para HAL y RN. Los cerdos Yorkshire presentaron mayor frecuencia en alelos favorables para CAST, LIPE, MCR4 y MYOG, menor frecuencia de DECR1, HFABP4, CHX, y moderada en LEP. La heterocigosidad promedio para todos los genes fue mayor en CPM (0,42±0,05) y similar en CC (0,33±0,06) y Yorkshire (0,35±0,05). Al calcular distancias genéticas con todos los genes, los CC se encuentran más distantes de los Yorkshire.

The objective of this work was to validate, by quantitative PCR in real time (RT-qPCR), genes to be used as reference in studies of gene expression in soybean in drought-stressed trials. Four genes commonly used in soybean were evaluated: Gmβ-actin, GmGAPDH, GmLectin and GmRNAr18S. Total RNA was extracted from six samples: three from roots in a hydroponic system with different drought intensities (0, 25, 50, 75 and 100 minutes of water stress), and three from leaves of plants grown in sand with different soil moistures (15, 5 and 2.5% gravimetric humidity). The raw cycle threshold (Ct) data were analyzed, and the efficiency of each primer was calculated for an overall analysis of the Ct range among the different samples. The GeNorm application was used to evaluate the best reference gene, according to its stability. The GmGAPDH was the least stable gene, with the highest mean values of expression stability (M), and the most stable genes, with the lowest M values, were the Gmβ-actin and GmRNAr18S, when both root and leaves samples were tested. These genes can be used in RT-qPCR as reference gene for expression analysis.
