The DNA extraction is a critical step in Genetically Modified Organisms analysis based on real-time PCR. In this study, the CTAB and DNeasy methods provided good quality and quantity of DNA from the texturized soy protein, infant formula, and soy milk samples. Concerning the Certified Reference Material consisting of 5% Roundup Ready® soybean, neither method yielded DNA of good quality. However, the dilution test applied in the CTAB extracts showed no interference of inhibitory substances. The PCR efficiencies of lectin target amplification were not statistically different, and the coefficients of correlation (R²) demonstrated high degree of correlation between the copy numbers and the threshold cycle (Ct) values. ANOVA showed suitable adjustment of the regression and absence of significant linear deviations. The efficiencies of the p35S amplification were not statistically different, and all R² values using DNeasy extracts were above 0.98 with no significant linear deviations. Two out of three R² values using CTAB extracts were lower than 0.98, corresponding to lower degree of correlation, and the lack-of-fit test showed significant linear deviation in one run. The comparative analysis of the Ct values for the p35S and lectin targets demonstrated no statistical significant differences between the analytical curves of each target.

The consumption of Brazilian cassava has been reduced due to a lack of adjustment to the modern lifestyle. To reverse this trend, new products could be developed specifically targeted to high-value niche markets. Cereal bars stand out as fast food high in nutritional value. A bar formula mimicking cereal bars was prepared using a mixture of Brazilian cassava flour, hydrogenated vegetable fat, dried bananas, ground cashew nuts, and glucose syrup. After being pressed, the bars were dried for 1 hour at 65 °C, packaged in films, and stored under ambient conditions. Its stability was continuously monitored for 210 days in order to ensure its safety and enable its introduction to the market. Texture loss was observed in the packed bars after 90 days of storage, but the sensory characteristics allowed the testers to perceive this tendency after only 30 days of storage. However, chemical, physical, and microbial analyses confirmed that the bars were safe for consumption for 180 days. The results showed that a 45 g cassava flour-based bar enriched with nuts and dried fruits can meet 6% of the recommended daily fiber intake with a caloric value between that of the common cereal bar and that of an energy bar. Adapting the formula with ingredients (fruits, nuts) from different regions of Brazil may add value to this traditional product as a fast food.

The purpose of this study was to introduce yam in the development of two new composite flours containing soy and cassava. Two composite flours were obtained after fermentation of yam, soybean, and cassava in respectively 60, 30, and 10% proportions. Two varieties of yam were used: Dioscorea alata (variety "Bete bete") and Dioscorea cayenensis (variety "Lokpa"). Proximate composition, mineral content, some anti-nutritional factors (oxalates, phenols), microbiological quality, and α-amylase digestibility were determined for the fermented and unfermented composite flours. The results indicated that for the composite flours made of D. alata and D. cayenensis, fermentation increased ash and titrable acidity. Carbohydrates, pH, and energy decreased. Crude fat content was not affected by the fermentation process. Anti-nutritional factors such as oxalates and phenols were found to decrease significantly after the fermentation of the composite flours. Fermentation increased the mineral content (Mg, K, Fe, and Ca) of the composite flours. A decrease in P and Na was observed after fermentation. The microbiological study showed that safety flours contain no potential pathogenic germs. The in vitro α-amylase digestibility of the composite flours was significantly improved after fermentation. The biochemical characteristics and good hygienic quality of the obtained flours suggest that these flours can be considered as a feeding alternative for children in poor areas where yam is produced.

The objective of this study was to prepare roasted and lyophilized tempeh flour with soybean cultivar BRS 267 to apply them in the formulation of coconut biscuits. The cookies produced with whole soy flour and mixed flour of soybean and tempeh were evaluated for proximate composition, fatty acid profile, and isoflavone aglycones in order to verify the effects of inoculation with the fungus Rhizopus oligosporus and those of the drying processes of roasting and lyophilization on the chemical characteristics of the final product. Sensory acceptance and purchase intention of the formulated products were also evaluated. The results indicate the maintenance of linolenic acid, which is important in the prevention of coronary diseases, and an increase in the aglycones levels when the tempeh flour was used. Lipids and proteins showed differences, and the sensory analyses demonstrated similarity between the cookies with satisfactory scores for aroma, flavor, texture, and overall acceptability for both samples. when compared to the control. Purchase intent was also positive for the lyophilized and toasted tempeh flours, thus enabling the use of the roasting process as a simple drying method, for processing tempeh and obtaining a flour rich in proteins and aglycones that can be used as a partial substitute for soy flour in cookies and other bakery products.

Food industry has been developing products to meet the demands of increasing number of consumers who are concerned with their health and who seek food products that satisfy their needs. Therefore, the development of processed foods that contain functional components has become important for this industry. Microencapsulation can be used to reduce the effects of processing on functional components and preserve their bioactivity. The present study investigated the production of lipid microparticles containing phytosterols by spray chilling. The matrices comprised mixtures of stearic acid and hydrogenated vegetable fat, and the ratio of the matrix components to phytosterols was defined by an experimental design using the mean diameters of the microparticles as the response variable. The melting point of the matrices ranged from 44.5 and 53.4 ºC. The process yield was melting point dependent; the particles that exhibited lower melting point had greater losses than those with higher melting point. The microparticles' mean diameters ranged from 13.8 and 32.2 µm and were influenced by the amount of phytosterols and stearic acid. The microparticles exhibited spherical shape and typical polydispersity of atomized products. From a technological and practical (handling, yield, and agglomeration) points of view, lipid microparticles with higher melting point proved promising as phytosterol carriers.

Okara is a by-product generated during the manufacture of soymilk and tofu. Wet okara was added to beef burgers at 0%, 20%, and 25%. The effects of okara on certain physicochemical, textural, and sensory properties of reduced fat beef burgers were investigated. The beef burgers formulated with okara (104.0-106.0 kcal/100 g) had 60% less calories than commercial beef burgers (268.8 kcal/100 g). The texture profile analysis showed that the addition of wet okara led to a significant increase in hardness (p < 0.05) and a concomitant reduction in the values of chewiness, springiness, and cohesiveness. Lower sensory scores (p < 0.05) of flavour were observed in the beef burgers containing 25% wet okara. However, the sensory evaluation results showed that juiciness, appearance, tenderness, and overall acceptability of beef burgers formulated with okara did not differ statistically from that of the control (0% okara). Wet okara (20%) can be used as a non-meat protein source in the production of reduced-fat beef burgers without changing their sensory quality.

Chloropropanols, including 3-monochloropropane-1,2-diol (3-MCPD) and 1,3-dichloropropan-2-ol (1,3-DCP), comprise a group of chemical contaminants with carcinogenic and genotoxic properties. They have been found in a variety of processed foods and food ingredients, such as hydrolyzed vegetable protein, soy sauce, cereal-based products, malt-derived ingredients, and smoked foods. This study aimed to assess the dietary exposure to 3-MCPD and 1,3-DCP in Brazil and verify whether the presence of these substances in foods could represent health risks. The intake was calculated by combining data on food consumption, provided by the Consumer Expenditure Survey 2008-2009, with the levels of contaminant occurrence determined by gas chromatography-mass spectrometry. The exposure to 3-MCPD ranged from 0.06 to 0.51 µg.kg bw-1.day-1 considering average and high consumers, while the intake of 1,3-DCP was estimated to be 0.0036 µg.kg bw-1.day-1 in the worst case scenario evaluated. Based on these results, it was verified that the Brazilians' exposure to chloropropanols does not present a significant health risk. However, the consumption of specific foods containing high levels of 3-MCPD could exceed the provisional maximum tolerable daily intake of 2 µg.kg bw-1 established for this compound and, therefore, represent a potential concern.

Abstract The aim of this study was to evaluate the physical quality and sensory acceptance of gluten-free breads with different percentages of chia flour (Salvia Hispanica L .). The chia flour was used to substitute rice flour and soy flour in order to replace the gum required in this type of bread. Four formulations were developed; a standard made with gum, and three formulations with 2.5%, 5.0% and 7.5% of chia flour. Analyses of specific volume, cooking losses and the rise in dough of the breads were performed. Sensory analysis included tests for affective acceptability and purchase intent. The results showed that the bread with 2.5% chia flour had specific volume and cooking losses similar to the standard. In terms of the rise in dough, the standard showed the highest values, followed by the bread made with 5.0% chia flour. The substitution of soy and rice flour with 2.5% of chia flour produced bread with sensory characteristics similar to the standard in all of the analyzed attributes; it also received higher purchase intent. Using chia flour at a concentration of 2.5%, compared to rice flour and soy flour, proved that it was possible to replace gum in the bread formulation.

Abstract The objective of this work was to evaluate the antioxidant activity of protein hydrolysates obtained by the enzymatic hydrolysis of okara using an endopeptidase (Alcalase) and exopeptidase (Flavourzyme). The reaction was monitored by the pH-stat procedure in which five aliquots were collected during the hydrolysis by each enzyme, corresponding to different degrees of hydrolysis (DH). The antioxidant activities of the aliquots were evaluated by the ABTS, DPPH and FRAP methods. For the hydrolysates obtained using Alcalase, the antioxidant activities increased from: 68.6 to 99.5% (ABTS), 14.5 to 17.7% (DPPH) and 222.6 to 684.9 µM Trolox (FRAP), when the DH varied from 0 to 33.6%. With respect to Flavourzyme, the results were: 67.2 to 88.2% (ABTS), 9.5 to 18.5% (DPPH) and 168.0 to 360.3 µM Trolox (FRAP), when the DH increased up to 5.8%. The results showed that the protein hydrolysates had antioxidant capacities, which were influenced by the degree of hydrolysis and the type of enzyme.

Com o objetivo de avaliar o efeito do vigor de lotes de sementes na emergência em campo, no desenvolvimento vegetativo e reprodutivo de plantas de soja, foi conduzido o presente experimento durante o ano agrícola 98/99, em Jaboticabal, SP. Foram utilizados nove lotes de sementes de soja, da cultivar M-Soy 109, com padrões comercializáveis. Na caracterização dos lotes em laboratório, foram utilizados os seguintes testes: teor de água, germinação em areia, índice de velocidade de emergência, comprimento de plântulas, matéria seca de plântulas, condutividade elétrica e envelhecimento acelerado. Os dados obtidos por meio dos testes de laboratório permitiram classificar os nove lotes em três de alto, três de médio e três de baixo vigor. Para a semeadura desses nove lotes no campo, calculou-se, com os dados de germinação, a quantidade de sementes para a correção do estande, visando obter uma população de 400 mil plantas por hectare, num espaçamento de 0,50m entre linhas.As características avaliadas, no campo, foram o índice de velocidade de emergência, a emergência total, os estande inicial e final, a altura das plantas aos 18, 38 e 49 dias após a semeadura e na colheita, quantidade de plantas com flores em R1, o teor de água na colheita, a altura de inserção da primeira vagem, a sobrevivência de plantas, o número de vagens por planta, o número de sementes por vagem, a massa de 100 sementes e a produção, tanto em g.planta-1 quanto em kg.ha-1. O efeito do vigor das sementes foi maior no início do desenvolvimento das plantas (emergência total e velocidade de emergência); lotes de menor vigor tiveram quedas mais acentuadas de estande. Houve um atraso no início do florescimento, mostrando um efeito de tipo residual do vigor das sementes. As plantas que conseguiram se formar a partir de lotes de baixo vigor eram vigorosas e, em condições favoráveis de clima e manejo, em função do maior espaço por planta, produziram mais vagens por planta, resultando em maior produção por planta e isso levou-as a igualarem-se em produtividade às dos demais níveis de vigor.

A comercialização antecipada de soja é uma realidade em Mato Grosso. Isso implica na necessidade de testes rápidos, confiáveis e que permitam minimizar riscos futuros, principalmente, quando existe um agente complicador a armazenabilidade, como é o caso de sementes com retenção de clorofila. Mudanças associadas à deterioração com conseqüente perda de vigor e viabilidade são observadas nesse tipo de semente, fazendo-se necessário o conhecimento da localização e abrangência dos pontos deteriorativos. Objetivando avaliar testes que sejam factíveis e tenham potencial de determinar a qualidade de sementes esverdeadas, foi realizado este estudo. FMT-Tucunaré, ciclo semi-precoce, e EMGOPA 313, FMT-Arara Azul e M-SOY 9350, ciclo tardio, que apresentavam 25,7%, 26,8%, 22,6% e 15,6%, respectivamente, de sementes esverdeadas, foram as cultivares estudadas. Para cada cultivar foi utilizada uma amostra testemunha, isenta de pigmento verde. Cada amostra constituiu um tratamento. A qualidade foi avaliada em duas épocas (inicial, maio/2001 e final, novembro/2001), através dos testes de germinação, tetrazólio, emergência em areia, envelhecimento acelerado (24 horas e 48 horas), utilizando tetrazólio em substituição à germinação, e massa seca de plântulas. No teste de tetrazólio, a subclasse DU 4-5 indica a deterioração por umidade, como principal problema fisiológico evolutivo e o tetrazólio, em substituição à germinação no teste de envelhecimento acelerado, 24 horas, pode avaliar o risco do potencial fisiológico do lote com sementes esverdeadas, na época inicial de armazenagem.

O objetivo deste trabalho foi avaliar os efeitos de reduções na população de plantas sobre a produtividade e a qualidade fisiológica da semente de soja. Duas cultivares de soja (BRSMG 68 Vencedora e M-SOY 8001) foram cultivadas em áreas com e sem o controle de plantas daninhas. As populações estudadas foram: 400.000, 340.000, 280.000, 220.000 e 160.000 pl.ha-1, em um espaçamento entrelinhas de 0,43m. De maneira geral, a presença de plantas daninhas causa decréscimos na produtividade e no tamanho da semente produzida. Entretanto, variações na população não interferem na qualidade fisiológica, no tamanho e na massa de 100 sementes. A cultura da soja é capaz de suportar grandes reduções de população sem perdas significativas de produtividade. Essa capacidade é grandemente influenciável pela cultivar: a Vencedora suporta reduções de até 45% e a M-SOY 8001 de até 30%, em relação à população de 400.000 pl.ha-1.

Dentre os conceitos modernos de controle de pragas, o uso de inseticidas no tratamento de sementes constitui-se num dos métodos mais eficientes. Deve-se, entretanto, conhecer a influência desses produtos com relação à qualidade fisiológica das sementes tratadas. Objetivou-se através desta pesquisa avaliar a qualidade fisiológica de sementes de soja tratadas com inseticidas, sob quatro períodos de armazenamento (0, 15, 30 e 45 dias após o tratamento). O delineamento experimental foi o inteiramente casualizado, em esquema fatorial 4 x 7, com quatro repetições, onde as sementes do cultivar M-SOY 6101 foram tratadas com os inseticidas thiamethoxam (Cruiser 700 WS), na dose de 250 mL de produto comercial (p.c.)/100 kg de sementes; fipronil (Standak), na dose de 150 mL de p.c./100 kg de sementes; imidacloprid (Gaucho FS), na dose de 150 ml de p.c./100 kg de sementes; [imidacloprid + thiodicarb] (CropStar FS), na dose de 0,3 L. ha-1; carbofuran (Furadan 350 TS), na dose de 1,5 L/100 kg de sementes; acefato (Orthene 750 BR), na dose de 1 kg/ 100 kg de sementes e uma testemunha, sem tratamento. As variáveis analisadas foram: germinação, velocidade de emergência, comprimento de raiz e de plântula e porcentagem de plântulas normais no teste de envelhecimento acelerado. A aplicação dos inseticidas carbofuran e acefato é prejudicial à qualidade fisiológica das sementes de soja cultivar M-SOY 6101, por um período de armazenamento de até 45 dias. A redução da qualidade fisiológica das sementes, condicionada pelos inseticidas avaliados, intensifica-se com o prolongamento do armazenamento das sementes tratadas, recomendando-se, portanto, que o tratamento inseticida das sementes seja realizado próximo ao momento da semeadura.
