We determined the effect of fish oil (FO) ingestion on colonic carcinogenesis in rats. Male Wistar rats received 4 subcutaneous injections (40 mg/kg body weight each) of 1,2-dimethylhydrazine (DMH) at 3-day intervals and were fed a diet containing 18% by weight FO (N = 10) or soybean oil (SO, N = 10) for 36 weeks. At sacrifice, the colon was removed, aberrant crypt foci were counted and the fatty acid profile was determined. Intestinal tumors were removed and classified as adenoma or carcinoma. Liver and feces were collected and analyzed for fatty acid profile. FO reduced the mean (± SEM) number of aberrant crypt foci compared to SO (113.55 ± 6.97 vs 214.60 ± 18.61; P < 0.05) and the incidence of adenoma (FO: 20% vs SO: 100%), but carcinoma occurred equally in FO and SO rats (2 animals per group). The polyunsaturated fatty acid (PUFA) profile of the colon was affected by diet (P < 0.05): total ω-3 (FO: 8.18 ± 0.97 vs SO: 1.71 ± 0.54%) and total ω-6 (FO: 3.83 ± 0.59 vs SO: 10.43 ± 1.28%). The same occurred in the liver (P < 0.05): total ω-3 (FO: 34.41 ± 2.6 vs SO: 6.46 ± 0.59%) and total ω-6 (FO: 8.73 ± 1.37 vs SO: 42.12 ± 2.33%). The PUFA profile of the feces and liver polyamine levels did not differ between groups (P > 0.05). In conclusion, our findings indicate that chronic FO ingestion protected against the DMH-induced preneoplastic colon lesions and adenoma development, but not against carcinoma in rats.

The production of oxygen free radicals in type 2 diabetes mellitus contributes to the development of complications, especially the cardiovascular-related ones. Peroxiredoxins (PRDXs) are antioxidant enzymes that combat oxidative stress. The aim of this study was to investigate the associations between the levels of PRDX isoforms (1, 2, 4, and 6) and cardiovascular risk factors in type 2 diabetes mellitus. Fifty-three patients with type 2 diabetes mellitus (28F/25M) and 25 healthy control subjects (7F/18M) were enrolled. We measured the plasma levels of each PRDX isoform and analyzed their correlations with cardiovascular risk factors. The plasma PRDX1, -2, -4, and -6 levels were higher in the diabetic patients than in the healthy control subjects. PRDX2 and -6 levels were negatively correlated with diastolic blood pressure, fasting blood sugar, and hemoglobin A1c. In contrast, PRDX1 levels were positively correlated with low-density lipoprotein and C-reactive protein levels. PRDX4 levels were negatively correlated with triglycerides. In conclusion, PRDX1, -2, -4, and -6 showed differential correlations with a variety of traditional cardiovascular risk factors. These results should encourage further research into the crosstalk between PRDX isoforms and cardiovascular risk factors.

INTRODUÇÃO: A maioria dos pacientes com doença renal crônica terminal depende de hemodiálise (HD) para a manutenção de sua vida. A análise dos fatores que influenciam na sobrevida pode auxiliar na busca contínua por melhores resultados. MÉTODOS: Analisamos 1.009 pacientes tratados por HD crônica em três unidades de diálise de Santa Maria, RS, Brasil, durante 25 anos (1982-2007). RESULTADOS: A sobrevida (método de Kaplan-Meier) em 1, 2 e 5 anos foi de 91%, 84% e 64%, respectivamente. No modelo proporcional de Cox, tiveram influência estatisticamente significativa sobre o risco de mortalidade: idade ao iniciar HD (aumento de 4,5% por ano a mais; p = 0,0001), presença de diabetes (aumento de 56%; p = 0,001) e ano de início da HD (redução de 5,2% por ano mais tarde; p = 0,0001). A sobrevida foi significativamente melhor para pacientes que iniciaram HD de 1997 a 2007 do que para os que iniciaram de 1982 a 1996, tanto em diabéticos (54% versus 41% em 5 anos; p = 0,01) como não diabéticos (72% versus 65% em 5 anos; p = 0,045), embora, nestes, a idade tenha sido significativamente maior no período mais recente. CONCLUSÕES: A presença de diabete e cada ano a mais na idade determinaram risco significativamente aumentado. Cada ano subsequente do calendário trouxe um risco significativamente menor. Nos anos mais recentes, a melhora de sobrevida foi maior para pacientes diabéticos e idosos, sendo atribuída a avanços diagnósticos e terapêuticos e melhor qualidade global do programa dialítico.
