Movement of transmigrants and livestock from western Indonesia to southeastern areas of Irian Jaya near the border with Papua New Guinea may pose a risk of introducing Trypanosoma evansi into Papua New Guinea via feral Rusa deer (Cervus timorensis russa) and wild pigs which inhabit these areas in large numbers. Pilot experimental studies were conducted to observe infection in pigs and Rusa deer with a strain of T. evansi isolated in Indonesia. Parasitaemia and signs of clinical disease were monitored each second day for 120 days. Trypanosomes were observed in haematocrit tubes at the plasma-buffy coat interface of jugular blood of deer and pigs on 86% and 37% of sampling occasions respectively. Parasitaemia was at a high level in deer for 35% of the time but for only 11.5% of the time in pigs. Results indicate that both Rusa deer and pigs have a high tolerance for infection with T. evansi. The deer suffered mild anaemia evidenced by a 25% reduction in packed cell volume (PCV) 14 days after infection which coincided with the initial peak in parasitaemia. However, PCV had returned to pre infection values by the end of the experiment. The pigs showed no change in PCV. There were no visual indications of disease in either species and appetite was not noticeably affected. It was concluded that both Rusa deer and pigs were capable reservoir hosts for T. evansi but that Rusa deer, with their more persistent higher levels of parasitaemia, have more potential to spread T. evansi into Papua New Guinea from West Irian than pigs.

We present here three expression plasmids for Trypanosoma cruzi adapted to the Gateway® recombination cloning system. Two of these plasmids were designed to express trypanosomal proteins fused to a double tag for tandem affinity purification (TAPtag). The TAPtag and Gateway® cassette were introduced into an episomal (pTEX) and an integrative (pTREX) plasmid. Both plasmids were assayed by introducing green fluorescent protein (GFP) by recombination and the integrity of the double-tagged protein was determined by western blotting and immunofluorescence microscopy. The third Gateway adapted vector assayed was the inducible pTcINDEX. When tested with GFP, pTcINDEX-GW showed a good response to tetracycline, being less leaky than its precursor (pTcINDEX).

Odontopus brevirostris (Hustache, 1936) feeding on Annona squamosa L., A. cherimola Mill., A. glabra L., and A. muricata L. was observed. The last three host plants are recorded for the first time. The endophitic oviposition occurs in the veins of the ventral surface of the young leaves. The larvae, leaf miners, eat the parenchyma and the adults make small holes in the leaves. The pupation occurs in spherical cocoons protected by a sort of nest (pupation chamber) between the two epidermal layers.

Paty do Alferes (RJ) é um município com tradição agrícola, sendo a olericultura a sua principal atividade econômica. A região apresenta uma topografia e regime de chuvas que favorecem a erosão. A falta de um manejo adequado do solo e o uso excessivo de agroquímicos favorecem os processos de degradação física e química do solo, acarretando sérios impactos ao ambiente. Com o objetivo de avaliar o acúmulo de metais pesados no solo e em plantas de tomate (Lycopersicon esculentum Mill), cultivadas sob diferentes sistemas de preparo do solo, foi realizado um trabalho no município de Paty do Alferes (RJ), entre os meses de agosto e dezembro de 1999. Foram instaladas parcelas do tipo Wischmeier (com coletores de enxurrada e sedimentos), com dimensões de 22,0 x 4,0 m, em um Latossolo Vermelho-Amarelo com 30 % de declividade. Cada tratamento consistiu de um tipo de sistema de preparo do solo: (a) preparo convencional: aração no sentido morro abaixo e queima dos restos vegetais (PC); (b) preparo em curvas de nível: aração com tração animal em nível com faixas de capim-colonião a cada 7,0 m (PN); (c) cultivo mínimo: apenas abertura de covas para plantio e conservação dos restos vegetais (CM). Foram coletadas amostras de solo e plantas de tomate, sendo analisados os teores de metais pesados nas amostras coletadas, a produtividade das plantas e a fertilidade do solo. A produtividade no cultivo mínimo foi significativamente inferior à de outros manejos, apesar de tal sistema favorecer a maior fertilidade do solo. Esse manejo também proporcionou maior acúmulo de metais pesados no solo, principalmente quando comparado ao plantio convencional. Com relação às plantas, a aplicação foliar de defensivos agrícolas favoreceu o maior acúmulo de metais pesados na parte aérea das plantas, e os teores de Cu e Zn estiveram acima dos limites permitidos para metais pesados em plantas. Os frutos produzidos nos três sistemas de preparo do solo apresentaram concentrações de Pb próximas ao limites permitidos para alimentos frescos.

A produção de biomassa, a manutenção dos resíduos vegetais sobre o solo e sua posterior decomposição são fatores de grande importância no estudo da ciclagem de nutrientes. Este estudo foi desenvolvido na área experimental do CEFET-Uberaba-MG, onde foram avaliados oito tipos de coberturas vegetais: milheto (Pennisetum americanum sin. tiphoydes), braquiária (Brachiaria brizantha), sorgo-forrageiro (Sorghum bicolor L. Moench), guandu (Cajanus cajan (L.) Mill sp.), crotalária (Crotalarea juncea), aveia-preta (Avena strigosa Schreb), pousio e área em preparo convencional de solo (testemunha) em área de Cerrado, na região do Triângulo Mineiro. Avaliaram-se a fitomassa seca (FS), a decomposição dos resíduos em bolsas de decomposição, e a liberação de K. Utilizou-se um modelo matemático para descrever a decomposição dos resíduos e a liberação de K, e calcularam-se a constante de decomposição (k) e o tempo de meia-vida (T½). O milheto, o sorgo e a crotalária foram as coberturas que apresentaram maiores produções de matéria seca. O maior acúmulo de K ocorreu em gramíneas e a maior liberação de K ocorreu no milheto, aveia, braquiária e crotalária nos primeiros 42 dias após manejo, nos dois períodos avaliados. A braquiária apresentou o menor T½ vida e a maior taxa de liberação de K.

Microbial processes have been used as indicators of soil quality, due to the high sensitivity to small changes in management to evaluate, e.g., the impact of applying organic residues to the soil. In an experiment in a completely randomized factorial design 6 x 13 + 4, (pot without soil and residue or absolute control) the effect of following organic wastes was evaluated: pulp mill sludge, petrochemical complex sludge, municipal sewage sludge, dairy factory sewage sludge, waste from pulp industry and control (soil without organic waste) after 2, 4, 6, 12, 14, 20, 28, 36, 44, 60, 74, 86, and 98 days of incubation on some soil microbial properties, with four replications. The soil microbial activity was highly sensitive to the carbon/nitrogen ratio of the organic wastes. The amount of mineralized carbon was proportional to the quantity of soil-applied carbon. The average carbon dioxide emanating from the soil with pulp mill sludge, corresponding to soil basal respiration, was 0.141 mg C-CO2 100 g-1 soil h-1. This value is 6.4 times higher than in the control, resulting in a significant increase in the metabolic quotient from 0.005 in the control to 0.025 mg C-CO2 g-1 Cmic h-1 in the soil with pulp mill sludge. The metabolic quotient in the other treatments did not differ from the control (p < 0.01), demonstrating that these organic wastes cause no disturbance in the microbial community.

Currently, sugarcane plays an important global role, particularly with a view to alternative energy sources. Thus, in a sugarcane field of the mill Vale do Paraná S/A Álcool e Açúcar, Rubineia, São Paulo State, managed under two green cane harvest systems (cane trash left on and cane trash removed from the soil), Pearson and spatial correlations between the sugarcane yield (variety RB855035 in the third cut) and soil physical and chemical properties were studied to identify the property best correlated with stalk yield and the best harvest method. For this purpose, two geostatistical grids (121 sampling points on 1.30 ha) were installed on a eutrophic Red Argisol (homogeneous slope of 0.065 m m-1), in 2011, to determine the properties: stalk yield and sugarcane plant population, and soil resistance to penetration, gravimetric moisture, bulk density, and carbon stock, in the layers 0-0.20 and 0.20-0.40 m. The data were analyzed by descriptive, linear correlation and geostatistical analysis. In both treatments, the property stand density was best correlated with sugarcane yield (r = 0.725 in the trash mulching treatment - TM and r = 0.769 in the trash removal treatment - TR). However, in relation to the soil properties, bulk density (0-0.20 m) was best correlated (r = 0.305 in TM, r = 0.211 in TR). Similarly, from the spatial point of view, stand density was the property that best explained the sugarcane yield. However, in the TM treatment the density (0.20-0.40 m) was the only soil property spatially correlated with stalk yield. The carbon stock in the soil of the TM was 11.5 % higher than in the TR treatment. Results of the TM treatment were best, also with regard to soil management and conservation.

Soil sampling should provide an accurate representation of a given area so that recommendations for amendments of soil acidity, fertilization and soil conservation may be drafted to increase yield and improve the use of inputs. The aim of this study was to evaluate the variability of soil fertility properties of Oxisols in areas planted to sugarcane in the State of Goias, Brazil. Two areas of approximately 8,100 m² each were selected, representing two fields of the Goiasa sugarcane mill in Goiatuba. The sugarcane crop had a row spacing of 1.5 m and subsamples were taken from 49 points in the row and 49 between the row with a Dutch auger at depths of 0.0-0.2 and 0.2-0.4 m, for a total of 196 subsamples for each area. The samples were individually subjected to chemical analyses of soil fertility (pH in CaCl2, potential acidity, organic matter, P, K, Ca and Mg) and particle size analysis. The number of subsamples required to compose a sample within the acceptable ranges of error of 5, 10, 20 and 40 % of each property were computed from the coefficients of variation and the Student t-value for 95 % confidence. The soil properties under analysis exhibited different variabilities: high (P and K), medium (potential acidity, Ca and Mg) and low (pH, organic matter and clay content). Most of the properties analyzed showed an error of less than 20 % for a group of 20 subsamples, except for P and K, which were capable of showing an error greater than 40 % around the mean. The extreme variability in phosphorus, particularly at the depth of 0.2-0.4 m, attributed to banded application of high rates of P fertilizers at planting, places limitations on assessment of its availability due to the high number of subsamples required for a composite sample.

Lime application recommendations for amendment of soil acidity in sugarcane were developed with a burnt cane harvesting system in mind. Sugarcane is now harvested in most areas without burning, and lime application for amendment of soil acidity in this system in which the sugarcane crop residue remains on the ground has been carried out without a scientific basis. The aim of this study was to evaluate the changes in soil acidity and stalk and sugar yield with different rates of surface application of calcium, magnesium silicate, and gypsum in ratoon cane. The experiment was performed after the 3rd harvest of the variety SP 81-3250 in a commercial green sugarcane plantation of the São Luiz Sugar Mill (47º 25' 33" W; 21º 59' 46" S), located in Pirassununga, São Paulo, in southeast Brazil. A factorial arrangement of four Ca-Mg silicate rates (0, 850, 1700, and 3400 kg ha-1) and two gypsum rates (0 and 1700 kg ha-1) was used in the experiment. After 12 months, the experiment was harvested and technological measurements of stalk and sugar yield were made. After harvest, soil samples were taken at the depths of 0.00-0.05, 0.05-0.10, 0.10-0.20, 0.20-0.40, and 0.40-0.60 m in all plots, and the following determinations were made: soil pH in CaCl2, organic matter, P, S, K, Ca, Mg, H+Al, Al, Si, and base saturation. The results show that the application of gypsum reduced the exchangeable Al3+ content and Al saturation below 0.05 m, and increased the Ca2+ concentration in the whole profile, the Mg2+ content below 0.10 m, K+ below 0.4 m, and base saturation below 0.20 m. This contributed to the effect of surface application of silicate on amendment of soil acidity reaching deeper layers. From the results of this study, it may be concluded that the silicate rate recommended may be too low, since the greater rates used in this experiment showed greater reduction in soil acidity, higher levels of nutrients at greater depths and an increase in stalk and sugar yield.

Biochar (carbonized biomass for agricultural use) has been used worldwide as soil amendment and is a technology of particular interest for Brazil, since its "inspiration" is from the historical Terra Preta de Índios(Amazon Dark Earth), and also because Brazil is the world's largest charcoal producer, generating enormous residue quantities in form of fine charcoal and due to the availability of different residual biomasses, mainly from agroindustry (e.g., sugar-cane bagasse; wood and paper-mill wastes; residues from biofuel industries; sewage sludge etc), that can be used for biochar production, making Brazil a key actor in the international scenario in terms of biochar research and utilization). In the last decade, numerous studies on biochar have been carried out and now a vast literature, and excellent reviews, are available. The objective of this paper is therefore to deliver a critical review with some highlights on biochar research, rather than an exhaustive bibliographic review. To this end, some key points considered critical and relevant were selected and the pertinent literature "condensed", with a view to guide future research, rather than analyze trends of the past.

Este trabalho foi realizado na zona semi-árida de Pernambuco e teve como objetivo investigar o efeito de espaçamento, e a freqüência e intensidade de colheitas da palma-forrageira (Opuntia ficus-indica Mill.) consorciada com sorgo granífero (Sorghum bicolor (L.) Moench). O delineamento experimental foi o de blocos ao acaso em parcelas subdivididas, sendo os espaçamentos alocados nas parcelas principais, e as freqüências e intensidades de colheitas, nas subparcelas. Os resultados são de um período de 12 anos, e as produções de matéria seca de palma, de grãos e restolhos de sorgo foram: 5,23, 1,65 e 2,07; 4,51, 1,30 e 2,10; 2,75, 1,97 e 3,51 t/ha/ano, em espaçamentos de 2,0 m x 1,0 m; 3,0 m x 1,0 m x 0,50 m e 7,0 m x 1,0 m x 0,50 m, respectivamente. A produção de matéria seca foi diferente entre as freqüências de corte, quando foram conservados os artículos primários: 4,08 t/ha/ano na freqüência de quatro anos, e de 3,43 t/ha/ano na freqüência de dois anos. A produção de palma aumentou com o período de crescimento da planta, nas duas intensidades de corte estudadas. A composição química dos artículos de palma e dos restolhos de sorgo foi pouco afetada pelos tratamentos.

Tomato (Lycopersicon esculentum Mill.) cv. Santa Clara was grown on a silt clay soil with 46 mg dm-3 Mehlich 1 extractable K, to evaluate the effects of trickle-applied K rates on fruit yield and to establish K critical concentrations in soil and in plant petioles. Six potassium rates (0, 48, 119, 189, 259 and 400 kg ha-1 K) were applied in a randomized complete block design with four replications. Soil and plant K critical levels were determined at two plant growth stages (at the beginning of the second and fourth cluster flowering). Total, marketable and weighted yields increased with K rates, reaching their maximum of 86.4, 73.4, and 54.9 ton ha-1 at 198, 194, and 125 kg ha-1 K , respectively. At the first soil sampling date K critical concentrations in the soil associated with K rates for maximum marketable and weighted yields were 92 and 68 mg dm-3, respectively. Potassium critical concentrations in the dry matter of the petioles sampled by the beginning of the second and fourth cluster flowering time, associated with maximum weighted yield, were 10.30 and 7.30 dag kg-1, respectively.

Este trabalho teve por objetivo avaliar a capacidade de regeneração das cultivares de tomateiro industrial (Lycopersicon esculentum Mill) IPA-5 e IPA-6, utilizando quatro composições de meio de cultura descritos na literatura e cinco variações de inoculação. Foi testada uma nova variação de inoculação, denominada cotilédone fendido. A maior freqüência de formação de gemas vegetativas foi 100% no caso de IPA-5, e 65% no caso de IPA-6. Para induzir o alongamento de brotos, foram necessários três subcultivos dos explantes apresentando gemas. No caso de IPA-5, o número de brotos obtidos foi maior quando a indução de gemas foi realizada em meio contendo BAP (2,5 mg L-1) e AIA (0,2 mg L-1) seguido de três subcultivos, em meio como zeatina (0,5 mg L-1). Usando esse protocolo, a cultivar IPA-5 produziu uma média de 5,45 brotos alongados a partir do cotilédone fendido. Essa capacidade excedeu significativamente o cotilédone aparado, que produziu 4,4 brotos alongados por explante. No caso de IPA-6, a melhor combinação de meios e método de inoculação produziu 0,87 broto alongado por explante. Os brotos alongados foram enraizados e transferidos para casa de vegetação.

O abacaxi (Ananas comosus Mill.) está sujeito a danos causados pelo frio durante o armazenamento refrigerado. A aplicação, pós-colheita, de Ca pode contribuir para reduzir vários tipos de desordens fisiológicas. Neste trabalho, verificou-se a influência da aplicação, pós-colheita, de CaCl2 a 2%, associada ao tratamento hidrotérmico (38ºC e 40ºC) por 10 e 20 minutos de imersão, na composição química (fenólicos e enzimas), e na suscetibilidade ao escurecimento interno do abacaxi (Ananas comosus Mill.) cultivar Smooth Cayenne. Os frutos foram armazenados a 9ºC e umidade relativa de 90% por um período de 15 dias. As avaliações foram efetuadas sete dias após a retirada dos frutos das condições de refrigeração. O tratamento dos frutos com CaCl2 reduziu o índice de escurecimento interno, conferindo menor atividade das enzimas polifenoloxidase, peroxidase e fenilalanina amônio liase e reduzindo o teor de compostos fenólicos na polpa quando associado ao tratamento hidrotérmico, independentemente do tempo de imersão.
