AbstractBackground:Although nutritional, metabolic and cardiovascular abnormalities are commonly seen in experimental studies of obesity, it is uncertain whether these effects result from the treatment or from body adiposity.Objective:To evaluate the influence of treatment and body composition on metabolic and cardiovascular aspects in rats receiving high saturated fat diet.Methods:Sixteen Wistar rats were used, distributed into two groups, the control (C) group, treated with isocaloric diet (2.93 kcal/g) and an obese (OB) group, treated with high-fat diet (3.64 kcal/g). The study period was 20 weeks. Analyses of nutritional behavior, body composition, glycemia, cholesterolemia, lipemia, systolic arterial pressure, echocardiography, and cardiac histology were performed.Results:High-fat diet associates with manifestations of obesity, accompanied by changes in glycemia, cardiomyocyte hypertrophy, and myocardial interstitial fibrosis. After adjusting for adiposity, the metabolic effects were normalized, whereas differences in morphometric changes between groups were maintained.Conclusion:It was concluded that adiposity body composition has a stronger association with metabolic disturbances in obese rodents, whereas the high-fat dietary intervention is found to be more related to cardiac morphological changes in experimental models of diet-induced obesity.

A influência do tamanho de sementes de milho (Zea mays L.) sobre a germinação, o vigor e a produção de grãos foi estudada mediante a instalação de testes de laboratório e de um ensaio de campo. Em laboratório foram conduzidos testes de germinação, primeira contagem de germinação, envelhecimento rápido e cloreto de amônio. No campo foram determinados a porcentagem de emergência das plântulas, "stand final", número e peso das espigas. As análises dos dados e as interpretações dos resultados permitiram concluir que o tamanho das sementes não influenciou a germinação, o vigor e a produção.

O objetivo principal do presente trabalho foi determinar através do teste de germinação, a importância do peso das sementes de arroz (Oryza sativa L.). Utilizaram-se sementes das variedades Batatais, Pratão Precoce, IAC-120, IAC-435 e IAC-1246. O trabalho foi conduzido no Laboratório de Sementes do D.A.H. da ESALQ no período de novembro de 1973 a dezembro de 1974. As sementes das cinco variedades foram submetidas à ação de ventilação controlada através de um assoprador, permitindo a separação de cada variedade em três frações com pesos diferentes, isto é, leve, média e pesada. As sementes leves, médias e pesadas, assim obtidas, foram armazenadas em câmara seca e laboratório. Os testes de germinação foram efetuados em três épocas (março/abril, julho e dezembro de 1974), utilizando-se como delineamento experimental um fatorial 3x2, cujas variáveis foram três pesos e dois ambientes de conservação das sementes, com quatro repetições. As análises dos dados e a interpretação dos resultados obtidos permitiram que se chegassem às seguintes conclusões: a) as sementes pesadas e médias apresentaram uma maior porcentagem de germinação que as leves em todas as variedades estudadas; b) este efeito foi menos intenso no caso das sementes das variedades Batatais e IAC-1246; c) o ambiente de câmara seca mostrou-se superior ao de laboratório para a conservação das sementes.

No Estado de São Paulo, a coleta de sementes de seringueira se inicia em fevereiro e se prolonga até os meses de março/abril; os meses subsequentes se caracterizam por apresentarem um período relativamente seco e frio, inadequado para a instalação de viveiros. Desta forma, seria importante o armazenamento das sementes até os meses setembro/outubro. As sementes de seringueira perdem a viabilidade, em condições normais de ambiente, num período relativamente curto e exigem determinadas condições para o seu armazenamento; assim, estudaram--se no presente trabalho os efeitos do tratamento fungicida e de três ambientes de armazenamento sobre a qualidade das sementes acondicionadas em sacos plásticos. Foram conduzidos 2 experimentos com sementes provenientes de seringais de cultivo, nas safras de 1983 e 1984. Os testes de emergência em areia, altura média, peso fresco médio e peso seco médio de plantas foram conduzidos em épocas (1983) e 5 épocas (1984) espaçadas de 2 meses. Observou -se que os fungicidas Benlate e Captan não se mostraram adequados ao tratamento das sementes de seringueira; sementes de boa qualidade fisiológica e apresentando grau de umidade acima de 30% foram bem conservadas por um período de 6 meses, quando armazenadas em condições normais de ambiente (sem controle de umidade e de temperatura).

Este trabalho foi conduzido com o objetivo de estudar o efeito da escolha manual, da seleção eletrônica pela cor e do tratamento fungicida sobre a qualidade de sementes de amendoim (Arachis hypogaea L.). O lote, após o descascamento, limpeza, classificação em peneira 22/64 de polegada e separação da porção mais pesada obtida pelo processamento em mesa gravitacional foi submetido aos seguintes tratamentos: 1. material original (testemunha), 2. material submetido a uma seleção eletrônica, 3. material submetido a duas seleções eletrônicas, 4. material submetido a três seleções eletrônicas, 5. escolha manual por oito pessoas, 6. escolha manual por quatro pessoas seguida de uma seleção eletrônica, 7. material descartado pela selecionadora eletrônica. Todos os tratamentos foram divididos em duas partes e uma delas foi tratada com o fungicida thiran, (2 g.i.a por kg de sementes). Para avaliar a qualidade das sementes foram conduzidos os seguintes testes: análise de pureza física, exame de sementes com tegumento danificado, germinarão, envelhecimento acelerado, emergência em areia, emergência em campo e exame de sanidade das sementes. Conclui-se que as sementes descartadas pela sele cionadora eletrônica, composta predominantemente de sementes de amendoim de coloração mais clara e mais escura que a cor característica do cultivar e de sementes com tegumento danificado expondo o embrião foram de menor qualidade física, fisiológica e sanitária, entretanto, a remoção destes materiais através da escolha manual e/ou seleção eletrônica melhorou apenas a qualidade física, não alterou a qualidade fisiológica e sanitaria do lote de sementes. O tratamento fungicida proporcionou melhoria na qualidade fisiológica e sanitária das sementes e exerceu efeito protetivo no momento da semeadura.

Homologies of minicircle kDNA of 27 Mexican stocks were studied by cross-hybridization with four kDNA probes derived from three reference stocks belonging to groups Trypanosoma cruzi I (SO34 cl4 and Silvio) and T. cruzi II (MN) and one Mexican stock. High homologies were only observed with Silvio (six stocks) and Mexican probes (11 stocks). After 30 min exposure (low homology) additional stocks were recognized with SO34 cl4 (three stocks) and Silvio (six stocks) probes; with the Mexican probe only five stocks remained non-reactive. All the stocks were typed by isoenzyme (16 loci) and Mexican stocks belonged to T. cruzi I. Hybridization patterns were not strictly correlated with the observed clustering and cross-hybridization of kDNA minicircles is not available to distinct Mexican stocks.

From 2002 to 2005, a program of active search for patients with hepatosplenic schistosomiasis and schistosomal myeloradiculopathy has been implemented in the state of Minas Gerais by the local Health Department. The state was divided in 28 regional health centers and the local representatives have been trained to identify and direct patients with hepatosplenic schistosomiasis and neuroschistosomiasis to a reference center in Belo Horizonte, the capital of the state of Minas Gerais. Seventy five patients with hepatosplenic schistosomiasis and 54 with schistosomal myeloradiculopathy have been referred and examined in the reference center in a period of time of 3 years. Schistosomal myeloradiculopathy should be emphasized because the number of cases reported is increasing rapidly and when timely diagnosed and treated, they respond promptly to treatment. Left untreated, they die or become invalid for life. In our view, the time has come for more active investigation of the different aspects of morbidity caused by schistosomiasis mansoni in Brazil.

Chemokines are a superfamily of low-molecular-weight cytokines that were initially described for their chemoattractant activity. It is now clear chemokines have several other activities that modulate immune processes. More than 50 chemokines ligands and at least 19 receptors have been described to date. Depending on the number of N-terminal cysteine residues, chemokines are grouped in the subfamilies CXC, CC, C or CX3C. A growing body of evidence suggests a role for chemokines in the pathogenesis of several inflammatory diseases. Our studies involving mice and humans infected with Schistosoma mansoni suggest an important role of the chemokine CCL3 and its receptors (CCR1 and CCR5) in the pathogenesis of severe schistosomiasis. We suggest that the differential activation of CCR1 or CCR5 during the course of schistosomiasis may dictate the outcome of the disease.

The leishmanicidal activity of four batches of meglumine antimoniate, produced in Farmanguinhos-Fiocruz, Brazil (TAMs), was assessed and compared to Glucantime®-Aventis Pharma Ltda. Using the amastigote-like in vitro model, the active concentrations of Sb v varied from 10µg/ml to 300 µg/ml for L. (L.) chagasi and from 50µg/ml to 300µg/ml for L. (L.) amazonensis, with no statistically significant differences among the four batches of TAMs and Glucantime®. The inhibitory concentrations (IC50) determined by the amastigote-infected macrophage model for TAM01/03 and Glucantime® were, respectively: 26.3µg/ml and 127.6µg/ml for L. chagasi, 15.4µg /ml and 22.9µg/ml for L. amazonensis, and 12.1µg/ml and 24.2µg/ml for L. (V.) braziliensis. The activities of the four batches of TAMs were confirmed in an in vivo model by assessing, during eight weeks skin lesions caused by L. braziliensis in hamster that were treated with 20mg Sb v/Kg/day for 30 consecutive days. The meglumine antimoniate produced by Farmanguinhos was as effective as the reference drug, Glucantime®-Aventis, against three species of Leishmania that are of medical importance in Brazil.

We assessed the risk classification of dengue fever based on the capture of Aedes aegypti adults using MosquiTRAP, a type of sticky trap, in comparison with traditional larval infestation indices. A total of 27 MosquiTRAPs were installed, with one trap per block, and were inspected weekly between November 2008-February 2009. Infestation baseline data were obtained from a survey conducted prior to trap installation. The index generated by MosquiTRAP and house index (HI) classified the area "in alert situation". The set for risk of dengue occurrence proposed by the use of MosquiTRAP classify areas in the same way of the traditional HI.

Leptospirosis is a zoonotic disease affecting most mammals and is distributed throughout the world. Several species of domestic and wild animals may act as reservoirs for this disease. The purpose of this study was to assess the exposure of free-ranging wild carnivores, horses and domestic dogs on a private reserve located in the northern Pantanal (Brazil) and the surrounding areas to Leptospira spp from 2002-2006, 75 free-ranging wild carnivores were captured in the Pantanal and serum samples were collected. In addition, samples from 103 domestic dogs and 23 horses in the region were collected. Serum samples were tested for the presence of Leptospira antibodies using the microscopic agglutination test. Thirty-two wild carnivores (42.7%) were considered positive with titres > 100, and 18 domestic dogs (17.5%) and 20 horses (74.1%) were also found to be positive. Our study showed that horses, dogs and several species of free-ranging wild carnivores have been exposed to Leptospira spp in the Pantanal, suggesting that the peculiar characteristics of this biome, such as high temperatures and an extended period of flooding, may favour bacterial persistence and transmission. In this region, wild carnivores and horses seem to be important hosts for the epidemiology of Leptospira species.

This work reports a survey of Leptospira spp in pampas deer (Ozotoceros bezoarticus) in the Pantanal wetlands of the state of Mato Grosso do Sul, Brazil by serology and polymerase chain reaction (PCR). Seventy pampas deer were captured in the dry season and surveyed using PCR, microscopic agglutination test (MAT) (n = 51) and by both techniques (n = 47). PCR detected infections in two pampas deer and MAT detected infections in three. Through sequencing and phylogenetic analyses, the PCR-amplified fragment detected in deer was identified as Leptospira interrogans. Serovars Pomona and Butembo were detected using MAT and the highest titre was 200 for serovar Pomona. Epidemiological aspects of the findings are discussed.

Vaccination is the method of choice for the prevention of influenza infection. However, the quantity of the antigen available, especially in the case of pandemics, often fails to meet the global demand. However, improved adjuvants can overcome this problem. Preliminary results obtained in this study revealed that one year after a single subcutaneous immunisation with influenza A H3N2 virus in an oil-based carrier, VaxcineTM, outbreed mice produced a high immunoglobulin G response that lasted for up to one year and exhibited less variation in titre compared with the response of the control group treated with alum. The haemagglutination-inhibition titres induced by VaxcineTM were also higher than those generated by alum. These data indicate that VaxcineTM is a good adjuvant candidate for seasonal influenza vaccines.
