Two adjacent tracts of tropical secondary forest, situated in Itambé do Mato Dentro, south-eastern Brazil, which had been regenerating for 15 and 40 years after clearing, were compared with the purpose of detecting differences in species diversity and composition, species guild composition (regeneration, stratification and dispersion), and stand structure. Four and three 1,125 m² plots laid on the 15- and 40-year-old stands, respectively, sampled 2,430 trees with diameter at the base of the stem > 5 cm. The number of species (S = 199) was high for this forest type and significantly higher for the older stand. Tree density was significantly higher in the younger stand, particularly for smaller trees, whereas the two stands did not differ in both basal area and volume per hectare. Trees of shade-tolerant and understory species were significantly more abundant in the older stand. Though sharing a large proportion of species (49%), the two stands differed significantly in the abundance of many species. Live stumps probably contributed to the relatively quick restoration of some forest characteristics, particularly species diversity, basal area and volume.

O presente estudo teve como objetivos caracterizar a composição da regeneração natural e da chuva de sementes presentes sob a copa de três espécies de início de sucessão - Centrolobium tomentosum Guill. ex Benth. (Fabaceae), Cordia myxa L. (Boraginaceae) e Melia azedarach L. (Meliaceae) em um plantio misto de 15 anos. Para tanto, 15 indivíduos de cada espécie foram marcados e sob suas copas foram instalados dois coletores de sementes (1 m² cada), de onde se coletou propágulos mensalmente, por um ano. A regeneração foi avaliada em parcelas circulares (17,6 m²) sob as copas das árvores, onde foram amostrados os indivíduos arbustivo-arbóreos de 0,3 a 2,0 m de altura. A chuva de sementes sob M. azedarach apresentou maior densidade média e sob C. myxa maior riqueza. Quanto à regeneração natural, as comunidades diferenciaram-se em relação à diversidade de espécies (maior sob M. azedarach) e à estrutura (indivíduos maiores sob C. myxa) entre as espécies do dossel. A maioria dos indivíduos e espécies amostradas pertence aos estádios iniciais da sucessão e muitas, dentre as mais abundantes, são exóticas. Discute-se a formação de nichos de regeneração sob elas e a influência dessas na estrutura e composição dessas comunidades.

An area's innate potential to regenerate represents a crucial factor for its conservation and management. The seed rain and seed bank are important agents in the regeneration process. Seed banks are particularly important in communities where there is a high proportion of obligate seeders. Rocky outcrops are habitats where most part of the plant species depends on their seeds to reproduce and maintain viable populations. Therefore, seed banks ought to be important in this vegetation physiognomy. We test the hypotheses that the seed bank of the rocky outcrops found in the rupestrian fields of "Serra do Cipó", Brazil, is richer in species and denser than those formed on different vegetation physiognomies neighboring the outcrops. We then compared species abundance, species richness and composition in the rocky outcrops' seed banks with those of sandy and peaty bogs, forests, gallery forests, and "cerrados". Furthermore, we report on the natural regeneration potential of these soils by assessing a greenhouse study on seedling emergence. Soil samples were collected from 0 to 5 and 5 to 10 cm of depth. Rocky outcrops had the poorest in species and less dense seed bank and showed segregation in species composition. Emergence was greater in the most superficial layer. However, soils on rocky outcrops showed the greatest proportion of endemic threatened species in their seed banks, demonstrating their importance for biodiversity conservation of the "Serra do Cipó" rupestrian fields.

The crude latex of Crown-of-Thorns (Euphorbia milii var. hislopii) is a potent plant molluscicide and a promising alternative to the synthetic molluscicides used in schistosomiasis control. The present study was undertaken to investigate the embryofeto-toxic potential of E. milii latex. The study is part of a comprehensive safety evaluation of this plant molluscicide. Lyophilized latex (0, 125, 250 and 500 mg/kg body weight) in corn oil was given by gavage to Wistar rats (N = 100) from days 6 to 15 of pregnancy and cesarean sections were performed on day 21 of pregnancy. The numbers of implantation sites, living and dead fetuses, resorptions and corpora lutea were recorded. Fetuses were weighed, examined for external malformations, and fixed for visceral examination, or cleared and stained with Alizarin red S for skeleton evaluation. A reduction of body weight minus uterine weight at term indicated that E. milii latex was maternally toxic over the dose range tested. No latex-induced embryolethality was noted at the lowest dose (125 mg/kg) but the resorption rate was markedly increased at 250 mg/kg (62.5%) and 500 mg/kg (93.4%). A higher frequency of fetuses showing signs of delayed ossification (control: 17.4%; 125 mg/kg: 27.4% and 250 mg/kg: 62.8%; P<0.05 vs control) indicated that fetal growth was retarded at doses ³ 125 mg latex/kg body weight. No increase in the proportion of fetuses with skeletal anomalies was observed at the lowest dose but the incidence of minor skeletal malformations was higher at 250 mg/kg body weight (control: 13.7%; 125 mg/kg: 14.8%; 250 mg/kg: 45.7%; P<0.05 vs control). Since a higher frequency of minor malformations was noted only at very high doses of latex which are embryolethal and maternally toxic, it is reasonable to conclude that this plant molluscicide poses no teratogenic hazard or, at least, that this possibility is of a considerably low order of magnitude

Interleukin-15 (IL-15) is a newly-discovered cytokine that is produced by activated monocytes early in the course of the innate immune response. IL-15 is able to bind to components of the interleukin-2 receptor (IL-2R) despite the fact that it has no sequence homology with IL-2. IL-15 stimulates human natural killer cell proliferation, cytotoxicity, and cytokine production and can substitute for IL-2 under most conditions. In vitro studies indicate that monocyte-derived IL-15 may be an important determinant of IFN-gamma production by NK cells. In addition, IL-15 is able to promote the survival of natural killer cells under serum-free conditions. The IL-15 receptor is a heterotrimeric complex which is composed of the IL-2Rß and g chains in combination with a unique alpha chain (IL-15a). The IL-15Ra chain has strong sequence homology to the IL-2Ra chain and confers high affinity binding to the IL-15R. In contrast to IL-2, transcript for IL-15 and IL-15a is expressed in a number of tissues and indicates that IL-15 may be an important ligand for cells that express components of the IL-2R

In order to study the role of natural killer (NK) cells during the early period of Leishmania infection, BALB/c mice were selectively and permanently depleted of NK cells by injection with 90Sr and subsequently infected with Leishmania (Leishmania) amazonensis (HSJD-1 strain). 90Sr is known to selectively deplete NK cells, leaving an intact T- and B-cell compartment and preserving the ability to produce both interferon alpha and IL-2. This method of depletion has advantages when compared with depletion using anti-NK cell monoclonal antibodies because the effect is permanent and neither activates complement nor provokes massive cell death. In the present study, after one month of treatment with 90Sr, the depletion of NK cells was shown by a more than ten-fold reduction in the cytotoxic activity of these cells: 2 x 106 spleen cells from NK-depleted animals were required to reach the same specific lysis of target cells effected by 0.15 x 106 spleen cells from normal control animals. The histopathology of the skin lesion at 7 days after Leishmania infection showed more parasites in the NK cell-depleted group. This observation further strengthens a direct role of NK cells during the early period of Leishmania infection.

The aim of the present study was to compare healing obtained with biomembranes with the natural healing process (sham) using biochemical and immunohistological assays. C57BL/6 mice were divided into 4 groups of 15 mice each and received different subcutaneous implants: natural latex biomembrane (NLB), denatured latex (DL), expanded polytetrafluorethylene (ePTFE), or sham. On the 2nd, 7th, and 14th days post-treatment, 5 mice per group were sacrificed and biopsied for the following measurements: oxidative stress based on malondialdehyde (MDA), myeloperoxidase (MPO) and hydrogen peroxide by the method of ferrous oxidation-xylenol orange (FOX), as well as glutathione and total proteins; histological evaluation to enumerate inflammatory cells, fibroblasts, blood vessels, and collagen, and immunohistochemical staining for inducible nitric oxide synthase, interleukin-1β, vascular endothelial growth factor (VEGF), and transforming growth factor-β1 (TGF-β1). On day 2 post-treatment, NLB stimulated a dense inflammatory infiltrate mainly consisting of polymorphonuclear cells, as indicated by increased MPO (P < 0.05), but oxidative stress due to MDA was not observed until the 7th day (P < 0.05). The number of blood vessels was greater in NLB (P < 0.05) and DL (P < 0.05) mice compared to sham animals on day 14. NLB induced fibroplasia by day 14 (P < 0.05) with low expression of TGF-β1 and collagenesis. Thus, NLB significantly induced the inflammatory phase of healing mediated by oxidative stress, which appeared to influence the subsequent phases such as angiogenesis (with low expression of VEGF) and fibroplasia (independent of TGF-β1) without influencing collagenesis.

A presença de Vibrio parahaemolyticus foi avaliada em 50 amostras de moluscos bivalves marinhos compostas por 40 amostras de ostras coletadas em 15 restaurantes do Rio de Janeiro e 10 amostras de mexilhões capturados de banco natural em Ponta de Itaipú - Niterói. Foram empregadas a técnica do Número Mais Provável (NMP) para a enumeração de V. parahaemolyticus utilizando Caldo Glicosado Salgado com Teepol (GSTB) e Água Peptonada Alcalina (APA) com 3% de cloreto de sódio (NaCl). Paralelamente foi realizada técnica de enriquecimento em APA com 1 e 3% de NaCl. Decorrido o período de incubação de ambas as técnicas, foi realizado plaqueamento em ágar TCBS (Tiossulfato Citrato Bile Sacarose). Todas as cepas de V. parahaemolyticus isoladas através das duas técnicas foram testadas para o fenômeno de Kanagawa e, quanto à produção de urease. Do total de 141 cepas de V. parahaemolyticus isoladas, 62% revelaram-se urease positivas e, dentre estas, os sorotipos predominantes foram O10:K?, O11:K? e O3:K57 dentre o total de 24 sorotipos urease positivos identificados. Embora todas as cepas de V. parahaemolyticus tenham sido Kanagawa negativas, os resultados apontam elevada incidência desta espécie em ostras comercializadas em restaurantes.

A elaboração de iogurtes pode ser considerada uma excelente alternativa para a utilização industrial do leite de búfala. Entretanto, alguns problemas de aceitação por parte dos consumidores podem ocorrer devido ao elevado conteúdo de gordura do leite original. Este trabalho teve por objetivo avaliar algumas características do iogurte natural batido elaborado com leite de búfala com os seguintes teores de gordura: integral, 3,0% (padronizado) e 0,5% (desnatado). Os iogurtes foram embalados em frascos de polietileno de 1L e estocados a 5°C por 30 dias. Amostras foram colhidas nos dias 1, 15 e 30 e submetidas à análise de acidez, pH, viscosidade e avaliação sensorial. Os valores de acidez e pH não apresentaram diferenças (P > 0,05) entre os tratamentos, durante o período de estocagem. A viscosidade também não foi influenciada (P > 0,05) pelos níveis de gordura nos dias 1, 15 e 30. O iogurte produzido com leite padronizado apresentou notas maiores (P < 0,05) quanto ao sabor na análise sensorial nos dias 15 e 30. Conclui-se que, o desnatamento do leite de búfala não altera as propriedades físico-químicas estudadas no iogurte durante o armazenamento por 30 dias, e que o iogurte padronizado a 3,0% de gordura apresenta uma melhor qualidade sensorial.
